This study was aimed to investigate the effect of baicalin on the proportion of Th22 cells and the concentration of IL-22 bothin vivo andin vitro, in order to explore the immune mechanism of baicalin on inflammatory bowel disease mice model. The 3.5% dextran sodium sulfate (DSS) was used on C57BL/6 mice for the establishment of colitis mice model. Mice were randomly divided into the blank control group, model group, and baicalin group. Flow cytometry and ELISA were used in the detection of the proportion of Th22 cells and concentration of IL-22 in peripheral blood serum, respectively. The spleen lymphocytes of mice were isolated and cultured by baicalin medium (0, 10, 20, 40μM) for 48 h. Flow cytometry was used in the detection of the proportion of Th22 cells. The results showed that baicalin reduced the proportion of Th22 cells and the expression of IL-22 bothin vivo andin vitro experiments. It was concluded that baicalin can inhibit Th22 cell differentiation and expression of IL-22in vitro and DSS-induced colitis mice. It indicated that baicalin had a good treatment potential in Th22 cell-mediated inflammatory diseases.

A real-time polymerase chain reaction ( PCR ) micro total analysis system (μ-TAS ) integrated nucleic acid extraction, PCR amplification and real-time-fluorescent PCR detection on a same microfluidic chip was prepared for the fully automated and on-chip analysis of nucleic acid. The proposed method had the advantage such as low sample consumption, fast analysis and simple operation and so on. Micromachining technology was used to fabricate the anodic molds of integrated nucleic acid extraction microfluidic chip. A polydimethylsiloxane (PDMS) substrate with 3D channels was manufactured by a combination of molds and an injection molding method. The glass substrate and the chip were bonded together using a plasma treatment. The μ-TAS included a microfluidic control device whose micro fluidic velocity ( 0-10 mL/min ) could be adjusted, a TEC platform which the precision of temperature control was 0. 1℃ and a CCD detection module. The DNA of human blood was extracted by using a silica gel membrane method on the microfluidic chip. The processes of DNA extraction and detection were preset in the μ-TAS. Human blood lysate ( 20 μL ) were driven to the extraction chamber and was then washed. The fluidic drive speed was 2 mL/min. DNA and PCR reagents were mixed and then were driven into the PCR chamber. The fluidic drive speed was 1 mL/min. The GAPDH gene in extracted genome DNA was amplified by PCR and detected. The amplified product was verified by melting analysis. The results of nucleic acid extraction method on the chip were compared to those obtained using a standard manual centrifuge extraction method. The amplification curves were obvious. Ct values of the chip method were 25 . 3 and 26 . 9 . The denaturation temperature of all the melting was 89 . 9 ℃. The results validated that the chip-based method and device realized the extraction of nucleic acid, amplification and detection automatically.

Chinese Journal of School Health 2020;41(2):243-246
Objective:
To explore the relationship among dark triad, peer relationship, and cyber bullying of middle school students, and to provide suggestions for intervention of cyberbullying behavior of middle school students.
Methods:
Peer Relationship Scale (PRS), Dirty Dozen (DD) and Cyberbullying Questionnaire (CBQ) were administrated to 1 934 middle school students in Beijing and Yunnan by cluster sampling.
Results:
Middle school boys scored significantly higher on the dark triad and cyber-bullying than girls ( F =13.45, 50.20, P <0.01). The dark triad of middle school students was positively correlated with cyber-bullying ( r =0.38, 0.40, 0.21, P <0.01), while negative dimensions of peer relationship were positively correlated with cyber-bullying ( r = -0.10 , -0.22, -0.16, P <0.01). Peer relationship had a moderating effect on the relationship between dark triad and cyber-bullying ( β =-0.07, t =-3.24,Δ R 2=0.00, P <0.01).
Conclusion
Positive peer relationship can reduce the cyber-bullying behavior of middle school students with high dark triad which should be emphasized among middle school students.

Chinese Journal of Geriatrics 2018;37(10):1156-1160
Objective To explore the characteristics and influencing factors for concurrent anxiety and depression in geriatric patients. Methods A total of 1 318 older people aged over 60 from Beijing were investigated and divided into four groups:high anxiety with high depression, high anxiety with low depression, low anxiety with high depression, and low anxiety with low depression, based on results of Geriatric Depression Scale(GDS)and Geriatric Anxiety Inventory(GAI).Results Logistic regression analysis found that, compared with institution-dwelling elderly,home-dwelling patients were more prone to anxiety (βlow depression =1.66,P =0.01;βhigh depression =2.41,P =0.03),regardless of depression levels, but when anxiety levels were high, depression was easier to relieve in home-dwelling elderly than in their institution-dwelling counterparts(β=-1.50,P =0.01);Compared with patients with low anxiety and low depression, family support (β =-0.16,P =0.06) and friend support(β=-0.23,P =0.01)could reduce the incidence of high anxiety and high depression.Self-care abilities could predict the occurrence of high anxiety and high depression, rather than low anxiety and low depression,in the elderly (β=-1.81,P =0.01).Comorbidities(β=0.28,P =0.09)and neurological disorders were importan t risk factors for concurrent anxiety and depression. Conclusions In addition to physical health and self-care abilities, strong social support and quality elderly care (especially for neurological patients)can help reduce the incidence of concurrent anxiety and depression in elderly people.

Objective To investigate the efficacy and influential factors of post-resection 131 I thera-py in DTC patients with metastases can′t be removed by surgery. Methods A total of 112 DTC patients ( 41 males, 71 females, age range 8-80 years) with metastases from January 2012 to October 2016 in Affil-iated Hospital of Qingdao University were retrospectively analyzed. All patients underwent post-resection 131 I therapy, and the therapeutic effect was assessed. Univariate analysis (χ2 test, rank sum test) and the logis-tic regression analysis were performed to select the influential factors for treatment effect. The ROC curve a-nalysis of significant factors to predict the ineffectiveness of 131 I therapy was performed. Results Patients underwent single or repeated 131I therapy (1-9 times), and the effective rate was 75.89%(85/112). Univa-riate analysis showed that the tumor diameter, the location of metastasis, preablation sTg and sTg/TSH ratio were influential factors for 131 I therapy on metastases(χ2 values:7.187, 15.381;z values:-5.053,-5.187, all P<0.05) . Logistic regression analysis showed that the tumor diameter, preablation sTg were independent influential factors for 131 I therapy ( both P<0.05) . The areas under ROC curve for sTg, sTg/TSH ratio, and tumor diameter to predictive ineffectiveness were 0.824, 0.832, and 0.718, respectively. The cutoff values were 29.825μg/L, 0.298, 3.250 cm, respectively, the sensitivities were 92.60%, 96.30%, 40.70%, and the specificities were 62.40%, 55.30%, 91.80%. Conclusions The therapeutic effect of 131I therapy on metastases from DTC is significant. The longer tumor diameter, the distant metastasis, and the higher preab-lation sTg are negative influential factors for131 I therapy, which could be used to predict the therapeutic effect.
