1.Change of puerarin on the dynamic equilibrium of ET-NO and ET-ANF of diabetic vascular complicatioin induced by streptozotocin

AIM: To investigate the effect of puerarin on the alteration of ET and ANF in plasma, NO and NOS in serum of diabetic rats induced by streptozotocin(STZ). METHODS: Rat modeling was set up by means of intraperitoneal injection of ST2 with a dose of 60mg?kg -1. Four days later, blood glucose was measured by glucose oxygen kit. On the basis of present documents, the rats that blood glucose was over 16.7mM were accepted as diabetic ones, and then were divided randomly into five groups: diabetic mellitus (DM), aminoguanidine (AG 0.1g?kg -1,ig.), puel (0.5g?kg -1,ig.),pue2(0.25g?kg -1,ig.), pue3(0.125g?kg -1,ig.) and a normal group additionally. In order to set up a chronic model, we observed 12 weeks according to our past experiments. The FBS was measured at the twelfth week using the test kit. NO and NOS in serum were determined by checking their OD values, ET and ANF in plasma were determined by radio-immunization, respectively. 2.VITEK-32 and Microscan AS-4 in External Quality Assessment of Microbes: Their Application and Comparison

OBJECTIVE By using the VITEK32 and Microscan AS-4 microbiology identification system to identify the external quality control(QC) strains of microbes,to find out the difference and the coincidence degree with quality assessment result of the two systems. METHODS Thirty nine various strains of QC bacteria were isolated and cultured according to operation rule,and were identified by VITEK-32 and Microscan AS-4 and compared within them. RESULTS The coincidence rate of VITEK-32 and Microscan AS-4 to identify directly their species was 76.9% and 69.5% for 39 strains of QC bacteria,It was 89.7% and 76.9% after the serum or further tests,and the coincidence rate was 90.9% and 81.8% in 22 strains of Gram negatives,and 88.2% and 58.8% in 17 strains of Gram positives. CONCLUSIONS The detectable rate and coincidence rate of VITEK-32 are superior to those of Microscan AS-4,especially in the identification of Enterococcus and Streptococcus,the accuracy of Gram negatives is good,and of Gram positives is short,both them could not identify Salmonella and Shigella to their species.
3.Experimental effect of Puerarin on the formation of advanced glycation end products and expression of RAGE in the arota of diabetic rats

AIM To investigate the effect of puerarin on the formation of advanced glycation end products (AGEs) and RAGE(receptor for advanced glycation end products)mRNA expression in the arota of diabetic rats induced by streptozotocin (STZ). METHODS Rats were injected streptozotocin into intraperitonel with a dose of 60 mg?kg -1 . Four days later, blood glucose was measured using glucose oxygen kit. The rats with over 16 7 mmol?L -1 blood glucose were considered as diabetic ones, and then divided randomly into five groups: diabetic mellitus(DM), aminoguanidine(AG 0 1 g?kg -1 , ig), pue1(0 5 g?kg -1 , ig), pue2(0 25 g?kg -1 , ig) and pue3(0 125 g?kg -1 , ig). In order to set up a chronic model, we observed 12 weeks according to our past experiments. The fasting blood glucose(FBG) was and the contents of serum fructosamine (FMN) were measured using the test kit. The quantity of AGEs accumulation, the expression of RAGE in the arota was detected using fluorescence method and reverse transcription polymerase chain reaction(RT PCR). 4.Gestational trophoblastic diseases presented with persistent low levels of hCG

Objective To study the clinical characteristics, diagnosis, and treatment of patients with persistent low levels of β-hCG. Methods To analyze the clinical data of 6 patients with persistent low level of β-hCG retrospectively and review the relevant literature. Results The periods of low levels of β-hCG in the 6 patients were 10-53 months. The range of β-hCG was 3~637 U/L. The last pregnancy were hydatidiform mole in 5 cases and abortion in 1 case. After hydatidiform mole 5 cases were received multi-chemotherapy. Normal level of β-hCG maintained 1-12 months after chemotherapy, then β-hCG titer has gone up again, One patient developed lung metastases after 30 months and then cured by surgery and chemotherapy. There were no clinical evidence of malignancy in the other 4 cases, and 1 case received no therapy after abortion during course of therapy and following up. Conclusion Persistent low level of hCG was a special kind of disease with poor response to chemotherapy, but the majority were with stable status. Therapy should be restrained if there was no evidence of malignancy.
5.Effect of Shengeyifei Capsule on Inflammatory Mediators in Rat with Chronic Obstructive Pulmonary Disearse

