1.The effects of CD28 superagonist antibody in rat obliterative airway disease

Objective:To investigate the effects of superagonistic CD28~- specific monoclonal antibody JJ316 (supCD28 MAb) on preferentially expanded rat CD4~+CD25~+Treg (Treg) cells in vivo and its applicability in obliterative airway disease (OAD).Methods:The heterotopic tracheal transplantation model in rats was used.One group received mIgG-treatment(0.5 mg/rat) as control.The experimental group was treated with supCD28 Mab(0.5 mg/rat) via intraperitoneal injection on the day of transplantation.The changes of Treg cell population in cervical lymph nodes were monitored by flow cytometry after 7 days.Tracheas were harvested after 21 days for further histologic evaluation.Results:SupCD28 MAb administration was revealed with a significant increase in the CD4~+CD25~+ T and CD4~+Foxp3~+ T cells population in cervical lymph nodes compared to treatment with mIgG group on day 7 after transplantation,[8.5%±3.4% and 11.5%±2.7% (P<0.05 vs mIgG group)in the supCD28 Mab group,1.8%±1.9% and 3.2%±2.1% in the mIgG group,respectively].Furthermore,the allografts from animals treated with supCD28 MAb were significantly less airway obliteration and destruction of the epithelium compared with that of control group animals on day 21 after transplantation.Conclusion:SupCD28 MAb targets expansion of Treg cells and attenuates airway lumen obliteration in rat obliterative airway disease.
3.Application of real-time intraoperative neuronavigation integrated with ultrasound in resection of deep seated brain tumor

Objective To assess the value of real-time intraoperative neuronavigation integrated with ultrasound in the resection of deep-seated brain tumor. Methods Thirty patients with deep-seated brain tumor were treated with microneurosurgery guided with real-time ultrasound integrated with the Brain Lab IGSonic navigation. During the ultrasound based operation, the degree of brain shift and the tumor border was timely observed, and then the tumor was resected totally. Results Guiding with navigation integrated ultrasound, we noticed the brain shift with various degrees happening in 30 patients,and the border of tumor was exposed and the tumor was resected totally without serious complications.Conclusions Intraoperative ultrasound integrated with navigation is a reliable guidance which can accurately re-localize the border of deep-seated brain tumor even when the tumor is shifting, timely delineate the reformatted images from ultrasound and totally resectcd the tumor, thus decrease the surgical time and increase the safety of surgical procedure.
4.Application of self-made health education video in health education for patients with liver cancer:evaluation of its clinical effect

Objective To discuss the health education effect of a self-made health education video for patients with hepatic carcinoma who are receiving transcatheter arterial chemoembolization (TACE)therapy.Methods Based on the needs of patients,a TACE health education video was designed and produced,which was input into the tablet personal computer,such as iPad,to let patients watch.A total of 451 patients with liver cancer,who were admitted to the Department of Intervention,First Affiliated Hospital of Henan University of Technology and Science,China,to receive TACE during the period from June 2014 to June 2016,were included in this study.By using the random number table method,the patients were divided into the observation group (n=226) and the control group (n=225).The self-made health education video together with regular education was used to educate the patients of the observation group,while routine oral and written education was executed for the patients of the control group.The cognition to TACE-related knowledge,the compliance with postoperative treatment and nursing,and the patient's satisfaction were assessed and the results were compared between the two groups.Results The cognition in the following three aspects,including TACE-related knowledge,perioperative education and discharge education,in the patients of the observation group was significantly higher than that in the patients of the control group (P<0.05).The compliance with postoperative treatment and nursing in the patients of the observation group was remarkably higher than that in the patients of the control group (P<0.01).The patient's satisfaction in the patients of the observation group was strikingly higher than that in the patients of the control group (P<0.01).Conclusion Health education by using a self-made health education video can improve liver cancer patient's cognition to TACE as well as the patient's compliance with postoperative treatment and nursing.The health education in the form of watching video can significantly improve health education effect and the satisfaction of the patients.Thus,it is worthy of clinical application.
5.Inhibition of chromosomal region maintenance 1 suppresses the migration and invasion of glioma cells via inactivation of the STAT3/MMP2 signaling pathway

Chromosomal region maintenance 1 (CRM1) is associated with an adverse prognosis in glioma. We previously reported that CRM1 inhibition suppressed glioma cell proliferation both in vitro and in vivo. In this study, we investigated the role of CRM1 in the migration and invasion of glioma cells. S109, a novel reversible selective inhibitor of CRM1, was used to treat Human glioma U87 and U251 cells. Cell migration and invasion were evaluated by wound-healing and transwell invasion assays. The results showed that S109 significantly inhibited the migration and invasion of U87 and U251 cells. However, mutation of Cys528 in CRM1 abolished the inhibitory activity of S109 in glioma cells. Furthermore, we found that S109 treatment decreased the expression level and activity of MMP2 and reduced the level of phosphorylated STAT3 but not total STAT3. Therefore, the inhibition of migration and invasion induced by S109 may be associated with the downregulation of MMP2 activity and expression, and inactivation of the STAT3 signaling pathway. These results support our previous conclusion that inhibition of CRM1 is an attractive strategy for the treatment of glioma.

