1.PI3K-Akt signaling pathway and chemotherapy drug resistance in cancer

PI3K-Akt signaling pathway plays a critical role in the regulation of cell proliferation,differentiation and apoptosis,and is closely related to the physiological function of cells and thus may cause certain diseases.The excessive activation of this pathway is found closely associated with the formation of drug resistance of tumors.Recent studies in vitro indicate that the combined chemotherapy drugs with PI3K-Akt signaling pathway inhibitors,can significantly enhance the efficiency of chemotherapy drug and lower the value of IC50.Therefore,PI3K-Akt signaling pathway has become an important target to inhibit tumor cell growth and reverse tumor drug resistance.
2.Clinical comparison of phacoemulsification and non-phacoemulsification for cataract

AIM: To contrast the effect of small incision phacoemulsification and non-phacoemulsification to treat cataract.
METHODS: Totally 172 patients with cataract were divided into 2 groups ( n = 86 ) randomly. Patients in Group Phaco were treated with phacoemulsification and, while those in Group Siecs were treated with small incision extracapsular cataract surgery and intraocular lens implantation. They were all followed up for 1-3y. Functional examinations were made at 3d;1,6mo and last follow - up. The visual acuity, intraocular pressure, anterior chamber depth, average corneal power ( ACP ) , cylinder ( CYL ) , surface asymmetry index ( SAI ) and complications were contrasted between groups.
RESULTS: At 3d after operation, the visual acuity of Group Phaco was better than that of Group Siecs ( P<0-05), and CYL, SAI and intraocular pressure of Group Phaco were all significantly lower than those of Group Siecs (P<0. 05). At 1mo after operation, the visual acuity of Group Phaco was better than that of Group Siecs ( P<0-05), and CYL, SAI and intraocular pressure of Group Phaco were slightly lower than those of Group Siecs ( P<0. 05);there was no difference between the two group. At 6mo after operation, the visual acuity of Group Phaco was still better than that of Group Siecs (P<0. 05), and CYL, SAI and intraocular pressure of Group Phaco were flat as those of Group Siecs (P<0. 05); there was no difference between groups in those indexes above ( P > 0. 05 ). Furthermore, the APC and anterior chamber depth of each point- in - time had no significant difference between groups (P>0. 05). The incidences of complication were similar in two groups ( P> 0. 05 ); but with individual differences, patients of GradeII and Ⅲ in Group Phaco got lower complication rate (P<0. 05), while those of Grade Ⅳ higher (P<0. 05) than those of Group Siecs.
CONCLUSION: Our research shows that phacoemulsification has high effect in the treatment of patients with GradeII and Ⅲ nuclear hardness, while small incision extracapsular cataract surgery has high effect in the treatment of patients with Grade Ⅳ nuclear hardness.
3.Protective action of grape seed proanthocyanidins combined With casein on radiation damage of rats

BACKGROUND: A great deal of free radicals can be induced,which results in peroxidation of the whole body. The antioxidant activity and the abilities of scavenging free radicals of proanthocyanidins(GSP) have been well documented,but related researches about the protective role in radiation damage were less reported.OBJECTIVE: To investigate the protective role of GSP combined with casein in 60Co-y radiation-induced peroxidative damage.DESIGN: Observing and comparative study based on animal model.SETTING: Nutrition Department of Daping Hospital Affiliated to the Third Military Medical College of Chinese PLA.MATERIALS: This study was carried out in the Third Institution of Field Operation Surgery, the Third Military Medical College of Chinese PLA.Eighty healthy Wistar rats,clearage,were adopted,ith weight from 180 to 220 g,half male and half female were adopted.METHODS: Rats were divided into four groups: normal control group,irradiation group,GSP group and GSP + casine group with 20 rats each. Rats in GSP group were fed routinely and supplemented with 1% GSP for 10days,which was replaced by 1% GSP and 5% casein. All rats except those of control group were exposed to whole-body 60Co-g amma-radiation at a dose of 7 Gy. Seven days later,6 survival rats randomly selected from each group were killed.MAIN OUTCOME MEASURES: The level of serum malondialdehyde (MDA), activities of serum superoxide dismutase(SOD),spleen index and the number of peripheral white blood cell(WBC).RESULTS: Serum MDA was obviously increased,accompanied by obviously attenuated SOD activity after irradiation; the level of MDA in both GSP group and combined group was found lower than that in irradiation group with statistical significance between them(P<0.01), the activity of SOD was significantly higher than that of irradiation group(P<0.01 ) . Markedly atrophic pathological change in spleen could be observed in irradiated rats,and meanwhile spleen index declined with the number of WBC notably decreased. But spleen index in both GSP group and combining group was markedly higher than irradiation group(P<0.05),and the same comparing result could also be found in the number of WBC(P<0.01 ).CONCLUSION: Peroxidative impairments could be obviously attenuated,and decrement of WBC could also be improved by single GSP or alternatively GSP combined with casein,with the latter showing greater effects in respect of improving immune function following irradiation,cellular chemical imbalance,and consequently the life quality.
5.Effects of Grape Seed Proanthocyanidin and Casein Peptide on the liver function and protein nutritional status in rats with radiation injury

