RAPD molecular marker was widely applied to the studies of edible fungi due to it’s simplicity , rapidity and economy. The principle of RAPD molecular marker and its applications to edible fungi were summarized. The applications of RAPD to edible fungi were introduced in species and parental strain identification,genetic diversity, gene clone, gene isolation and the construction of gene linkage map. RAPD molecular marker provids a powerful tool for the studies of edible fungi.
3.The interference effects on UL29 shRNA expression vectors on HSV-2

Objective To construct short hairpin RNA (shRNA) recombinant expression vector for herpes simplex virus typeⅡ(HSV-2) UL29 gene and observe its inhibitory effect on HSV-2. Methods Four interference target sites of HSV-2UL29 gene were selected to construct 4 groups of small hairpin RNA respectively,named shRNA recombinant expression vector. The expression vectors were transfected into HEK293 cells with liposome. HEK293 cells were infected with HSV-2 after expression vector being transfected. The viral titer was estimated by end-point titration assay. The level of transcription was estimated by Real-Time PCR method. The expressing effect of protein was detected by Western-blot. Results Recombinant expression vector pGPU6/GFP/Neo-shRNA was constructed successfully. The result of end-point titration assay showed that the viral titer was reduced comparing with blank control (P<0.05). The result of RT-PCR showed that inhibition rates were respectively 28.80%, 59.95%, 66.08%and 36.27% comparing with blank control, and there were significant differences (P < 0.05). The effect of UL29shRNA1461 group was the best one. The result of Western-blot showed that the expressing quantity of ICP8 was reduced. Conclusion Recombinant expression vector pGPU6/GFP/Neo-shRNA can interfere HSV-2 UL29 gene expression from different cell level in vitro, which can inhibit the replication of HSV-2 genome in HEK293 cells. Thus, RNA interference (RNAi) is conducive to the further exploration of viral therapy.
4.Analysis of Risk Factors on Coronary Artery Lesions Secondary to Kawasaki Disease

Objective To investigate the risk factors on children with coronary artery lesions (CAL) secondary to Kawasaki di-sease(KD). Methods One hundred and forty - five patients with KD from January 1999 to December 2001 were collected. Among them, 93 cases without CAL and 52 cases with CAL. The test results, therapeutic methods and prognosis were analyzed in two groups. Results The duration of fever was longer and mean value of erythrocyte sedimentation rate (ESR) higher in group with CAL than those m the group without CAL(P0.05).The incidence of CAL was 18.8% (18/96) in patients of IVIG treatment within 10 days from onset, and 69.4% (34/49) in patients without IVIG treatment (P

Objective To explore the procedures of 3Q validation for blood coagulation analyzer under the good laboratory prac‐tice(GLP) system based on the Sysmex CA7000 automatic blood coagulation analyzer .Methods Four test indicators of PT ,APTT , TT and FIB were chosen to conduct the performance verification in the within‐run precision ,between‐day precision ,accuracy ,linear‐ity ,carry‐over contamination rate of the automatic blood coagulation analyzer .They were prothrombin time(PT) ,activated partial thromboplastin time(APTT) ,thrombin time(TT) ,and fibrinogen(FIB) .Results The within‐run precision CV values of PT , APTT ,TT and FIB were 1 .33% ,1 .57% ,1 .47% and 1 .90% ,respectively ;the inter‐day precision CV values of PT ,APTT ,TT and FIB were 1 .73% ,1 .52% ,1 .55% and 2 .14% respectively ;in terms of accuracy ,the normal quality control CV values were 7 .45% ,3 .88% ,-4 .98% and 4 .36% respectively ;the abnormal quality control CV values of PT and APTT were 8 .11% and 8 .77% respectively ;the r value of FIB linear correlation coefficient was 0 .999 3 ,the a value was 1 .02 ;the highest CV value of car‐ry‐over contamination rate was 2 .15% ;the detected results all conformed to the general industry standards and requirements of in‐strument manufacturer .Conclusion The 3Q validation under the GLP system proves that the Sysmex CA7000 automatic blood co‐agulation analyzer is good in performance and is appropriate for the laboratory work of clinical laboratory department .
7.Effect of salt loading on thoracic aortic stress relaxation properties of rat models of spontaneous hypertension

