1.Current status and future about maintenance treatment of advanced colorectal cancer

Through PubMed, CNKI and other databases to retrieve original documents about the maintenance treatment of advanced colorectal cancer, collect and analyzed research theory and progress in the maintenance treatment of advanced colorectal cancer. The clinical efficacy of maintenance therapy are evaluated and the patient's disease-free survival and quality of life are observed. The Result shows that the maintenance treatment can improve disease-free survival of advanced colorectal cancer patients, and the toxicity of maintenance treatment can still be tolerated. So the maintenance treatment of advanced colorectal cancer has better clinical efficacy, it is worth to study in the future.
2.Effects of Bevacizumab on the proliferation and epithelial-mesenchymal transition in human retinal pigment epithelial cells in vitro

?AIM:To investigate the effects of Bevacizumab on the proliferation and the expression of E -Cadherin and fibronectin in human retinal pigment epithelial cell ( ARPE-19) in vitro.?METHODS: Different concentrations (0, 0.625, 1.25, 2.5, 5.0mg/mL) of bevacizumab were exposed to ARPE-19 cells, then cell viability was analyzed by CCK-8, cell cycle was determined by flow cytometry, and the expression of E-Cadherin and fibornectin was detected by Western blot and RT-PCR.?RESULTS:The concentration as 2.5mg/mL or 5.0mg/mL of bevacizumab was shown to effectively suppress the proliferation and cell cycle of ARPE-19 cell (P<0.05). In addition, 2.5mg/mL or 5.0mg/mL of bevacizumab could downregulate the expression of E-cadherin and promote the transcription of fibronection gene (P<0.05).?CONCLUSION:High concentration of bevacizumab was able to inhibit ARPE-19 proliferation, downregulate E-Cadherin expression and promote fibronectin expression, indicating epithelial-mesenchymal transition induced by bevacizumab in ARPE-19 cell.
3.Study on Changes of Microelement Content in Crude and Calcined Pyritum

Objective To investigate the changes of microelement in curde and calcined pyritum, and approach the process mechanism of pyritum. Methods The crude and calcined pyritum were extracted in water decocting. Subsequently, the content of ten elements was determined by inductively coupled plasma emission spectrometry (ICP) and compared. Results After being calcined, the content of Pb in pyritum decreased, while the content of the other nine elements investigated all increased in different degrees. Conclision After being calcined, the content of ten elements in pyritum changed largely.
4.Partial characterization of genes encoding the ATP-binding cassette proteins of Cryptosporidium parvum.

The present study aims to explore the possible mechanisms underlying the multidrug resistance characteristic of Cryptosporidium parvum by detecting the presence of ATP-binding cassette (ABC) protein encoding genes, especially one that shows high similarity to members belonging to the multidrug resistance protein (MDR) and multidrug resistance associated protein (MRP) subfamilies. PCR using ABC-specific degenerate primers successfully amplified two unique fragments, designated Cpnbd1 and Cpnbd2, from C. parvum genomic DNA. Cpnbd1 exhibited high degree of homology (99-100%) with the nucleotide- binding domains (NBDs) at the NH2 -terminal halves of two previously reported ABC proteins (CpABC and CpABC1) of human and bovine origin C. parvum isolates. It is likely that CpABC, CpABC1 and Cpnbd1 were encoded by homologous genes of a type of ABC transporter protein found in different C. parvum isolates. However, Cpnbd2 showed moderate levels of similarities (28-49%) to the NBDs of four ABC proteins characterised in C. parvum to date. Therefore, Cpnbd2 could be a novel member of an ABC superfamily of proteins in C. parvum. Phylogenetic analyses on a list of ABC transporters known to associate with MDR phenotype has significantly linked Cpnbd1 and Cpnbd2 to these transporters, thus suggesting that Cpnbd1 and Cpnbd2 proteins may contribute to the intrinsic multidrug resistance phenotype of C. parvum.

5.The clinical spectrum of localized peripapillary detachment in pathologic myopia

AIM: To report the clinical manifestations of localized peripapillary detachment in pathologic myopia and to evaluate its underlying associations and causes.METHODS: From December 2002 to January 2004, 10eyes from 7 patients with high myopia were identified to have localized peripapillary detachment by optical coherent tomography (OCT). The features were described together with the fundus fluorescein angiography, indocyanine green angiography and multifocal electroretinogram.RESULTS: Localised peripapillary detachments did not cause any symptoms by themselves and all the lesions were recognized because of other ocular problems. The areas of peripapillary detachment were all located within the posterior staphyloma. In the 5 eyes with type 1 staphyloma, the locations of detachment were all in the nasal half of the peripapillary area. In the other 5 eyes with type 2 and 3 staphyloma, the locations of detachment were all in the non-nasal peripapillary area. The difference between these two groups was statistically significant (Fisher's exact test, ,P= 0.01).CONCLUSION: Peripapillary detachment is probably a benign complication of posterior staphyloma in pathologic myopia. The site of peripapillary detachment is affected by the location of staphyloma and OCT is important in making the diagnosis.
6.Molecular quantification for quality control of Jinqian Baihuashe in Jinlong Capsule

