1.Related Factors of Ischemic Cerebrovascular Disease in Patients with Patent Foramen Ovale

ObjectiveTo investigate the related factors of ischemic cerebrovascular diseases in patients with patent foramen ovale (PFO). Methods42 patients with PFO,aged ≤55 years old, with cerebral infarction or transient ischemic attack and treated with aspirin were involved. All patients were examined with transesophageal echocardiography. 6 cases relapsed and 36 cases did not. The size of PFO, serum fibrinogen level, and combination of atrial septal aneurysm (ASA) were compared. ResultsThey were followed up for (512.51±127.38) d. The serum fibrinogen level was (3.33±1.11) g/L in no relapse group, and was (4.40±1.49) g/L in relapse group (P=0.032). There was not significant difference in the size of PFO (P=0.618) and combination ASA (P=0.554) between these groups. Logistic regression showed that patients with ischemic cerebrovascular diseases history involved in relapse (OR=46.913,95%CI:2.371~928.254), but the serum fibrinogen level did not (OR=2.656,95%CI:0.977~7.219). ConclusionThe PFO patients with history of ischemic cerebrovascular diseases incline to relapse. It is not sure the influence of the serum fibrinogen level on it.
2.Bcl-2 and Bax expression in mice with collagen-induced arthritis following heterogenic umbilical cord blood stem cell transplantation

BACKGROUND: The occurrenceand development of rheumatoid arthritis were strongly associated with unbalance proliferation and apoptosis of synovial cells and lymphocytes. Some synovial cell apoptosis was abnormal. OBJECTIVE: To observe effects of heterogenic double umbilical cord blood stem cell transplantation on Bcl-2 and Bax expression in mice with type Ⅱcollagen-induced arthritis. METHODS: C57BL/6(H-2b) mice were induced with Freund's complete adjuvant and type Ⅱcollagen to establish mouse models of type Ⅱcollagen-induced arthritis.At 2days after secondary immunity incubation, mice were injected with saline via tail vein in the model and normal control groups. Umbilical cord blood hemopoietic stem cells were injected into mouse tail vein in the UBSCs transplantation groups (single dose:2×106/50 g; double dose: 1×106/50 g each, totally 2×106/50 g). Mice were intragastrically administrated methotrexate in the methotrexate positive control group, 0.017 5 g/kg once, once every 5 days, totally six times. RESULTS AND CONCLUSION: Joint tissue below knee and elbow was obtained at42 days following transplantation. Histopathology displayed that smooth and glossy articular surface, no inflammatory cell infiltration in the synovial layer and normal chondrocytes in the normal control group. Hyperplasia, a lot of inflammatory cell infiltration and damaged cartilage surface were visible in the model group. Slight hyperplasia and a little inflammatory cell infiltration were detectable in the methotrexate positive controlgroup and single UBSCs transplantation group. Double UBSCs transplantation group exhibited smooth and glossy articular cartilage surface, no damage, a little inflammatory cell infiltration. Immunohistochemistry demonstrated that Bcl-2 and Bax expression was lower in the double UBSCs transplantation group compared with single UBSCs transplantation group (P < 0.05). Compared with normal control group, no significant difference was detected (P > 0.05). Results suggest that double umbilical cord blood stem cells can induce synovial cell apoptosis in mice with type Ⅱcollagen-induced arthritis in a certain number and action time, and protect synovial membrane against damage.
3.Dosimetric study of different intensity-modulated modalities in the radiotherapy for mid and upper thoracic esophageal carcinoma

Objective To compare the static intensity-modulated radiotherapy (IMRT) and volumetric modulated arc therapy (VMAT) for mid and upper thoracic esophageal cancer.Method The data of twenty esophageal cancer patients were retrospectively re-planned with VMAT(single arc and double arcs) modality using Pinnacle treatment plan system.Five of these patients were selected again to simulate single arc plans with different segment intervals (4°,3°,2°) and re-planned on other treatment planning systems (Monaco and MasterPlan).Differences of dose distribution and treatment parameters were compared.Results In comparison to IMRT and single-VMAT (S-VMAT),Double-VMAT (D-VMAT) significantly improves the dosimetric parameters for targets(P < 0.05),dose homogeneity(P < 0.05) and conformity(P < 0.05).Though VMAT plans were slightly better than IMRT in reducing the doses to the organs at risk (OARs),no advantage was observed in the low-dose protection of lung and E-P (P < 0.05).For the VMAT plans with different segment intervals,lower OAR doses were observed using an interval of 2°(P < 0.05),except for the mean dose of the heart.For the VMAT plans on different treatment planning systems,Monaco-based plans protected OARs better (P < 0.05).The number of monitor units (MU) and treatment time were less in VMAT cases.Conclusions VMAT plans perform better in target coverage,dose homogeneity and conformity,and can reduce the radiation dose to the spinal cord,lungs,heart and other normal tissue than IMRT plans.The VMAT plan quality could be further improved by using double arcs and smaller segment interval.Monaco-based plans provide better OAR protections under the same conditions of physical and optimization parameters.
4.Analysis on correlation between CYP17 gene polymorphism and hyperandrogenmia in Xinjiang Uyghur patients with polycystic ovary syndrome

