1.Study on dissolution method of Vitamin B4 tablets

Objective To establish a test method for determining the vitamin B4 tablets dissolution.Methods Paddle method was used to 900 mL pH6.8 phosphate buffer solution as the dissolution medium, the temperature of 37℃, the speed of 50 r/min, take time for 45 min.Taking the filtrate through the 0.45μm microporous membrane filtration, vitamin B4 tablets dissolution quantity determined by ultraviolet spectrophotometry, determined the wavelength of 262 nm, dissolution limit for 80% of the labeled amount.Results Vitamin B4 concentration had a good linear relationship with the absorbance in 4~16μg/mL, r=0.9999 (n =5), the average recovery was 100.1%, RSD =1.0% (n=9).Conclusion The method is simple, sensitive and accurate, has certain effect on prescription and study of the process of different production enterprises, can be used for the dissolution determination of vitamin B4 tablets.
2.The Effects of 4 Traditional Chinese Medicinal Compounds on Murine DNFB Dermatitis

Objective To study the relationship between the therapeutic principle and prescription of traditional Chinese medicine and the experimental pharmacologic effects in relation to the treatment of eczema. Methods Murine DNFB allergic contact dermatitis was used as an animal model, and was treated with 4 traditional Chinese medicinal compounds which were composed of Chinese materia medica with activities to suppress type Ⅳ allergic reaction. Results The results showed that the effect of compound Ⅲ composed of Paeoniae obovata Maxim(赤芍), Fructus Gardeniae(栀子), Herba Schizonepetae(荆芥), Herba Spirodelae(浮萍) and Poria(茯苓), possessing multiple potencies of cooling and promoting blood, clearing heat, expelling wind and eliminating dampness, was strongest among the compounds tested, and the compound up regulated the serum level of calcitonin gene related peptide which was lowered in the mice with DNFB induced dermatitis. Conclusion It is suggested that the compounds with suppressive effect on type Ⅳ allergic reaction might improve the therapeutic effect in clinical practice.
3.Studies on modern mechanism of syndrome and treatment of abdominal fullness in Taiyin disease

Abdominal fullness in Taiyin disease,marked by being aggravated after vomiting,is mainly caused by internal exuberance of cold dampness,which indicates that the pathogenic cold dampness is accumulated in not only the interior of intestines,but also the exterior of intestine in the abdominal cavity.According to the description about hardness beneath chest in outline syndrome of Taiyin disease,Taiyin disease may be considered as such diseases as cirrhosis,congestive splenomegaly due to portal hypertension.Sini decoction should be used to treat abdominal fullness in taiyin disease with the function of warming and eliminating cold dampness,which shows the decoction has the function of recovery of cirrhosis of the liver decompensation.
4.Effect of Acupuncture on Apoptosis and Protein Caspase-9 Expression in Hippocampus of Cerebral Ischemia Rats

5.Therapeutic Observation of Warm Needling for Neck-back Myofascial Pain Syndrome

Objective To observe the clinical efficacy of warm needling in treating neck-back myofasical pain syndrome. Method Sixty patients with neck-back myofasical pain syndrome were randomized into a treatment group and a control group, 30 cases in each group. The treatment group was intervened by warm needling therapy, while the control group was by electroacupuncture. The short-form McGill Pain Questionnaire (SF-MPQ) and Physion MD muscle resistance detector (Japan) were adopted to measure the count of tender points (red points and yellow points), Pain Rating Index (PRI), Visual Analogue Scale (VAS), and Present Pain Intensity (PPI) before and after the treatment, and the clinical efficacies were also compared.Result The total effective rate was 83.3% in the treatment group versus 66.7% in the control group, and the difference was statistically significant (P<0.05). The SF-MPQ scores (PRI, VAS, and PPI scores) were significantly changed in the treatment group after the intervention (P<0.05). In the control group, VAS and PPI scores were significantly changed after the treatment (P<0.05). After treatment, there were significant differences in comparing the SF-MPQ scores between the two groups (P<0.05). The counts of red and yellow tender points were significantly changed in the treatment group after the intervention (P<0.05). The count of red tender points was significantly changed in the control group after the intervention (P<0.05). After the treatment, there were significant differences in comparing the number of red and yellow tender points between the two groups (P<0.05).Conclusion Warm needling is an effective approach in treating neck-back myofascial pain syndrome.
6.Levels of Soluble Interleukin-2 Receptor in the Sera and Skin Tissue Fluids of Patients with Vitiligo

In order to study the possible role of soluble interleukin-2 receptor (slL-2R) in the pathogenesis of vitiligo, we measured the levels of slL-2R in the sera and the tissue fluids from skin lesions and uninvolved skin from 41 patients with vitiligo, using the method of sandwich ELISA with monoclonal and polyclonal antibodies. 7.Role of perforin in severe preeclampsia