The quality control of Chinese patent medicines containing animal-derived crude drugs is relatively difficult, because the effective constituents of most animal-derived crude drugs remain unknown. Even if there are relevant methods, they are usually qualitative, and quantitative indicators are either lacking or have poor specificity. This paper has proposed to use molecular quantitative technology to control the quality of Chinese patent medicines containing animal-derived crude drugs. In this study, a molecular quantitative method based on fluorescence quantitative PCR was established for the determination of Jinqian Baihua She in Jinlong Capsule. Objective To explore the experience of mothers who provide the first direct breast-feeding for preterm infants in hospital. Methods Using qualitative phenomenological research methods, eleven mothers who provided the first direct breast-feeding for preterm infants in hospital participated in semi-structured interviews. Results Four themes were concluded: the motivation of direct breast feeding in hospital, the lack of experience in breastfeeding, the psychological experience of breastfeeding, and the breast feeding health guidance for premature infants in the hospital expected by mothers. Conclusions Mothers who provide premature infants the first direct breast-feeding in hospital were facing many problems ,there is an urgent need to develop the hospital health guidance , to provide effective social support, helping them transit to direct breastfeeding smoothly and safely after discharge.
8.Determination of carbocisteine concentration in human plasma by HPLC-MS

Objective To establish a sensitive,valid and rapid high performance liquid chromatography-trap mass spectrometry (HPLC-MS) method for determining carbocisteine concentration in human plasma. Methods Plasma samples were pretreated by 100 mL/L perchloric acid to remove the interference protein. The supernatant was separated by reverse phase HPLC,electrospray MS was used to detect carbocisteine in plasma samples,and selective ion method of multiple reaction modes was used to determine the concentration of carbocisteine. Results The limit of detection was 0.01 ?g/mL and the linear range was 0.2-200.0 ?g/mL. The recovery rate was within the range of 89.2%-105.6%. Conclusion The proposed method could be used to determine carbocisteine in biological samples.
9.Evaluation of imaging performance of major image guidance systems

Purpose: The imaging characteristics of two popular kV cone-beam CT (CBCT) and two MVCT systems utilised in
image-guided radiation therapy (IGRT) were evaluated.
Materials and methods: The study was performed on Varian Clinac iX, Elekta Synergy S, Siemens Oncor, and
Tomotherapy. A CT phantom (Catphan-504, Phantom Laboratory, Salem, NY) was scanned for measurements of image
quality including image noise, uniformity, density accuracy, spatial resolution, contrast linearity, and contrast resolution.
The measurement results were analysed using in-house image analysis software. Reproducibility, position correction,
and geometric accuracy were also evaluated with markers in a smaller alignment phantom. The performance evaluation
compared volumetric image properties from these four systems with those from a conventional diagnostic CT (CCT).
Results: It was shown that the linearity of the two kV CBCT was fairly consistent with CCT. The Elekta CBCT
with half-circle 27-cm FOV had higher CT numbers than the other three systems. The image noises of the Elekta kV
CBCT, Siemens MV CBCT, and Tomotherapy fan-beam CT (FBCT) are about 2-4 times higher than that of the Varian
CBCT. The spatial resolutions of two kV CBCTs and two MV CBCTs were 8-11 lp/cm and 3-5 lp/cm, respectively.
Conclusion: Elekta CBCT provided a faster image reconstruction and low dose per scan for half-circle scanning.
Varian CBCT had relatively lower image noise. Tomotherapy FBCT had the best uniformity.
10.Prognostic value of the ABCD2 score on long-term follow-up of transient ischemic attack using the new tissue-based definition

score is validated for evaluating short-term stroke risk after transient ischemic attack
(TIA); however, whether it is able to predict the long-term risk of vascular outcome remains uncertain.
Recently a new tissue-based definition of TIA has been proposed. The ABCD2
scores of 145 TIA
patients admitted to our hospital were retrospectively calculated and stratified into two categories:
≤ 3 points (low risk); 4-7 points (moderate-high risk). At a median follow-up of 81 months, new
vascular events were recorded. Follow-up data were available in 107 patients. Seventy one patients
had a moderate-high ABCD2
score. Sixty six patients experienced a cerebral ischemic event; 8 a
myocardial infarction; 7 died of cerebrovascular or cardiovascular cause. Moderate-high ABCD2
score
was significantly associated with the further cerebral ischemic events (hazard ratio [HR], 1.755; 95%
confidence interval [CI], 1.019 to 3.024) and with the combined endpoint (HR, 1.818; 95% CI, 1.079
to 3.063). Our study shows that the ABCD2
score may also be used to predict long-term vascular
outcome after tissue-based definition of TIA. Moderate-high ABCD2
score is associated with an
increased general vascular risk in the long-term follow-up after TIA.
