2.Effective activation of CD11c and anti-tumor immune in immunized mice with recombinant E.coli. LLO/OVA

Objective To investigate the anti-tumor pathway and the effect of recombinant E.coli. LLO/OVA in C57BL/6 mice. Methods Magnetic sorting of splenic CD11c, CD4+ and CD8+ T cell from immunized mice with E.coli. LLO/OVA and E.coli. OVA vaccination were performed. In addition, the lymphocytes mixed reaction and IL-2 and IFN-? in the supernatant were detected. The percentage of OVA257-264 SIINFEKL specific CD8+ T cell were checked by flow cytometry. B16-OVA melanoma protection and inhibition response in E.coli. vaccinated mice were compared. Results Compared to E.coli. OVA, E.coli. LLO/OVA-vaccinated splenic CD11c cells stimulated proliferation of autogenic CD4+ T cells and IL-2 production of these cells. CD11c cells induced autogenic CD8+T cells proliferation and IFN-? secretion. A great number of OVA257-264 SIINFEKL-specific splenic CD8+ T cells were induced and the number of lung tumor nodules was significantly reduced in E.coli. LLO/OVA vaccinated mice. Conclusion Recombinant LLO, as an effective adjuvant of E.coli. OVA, can induce murine splenic CD11c cells activation and play potential roles on CD4+ and CD8+ T cells proliferation and IL-2 and IFN-? secretion of these cells, induce more OVA-specific CD8+ T cells and stimulate stronger tumor inhibition in vivo of vaccinated C57BL/6 mice.
3.The current situation and problem of mental health and social support on clinical and English undergraduates of Chongqing medical university

Objective To understand the status and existing problems of psychological health and social support of medical students ,analyze its influencing factors and provide basis for targeted education and management measures .Methods The social support scale(SSRS) ,symptom self-assessment scale (SCL-90) ,the quality of life questionnaire (SF -36)were used as question-naire survey for 120 college students .Results Positive rate of psychological problems in 91 undergraduates was 24 .18% ,obsessive compulsive disorder appeared the most ,phobia appeared the least .Positive rate of psychological problems in 29 English major un-dergraduates was 41 .38% ,the most frequent symptom was forcing symptoms ,followed by human and depressive symptoms .The interpersonal score of English major undergraduates was higher than that of clinical undergraduates ,with a statistically significant difference (P=0 .039) .Mental health between medical and English major students had a statistical difference (P=0 .010) .Objec-tive support scores between medical and English major students had a statistical significance (P=0 .014) .The highest score was the subjective support ,and the lowest scores was the use of social support .Conclusion The mental health of most of the medical un-dergraduates is not optimistic and there exist different psychological problems .At the same time ,the lower level of social support comes along with the higher level of health problems and vice versa .Therefore ,we suggest that society ,schools ,families and relat-ed management departments should further carry out mental health intervention for medical students .
4.Improvement of Detection Sensitivity of Triglyceride with Methylamine Formate as Ionization Enhancer in Reversed Phase Liquid Chromatography-Electrospray Ionization Mass Spectrometry

A novel ionization enhancer, methylamine formate, was proposed for improving the detection sensitivity of triglyceride in edible oil by reversed phase liquid chromatography electrospray ionization mass spectrometry (LC-ESI-MS).The commonly used isopropanol-acetonitrile-methanol-water and isopropanol-acetonitrile were selected as the mobile phase.By using a reversed phase C18 column, and taking the corn oil in isopropyl alcohol as sample solution, we compared methylamine formate with ammonium formate, as ionization enhancers, for their effect on the detection sensitivity of triglyceride by LC-ESI-MS after screening other different ionization enhancers, such as formic acid, acetic acid, ammonium formate, ammonium acetate, butyl formate, dibutylamine formate, triethylamine formate, diethylamine formate, methylamine formate, and ethylamine formate.The result indicated that, by using methylamine formate, the mass spectral peak response and the signal to noise ratio for trilinoleic glyceride component were 5 times higher than that of ammonium formate.The effect of the concentration of methylamine formate ionization enhancer, the flow rate of the mobile phase and the flow rate of nebulizing gas on the detection of methylamine formate were investigated.The concentration of triglyceride components in corn oil starting to form aggregates was similar in different mobile phases in the electrospray process according to measurement of the relationship between corn oil concentrations and the total ion chromatogram peak area of triglyceride.In particular, the peak area of trilinolein was linear with its concentration in the range of 7×10-7-2×10-4 mol/L (R2=0.9997), but increased slower in the higher concentration range.According to the experimental data, the mechanism for improvement of detection sensitivity of methylamine formate was suggested as that the addition of methylamine mono-charged ions with hydrophobic groups had lower solvation energy, which made the enriched addition ions easily evaporation from the droplet surface, thus improving the electrospray ionization efficiency.This method provided an effective way to improve the detection sensitivity of triglyceride in edible oil by LC-ESI-MS.

