1.Radix Astragali injection at appropriate concentrations enhances surface adhesion of bone marrow stromal cells

BACKGROUND:Several studies have demonstrated that many kinds of Chinese herbs for benefiting vital energy and enriching the blood secrete some cytokines by influencing bone marrow stromal cel s to promote the differentiation and proliferation of hemopoietic stem cel s or exert effects by promoting the adhesion of bone marrow stromal cel s and hemopoietic stem cel s.
OBJECTIVE:To investigate the effects of Radix Astragali injection on the expression of intercel ular adhesion molecule 1 and vascular cel adhesion molecule 1 on the surface of mouse bone marrow stromal cel s.
METHODS:Mouse bone marrow stromal cel s were isolated by adherent culture of whole bone marrow. The morphology of mouse bone marrow stromal cel s were observed using inverted phase microscopy, hematoxylin-eosin staining, optical microscopy and transmission electron microscopy. The optimal concentration at which Radix Astragali injection exhibited the strongest effects on promoting the proliferation of bone marrow stromal cel s was detected using MTT assay. Expression of intercel ular adhesion molecule 1 and vascular cel adhesion molecule 1 on the surface of mouse bone marrow stromal cel s was detected using flow cytrometry after intervention by Radix Astragali injection.
RESULTS AND CONCLUSION:Inverted phase microscopy and optical microscopy showed that bone marrow stromal cel s adhered to the wal of culture flask, exhibited a shuttle-shaped or irregular appearance with processes. Through transmission electron microscopy, organel es were abundant, such as rough endoplasmic reticulum, secretory vesicle, and mitochondria. Radix Astragali injection at 400 and 600 mg/L significantly promoted the proliferation of mouse bone marrow stromal cel s (P<0.05) and there was no significant difference in the cel proliferation between these two concentrations. Radix Astragali injection at 600 mg/L increased the expression of intercel ular adhesion molecule 1 and vascular cel adhesion molecule 1 on the surface of mouse bone marrow stromal cel s. These findings suggest that Radix Astragali injection at appropriate concentrations can increase the surface adhesion of bone marrow stromal cel s, thereby improving hemopoietic microenvironment.
2.Studies on modern mechanism of syndrome and treatment of abdominal fullness in Taiyin disease

Abdominal fullness in Taiyin disease,marked by being aggravated after vomiting,is mainly caused by internal exuberance of cold dampness,which indicates that the pathogenic cold dampness is accumulated in not only the interior of intestines,but also the exterior of intestine in the abdominal cavity.According to the description about hardness beneath chest in outline syndrome of Taiyin disease,Taiyin disease may be considered as such diseases as cirrhosis,congestive splenomegaly due to portal hypertension.Sini decoction should be used to treat abdominal fullness in taiyin disease with the function of warming and eliminating cold dampness,which shows the decoction has the function of recovery of cirrhosis of the liver decompensation.
3.Analysis of emergency obstetric hysterectomy:the change of indications and the application of intraoperative interventions

