1.Aberrant methylation of Wnt signaling pathway in lymphoblastic leukemia
International Journal of Pediatrics 2011;38(5):494-496
Wnt signaling plays an important role in blood tumorigenesis through the complex signal transduction pathways.Aberrant methylation of Wnt signaling inhibitor gene promoter leads to loss of function, which is involved in leukemia incidence and affects the relapse rate, disease free survival and overall survival.Demethylating agents or specific inhibitors of Wnt pathway, which was added in cell culture, increased apoptosis.Analysis of Wnt signaling pathway by aberrant methylation is important to guide lymphocytic leukemia prognosis and individual treatment.
2.Observation on the Expression of HBV-DNA in the Umbilical Cord Tissuesof the Died Fetus Delivered From Puerpera with Hepatitis Virus B
Journal of Chinese Physician 2001;0(10):-
Objective To observe the expression of HBV-DNA and whether there were copy of HBV in the umbilical cord tissues of the died fetus delivered from puerpera with hepatitis virus B. Methods 40 cases such died fetus were collected by routine autopsy to obtain umbilical cord tissues. And using in situ molecular hybridization technique detected HBV-DNA. Results For the umbilical cord tissues, there were 40%(16/40) cases detected out HBV-DNA.HBV-DNA mainly localization in the surface of the umbilical cord vessel and in the cytoplasma of the cord vessel's endothelial cells. They were not in the cord vessel's endothelial cells nuclei. Conclusions There were HBV replication in the umbilical cord tissues of the died fetus. But the expression of HBV-DNA in the umbilical cord tissues of the died fetus is not related with the HBV replication status in the pregnant woman veins.

4. Effect of double chain method on the closure of orthodontic teeth
Acta Anatomica Sinica 2020;51(1):66-71
Objective To study the safety and effectiveness of the double chain method on the closure of the space of the teeth. Methods Totally 72 cases of different maxillary and mandible dentition space were included in the study. The pulp vitality was measured before and after treatment, the maxillary and mandible first molar teeth. All values were compared to assess the safety of dental pulp vitality. There were 36 cases in tooth extraction group and non-tooth extraction group. The observation period was 4 weeks, the changes of the alveolar space between maxillary and mandible teeth in adult and minor groups were measured before and after treatment. The difference of the total groove spacing before and after treatment were calculated and compared, to assess the effectiveness of space closure. Results In all 72 patients, there was no significant difference in the vitality of the pulp before and after treatment (P>0. 05). Compared to before treatment, the sum of the distance between the upper and lower mandibular groove using the double chain method was significantly reduced for both adult and juvenile patients in the four week tooth extraction group and the nontooth extraction group, the difference was statistically significant (P<0. 05). Conclusion Double chain closed teeth space does not cause pulp degeneration and necrosis. It is safe in clinical practice. Double chain method is effective, and can simplify the clinical operation.

To investigate whether accelerated blood clearance (ABC) phenomenon could be induced after repeated injection of mitoxantrone thermosensitive liposomes, LC-MS/MS and enzyme linked immunosorbent assay (ELISA) were used to measure the concentration of mitoxantrone and the anti-polyethylene glycol (PEG) IgM levels in rat plasma, separately. The drug was rapidly cleared away after the second administration. The anti-PEG IgM was detected after the first dose which was neutralized quickly after the second dose. It is proved that repeated administration of mitoxantrone thermosensitive liposomes in rat caused the ABC phenomenon.

Objective To investigate the value of the intraoperative real-time three-dimensional transesophageal echocardiography (RT 3D-TEE) in therapeutic application of surgery for the mitral valve prolapse. Methods Thirty-five patients underwent surgical treatment were diagnosed as mitral valve prolapse by 2-dimensional transthoracic echocardiography (2D-TTE),with or without ruptured chordae tendineae.RT 3D-TEE was performed for assessing the segment of prolapse and its complications before cardiopulmonary bypass and after heart resuscitation respectively. The echocardiographic results were compared with the surgical findings. Results The accuracy of RT 3D-TEE to identify segments with prolapse was significantly higher than that of 2D-TTE (91.4% vs 82.9%,P =0.003).RT 3D-TEE was more accurate than 2D-TTE for identifying ruptured chordae tendineae, vegetation and thrombus since higher sensitivity, specificity and Youden index.Among all 35 patients, valvuloplasty was performed in 28 cases, whereas, only 7 cases underwent replacement of prosthetic valves.Three patients were benefited by accurate evaluation of therapeutic effect using RT 3D-TEE in time.ConclusionsIntraoperative RT 3D-TEE can be used to provide “ surgical view”,acquired adequate valuable information of mitral valve, and more reliable functional and anatomical assessment of the mitral valve components and its geometry.Suitable surgical intervention can be designed for improving outcomes of patients.

Objective To determine the polymorphism in CXC chemokine SDF-1α transcript and its effects on HIV infection. Methods Three groups of study subjects lived in Hang Kong were recruited:278 HIV-heahhy donors of Chinese origin.49 HIV+Caucasians and 13 Chinese with high risk behavior to HIV but kept uninfected.Genomic DNA and RNA were extracted from eripheral blood mononucleal cell. The PCR and RT-PCR reaction were set up accordingly.Sequence of the SDF-1α promoter, the open reading frame(ORF)and the 3'untranslate region(3'UTR)were analyzed. Two steps PCR reaction using two reverseprimers with mismatched nucleic acid were employed to screen the frequency of a novel mutation. Results equencing analysis from 100 subjects indicated that non mutation happened in tlle promoter and ORF of SDF-1α.A novel mutation Was detected from 3'UTR of SDF-1α.It is a "GA" insertion in "G" rich region near the stop code of SDF-1α.The mutation Was named as SDF-1-3’GA+and submitted to GenBank (AY874118).The mutation happened in three roups.with allele frequency of 15.1% in the healthy Chinese.of 30.7% in the high risk Chinese group. Conclusions our results confirm that SDF-1 genes arerelatively conserved.None noteworthy mutation is identified in the promoter and ORF regions of SDF-1α However.a novel mutation is identified from the 3'UTR of SDF-1α. It would be worthwhile to etermine effect of the novel mutation on HIV infection.