Objective:To investigate the role of recombinant E.coli LLO/OVA on regulating the function of murine CD4+ CD25+ Treg cells.Methods:After E.coli LLO/OVA or E.coli OVA vaccination,the murine spleen CD4+ CD25+ Treg,CD4+ CD25- T and CD11c cells were collected respectively by magnetic beads sorting.The concentration of IL-10 in the supernatant of mix cocultured CD4+ CD25+ Treg and CD11c cells,and the suppression role of CD4+ CD25+ Treg cells on the proliferation of CD4+ CD25- T cells were determined.The percentage of OVA specific CD8+ T cells in mouse spleen was analyzed by flow cytometry.The number of metastatic tumor nodules in lungs of the mice transplanted with B16-OVA subcutaneouly was compared before and after dilition of CD4+ CD25+ Treg cells in mice.Results:Compared to E.coli OVA,E.coli LLO/OVA significantly downregulated IL-10 secretion of CD4+CD25+Treg cells and attenuated the suppressive effect of CD4+ CD25+ Treg on the proliferation of CD4+ CD25- T cells(P
6.The cagG gene expression of Helicobacter pylori in various digestive diseases and its clinical signifi-cance

Conclusion The cagG gene of Hp was quite conservative, most of the Hp strains in Chinese patients were cagG positive, it has no specific predisposition to different disorders and shows no significant correlation to the extent of gastric mucosal inflammation. Therefore cagG can' t be served solely as a related pathogenic gene for certain diseases.
7.Expression of DOG1 protein in gastrointestinal stromal tumors and the relationship between them

Objective To analyze the association of DOG1 protein with gastrointestinal stromal tumors(GIST). Methods Seventy-seven patients diagnosed with GIST between January 2005 and June 2010 were enrolled. The DOG1 expression in GIST was determined using immunohistochemistry.The result was compared with that of CD117 and CD34. Results Of 77 GIST patients, the DOG1 was expressed in 74 (96.10%) patients and CD117 in 70 (90. 9%) patients, with no significant difference between DOG1 and CD117 (P>0.05). DOG1 expression was not related to the risk stratification of the tumor, site, age and gender (P>0. 05). Of 7 GIST patients with negative CD117,6 patients were positive for DOG1 expression. Whereas 2 of 3 DOG1 negative patients were positive for CD117 expression. Conclusions DOG1, as a novel marker for GIST,has high sensitivity as CD117. It is a good complementarity in diagnosis of GIST when combine DOG1 with CD117, especially in diagnosis of CD117 negative patients. However, the DOG1 can not be used as an index in eveluating the risk stratification of the GIST.
8.Application of Endoscopic Ultrasonography in Diagnosis and Treatment of Early Esophageal Cancer

Early esophageal cancer is derived from mucosa and submucosa without lymph node metastasis. They are detected generally during endoscopy or physical examination without clinical symptoms. Main treatment of early esophageal cancer is endoscopic resection,and the 5-year survival rate is over 90% . Once symptoms occur,local lymph node metastasis is often present and the prognosis is poor with a markedly reduced 5-year survival rate. Thus it is vital to diagnose and treat esophageal cancer at an early stage. With the development of endoscopic techniques,especially the proficiency of endoscopic ultrasonography(EUS),the detection rate of early esophageal cancer has been increased greatly. EUS also plays an important role in the staging of early esophageal cancer. This article reviewed the application of EUS in diagnosis and treatment of early esophageal cancer.

The function of metabotropic glutamate receptor(mGluRs) in stress response was increasingly gaining recognition. The mGluRs can regulate the level of glucocorticoid(GC), the neurotoxin of glutamate and synaptic plasticity (LTP,LTD). So it may play an important role in stress response. The function of distinct type of mGluRs is different. Their mechanisms in stress response are very complex. So it is very essential to study the function of mGluRs in stress. Furthermore we can explore a new type drug targeting at mGluRs to cure the stress damage.-
10.Construction of a recombinant attenuated Salmonella typhimurium nucliec acid vaccine carrying Helicobacter pylori HpaA and detecton of its immunogenicity

Objective To construct a recombinant live attenuated Salmonella typhimurium nucleic acid vaccine carrying Helicobacter pylori ( H. pylori ) hpaA gene and to detect its immunogenicity. Methods hpaA gene fragment was amplified by polymerase chain reaction (PCR) from the genomic DNA of the standard H. pylori strain and cloned into pUCmT vector. DNA sequence of the amplified hpaA gene was assayed,then cloned into the eukaryotic expression vector pIRES through enzyme digestion and ligation reactions. The recombinant plasmid was used to transform Escherichia coli DH5?,and the positive clones were screened with PCR reaction and restriction enzyme digestion. Then,the recombinant pIRES-hpaA was transformed to LB5000,then the recombinant plasmid was transformed to SL7207,and the recombinant strain was passaged repeatedly. Finally,recombinant pIRES-hpaA was transfected to COS-7 cells with Lipofectamine TM 2000. The immunogenicity of expressed hpaA protein was determined with SDS-PAGE Western blot. Results The hpaA gene fragment of the 750 base pair was amplified from the genomic DNA,and it was consistent with the sequence of the H. pylori hpaA by sequence analysis. It was confirmed by PCR and restriction enzyme digestion that Helicobacter pylori hpaA gene was inserted into the eukaryotic expression vector pIRES and a stable recombinant live attenuated Salmonella typhimurium nucleic acid vaccine carrying Helicobacter pylori hpaA gene was successfully constructed,and specific strip of hpaA expressed by pIRES-hpaA could be detected with Western blotting. Conclusion The recombinant attenuated Salmonella typhimurium nucleic acid vaccine strain expressing HpaA protein with immunogenicity was constructed,it might be helpful for further investigation concerning the immune action of the nucleic acid vaccine in vivo .
