1.Roles of THEM4 in the Akt pathway:a double-edged sword

The protein kinase B(Akt)pathway can regulate the growth,proliferation,and metabolism of tumor cells and stem cells through the activation of multiple downstream target genes,thus affecting the development and treatment of a range of diseases.Thioesterase superfamily member 4(THEM4),a member of the thioesterase superfamily,is one of the Akt kinase-binding proteins.Some studies on the mechanism of cancers and other diseases have shown that THEM4 binds to Akt to regulate its phosphorylation.Initially,THEM4 was considered an endogenous inhibitor of Akt,which can inhibit the phosphorylation of Akt in diseases such as lung cancer,pancreatic cancer,and liver cancer,but subsequently,THEM4 was shown to promote the proliferation of tumor cells by positively regulating Akt activity in breast cancer and nasopharyngeal carcinoma,which contradicts previous findings.Considering these two distinct views,this review summarizes the important roles of THEM4 in the Akt pathway,focusing on THEM4 as an Akt-binding protein and its regulatory relationship with Akt phosphorylation in various diseases,especially cancer.This work provides a better understanding of the roles of THEM4 combined with Akt in the treatment of diseases.
2.Non-contact vital signs monitoring systems in the ICU based on imaging photoplethysmography technology

Objective:To explore the application of photoplethysmography (iPPG) for contactless vital signs monitoring in the intensive care unit (ICU).Methods:Ten tracheostomy patients in intensive care had their heart rate, oxygen saturation, and diastolic and systolic pressures monitored using iPPG technology and a 24-hour bedside monitor. The readings included periods at rest, during turning, during suctioning, and when undergoing vigorous physical therapy and occupational therapy. The monitoring lasted 3 consecutive days. The data collected by the two methods were compared to analyze the accuracy of the contactless vital signs monitoring system.Results:The oxygen saturation readings of the two systems showed no significant differences. The heart rates, diastolic pressures, and systolic pressures did, however, differ significantly.Conclusions:In the situations tested, contactless monitoring of oxygen saturation is effective, but there is still significant room for improvement in the three indicators of heart rate, systolic pressure, and diastolic pressure.
3.Correlation analysis between interferonγ -inducible protein -10 and expression of PPAR and FABP-4 in pregnant women with preeclampsia

Objective To investigate the correlation between interferon γ-inducible protein-10 and expression of peroxisome proliferator-activated receptor (PPAR) and fatty acid-binding protein-4 (FABP-4) in pregnant women with preeclampsia. Methods 62 PE pregnant women who were treated in our hospital from January 2016 to January 2017 were enrolled in this study. They were divided into mild PE group (34 cases) and severe PE group (28 cases) according to their severity of disease. In addition, 30 healthy pregnant women in our hospital were selected as the control group. The levels of IP-10 in the three groups were measured by enzyme-linked immunosorbent assay (ELISA), and the levels of PPAR and FABP-4 and the correlation between them were analyzed using western blot assay. Results The levels of IP-10 in mild and severe PE pregnant women were significantly higher than those in the control group, and the mild group was lower than the severe group(P<0. 05). Compared with the control group, the level of PPAR in mild and severe PE pregnant women decreased significantly, and the severe group was lower than the mild group (P<0. 05). Compared with the control group, FABP-4 levels in mild and severe PE groups were significantly elevated, and the mild groups was lower than the severe group(P<0. 05). There was a negative correlation between the level of PPAR and IP-10 (r=-0. 529, P<0. 05), and a positive correlation between FABP-4 and IP-10(r=0. 692, P<0. 05). Conclusion There is a certain correlation between PPAR and FABP-4 and the level of IP-10 in pre-PE pregnant women. lt can be used to determine the condition of pre-PE in pregnant women.
4.Expression of microRNA-3620 in blood plasma and its clinical significance in patients with anti-tuberculosis drug-induced hepatotoxicity

Objective To investigate the expression of microRNA (miRNA)-3620 in the plasma of patients with anti-tuberculosis drug-induced hepatotoxicity (ATDH).Methods A total of 35 patients with ATDH and 35 non-ATDH paired individuals were included in this study.Plasma miRNA-3620 levels were detected using real-time Polymerase chain reaction.Comparison between two groups was done with t test.Receiver operation characteristic (ROC) curve analysis was performed to determine the diagnostic value of miRNA-3620 in ATDH.Results The relative expression of plasma miRNA-3620 of patients with ATDH and non-ATDH were 1.65±1.43 and 0.71±0.45, respectively, with significantly statistical difference (t=3.703, P<0.01).The cut off value of miRNA-3620 expression was 1.15 and the area under ROC curve were 0.71(95% CI: 0.43-1.45).Based on this cutoff value, the sensitivity and specificity of miRNA-3620 in diagnosing ATDH were 60.0% and 82.9%, respectively;the positive predictive value was 77.8% and the negative predictive value was 67.4%.Twenty-one ATDH cases and 29 non-ATDH cases was correctly diagnosed, with the accuracy of 71.4%.Conclusion The expression of miRNA-3620 in plasma is significantly increased in ATDH patients.
5.Observations on the Efficacy of Combined Acupuncture and Medicine in Treating Polycystic Ovarian Syndrome Infertility

