1.Characterization of Human Papillomavirus Types in Cervical Epithelial Neoplasia by in Situ Hybridization.

An in situ DNA hybridization technique was applied to detect human papillomavirus(HPV) DNA, HPV types 6/11 and 16/18, on paraffin sections of 36 cervical condylomatous lesions associated with cervical intraepithelial neoplasia and invasive squamous cell carcinoma. 1) HPV DNA sequences were identified in 14 of 36 cervical condylomatous lesions(39.0%); HPV 6/11 in 7 cases(19.0%) and HPV 16/18 in 7 cases(19.0%). 2) With the use of biotinylated HPV 6/11 DNA probes, 5 of 5 condyloma acuminata(100%), 1 of 5 flat condylomata(20%), and 1 of 7 mild dysplasias(14.3%) were positive. 3) With the use of HPV 16/18 DNA probes, 1 of 7 mild dysplasias(14.3%), 2 of 5 moderate dysplasias(40%), 2 of 4 severe dysplasias(50%), and 2 of 5 invasive squamous cell carcinomas(40%) were positive. 4) The positive stainings to HPV DNA probes were primarily detected in koilocytotic nuclei of the superficial epithelium. No positive signals were found in the normal, dysplastic or carcinoma cells. 5) The numbers positively stained cells were decreased with increasing severity of the lesions from benign condylomas to invasive squamous cell carcinomas. In conclusion, HPV types 6/11 were more commonly identified in benign condylomatous and low grade intraepithelial lesions than high grade lesions. However, HPV types 16/18 were identified in high grade CIN and invasive squamous cell carcinomas. The present results while supporting the concept on HPV 16/18 as the high risk of HPV types in cerivical carcinogenesis also emphasize the applicability of the situ DNA hybridization as a tool in analysis of the specific HPV DNA sequences in routine biopsies of these lesions.

Gross deciduosis in pregnancy is rare. A case of disseminated deciduosis peritonei studied by light and transmission electron microscopy is reported. The pathogenesis of deciduosis peritonei and relationship with leiomyomatosis peritonealis disseminata are discussed in view of the present findings and those previously reported.

3.Human Papillomavirus infection in Cervical Intraepithelial Neoplasia: A Comparative Histopathological and Immunohistochemical Study.

One hundred twelve cases of cervical intraepithelial neoplasia (CIN) were reviewed to detect the condylomatous lesion characterized by koilocytotic cells. Condylomatous lesions were present in 12 cases (30.8%) of 39 cases with mild dysplasia, 3 cases (15.0%) of 20 moderate dysplasia, 2 cases (13.3%) of 15 severe dysplasia and 4 cases (10.5%) of 38 cases with carcinoma in situ. These 21 cases with condylomatous lesions were subjected to the study on demonstration of human papillomavirus (HPV) antigen with peroxidase antiperoxidase reaction and 6 simple condylomatous lesions of the uterine cervix were included for control. Twenty seven cases with simple cervical condyloma and condylomatous lesions associated revealed 22 cases of flat type, 3 cases of exophytic type, 1 spiked type, and 1 inverted type. Cells showing a positive reaction for HPV antigen were identified in 5 cases (55.6%) of 27 cases with condylomatous lesions. HPV antigen positive nuclei were found chiefly in koilocytotic cells. The prevalence of HPV antigen positive cells was well correlated with the extent and intraepithelial level of the koilocytotic changes. The results of this study provide a confirmation of the presence of HPV antigens in CIN, and suggest that the HPV may be an important factor in the etiology of CIN.

5.The prognostic significance of tumor angiogenesis, proliferating cell nuclear antigen(PCNA), and the Ki-67 index in carcinoma of the uterine cervix.

Angiogenesis, the induction of new capillaries and venules, is associated with tumor growth. This study was designed to determine whether cervical carcinomas are angiogenic, and to investigate whether tumor angiogenesis can serve as a prognostic factor in cervical carcinoma. Surgical specimens of 47 cervical carcinomas were immunohistochemically stained specifically for endothelial cells with factor VIII-related antigen to identify all vessels. Microvessels were counted from photographs of 200x microscopic fields. In addition, thirty-seven cases were studied by immunohistochemical means using the monoclonal antibodies for PCNA and for Ki-67 to determine tumor cell proliferation rates in cervical carcinomas. The microvessel count(MVC), the PCNA labelling index, and the Ki-67 index were calculated and compared with known prognostic factors and disease free survival rates in cervical carcinomas. A wide range in the MVC count(range 12-100 mean=38.2+/-19.2), the PCNA labeling index(8-69% mean=33.6+/-15.2%), and in the extent of Ki-67 staining(0-43% mean=10.3+/-10.5%) was observed, indicating considerable variation of tumor angiogenic activity and tumor growth rates. This study showed statistically significant correlations in disease free survival rates with both lymph node status and the microvessel count. However, there was no significant difference in disease free survival rates between tumor stage, age, the PCNA labelling index, and the Ki-67 index.

6.The Effect of Copper on 3'-Methyl-4-dimethylaminoazobenzene Induced hepatic Carcinogenesis.