BACKGROUND:In recent years, great progress has been achieved in the mechanical and viscoelastic properties of arterial blood vessels of normal human corpses and animals. OBJECTIVE: To quantitatively analyze the tensile mechanical properties of thoracic aorta in normaly fed spontaneously hypertensive rats and salt-loaded spontaneously hypertensive rats. METHODS: Twenty spontaneously hypertensive rats were obtained and randomly divided into experimental and control groups (n=10/group). Rats in the experimental group were subjected to continuous salt loading intervention for 16 weeks. Rats in the control group were fed with normal diet and ordinary tap water. At the 16th week, 10 specimens of thoracic aorta of rats from these two groups were harvested to conduct stress relaxation experiments.RESULTS AND CONCLUSION: The decrease in stress at 7 200 seconds and the decrease in normalized stress relaxation function value at 7 200 seconds of thoracic aortic specimens of rats in experimental group were both lower than those in the control group (P< 0.05). These results confirm that the stress relaxation properties of thoracic aorta of normaly fed and salt-loaded spontaneously hypertensive rats change, wherein the changes in salt-loaded spontaneously hypertensive rats are more obvious.
8.Role of polycyclic aromatic hydrocarbons and aryl hydrocarbon receptor in the development of tumor

Polycyclic aromatic hydrocarbons ( PAHs) are ubiquitous environmental pollutants, whose carcinogenicity is determinated.The mechanism of their carcinogenicity: PAHs are able to combine with aryl hydrocarbon hydrocarbon receptor ( AhR ) , resulting in some toxicity and carcinogenicity.AhR is a ligand-dependent activation transcription factor, which is activated by a large variety of ligands, regulating the expression of a series of gene involved in metabolism, and participating in important biological processes, such as singal transduction, cell proliferation,differentiation and apoptosis,and so on.Besides, it's closely related with the tumor development.Thus, it will provide a new approach for cancer prevention and treatment to study the role of PAHs and AhR in the development of tumor.
9.Investigation of Mosquito Abundance and Composition around the Rare Birds National Nature Reserve of Yancheng,Jiangsu Province

Objective To investigate the mosquito abundance and their relative species composition within and outside the Rare Birds National Nature Reserve of Yancheng,Jiangsu Province. Methods Sampling was carried out between May and Oct. 2004 at two weeks interval in two foci (the Reserve and nearby residential district) in Sheyang County. Mosquitoes were collected with the modified CDC light trap. Density was calculated,and species were identified. Environmental temperatures,rainfall and relative humidity were monitored during the study. Results A total of 40 912 mosquitoes were captured in the two foci. The sampled mosquitoes were identified as 4 species belonging to three genera (Anopheles sinensis,Culex pipiens pallens,Cx. tritaeniorhynchus,and Armigeres subalbatus). The most abundant mosquito species was An. sinensis and Cx pipiens pallens,which accounted for 97.7% of the whole number. 92% and 8% of the total amount of mosquitoes were collected from the nature reserve and residential district respectively. The most abundant species in the nature reserve and residential district was An. sinensis(60.6%) and Cx. pipiens pallens(76%),respectively. Within the nature reserve,there were two peaks occurred in adult abundance,in mid-and late July and mid-Sept. The abundance of mosquitoes in the area was positively correlated to the temperature (r=0.765,P=0.005). Conclusion The wetland is an ideal breeding place for An. sinensis and Cx pipiens pallens. The peaks of mosquito abundance are in mid-and late July and mid-Sept. It is of importance to carry out surveillance on mosquito vectors with pathogen-transmitting potential.
10.Rehabilitation of Knee Osteoarthritis in Stroke Patients

Objective To explore the effective method on the knee osteoarthritis (KOA) of stroke patients.Methods Twenty-four stroke patients with KOA were randomly divided into the treatment group and control group with 12 cases in each group. All patients received facilitation technique exercise, the patients in the treatment group were added with an extra complex therapy of electronic acupuncture, physical and exercise thepapies. All patients of two groups were assessed with visual analogous score (VAS) and Fugl-Meyer Motion Scale (FMMS) before treatment and one course after treatment.Results After treatment, the scores of VAS and FMMS of the patients in the treatment group were significantly better than that in the control group ( P<0.01).Conclusion Multidisciplinary rehabilitation including electronic acupuncture, physical, and exercise is efficiancy to KOA of stroke patients.
