Dishevelled (Dvl) is a kind of protein widely existing in human tissue, containing three structural domains:Dishevelled and Axin (DIX);postsynaptic density protein-95, disc large tumour suppressor, zonula occludens-1 (PDZ);and Dishevelled, Egl-10 and pleck-strin (DEP). Dvl participates in the Wnt signaling through different structure domains to play a role in nervous system development and neu-rogenesis after nerve injury.
2.Assemble of magnetic nanoparticles into the structure of cisplatin liposome.

Effects of different procedures of magnetic nanoparticles into the liposome structure on the distribution of magnetic particles in the liposome were investigated. Magnetic liposomes with high-encapsulating rate of cisplatin (CDDP) were obtained. Fe3O4 magnetic nanoparticles which was modified by organic functional group on surface was synthesized by an one-step modified hydrothermal method. The CDDP magnetic liposomes were prepared by a film scattering-ultrasonic technique and the concentrations of CDDP in the liposomes were measured by graphite furnace atomic absorbance spectroscopy. Magnetic liposomes with different microstructure were prepared by the two different procedures, where the magnetic particles were combined with phospholipid before the film preparation to form liposome in procedure I, and drug solution and the magnetic particles were mixed before hydrating the lipids film to form liposome in procedure II. The liposome structure was observed by transmission electron microscope (TEM). The CDDP magnetic liposomes were prepared by the optimized method which was selected by orthogonal test. Encapsulation rate of the magnetic particles distributed in the phospholipid bilayer through the procedure I was 34.90%. While liposome, produced by the procedure II technique, contained magnetic particles in the interior aqueous compartment, which encapsulation rate was 28.34%. Encapsulation rates of both I and II were higher than that of conventional liposome. The release profile of all the three different liposomes in vitro fitted with a first-order equation. Because of distribution of magnetic particles in the phospholipid bilayer, the skeleton of phospholipid bilayer was changed. The releasing tl/2 of magnetic liposomes produced by the procedure I technique is 9 h, which is shorter than that of the other two liposomes. Assemble of magnetic nanoparticles into the structure of liposome was succeeded by the procedure I, which showed superiority than by procedure II whatever in CDDP liposome encapsulation efficiency and content of the magnetic particles and would ensure sustained-release character.
3.Effect and significance of salidroside on Eag1 expression in mouse cervical cancer xenograft

Objective To investigate the effect and significance of salidroside on Eag1 expression in mouse cervical cancer xenograft.Methods The mouse subcutaneous xenograft model of cervical cancer U14 cells was established.Forty female Kunming mice were divided into 4 groups according to the random principle,the xenograft group,cisplatinum group,salidroside group and cisplatinum combined salidroside group.Then the mass size was measured by using the vemier caliper everyday.The xenograft tumor weight was measured by using the electronic scale and the tumor inhibiting rate was calculated.The immunohistochemistry method was adopted to verify that the xenograft tumor was undifferentiated squamous cell carcinoma.The tumor surrounding tissue in the xenograft group was taken as the control group.RT-PCR and Western blot were adopted to detect the expression levels of Eag1 mRNA and protein in mouse cervical cancer xenografts in each group.Results Kunming mice renograft tumor model was established by subcutaneous injection of cervical cancer U14 cells,the tumor formation rate was 100%.The tumor volume and tumor weight in the cisplatinum group,salidroside group and combined group were lower than those in the senograft group,the xenograft tumor volume and tumor weight in the combined xenograft group were significantly lower than those in the cisplatinum group and salidroside group (P<0.05);the expressions of Eag1 mRNA and protein in the xenograft group,cisplatinum group and combined group were significantly higher than those in the control group,while which in the cisplatinum group,salidroside group and cisplatinum combined salidroside group were significantly lower than those in the xenograft group,the Eag1 expression level in the combined group was lower than that in the cisplatinum group and salidroside group,the difference was statistically significant(P< 0.05).Conclusion Salidroside can inhibit the growth of cervical cancer xenograft and Eag1 expression,and the effect in the combined use with cisplatinum is more significant.
4.Effects of Danhong mixture on platelet aggregation of rabbits