Objective To investigate the change of indications of emergency obstetric hysterectomy and the clinical application of intraoperative interventions. And to provide evidence for prevention of hysterectomy and improvement of obstetric quality. Methods Clinical data were collected from 97 patients who received emergency obstetric hysterectomy at Shengjing Hospital of China Medical University between January 1st, 2004 and December 31st, 2013. The patients were divided into two groups by the time point of January 1st, 2009: the first group was cases treated between January 1st, 2004 and December 31st, 2008, while the second group was cases treated between January 1st, 2009 and December 31st, 2013. The clinical indicators, surgical indications, intraoperative interventions, and blood loss between the two groups were analyzed retrospectively. Results (1) Incidence:54 857 women delivered at Shengjing Hospital of China Medical University between January 1st, 2004 and December 31st, 2013. Of them, 97 patients received emergency obstetric hysterectomy, with an incidence of 0.177% (97/54 857). (2) The 17 patients delivered vaginally (18%,17/97) and 80 by caesarean section (83%,80/97). Forty-nine patients experienced repregnancy with scar uterus (51%, 49/97). About 41 patients underwent abdominal total hysterectomy (42%,41/97) and 56 received subtotal hysterectomy (58%,56/97). (3) The number of patients were comparable between the two groups (50 vs 47;P>0.05). (4) The main surgical indication was uterine inertia (45%, 44/97). The main causes of uterine inertia were excessive uterine tension (45%, 20/44) and placental abruption due to gestational hypertension (32%, 14/44). Of all the indications, 29 patients in the first group (58%, 29/50) and 15 patients in the second group (32%, 15/47) suffered from postpartum hemorrhage. Pathological placenta embedment occurred in 15 patients in the first group (30%, 15/50) and 25 patients in the second group (53%, 25/47). The incidences of postpartum hemorrhage due to uterine inertia or pathological placenta embedment were significantly different between the two groups (both P<0.05), respectively. (5) In the first group, the average preoperative blood loss was (2 900±1 900) ml, and the average intraoperative amount of infused white&red blood cells was (5.9±3.5) U, with the average operation time of (2.2 ± 1.8) hours and the average in-hospital duration of (7.8 ± 2.3) days. In the second group, the average preoperative blood loss was (3 100± 2 200) ml, and the intraoperative amount of infused white&red blood cells was (6.2± 5.2) U, with the average operation time of (2.5± 2.1) hours and the average in-hospital duration of (7.9 ± 2.9) days. There was no significant difference between the two groups in any of these indicators (P>0.05). Postpartum hemorrhage was usually treated with uterine packing in the first group, but was preferentially treated with potent uterine contraction agents, arterial ligation, uterine balloon compression or B-Lynch suture in the second group. The therapeutic effects of these new treatments were significantly better than uterine packing (P<0.05). Conclusions The incidence of emergency obstetric hysterectomy did not change significantly in the past decade. However, the indications and intraoperative interventions have changed significantly in the second five years compared with the first five years. The main surgical indications were uterine inertia and postpartum hemorrhage due to pathological placenta embedment. Therefore, strict control of caesarean section indications was important to reduce emergency obstetric hysterectomy.
5.Experimental study of dynamic diffusion tensor imaging in spinal cord of goats under persistent compression

Objective To explore the dynamic changes of diffusion tensor imaging(DTI) in spinal cord of goats with persistent compression injury. Methods Eighteen goats weighted 20--25 kg were divided into three groups with completely random design: A, B and C. A balloon catheter was inserted into the epidural space at C3-4 level via intervertabral foramen for each goat. The balloon was inflated by injection of variable volumes of saline in group A and B 10 days following operation. The volume of saline was 0. 3 ml in group A and 0. 2 ml in group B,respectively. The compression sustained for 40 days. Group C served as uncompressed control without injection of saline. The locomotor rating score was applied to each group. Conventional MRI and DTI were performed. The apparent diffusion coefficient (ADC)and fractional anisotropy (FA) values were measured. Histopathological assessments of the compressed spinal cord were performed 50 days following operation with light microscope and transmission electron microscopy. Results Before operation, the locomotor rating score was 5, the ADC value was ( 1.23 ± 0. 05 ) × 10-3 mm2/s and the FA value was (0. 72 ± 0. 05 ) each group. Of six goats in Group A, the locomotor rating score severely decreased and reached( 1.5±0. 4)on the 40 th day after compression. The ADC value at compression site decreased soon and reached the minimum (0. 75±0. 04) × 10-3mm2/s on the 5 th day after compression. Then the ADC value increased gradually, restored normal on the 10 th day or so, then became markedly higher than normal and reached( 1.61±0. 05) × 10-3mm2/s on the 40 th day. The FA value at compression site decreased soon, reached(0. 54±0. 04)on the 1st day, then decreased gradually and reached(0. 43 ± 0.05) on the 40 th day. It appeared high signal intensity on T2WI on the 10 th day. In Group B, the locomotor rating score was moderately decreased and reached(3.4±0. 5 )on the 40 th day. The ADC value at compression site decreased slightly firstly, reached( 1.08±0. 04) × 10-3mm2/s on the 1st day, restored normal on the 20 th day or so, then increased gradually, became higher than normal and reached ( 1.27 ± 0. 05) × 10-3mm2/s on the 40 th day. The FA value increased slightly firstly, reached (0. 78±0. 05 )on the 1st day, then decreased gradually, restored normal on the 15 th day or so, became lower than normal and reached(0. 67±0. 05) on the 40 th day. There was no abnormality on conventional MRI. In Group C, the locomotor rating score, ADC value and FA value remained unchanged. There was no abnormality on conventional MRI. There were dynamic changes over time of the ADC value and FA value in Group A and B, which was more marked in Group A than that in Group B ( repeated measurements analysis of variance, F=426. 7 for the ADC value and F =7895.2 for the FA value, P < 0. 01 ). Histopathologically, swelling and degeneration of axons and neurons as well as the disarrangement of myelin sheathes could be seen. The pathological changes were more marked in Group A than in Group B. In Group C, no abnormality could be seen. Conclusion There are dynamic changes of DTI in cervical spinal cord with compressive injury that correlated with the degree and duration of compression. The ADC value decreased firstly, restored normal and then increased. The FA value increased firstly, restored normal and then decreased in mild compression while solely decreased in serious compression.
6.Mediation effects of attachment, social support and coping style on the links between life events and mental health