Objective To investigate the clinical efficacy and safety of combined acupuncture and medicine in treating polycystic ovarian syndrome infertility.Methods Ninety patients with polycystic ovarian syndrome infertility were randomly allocated to treatment and control groups, 45 cases each. The control group received oral administration of clomifene citrate tablets plus muscular injection of chorionic gonadotropin and the treatment group, electroacupuncture in addition. The ovulation rates, pregnancy rates, incidences of ovarian hyperstimulation syndrome (OHSS), ectopic pregnancy rates and early miscarriage rates were compared between the two groups of patients. Results The ovulation rate and the pregnancy rate were 97.8 % and 37.8%, respectively, in the treatment group and 75.6% and 17.8%, respectively, in the control group; there was a statistically significant difference between the two groups (P<0.05). Of 17 pregnant patients in the treatment group, 2 had early miscarriage, accounting for 11.8% and none had ectopic pregnancy, accounting for 0.0%. Of 8 pregnant patients in the control group, 4 had early miscarriage, accounting for 50.0% and none had ectopic pregnancy, accounting for 0.0%. The incidence of OHSS was 4.4% in the treatment group and 6.7% in the control group; there was no statistically significant difference between the two groups (P>0.05).Conclusions Combined use of acupuncture and medicine can increase the ovulation rate and the pregnancy rate in patients with polycystic ovarian syndrome infertility. It does not significantly cause ovarian hyperstimulation and ectopic pregnancy and may decrease the early miscarriage rate.
6.Study on the changes of gonadotropin releasing hormone agonist in pinopodes

Objective To study the changes of gonadotropin releasing hormone agonist (GnRH-a) in pinopodes during luteal phase and to explore the possible mechanism of GnRH-a in luteal phase support of assisted reproductive technology (ART).Methods Totally 40 primary infertility women who were treated with ART due to male factors were enrolled,according to the order of the group they were randomly divided into experimental group and control group.On the 7th day after ovulation,the experimental group received a subcutaneous injection of 0.1 mg of GnRH-a,while the control group received a subcutaneous injection of placebo only (0.9% salinc 2 ml),3 days later they came to the clinic again.Serum estradiol and progesterone levels were measured before and after treatment in each group.Pinopodes were collected for electron microscopic examination.Levels of ER and PR were detected by western blot.Results (1) There was no significant difference between the experimental group and the control group in the estrogen level before and after the treatment (all P>0.05).The level of progesterone in the experimental group after treatment [(66.8± 14.9) nmol/L] was significantly higher than that before treatment(P<0.05);also significantly higher than the same period of the control group (P<0.05).(2) There was no significant difference in the expression of ER protein in the experimental group before and after treatment (P>0.05).The expression of PR in the experimental group after treatment was significantly lower than that before treatment (P<0.05);also lower than the same period of the control group (P<0.05).(3) Expression amount of pinopodes in the experimental group after treatment was significantly higher than that before treatment [65% (13/20) versus 25% (5/20),P< 0.05],and the development trend was more mature [the percentage of maturation:75% (15/20) versus 35% (7/ 20),P<0.05].Expression amount of pinopodes after treatment and the percentage of maturation in the experimental group were significantly higher than those in the same period of control group (P<0.05).Conclusion GnRH-a in luteal phase support may play a role through the corpus luteum,which may promote the secretion of progesterone,downregulation of PR expression,promote the growth of pinopodes,and improve the endometrial receptivity.
7.Relationship between HDL subclass distribution and plasma glucose and lipid levels in patients with metabolic syndrome