Objective: To investigate the effects of grape seed proanthocyanidin(GSP) and casein peptide on the liver function and protein nutritional status in rays irradiated rats. Methods: Adult wistar rats were randomized into four groups with 20 rats in each group including normal control, radiation control, GSP and GSP+casein peptide combined groups. The GSP group was fed with basic diet containing 1% GSP, the combined group was fed with basic diet containing 1% GSP plus 5% casein peptide. Animals except normal control were irradiated by 60 Co? rays(7Gy) 10 days later. 6 survivors were randomly selected from each group and killed on the 7 th d after radiation. The levels of the total protein (TP) and the albumin (ALB), the activities of the glutamate pyruvate transaminase(GPT) and the glutamate oxaloacetale transaminase(GOT) in serum were measured. The change of the body weight of the animals was observed during the experiment time. Results: After radiation the body weight of the rats dropped sharply. 8.Changes in the pathological diagnosis rate of Graves'disease and Hashimoto's thyroiditis——analysis on the clinicopathologic data from 1978 to 2005 in General Hospital of Tianjin Medical University

Objective To observe the changes in the pathological diagnosis rate of Graves'disease(GD) and Hashimoto's thyroiditis(HT)in the past 28 years and to probe its relationship to the environmental factors. Methods Statistic analysis was performed on data of thyroid disease,GD,HT and the complications of HT in Department of Pathology(1978-2005)in General Hospital of Tianjin Medical University with Run test,Cox-stuart test,ANOVA,t or t'test and Chi-square test.Results In 28 years there were 243 403 biopsies(average 8 693/ year)including 6 771 cases of thyroid disease(27.8‰),216 cases of GD(0.89‰),352 cases of HT (1.45‰).The biopsy rate of thyroid disease and GD showed a descending tendency since 1982 but an ascending tendency from 1995,furthermore,with GD it showed a descending tendency from 2001 to 2005 once more.While the biopsy rate of HT and GD+HT showed a descending tendency from 1986 and an ascending tendency after 1996,being 1-4 years later than thyroid disease and GD.The biopsy rate of GD+HT and the proportion of HT showed a descending tendency in 2002-2005 and 2003-2005 respecitvely.The average age of total and female patients with GD showed a descending tendency from 1997.The average age of GD was 13.7 years younger than HT,the patients of GDⅠ-type andⅡ-type being younger thanⅢ-type.The average age of lymphoid type(L), oxyphilie epithelium type(O),pronounced epithelium destruction type(P)in HT increased sequentially.In 28 years,there were more female patients with GD and HT,especially with HT.There existed 7.95% hyperthyroidism,6.25% nodular goiter,2.27% carcinoma in HT(more in L-type)and 2.56% hypothyroidism in HT(more in P-type).Conclusion Within 28 years,according to the pathological diagnosis rate,GD,HT,and GD+HT showed phasic tendencies related to drinking low iodine water and the implement of universal salt iodization policies in 1994 and 2000.Therefore,on the levels of genetics and environment,the factors that influence autoimmunity of thyroid gland could not exclude the variations of iodine intake,infection,affection or emotional stress.The mechanism of the changes in the pathological diagnosis rate needs further study.
9. Determination of amifostine in human saliva by HPLC-MS/MS

OBJECTIVE: To establish an HPLC-MS/MS method (high performance liquid chromatography-tandem mass spectrometry) for the direct determination of amifostine in human saliva. METHODS: Saliva samples were collected from six adult healthy volunteers. After protein precipitation and addition of the internal standard (IS) huperzine-A (HupA), HPLC-MS/MS was used to analyze amifostine. The analysis was conducted using a ZIC-HILIC analytical column (2.1 mm × 100 mm, 3.5 μm). Electrospray ionization was used with multiple reaction monitoring (MRM) mode. RESULTS: The lower limit of quantification (LLOQ) of the method was 0.938 mg · L-1 (S/N<10). The standard curve was linear in the range of 0.938-30 mg · L (r=0.9991, n=6). The inter-day and intra-day RSDs were all less than 15% for the low, medium and high concentration quality control samples (1.0, 5.0 and 25 mg · L-1): The values of recovery were all more than 85%. CONCLUSION: The method is direct, rapid, simple and sensitive, and suitable for the determination of amifostine in saliva samples.