Objective To study the effect of attachment, coping style and social support on mental health in technical school students.Methods 372 technical school students were assessed by ECR(Experiences in Close Relationship's Inventory) ,Symptom Check Scale List-90 (SCL-90) and Coping Style Scale For Secondary School Students( CSS), Adolescent Self-Rating Life Events Checklist ( ASLEC), Perceived Social Support Scale (PSSS).Results ( 1 ) Life events had significant correlations with all indexes score of attachment anxiety ( r =0.293 ~ 0.356, P <0.01 ) ,the scale of coping style focus on emotion and the total score of SCL-90 ( r = 0.259 ~0.509, P<0.01 ) ,and the total score of perceived social support had significant correlations with the total score of SCL-90 ( r =-0.238, P < 0.01 ).( 2 ) Structural equation analysis ( model fit: χ2 = 540.593, RMSEA = 0.114,NFI = 0.864 ,CFI =0.884, P < 0.01 ) showed that life events affect on mental health through the mediation effects of coping style focus on emotion, attachment anxiety and the perceived social support and coping style focus on emotion had most mediating effect on mental health ( mediation effect = 33.2% ).Conclusion Life events had both direct effect and indirect effect through coping style focus on emotion, attachment anxiety and the Perceived Social Support on mental health.
7.Effects of Furong-Tongmai capsules on the expressions of interleukin-1 and tumor necrosis factor-α in the sciatic nerve in diabetic rats

Objective To investigate the effects of Furong-Tongmai capsules on the expressions of interleukin-1 (IL-1) and tumor necrosis factor-α (TNF-α) in the sciatic nerve in diabetic rats.Methods A total of 50 rats were randomly divided into the normal control group,model group,and low-,medium-,and high-dose of Furong-Tongmai groups,10 rats in each group.Diabetes mellitus in rats were induced by intraperitoneal streptozotocin injection (60 mg/kg).The rats in the low-,medium-,and high-dose of Furong-Tongmai groups were intragastric administrated with Furong-Tongmai suspension 0.7,1.4 and 2.8 g/kg daily for 8 weeks,respectively.The rots in the normal control group and model group were intragastric administrated with equal-volume normal saline daily for 8 weeks.The expression levels of IL-1 and TNF-α in the sciatic nerve were detected with immunohistochemistry staining.Results The expression levels of IL-1 (1.43% ± 0.17% vs.0.21% ± 0.09%;P<0.05) and TNF-α (1.98% ± 0.12% vs.0.35% ± 0.03%;P<0.05) in the model group were significantly increased than those in the normal control group.The expression levels of IL-1 (0.54% ± 0.14%,0.51% ± 0.13% vs.1.43% ± 0.17%;all P<0.05) and TNF-α (0.57% ± 0.17%,0.49% ± 0.15% vs.1.98% ± 0.12%;all P<0.05) in the medium-,and high-dose of Furong-Tongmai groups were significantly decreased than those in the model group and low-dose of Furong-Tongmai group (1.08% ± 0.18% in IL-1,1.11% ± 0.09% in TNF-α;all P<0.05).Conclusion Furong-Tongmai capsules can reduce the expressions of IL-1 and TNF-α in sciatic nerve in diabetic rats.
8.The role of arginine vasopressin played in proliferation and differentiation of osteoblast