Objective To investigate the correlation between the CYP17 gene polymorphisms and hyperandrogenemia(HA) in Uygur women patients with polycystic ovary syndrome (PCOS ) .Methods 59 Uygur patients with PCOS were selected as the Uygur PCOS group and re-divided into the Uygur PCOS complicating HA group and Uygur PCOS non-complicating HA group . Meanwhile 18 Han patients with PCOS complicating HA were selected as the Han PCOS group .The polymerasechain reaction-restrictive fragment length polymorphism method was used to examine the basic group polymorphisms in the promoter region of CYP17 gene .Then the correlation between basic group locus polymorphism and Uygur PCOS complicating HA was analyzed .Results There was no statistically significant difference between TT ,TC and CC genotypes and T ,C allele frequencies in the Uygur PCOS non-complicating HA group and Uygur PCOS complicating HA group (P> 0 .05) .There was no statistically significant difference between TT ,TC and CC genotypes and T ,C allele frequencies in the Uygur PCOS complicating HA group and Han P-COS group(P>0 .05) .The serum testosterone level in TT ,TC and CC genotypes patients of the Uygur PCOS complicating HA group was significantly higher than that in the Uygur PCOS non-complicating HA group(P<0 .05) .Serum testosterone level had no statistically significant difference between TC ,CC genotypes and TT genotype patients of Uygur PCOS group(P>0 .05) .Conclusion The distribution frequency of CYP17 genotypes does not increase the risk suffering from HA in Uygur women .Single nucleotide polymorphism in CYP17 gene promoter region had no obvious correlation with Uygur PCOS complicating HA occurrence .
5.Improvement effects of Dianxianqing granule on blood-brain barrier injury in Alzheimer ’s disease model mice

OBJECTIVE To study the impr ovement effects of Dianxianqing granule on blood-brain barrier (BBB)injury in Alzheimer’s disease (AD)model mice by regulating NLR family pyrin domain containing 3(NLRP3)inflammasome signaling pathway. METHODS Totally 125 mice were randomly divided into sham operation group (n=25)and modeling group (n=100) by body weight. AD model was induced by intracerebroventricular injection of β-amyloid 25-35 in model group. Sham operation group was given normal saline with same method. The 100 model mice were randomly divided into model group ,Donepezil hydrochloride tablets group (positive control 1,1.3 mg/kg,i.g.),MCC950 group [positive control 2(selective NLRP 3 inhibitor),10 mg/kg,i.p.] and Dianxianqing granule group (12.48 g/kg by crude drug ,i.g.)by body weight ,with 25 mice in each group. Second day after modeling ,administration groups were given relevant medicine ,once a day ,for consecutive 21 d. Sham operation group and model group were given intragastric administration of water and intraperitoneal injection of normal saline. At last administration,the learning and memory ability was determined by Y maze test ,and blood-brain barrier permeability was measured by Evans blue leakage assay. The expressions of NLRP 3,anti-ionized calcium-binding adapter molecule 1(IBA-1),nuclear factor kappa B (NF-κB)p65,p53 upregulated modulator of apoptosis (PUMA),occludin(ocln),zonula occluden- 1(ZO-1)and claudin-5 (cldn5) in cerebral tissue were determined. RESULTS Compared with model group , spontaneous alternate response rate ,protein expressions of ocln ,cldn5 lnzyxyqy2003@163.com and ZO- 1 in cerebral tissue were increased significantly in administration groups (P<0.05 or P<0.01);Evans blue E-mail:jiadg2003@126.com content and protein expressions of NLRP 3,IBA-1,PUMA and NF-κB p65 in cerebral tissue were decreased significantly (P<0.05 or P<0.01). CONCLUSIONS Dianxianqing granule can improve BBB injury of AD model m ice by inhibiting NLRP 3 inflammasome signaling pathway.