Objectives To investigate the possible role of perforin (PFN) in the pathogenesis of severe preeclampsia.Methods Thirty-two cases of severe preeclampsia were included in the study.Thirtytwo cases of normal pregnancy were selected as control group in random.The expression of PFN mRNA in the peripheral blood mononuclear cells (PBMC) was detected by reverse transcription (RT)-PCR, and its correlation with mean arterial pressure was analyzed in severe preeclamptic patients.The expression of PFN protein in the decidua was detected by immunohistochemistry.Results ( 1 ) The expression of PFN mRNA in PBMC:the PFN mRNA level in severe preeclamptic group was 1.19 ± 0.31, and that in normal pregnancy group is 0.82 ± 0.28.The PFN mRNA level in severe preeclamptic group was significantly higher than that of control group (P < 0.0l ).(2)Correlation analysis:the mean blood pressure in severe preeclampsia group was (133 ±5) mm Hg( 1 mm Hg =0.133 kPa).There was significant positive correlation between level of PFN mRNA in PBMC and mean blood pressure in severe preeclamptic patients ( r = 0.701, P = 0.000).(3)Decidual PFN protein expression:PFN protein was mainly expressed in lymphocytes and the cytoplasm of decidual stromal cells.The positive ratio of PFN in the decidua of severe preeclamptic patients was 84% ( 27/32), significantly higher than that of control group (53%, 17/32, P < 0.01 ).Conclusions Expression of PFN was significantly increased in severe preeclampsia, and it was of significant positive correlation with mean blood pressure.PFN may participate in the pathogenesis of severe preeclampsia.
8.The change and correlation of glycogen phosphorylase isoenzyme BB in neonates with asphyxia complicated with myocardial injury

Objective To study the change of glycogen phosphorylase isoenzyme BB(GPBB)in neonates with asphyxia complicated with myocardial injury and its correlation with various perinatal factors. Methods Sixty-four neonates with asphyxia(including 39 mild asphyxia and 25 severe asphyxiated neonates,30 neonates with and 34 without myocardial injures)were enrolled and 25 healthy neonates were studied as control. The plasma levels of GPBB were measured by enzyme-linked immunosorbent assay(ELISA). Myocardial enzymes,cardiac troponin I,electrocardiogram,chest X-ray were performed simultaneously. Results The plasma GPBB levels were significantly higher in neonates with myocardial injury(13.84,7.57 ng/ml)than those without myocardial injury(4.97,3.24 ng/ml)and control group(4.95,1.99 ng/ml)( P < 0.01). The sensitivities of GPBB,cTnI and CK-MB in diagnosing myocardial injury were 90%,66.7% and 83.3%,respectively. The sensitivity of GPBB was significantly higher than that of cTnI(χ~2 = 4.812,P < 0.05),with no statistical difference between GPBB and CK-MB(χ~2 = 0.577,P > 0.05). The specificities of GPBB,cTnI and CK-MB in diagnosing myocardial injury were 88.2%,91.2% and 67.6%,respectively. The specificity of GPBB was significantly higher than that of CK-MB(χ~2 = 4.191,P < 0.05),with no statistical difference between GPBB and cTnI(χ~2 = 0.159,P > 0.05). Plasma GPBB levels were significantly higher in neonates with severe asphyxia(14.67,6.09 ng/ml)than those with mild asphyxia(5.61,3.56 ng/ml)and control group(P < 0.01). GPBB levels were higher in mildly asphyxiated neonates than those in control neonates,but with no statistical significance(P > 0.05). Spearman rank correlation analysis showed that plasma GPBB levels positively correlated with the cloudiness of the amniotic fluid(r = 0.500,P < 0.001)and negatively correlated with Apgar score(r = -0.520,P < 0.001). Conclusions This study indicated that GPBB can be used as an ideal biomarker of myocardial ischemia injury in neonates with asphyxia. The determination of GPBB in early neonatal period is useful in detecting and assessing the severity of myocardial injury.

A solid-phase cell enzyme-linked immunoassay is described for screeningand analyzing monoclonal antibodies against the cell surface antigensof human colorectal adenocarcinoma.Horse-radish peroxidase conjugatedProtein A was used to detect the binding of mouse monoclonal antibodiesto human colorectal adenocarcinoma.HR_(8348) cells which had been cultured inand then fixed to the wells of microtiter plates by using glutaraldehyde.Thismethod was found to be specific,reproducible and practical,and especiallyto be advantageous for the large scale screening and analyzing of monoclo-nal antibodies with a panel of cell types.
10.Retroperitoneal laparoscopic partial nephrectomy in treatment of patients with T1b renal carcinoma

Objective To evaluate the clinical effect and safety of retroperitoneal laparoscopic partial nephrectomy in treatment of patients with T1b renal carcinoma.Methods Fourteen patients (11 males and 3 females) with T1b renal carcinoma were retrospectively performed.The age of patients was (54.5 ± 9.2)years old,with 8 cases on the left side and 6 cases on the right side.Tumor diameter was (5.1±1.3) cm.All the patients received retroperitoneal laparoscopic partial nephrectomy.Results None of the 14 cases was converted to open surgery.The operation time was (112.0 ± 24.7) min,the intraoperative blood loss was (64.6 ± 15.9) ml,the warm ischemia time was (26.5 ± 9.3) min.The 14 patients were not blood transfusion in intraoperative and postoperative.Postoperative negative pressure drainage placement time was (3.1 ± 1.5)d,lying in bed time was 72 h.Serum creatinine increase was found in 1 case postoperative 12 h,others were no severe complications.Postoperative pathology:the incisal margin of 14 cases were all negative,clear cell carcinoma was in 13 cases,the pathology stage was T1bNoM0;angiomyolipoma of kidney was in 1 case.All the patients were follow-up 3-16 (21.4 ± 9.6) months,all the patients had normal renal function and had no tumor recurrence or metastasis.Conclusion Retroperitoneal laparoscopic partial nephrectomy is safe and reliable for treatment of patients with T1b renal carcinoma.