The role of arginine vasopressin ( AVP) played in proliferation and differentiation of mouse primary osteoblast and its mechanism was investigated. 100 nmol/ L AVP was added into the medium containing primary mouse osteoblast: (1) After being cultured for 72 h, the proliferation of the cells was counted with a cell counter. (2) The media of cultured cells on 2,4,6,8,10 days were harvested and tested for the secreted ALP concentration by osteoblasts, and the cells were lysed in order to test the ALP concentration in cytosol. (3) The alizarin red staining was employed to detect the effect of AVP on calcium nodules formation on 8 th and 20 th days. (4) The osteoblast cells were incubated with AVP for 20 min, and then were lysed. Radioimmune assay was applied to test the change of cAMP in cytosol. These results showed that, compared to negative group, 100 nmol/ L AVP significantly promoted the proliferation of primary mouse osteoblast ( P<0. 01). ALP secretion was increased remarkably ( P <0. 01), and the number and area of calcium nodules were increased considerably(P<0. 01). The intracellular cAMP was increased after incubating cells with AVP for 20min ( P<0. 01). These results suggest that AVP may promote proliferation and differentiation of mouse primary osteoblasts by cAMP signal pathway.
9.The detecting value of CTCs in peripheral venous blood of patients with advanced metastatic breast cancer

Objective To detect the value of circulating tumor cells(CTCs)in peripheral venous blood in patients with metastatic breast cancer. Methods 50 female patients with advanced advanced metastatic breast can-cer hospitalized in our hospital from May 2016 to December 2016 were enrolled in the research. Patients were divid-ed into oligometastases group and extensive metastasis group through multi-department comprehensive analysis andimaging diagnosis. 10 cases with early stage breast cancer were selected randomly. 10 healthy female volunteers were recruited as control group. After obtaining written informed consent from research subjects ,7.5 mL peripheral blood was drew from patients and volunteers prior to starting a new line of chemotherapy ,surgery or other treat-ment. CTCs counts from Blood samples were detected density gradient centrifugation associate with flow cytometry. Results The base line was formulated as CTCs≥5/7.5 mL positive and CTCs<5/7.5 mL negative. By comparing the positive expression of CTCs in early and advanced metastatic breast cancer(Pa = 0.01,P < 0.05),positive CTCs was associated with advanced metastatic breast cancer. Comparision of the positive expression of CTCs between oligometastases group and the extensive transfer group showed significant difference in the CTCs count be-tween the two groups(Pb = 0.018,P < 0.05). In the corresponding period,no positive CTCs was detected in all healthy volunteers. Conclusion CTCs count was associated with metastatic breast cancer. There was a correlation between tumor metastasis and CTCs count (the more widely metastasis ,the higher the detection rate of CTCs). CTCs may be of relevant value in the diagnosis and treatment ,and prognosis evaluation of metastatic breast cancer.
10.Magnetic Resonance Urography in Diagnosing Congenital Renal Dysplasia in Children

Objective To approach the value of MR urography (MRU) in diagnosis of congenital renal dysplasia in children.Methods 13 children with congenital renal dysplasia and renal inadequacy were examined by routine MRI and MRU with heavy T_2 weighted single stimulation fast spin echo sequences.Results The congenital atopic dysplastic rens located at pelvis in one case and at below nephritic pit in one case were not detected by MR imaging and original MRU images,respectively.Multicystic dysplastic kidney could be well showed by MRU MIP images, but the cases with clubbed renal pelvis were not well demonstrated by MRU. All cases and the complications were correctly diagnosed by routine MRI, original MRU and MRU MIP images.Conclusion MR urography in combination with routine MRI is of important value in diagnosing congenital renal dysplasia in children.