Objective To study the effects of Danhong mixture on platelet aggregation of rabbits.Methods 40 rabbits were randomly divided into 5 groups,8 rabbits in each group,of blank control group (treated with 0.5% sodium carboxy methyl cellulose of the same volume),Danhong mixture of 5,2.5,1.25 g/kg group,aspirin group (treated with 0.025 g/kg aspirin).Each group was performed gastric lavage according to the ration of lml/kg,once daily,for continuous 8 days.Blood was obtained from heart 1 hour after last administration,then adenosine diphosphate (ADP),collagen,arachidonic acid (AA) were used for platelet aggregation test.Results Inhibition rate of Danhong mixture (5,2.5,1.25 g/kg) on ADP induced rabbit platelet aggregation were 9.4%,5.1%,and 0.6% respectively,on collagen induced platelet aggregation were 34.5%,11.1%,and 5.6%; on AA induced platelet aggregation were 21.4%,11.6%,and 2.3% respectively.Conclusion Danhong mixture had inhibitive action on platelet aggregation.
5.Influences of Polyethylene Glycol6000 on the Solubility of m-Nisoldipine

Objective To investigate the influences of polyethylene glycol on the solubility and in vitro dissolution of m-nisoldipine,which could provide guidance for chosing formulations of m-nisoldipine. Methods Solid dispersions of m-nisoldipine were prepared by solvent-melting method with polyethylene glycol6000 matrix. DSC and XRD spectroscopy were applied to identify the solid dispersions. The solubility and in vitro dissolution were detected by UV spectroscopy. Results The DSC and XRD map were different from the crude drug and their physical mixtures. The dissolution rates(13,15,17) were faster(35. 31%,38. 71%,41. 48%) than that of the crude drug(26. 80%),and the dissolution rates of the solid dispersions in the same ratio were higher than the physical mixtures. Conclusion DSC analysis indicated that eutectic compounds were produced by the m-nisoldipine and polyethylene glycol,in which polyethylene glycol6000 acts as a carrier. The solubility and in vitro dissolution of m-nisoldipine can be increased.
6.Preparation of m-Nisoldipine Solid Dispersion

OBJECTIVE:To prepare m-nisoldipine solid dispersion from poorly-soluble m-nisoldipine so as to improve its solubility and dissolution rate in vitro. METHODS: Solid dispersions of m-nisoldipine were prepared by coprecipitation method with poloxamer as carrier; Differential scanning calorimetry (DSC) was used to determine the status of nimodipine in carrier. The solubility and the dissolution rate of the solid dispersion in vitro were studied. RESULTS: DSC analysis indicated that eutectic mixture was formed from m-nisoldipine and poloxamer. The solubility of m-nisoldipine and the its solid dispersions prepared from m-nisoldipine and poloxamer at different ratio (1∶3, 1∶5, 1∶7) were 0.89, 4.50, 15.35, and 23.03 mg?L-1, respectively, and their 120 min dissolution rates were 26.80%, 38.57%, 41.38%, and 45.92%, respectively. In the same ratio, the dissolution rates of the solid dispersions were higher than those of their physics mixtures. CONCLUSIONS: The solid dispersion of m-nisoldipine prepared with poloxamer as carrier can increase the solubility and dissolution rate in vitro.
7.Analysis of monitoring data on congenital heart disease in Shenzhen city from 2009 to 2013

Objective To analyze monitoring data on congenital heart disease (CHD) in Shenzhen city from 2009 to 2013.Methods This study described epidemiological characteristics of perinatal CHD.Patients with CHD were divided into isolated CHD group and complex CHD group (the patients were found with malformations of other systems).The prognosis,social and demographic characteristics,and complications of pregnancy were analyzed.Results The average incidence rate of CHD was 7.25‰,of which 94.36% were diagnosed by ultrasound,of which 33.20% were diagnosed before delivery.The top three cardiovascular abnormalities were patent ductus arteriosus,patent foramen ovale,and ventricular septal defect.There were about 14.11% of CHD with other system malformations,mainly including musculoskeletal system,facial features,and nervous system.Compared isolated cardiac malformations and complex cardiacmalformations,significant differences were found in the age stratification,culture degree,prognosis,diagnosis time,incidence rate of complication of pregnancy,and childbirth complications rate (P <0.01).Conclusions Most CHDs were diagnosed by ultrasound.Simple cardiovascular malformation was common,especially CHD with two or more types of cardiovascular malformations.The incidence rate of CHD in 30 to 40-year-old women,stillbirth rate,incidence of prenatal diagnostic rate were significantly higher than the simple CHD group.
8.The application value of MSCT in diagnosis of acute appendicitis

