1.Effect of DNA vaccine on behavior and brain Aβ deposition in Alzheimer's disease transgenic mice

Objective To study on the effect of DNA vaccine, p(Aβ3-10) 10-mIL-4, immunization on behavior and brain Aβ deposition in APP/PS1 transgenic mice.Methods DNA vaccine p(Aβ3-10) 10-mIL-4 was constructed which expressed fusion protein of ten tandem repeats of Aβ3-10 and mouse IL-4.APP/PS1 transgenic mice were vaccinated with p (Aβ3-10) 10-mIL-4.Aβ42 peptide and pcDNA3.1 (+) were injected to the control groups.Anti-Aβ antibody titers were detected before and after vaccination by enzyme-linked immunosorbent assay (ELISA) ,the spatial learning and memory ability of the mice were evaluated by Morris water maze.The senile plaques in mouse brains were detected by immunohistochemistry.Results Compare with pcDNA3.1 (+) group,immunization with the vaccine p (Aβ3-10) 10-mIL-4 in APP/PS1 mice induced high-titer Anti-Aβ antibodies ((27.49±4.51) μg/ml, P<0.01) ,decreased Aβ deposition (reduced senile plaque number in the cortex and hippocampus by 52.86% and 58.29% respectively, P<0.01) and improved their cognitive ability.Conclusions The immunization with p(Aβ3-10)10-mIL-4 in APP/PS1 mice achieve an ideal effect of vaccination, and p(Aβ3-10) 10-mIL-4 can be an alternative Alzheimer's disease vaccine to further study.

The present situation of environment polluted by phthalate esters,the main kinds of phthalate esters in the air samples and water samples and the pretreatment and analysis technology of phthalate esters in different environment were reviewed in the present article. In the air samples ,di(2-ethylhexy) phthalate(DEHP),di-n-butyl phthalate(DBP) and dioctyl phthalate(DOP) were included,while diethyl phthalate(DEP),di(2-ethylhexy) phthalate(DEHP),dipropyl phthalate(DPrP),butyl benzyl phthalate(BBP),dioctyl phthalate(DOP) and dimethyl phthalate(DMP) could be mainly detected in the water samples.

Objective To summarize the perioperative nursing of undergoing radical cystectomy with ileal neobladder for the treatment of bladder cancer? Method The nursing measures included preoperative psychological care,bowel and body preparation as well as postoperative nursing of drainage tube,ileal bladder irrigation,bladder functional exercise and basic nursing care? Results No patients suffered from postoperative complications such as infections,urinary fistula and anastomotic leakage? Twenty patients recovered autonomous urination within 3-6 months after the operation? One patient got minor urinal incontinence? Conclusions Cystectomy with orthotopic ileal neobladder trauma contributes to more complications due to large surgical trauma? Therefore,good perioperative care is important for the successful operations?
7.In vivo integrated pharmacokinetics of four flavonoids of Abelmoschus manihot extract in rats

0.99.The quantitive restriction was 0.1 ?g/mL,the recovery rate was over 70%.The RSD of intra-and inter-day was less than 15%.The pharmacokinetic parameters of flavonoids were different from each other after ig and iv administration.The absolute bioavailability of flavonoids were 12.9%,10.8%,2.2%,10.2% and 5.9%,respectively.Conclusion The method is sensitive,specific,accurate,and is not only useful for guiding the bioavailability study on the corolla of A. manihot,but also for establishing a reasonable clinical dosage program.The four flavonols in the corolla of A.manihot can be rapidly distributed and eliminated in rats,the pharmacokinetics of two routes of administration are different.A novel integrated pharmacokinetic approach to describing the holistic pharmacokinetic properties of Chinese materia medica has been successfully developed and validated using four flavonols of A.manihot as a model herbal medicine.This study would be a new try for guiding the holistic pharmacokinetic study in consistence with the intrinsic theory and characteristics of traditional Chinese medicine.
8.Preparation and in vitro evaluation of dispersible tablet of total notoginseng saponin

AIM: To establish the preparative method of dispersible tablet of total notoginseng saponin (DTTNS) by powder direct compression,and to evaluate it pharmaceutical characteristics. METHODS: The effect of factors on the disintegration of DT-TNS was investigated by single factor method,and the formulation was optimized through orthogonal design. RESULTS: The disintegration time of DT-TNS containing 40% total notoginseng saponin was within 1 min while the formulation mainly consisted of 49% MCC as filler,12% of PVPP mixed with 3% L-HPC as disintegrating agent. In addition,the dissolution of DT-TNS was almost finished in 10-15 min. CONCLUSION: The preparative method of DT-TNS by powder direct compression is simple,with short disintegration time and high dissolution rate.
9.Relationship between activity and virulence of secretory acid proteinase and extracellular phosphlipase of Candida albicans

Objective To study the relationship between activity and virulence of secretory acid proteinase and extracellular phosphlipase of different Candida albicans isolated from various specimens.Methods 190 strains of Candida albicans were isolated from various specimens(sputum,blood,thrush,wound,secretion of vaginitis) of patients with Candida albicans infection in hospital.Milk-plate medium and egg-yolk medium were used respectively to test the activities of secretory acid proteinase and extracellular phosphlipase of Candida albicans.The suspension of two forms of Candida albicans(5?106CFU?mL-1) were injected to vein of tail in mice respectively in virulence test.The mortality and mean survival time of mice were observed in 1 month.The virulence was appreciated with the mean survival time of mice.Results All of 190 strains,the positive detectable rates of secretory acid proteinase and extracellular phosphlipase were 83.68% and 85.26%,respectively.Animal experimental results showed the activities of secretory acid proteinase and extracellular phospolipase in hyphal forms were significantly higher than that in spore form(P
10.Determination of adenosin content in Kudiezi Injection by HPLC

AIM: To determine adenosine content in Kudiezi Injection(Ixeris sonchifolia Hance). METHODS: HPLC was used with Shim-pack CLC-ODS(6.0 mm?150 mm,5 ?m). The mobile phase consisted of methanol-acelonitrile-0.025 mol?L -1 phosphoric acid (3∶4∶93). The flow rate was 1 mL?min -1 . The UV detection wavelength was at 260 nm. RESULTS: The adenosin content in Kudiezi Injection was 0.15/10 mL. The average recovery was 98.4%(n=5). CONCLUSION: HPLC can be used for the quality control of Kudiezi Injection Injection.