To elucidate the effect of copper on the 3'-methyl-4-dimethylaminoazobenzene(3'-MeDAB) induced hepatic carcinogenesis, Sprague-Dawley rats were divided into 4 groups according to 3'-MeDAB and copper administration: I. noraml control, II. copper only, III. 3'-MeDAB only, IV. 3'-MeDAB plus copper. The animals of groups III and IV were fed experimental diet containing 0.06% 3'-MeDAB. Copper was administrated intraperitoneally in a dose of 0.5 mg, twice a weak. Animals were sacrificed at different intervals. Liver weight, hepatic copper content and gross and microscopical changes of the liver were examined and the cell kinetics of various lesions in the hepatic carcinogenesis was studied by applying the immunohistochemical method for bromodeoxyuridine(BrdU). The hepatic copper content was significantly increased in animals given copper but returned to the normal value after cessation of adminstration. 3'-MeDAB administration caused oval cell proliferation and produced hyperplastic nodules, cholangiofibrosis and carcinoma of the liver. Simultaneous administration of copper did not alter the incidence of 3'-MeDAB induced lesions, except for carcinoma. The liver weight and the size of hepatic nodules and masses were smaller in group IV than in group III. The liver weight as well as the nodularity and the mass formation continued to increase affect cessation of 3'-MeDAB administration. Copper did not affect the BrdU labelling indices of the hepatic lesions induced by 3'-MeDAB. The oval cell proliferation and the BrdU labelling indices of the oval cell and the hyperplastic nodule were decreased, but the incidence of cholangiofibrosis and its BrdU labelling index were still elevated after cessation of 3'MeDAB administration. These findings indicate that copper could delay the developement of 3'-MeDAB induced hepatic lesions, but not suppress, since copper does not stay long enough to accumulate in the rat liver, and that copper could not affect the proliferation of 3'-MeDAB induced hepatic lesions once developed.

7.Assessment of Viability in Regional Myocardium with Reversed Redistribution by Thallium Reinjection in Patients with Acute Myocardial Infarction.

PURPOSE: The aim of this study was to evaluate whether T1-201 reinjection distinguishes viable from non-viable myocardium in patients with reverse redistribution after acute myocardial infarction. MATERIALS AND METHODS: We studied 42 patients with acute myocardial infarction (age, 55+/-12 years). Eighteen (43%) out of 42 showed reverse redistribution on dipyridamole stress-4 hour redistribution T1-201 single photon emission computed tomography (SPECT). T1-201 reinjection was performed at 24 hours. Reverse redistribution was defined as worsening of perfusion defect at 4 hour delayed scan. All patients underwent follow-up echocardiography in 4 months to assess regional wall motion improvement. T1-201 uptake on reinjection images were analyzed for the prediction of myocardial wall motion improvement. RESULTS: Of 36 segments with reverse redistribution, 17 segments showed normal wall motion on echocardiography, while 19 segments showed all motion abnormalities. Of 19 the segments with reverse redistribution, 11 (58%) showed enhanced uptake after 24 hour reinjection. Myocardial wall motion was improved in 10 of 11 segments (90%) with enhanced uptake on reinjection. Wall motion improvement was not seen in 5 of 8 segments (63%) without enhanced thallium uptake. When myocardial viability was assessed by the uptake on reinjection image, nine of 10 segments (90%) with normal or mildly decreased uptake showed improved wall motion. Wall motion was not improved in 5 of 9 segments (56%) with severely decreased uptake. CONCLUSION: In patients with acute myocardial ifarction, T1-201 reinjection imaging on myocardial segments with reverse redistribution has a high positive predictive value in the assessment of myocardial viability.

9.A Study for A New Method for Evaluation of Phototoicity.

The present study was carried out to find a simple and safe in. vitro test for phtotoxic drugs. Authors selected two strains of Salmonella typhimurium(TA98 and TA102) which have been used in Ames test for the detsction of mutagenecity af various chemical substances. Both strains are genuine products of genetic enzineering. The etrain TA98 should be highly vulnerable to ultraviolet radition because it lacks normal I)NA excision repair gene. The strain TA102 was chosen as control since it maintained the DNA repair gene. These strains were subjected to increasing dosea of UVA with or without pretreatment of 8 methoxypsoralsn(8-MOP) which is a prototype of photatoxic druge. The authors made use of a perforated stain-less steel template which provided a simple and eosy monitoring of ultraviolet irradiation effects i.e. clear zones due to inhibition of the atrains could be determined. By using this methad, the authors acquired the following results .' I. 8-MOP alone exerted no inhibition on both strains at concentration upto 100mg /ml. 2. UVA irradiation alone showed no growth inhibition at dose upto 5J/cm. 3. UVA irradiation after pretreatment with 8 MOP resulted varying growth inhibition in proportion to irradiation doses. 4. Authors found a suitable concentration of 8-MOP for this test is 10pg/ml. With this ccncentrstion, minimal phatatoxic dose of UVA were O.l J/cm for the strain TA98 and 1.0J/cm for the strain TA102 respectively.

10.A Case of Fabry ` s Disease with Generalized Anhidrosis.