Objective To discuss the application value of MSCT in the diagnosis of acute appendicitis.Methods 30 patients with appendicitis were enrolled as a research group,meanwhile 30 healthy adults without any diseases were as a control group.Some MSCT findings were observed or measured including diameter and position of appendix,fluid and gas density in appendix cavity, periappendicural fat density and peritoneal thickening,ascites and so on.Results The outside diameter of acute appendicitis appen-dix was (9.78±2.33)mm on average,whereas the diameter of normal adults appendix was (5.05±0.53)mm,exhibiting a signifi-cant difference (t=24.85,P <0.01).Rough periappendicural fat with increased density in 25 patients (83.33%)with the CT value of about -40 to 20 HU,exudation and peritoneal thickening around the appendix in 13 (43.33%),thickening and edematous appen-dix walls in 29 (96.67%),fecal stone in the appendix cavity with high density in 9(30.00%),the appendicitis in combination with intestinal obstruction in 1 (3.33%),and the appendicitis with peritoneal thickening in 80.00% and with peritoneal effusion in 23.33%were found in the study.Conclusion Full abdominal MSCT can help to the diagnosis of acute appendicitis,and some MSCT findings can be useful for the diagnosis including the appendix diameter≥6 mm,appendix cavity effusion,lumen stone,peripheral increased adipose density and peritoneal effusion.
9.The proliferative effect of CⅡ 260-272 peptide on lymphocytes of collagen-induced arthritis rat

ObjectiveTo examine the proliferative effect of synthetic C Ⅱ260-272 peptide on T cells of collagen-induced arthritis(CIA) rat,and to explore the role of C Ⅱ 260-272 in RA.MethodsThe rat model of collagen-induced arthritis(CIA) was established.The T lymphocytes were isolated and incubated with C Ⅱ 260-272 peptide.Proliferation of T cells was determined by cell counting kit-8.The data were analyzed with SPSS 17.0.The Mann-Whitney U test was used for proliferation levels comparison between the two groups,and the relationship of cell proliferation with other indicators was analyzed with Spearman's correlation.Antibody levels between the two groups were compared using t test.ResultsThree weeks after the first immunization,T cell response to C Ⅱ 260-272 in the CIA groups [0.963 (0.332-1.628)] was more significant than in controls[0.332 (0.331-0.357)](P<0.05),and the response was associated with disease course(r=0.624,P<0.01 ).Strong correlation between T cell proliferation and the antibodies,including antibC Ⅱ antibody and anti-Cit-bC Ⅱ antibody was also observed(P<0.01 ).Two weeks later,the levels of anti-C Ⅱ peptide antibody in each CIA group were significantly higher than those in the controls(P<0.01).The antibodies strongly correlated with pannus formation (P<0.01,P<0.05).ConclusionThe proliferative response of T cell to C Ⅱ 260-272 peptide can be found in the early stage of CIA rat,and the anti-C Ⅱ peptide antibody could be detected in the serum,which suggeststhat the C Ⅱ peptide may play an important role in activating T/B lymphocytes and causing immune reaction when CIA is induced by C Ⅱ.
10.Clinical significance of miR-26a-5p-regulated MCL-1 expression in pre-eclampsia

AIM:To investigate the clinical significance of microRNA-26a-5p (miR-26a-5p)-regulated mye-loid cell leukemia-1 (MCL-1) expression in the development of maternal preeclampsia.METHODS:Plasma and placen-tal tissues were collected from 21 cases of normal pregnancy, 13 cases of maternal gestational hypertension, 15 cases of mild preeclampsia and 26 cases of severe preeclampsia.The levels of plasma and placental miR-26a-5p and placental MCL-1 mRNA were detected by real-time PCR.Western blotting analysis was used to determine the protein expression of placen-tal MCL-1.The clinical significance of the above parameters was also analyzed.RESULTS:miR-26a-5p expression gradu-ally increased(P<0.01) in the 4 groups of maternal plasma and placentas with the disease development, and the mRNA expression of MCL-1 was significantly reduced in the placentas (P<0.01), both showing a significant negative correlation (P<0.01).Meanwhile, the expression of miR-26a-5p and MCL-1 protein in the placental tissues was negatively correla-ted (P<0.01).The miR-26a-5p up-regulation in maternal plasma and placental tissues was negatively correlated with ges-tational age, maternal plasma albumin levels and fetal weight, while it was positively correlated with maternal blood pres-sure and urinary protein level (P<0.01), which was in contrary to the down-regulation of placental MCL-1.CONCLU-SION:Up-regulation of miR-26a-5p is involved in the occurrence and development of preeclampsia by down-regulation of MCL-1.
