1.Content Comparison of Total Flavonoids in Cyperus Rotundus from Different Habitats

Objective:To establish the determination method for total flavonoids in Cyperus rotundus and investigate the total fla-vonoids content in Cyperus rotundus from different habitats. Methods:Total flavonoids in Cyperus rotundus from different habitats were detected by ultraviolet spectrophotometry with rutin as the standard substance. Results:The absorbance of rutin had a good linear cor-relation with the concentration within the range of 0.064 3-0.642 6 mg·ml-1(r =0.999 1). The average recovery was 99.74%(RSD=2. 16%,n=6). The order of total flavonoids content in Cyperus rotundus from different habitats was Henan province >Sichuan province> Guangdong province >Anhui province > Halnan province >Shandong province. Conclusion:The method is easy and ac-curate, which can be applied in the quality control of Cyperus rotundus. The total flavonoids content in Cyperus rotundus from Henan province is the highest.
2.Exploration on construction of faculty of medical informatics teaching

3. Expression of X-linked inhibitor of apoptosis protein and caspase-3 p17 after focal cerebral ischemia/reperfusion in rats

Objective: To study the dynamic expression of X-linked inhibitor of apoptosis protein (XIAP) and caspase-3 p17 after ischemia/reperfusion (IR) in rates. Methods: Thirty male Wistar rats were divided into sham-operated, ischemia 1.5 hours and reperfusion for 6, 12, 24, 48 and 72 hours (n = 5 in each group). A model of reperfusion after 1.5 hours of focal cerebral ischemia was established with the suture method. The expression of XIAP and caspase-3p17 in brains were observed respectively by immunohistochemical method. Results: A small amount of XIAP-positive cells were observed in the non-ischemic side in the sham-operated and IR groups, but no caspase-3 p17 positive cells were observed. The XI-APpositive cells expression began to increase (P = 0. 00) at 6-hour reperfusion, it reached to the peak at 12 hours and began to decrease at 24 hours, and then it gradually returned to normal at 48 hours (P = 0.549) as compared with the sham-operated group; a small amount of caspase-3 p17 positive cells were observed at 6-hour reperfusion, they reached the peak at 24 hours and remained for 72 hours, and they were significantly more than those at 6 hours after reperfusion (P = 0.004). Conclusion: Focal cerebral ischemia may induce the expression of apoptosis-inhibiting gene XIAP and apoptosis-promoting gene caspase-3 p17 to increase transiently, and the peak time of the former expression is earlier than that of the latter.
4.Urgency and Importance of the Establishment of TCM Clinical Pharmacy Training Base

Objective:To discuss the importance and significance of training base establishment for clinical pharmacy of traditional Chinese medicine ( TCM) . Methods:The urgency and necessity of training base establishment for TCM clinical pharmacy was stated based on the status and problems of TCM clinical pharmacy, and the important significance of training base establishment of TCM clini-cal pharmacy was clearly put forward. Results:The establishment of TCM clinical pharmacy training base with the aim of training pro-fessional talents for TCM clinical pharmacy can provide clinical pharmaceutical care for patients with higher quality, which surely will promote the development of TCM clinical pharmacy and the pharmaceutical care level. Conclusion:It is extremely urgent and signifi-cant to set up TCM clinical pharmacy training base, which is the important topic needed to be solved in TCM clinical pharmacy work.

6.Transfection of osteoprotegerin gene into Cos-7 cells

Objective:To transfect human osteoprotegerin( o pg) gene into Cos-7 cells.Methods:The primers were designed b ased on the human opg cDNA sequence,total mRNA was isolated from 293 cells and RT-PCR was performed.The fragment of opg cDNA was inserted into pSecTag 2/B vector and sequenced by automatic sequence analyzer. The recombined plasmid was transfected into Cos-7 cells by Lipofectamine 2000 and OPG protein expressi on in Cos-7 cells was determined by immunohistochemistry and Western blot. Results:The sequence of opg cDNA from 293 cells obtained by RT- PCR was identical to the sequence provided by GenBank [gi:33878056]. The fragm ents of the recombinant plasmid digested with Hind Ⅲ,EcoR I and BamH Ⅰ and exa mined by 10 g/L agarose electrophoresis were consistent with predicted size.OPG over-expressing Cos-7 cells was selected and confirmed by immunohistochemistry and Western blot.Conclusion:Human opg gene may be transfect ed into Cos-7 cells.

8.Enhancing Effect of Exogenous Gangliosides on Learning and Memory in Hypoxic-ischemic Brain Damage Rats

Objective To investigate the effect of exogenous GAs on learning and memory capability of hypoxic-ischemic(HI) brain damage rats. Methods After ligation of the right carotid artery of 7-day old male SD rats and waiting for 4 hours of recovery, the rats were exposed to 8% oxygen-92% nitrogen gas mixture for 2 hours. The HI animals were randomly divided into 3 groups. GAs-treated group 1: animals were injected i.p. GAs immediately after HI every 12 hours for 4 doses with 50 mg/kg, followed by a 2 weeks consecutive daily injections at dose of 30 mg/kg/day. GAs treated group 2: rats received GAs 50 mg/kg immediately after HI every 12 hours for 4 doses, followed by injection of saline in the same volums for 2 weeks. Control group: rats received saline in the equivalent volums and duration. The behavioral test starded when the animals were 24 days old. Discriminative learning ability and memory retention were tested in the tri-equal-arms maze . The locomotor activity and alternative behavior was observed. The degree of neuronal damage were assessed after behavioral test. Results (1)The number of trails to reach the learning criteria were (39?17), (32?14)and (20?13) for control group, GAs-treated group 2 and GAs-treated group 1, respectively. The percentages of memory retention for control group, GAs-treated group 2 and GAs-treated group 1 were (62?10)%, (64?11)% and (70?9)%, respectively. Two weeks consecutive daily injection of GAs had facilitatory effects on learning ability and memory retention of HI rats(P0.05). Conclusion Gangliosides can enhance the learning ability and memory retention in neonate rats with HI.
9.Effects of Wound Treatment Methods on Residual Ovarian Reserve Function after Laparoscopic Ovarian Cystectomy

Objective To investigate the influence of wound treatment methods on residual ovarian reserve function after laparoscopic ovarian cystectomy.Methods A total of 120 patients with ovarian cyst who underwent laparoscopic ovarian cystec‐tomy in the hospital were randomly divided into three groups in terms of different wound treatment methods:electric coagulation group ,ultrasound group and suture group ,with 40 cases in each group.The levels of follicle stimulating hormone(FSH) ,luteini‐zing hormone(LH) ,and estradiol(E2 ) were detected in the blood before and immediately after the operation ,2 days after men‐struation at postoperative 3 and 6 months ,and the number of ovarian reserve follicles determined by transvaginal ultra‐sound.The changes of menstruation and clinical manifestations of the patients were recorded at the same time.Results The lev‐els of LH ,E2 and FSH were dramatically changed in electric coagulation group ,which was followed by the ultrasonic group.Patients in the suture group had the smallest changes of these indices and recovered quickly(P<0.05).In the electric co‐agulation group relative to the other two groups ,the number of ovarian reserve follicles was significantly decreased and the inci‐dence of adverse reactions was profoundly increased.Conclusion Electric coagulation greatly adversely affects the residual ovar‐ian reserve function after laparoscopic ovarian cystectomy ,and ultrasonic and suture hemostasis techniques can help to protect o‐varian function.