Objective To observe the effect of Shengeyifei capsule on the inflammatory mediators in the airway and the pathology of the airway in the rat with chronic obstructive pulmonary diserse (COPD). Methods 60 SD rats were divided randomly into normal group, COPD model group, Jinshuibao control group and Shengeyifei groups. The COPD model was established by smoking and infusing lipopoly- saccharide (LPS). The level of interleukin-8 (IL-8) and tumor necrosis factor ? (TNF-?) in bronchoalveolar lavage fluid (BALF) were measured and the pathology of the airway and lung tissue was observed after treatment on the rats. Object To investigate the effect of puerarin (Pue) on renal protein kinase C (PKC) activity, kidney structure and function in diabetic rats. Methods STZ-induced diabetic rats were randomly divided into five groups: Diabetic rats model group (DM), Pue (500, 250, 125 mg/kg) treatment group, and VitE group, in addition, normal rats for control group. All rats were given by ig for 12 weeks. Kidney function and kidney index were determined; The PKC activity was measured by ELISA. The excretion of microalbuminuria (MAU) was measured by radio-immunoassay, and kidney tissue was observed by light-microscope and transmission electron microscope. Results The excretion of MAU, kidney index (kidney weight/body weight) and PKC activity in diabetic rats were significantly increased. The excretion of MAU, and PKC activity were markedly decreased in Pue treatment group, and kidney pathologic changes of diabetic rats in Pue treatment group were improved. Conclusion Pue can ameliorate early kidney hy-perdynamic abnormality in diabetic rats, possess protective effect on kidney of diabetic rats, whose mechanism may be associated partly with a down-regulation of PKC activity.
9.Distribution and drug resistance of pathogens in cancer patients with nosocomiai infection

Objective To investigate the distribution and drug resistance of pathogens in cancer patients with nosocomial infections. Methods Pathogens isolated from cancer patients with nosocomial infections in Zhejiang Cancer Hospital from 2004 to 2008 were analyzed. Results Totally 3454 strains of pathogens were isolated, including 1900 strains of Gram-negative bacilli (55.0%), 838 strains of fungi (24.3%), and 716 strains of Gram-positive bacteria (20.7%). Pathogenic bacteria Escherichia coli and Candida albicans were isolated mainly from surgical wound, urinary tract, respiratory tract and oral cavity. The drug resistant rates for Gram-negative Escherichia coli and Gram-positive staphylococci were high, and Candida albicans was sensitive to all antifungal drugs. Conclusion Opportunistic pathogens account for most nosocomial infections in cancer patients, and some of them are of high-level resistance to antimicrobial agents.
10.Analysis of the follow-up results concerning pregnancy, delivery and infants after assisted reproductive technique with GnRH-a for luteal support

Objective To investigate the results of follow-up visits of pregnancy course, delivery and infants of women who got clinically pregnant by assisted reproductive technique after gonadotropin-releasing hormone agonist (GnRH-a) added for luteal support, and to analyse the influence of adding GnRH-a in luteal support on the safety of mother and infant. Methods A retrospective analysis was carried out on the medical record from 215 patients who got clinically pregnant after luteal phase long regimen fresh-cycle transfer was operated. According to the differences in luteal support methods, the patients were assigned to Group A (124 patients, progesterone+dydrogesterone group), Group B (91 patients, GnRH-a added group). The patients′ pregnancy course, delivery time, and the growth and development of infants within 1-2 years were followed up. Results (1)There was no obvious difference between Group A and Group B in terms of the abortion ratio during the early pregnancy (8.1%, 12.1%), the rate of abortion villous deformity (50.0%, 9.1%), the rate of heterotopic pregnancy (10.5%, 5.5%) and rate of twin pregnancy (19.4%, 28.6%;all P>0.05).(2)Compared to group A, during the middle and late pregnancy of single or twin pregnancy in Group B , there was no obvious difference in the rate of fetal chromosomal abnormality, organ malformation incidence, late abortion rate and stillbirth rate (all P>0.05).(3)As to childbirth, in the case of twin pregnancy, there was a higher rate of premature delivery (60.0%, 39.1%;P=0.041), as well as rate of lower birth weight of newborn (56.0%, 34.8%; P=0.037) in group B.(4)The statistics on general growth and development as well as infantile common diseases within 2 years after birth indicated that there was no obvious difference between the two groups in single birth and twin birth subgroup (all P>0.05). Conclusion On the basis of controlling of implanted embryos and reducing the occurrence of twins, GnRH-a luteal support maybe relatively safe and effective.