AIM:To investigate high-density lipoprotein ( HDL) subclass distribution and to analyze the rela-tionship between HDL subclasses with plasma glucose and lipids in metabolic syndrome ( MS) .METHODS:Apolipopro-tein A-I ( apoA-I) contents of plasma HDL subclasses were determined by two-dimensional gel electrophoresis associated with immunodetection .The concentrations of lipids and apolipoproteins in the plasma were measured by an automated bio -chemical analyzer .RESULTS:Compared with the controls , the levels of fasting plasma glucose ( FPG) , total cholesterol (TC), triglyceride ( TG), low-density lipoprotein cholesterol ( LDL-C), LDL-C/high-density lipoprotein cholesterol (HDL-C), apolipoprotein B100(apoB100), apoB100/apoA-I, systolic blood pressure (SBP), body mass index (BMI) and HDL3b were increased in the MS patients (P<0.05).Meanwhile, HDL-C, apoA-I and preβ2-HDL, HDL2a and HDL2b were decreased in the MS patients (P<0.01).With the increase in the plasma glucose level , the contents of HDL2a and HDL2b were decreased in the MS patients (P<0.05), while preβ1-HDL was increased (P<0.05).With the decrease in the HDL-C level, the content of HDL2b was decreased in the MS patients (P<0.01), while preβ1-HDL was increased (P<0.01).With the increase in the TG level and the decrease in the HDL-C level, the content of HDL2b had a decrea-sing trend and the content of small-particle preβ1-HDL had an increasing trend , indicating that HDL maturation metabolism was disrupted.The correlation analysis showed that FPG was negatively correlated with the levels of HDL 2a and HDL2b, HDL-C was negatively correlated with the level of preβ1-HDL and positively correlated with the level of HDL 2b , and TG was positively correlated with the levels of preβ1-HDL and HDL3b .CONCLUSION:With the increases in the plasma glucose and TG, and the decrease in HDL-C in the MS patients, HDL particles have minifying tendency , and the maturation me-tabolism of HDL particles is disrupted .
8.The Infectious and Pathogenic Characteristics of Rabies Virus Strain CTNCEC25.

To investigate the phenotypic characteristics of the strain of the rabies virus CTNCEC25, the strain of the China rabies virus CTN-1 adapted to primary chicken embryo cells (CECs), Vero cells, and mouse neuroblastoma N2a cells was inoculated with CTNCEC25 and parental CTN-1 strains to explore the cytopathic effect (CPE) and growth kinetics of CTNCEC25 on cultured cells. To determine the pathogenicity of CTNCEC25, suckling mice, adult mice, guinea pigs and rabbits were inoculated with CTNCEC25 via the intracerebral route and their survival monitored every day. Furthermore, the CTNCEC25 strain was passed serially in CECs for 20 passages and then 3 passages in the brains of suckling mice to determine phenotypic stability. CTNCEC25 achieved similar growth kinetics in Vero cells and N2a cells compared with parental CTN-1, but CTNCEC25 replicated more efficiently in CECs than the CTN-1 strain with a titer 72 h after infection reaching 10(7.5-7.6) FFU/mL, which was significantly higher than the 10(5.8) FFU/mL achieved by its parental strain, CTN-1. Moreover, CTNCEC25 induced apparent CPE in Vero cells, CECs and N2a cells. Analyses of intracerebral inoculation demonstrated that CTNCEC25 was attenuated profoundly in adult mice and was completely apathogenic to guinea pigs and rabbits, though it caused death in suckling mice. The CTNCEC25 strain proliferated steadily after serial passage in CECs and the brains of suckling mice, and remained avirulent in adult mice. These results suggest that CTNCEC25 is a highly attenuated and genetically stable strain of the rabies virus. CTNCEC25 replicated stably and efficiently in cultured cells and achieved high titers, so it could be a promising and safe vaccine strain for rabies prevention in China.

9.Qualitative and quantitative study on the apoptosis of cervical cancer cells treated by tetrandrine

Objective To study the effects of tetrandrine on apoptosis of HeLa cervical cancer cells qualitatively and quantitatively. Methods We measured tetrandrine-induced inhibition of HeLa cell proliferation at different concentrations and time points by MTT assay. The rate of Hela cell apoptosis induced by tetrandrine was detected by flow cytometer and confocal laser scanning microscope (CLSM). Results Tetrandrine inhibited the proliferation of HeLa cells in dosage- and time-dependent manners. Flow cytometry showed that the apoptosis rate was (51.8±0.97)% at the concentration of 15μmol/L, which was significantly higher than that in the control group (24.3±1.23)% (P<0.05). The cells treated with tetrandrine showed typical apoptotic morphology under CLSM. Conclusion Tetrandrine can inhibit proliferation and induce apoptosis of HeLa cervical cancer cells.
10.Postnatal depression and anxiety state and its related factors of parturtents from migrant worker family

Objective To know the postnatal depression and anxiety state and its related factors of parturients from migrant worker family,and then reference to certain nursing counter-measures. Methods Sampled 73 parturients by spontaneous labor from migrant worker family and 45 local matched parturients,interviewed them by SAS and SDS to know their postnatal depression and anxiety state,and then analyzed the datum between them. Results The level of postnatal depression and anxiety in parturients from migrant worker family was significant higher than that in local parturienta.Different level of depression and anxiety was existed in different parturients from migrant worker family by different educational attainments,different family type and different financial state. Conclusions The state of postnatal depression and anxiety in parturients from migrant worker family is severely,related health education should be strengthened,associated social secu-rity and support system should be established to help them release their unhealthy emotion.