7.Effect of Internet addiction on adolescent's attention

Objective To explore the effect of Internet addiction on adolescent's attention.Methods Neuropsychological evaluations of attention function were done separately in 18 adolescents who met the diagnostic criterion for Internet addiction and in other 18 ones without Intemet addition tendency as a control group. With auditory and visual oddball paradigms, and stimulated by standard,target and novel stimuli, EEG was recorded and analyzed to get the event-related potential P300 and compare the latency and amplitude of P3a and P3b between the 2 groups. Results Compared with the controls, the attention of Internet addiction group was decreased significantly in neuropsychological evaluations. The latency of P3a potentials induced by novel stimulus was much shorter and the amplitude of it was higher in addiction group than in control group, but the latency of P3b potentials generated by target stimulus was prolonged and the amplitude decreased obviously. Conclusions Intemet addiction can cause damage to adolescent's attention function. There is a correlation between the attention impairment and the change of P300 potentials.
8. Environmental enrichment ameliorates lipopolysaccharide-induced cognitive impairment by inhibiting NOD-like recepter protein 3 in fla mmasome activation in mice

Objective To investigate the effect of environmental enrichment (EE) on lipopolysaccharide (LPS) induced cognitive dysfunction in mice. Methods A total of thirty six 3 weeks old Kunming mice experienced 8 weeks of EE or standard environment (SE) feeding. After 8 weeks, they were divided into three groups: standard environment+ normal saline (SE+NS) group, standard environment+lipopolysaccharide (SE+LPS) group, environmental enrichment+ lipopolysaccharide (EE+LPS) group. The open field test was used to measure the locomotive of mice, and the cognitive function was determined by novelty object recognition test. The expression of microglial marker ionized calcium binding adaptor molecule-1 (IBA-1) in hippocampus was determined by immunohistochemical staining. The expression of microglial activation marker CD68 and NOD-like receptor protein 3 (NLRP3) inflammasome related protein in the hippocampus was detected by Western blotting. Results In the open field test, there was no difference in the activity among the three groups. Compared with the SE + NS group, SE + LPS group showed decreased discrimination ratio in novelty object recognition task, with remarkably up-regulated expression of CD68 in the hippocampus (P< 0. 01) . In addition, SE+LPS group exhibited significantly enhanced expression of NLRP3, apoptosis associated speck-like protein (ASC), Caspase-1 and interleukin-1β(IL-1β) in the hippocampus compared with SE + NS group (P < 0. 05) . Compared with the SE + LPS group, EE+LPS group showed enhanced discrimination ratio in the object recognition task, with down-regulated expression of CD68, NLRP3, ASC, Caspase-1, IL-1β and IL-18 in the hippocampus (P < 0. 01) . Conclusion Environmental enrichment can alleviate LPS induced cognitive dysfunction, which might be attributed to the inhibiting of microglia and NLRP3 activation in the hippocampus.
9.Study on HPLC Fingerprint and Content Determination of 5 Components in Mahai Zhitan Capsule

OBJECTIVE: To establish the fingerprint of Mahai zhitan capsule, to determine the contents of main components, and to provide scientific basis for the stability and quality control of the preparation technology. METHODS: The determination was performed on Inertsil ODS-3 column with acetonitrile-0.1% phosphoric acid as mobile phase (gradient elution) at the flow rate of 1.0 mL/min. The detection wavelength was set at 250 nm (0-23 min and 31-120 min) and 230 nm (23-31 min). The column temperature was set at 30 ℃. HPLC fingerprint for 10 batches of Mahai zhitan capsule was established by using “similarity evaluation software for chromatographic fingerprint of traditional chinese medicine” (2012 edition) and the similarity was evaluated. The chromatographic peaks were assigned and identified with reference substance, negative samples without ingredient and substance control respectively, and the identified main components were quantitatively analyzed. RESULTS: The similarity of 10 batches of sample was more than 0.99; 20 common peaks were found, and 10 common peaks were identified. Among them, No. 1,13,14,15,16,17,18,19,20 chromatographic peaks originated from Rheum palmatum; No. 3,4,6,7 chromatographic peaks originated from processed Strychnos nuxvomica; No. 8 chromatographic peaks originated from Angelica sinensis; the corresponding source of medicinal materials was not found in No. 2,5,9,10,11,12 chromatographic peaks. By comparing the reference substances, No. 1,4,6,7,8,16,17,18,19 and 20 chromatographic peaks were identified as gallic acid, loganin acid, strychnine, brucine, ferulic acid, aloe-emodin, rhein, emodin, chrysophanol and emodin methyl ether, respectively. In the determination of identified five main components (loganin, strychnine, brucine, emodin and chrysophanol), the methodological investigation met the relevant standards. In 10 batches of samples, the contents of loganin, strychnine, brucine, emodin and chrysophanol were 2.477 1-2.785 9, 1.746 1-1.946 0, 1.374 6-1.505 8, 1.573 2-1.824 1 and 0.232 1-0.261 7 mg/g, respectively. CONCLUSIONS: The established method is reliable, accurate, stable and simple, which could provide reference for the preparation technology and quality control of Mahai zhitan capsule.

Lower electrode temperatures (38-41℃) can accurately measure blood carbon dioxide partial pressure in very low birth weight infants, and thus can be used to replace the electrode temperature of 42°C. Transcutaneous measurements at the lower electrode temperatures may be helpful for understanding the changing trend of blood oxygen partial pressure.
