2.Idea of medical equipment integrated management based on the theory of the full dimensional security

Objective: To explore the full-dimension security theory in the application of the medical equipment management practices. Methods: By analyzing the current problems existing in the process of medical equipment management, put forward based on the theory of the full-dimension guarantee medical equipment integrated management idea. Results:To clarify the theory of medical equipment integrated management ideas and implementation path. Under the guidance of the medical equipment management committee, Medical equipment full-dimension security management can be divided into state of equipment procurement management, file management, equipment management, equipment, inventory management module, guarantee full medical equipment at various stages. Conclusion: Based on the theory of full-dimension security thought established medical equipment integrated management idea, provides a new idea to improve the traditional equipment management from the concept, method and technology.
3.Correlation between micropowder processing method and ginsenoside Rb_1 content of Radix Panacis Quinquefolii

AIM: To compare the ginsenoside Rb 1 content in different granularity of Radix Panacis Quinquefolii in order to confine Chinese medicine by ultramicro powder technique. METHODS: HPLC was used to determine ginsenoside Rb 1 in micropowder of Radix Panacis Quinquefolii.RESULTS:The contents of ginsenoside Rb 1 in micropowder of Radix Panacis Quinquefolii pulverized in normal and low temperature were increasing with particle size reduction, and the content in low temperature pulverization was distinctly higher than that in normal temperature pulverization. CONCLUSIONS: Low temperature pulverization of Radix panacis Quinquefolii is superior to processing at temperature.
4.Infection of mutated mouse complement receptor Type Ⅱ by Epstein-Barr virus

Site-directed mutagenesis method was used to introduce two desired mutations, which were confirmed by DNA sequencing, into mouse complement receptor Type II gene(MCR2). Then the constructed eukaryotic expression vectors containing wild type mouse CR2/1(wtMCR2/1), mutant type mouse CR2/1 (mtMCR2/1) and human CR2 (hCR2) cDNA were transferred into mouse SP2/0 cells by electroporation. After two-week screening by G418, the stably transfected clones were obtained. Several ways including PCR, RT-PCR, and immunohistochemistry were utilized to screen those clones with interesting genes integrated and expressed. Then Epstein-Barr virus(EBV) was used to infect these transfected cells and EBER-1 (EBV encoded RNAs) hybridization results showed that only hCR2 and mtMCR2 transfected SP2/0 cells could be infected by EBV, but positive rate of the former was much higher than the latter. This study sets groundwork for elucidating the mechanism by which EBV enters the cells and for establishing the animal model of EBV-related nasopharyngeal carcinoma (NPC).
5.Establishment of nasopharyngeal carcinoma (NPC) cell lines stable expressing NPC-derived LMP1

AIM: To establish nasopharyngeal carcinoma(NPC) cell lines stable expressing NPC-derived latent membrane protein 1(LMP1) gene. METHODS: General expression vector and epithelium-specific expression vector of NPC-derived LMP1 gene were constructed by using recombinant techniques, then transfected these vectors into a poor differentiated NPC cell line named CNE-2 ,integration and expression of N-LMP1 in CNE-2 cells were detected by PCR,RT-PCR and Western blot. RESULTS: (1) General expression vector and epithelium-specific expression vector of NPC-derived LMP1 gene were constructed successfully.(2) It showed that N-LMP1 gene expressed in CNE-2 cells correctly. CONCLUSION: The first NPC cell lines which stable express NPC-LMP1 were established. The cell lines obtained will provide important basis for exploring the role of NPC-LMP1 in nasopharynx carcinogenesis.
6.Recovery of IgG binding capability of human FcγRⅡa refolded by rapid dilution expressed in E. coli

Objective To study the effect of soluble, refolded, recombinant extracellular domain of the human Fc gamma receptor Ⅱ a (huFcγRⅡa) on the binding of human IgG to cells. Methods Extra-cellular domain of the huFcγRⅡ a gene was amplified from recombinant plasmid pe3huR Ⅱ by PCR and then cloned into pET-28a vector. The recombinant plasmid pETshuR Ⅱ was transformed into E. coli BL21 (DE3) after identified by PCR and doubly digested. The inclusion bodies of fusion protein were extracted and purified by washing, dissolved in 6 mol/L guanidine buffer, and refolded by rapid dilution technique. The refolding protein activity was tested by ELISA and flow cytometry. Results Extraceilular domain of the huFcγRⅡa gene was successfully cloned into pET-28a. The results of SDS-PAGE showed that the molecular mass (Mr) of the expressed protein was 24.8 × 103, and the expression rate was 30%. The purity of recom-binant shuR Ⅱ was up to 90% after washing. ELISA showed that the recombinant shuR Ⅱ was able to bind human IgG in a dose dependent manner, shuRⅡ could competitively inhibit the binding of human IgG to huFcγRⅡa expressed on the surface of COS-7 cells by flow cytometry. Conclusion The results demon-strate that it is possible to obtain large quantities of recombinant shuR Ⅱ which has comparable binding prep-erties to those of the whole membrane bound huFcγR Ⅱ a.
7.Efficacy Evaluation of Vascular Intervention in Adjuvant Treatment of Uterine Scar Pregnancy after Cesarean Section

Objective:To observe the clinical efficacy of vascular intervention in the treatment of uterine scar pregnancy after cesarean section .Methods:A total of 50 patients with uterine scar pregnancy after cesarean section were selected and randomly divided into observation group and control group ,with 25 patients in each group .The patients in the control group were treated with dilatation and curettage , while the patients in the observation group were treated with uterine artery embolization combined with curettage with methotrexate .After treatment ,the clinical efficacy ,the blood loss ,the time of serum β‐human chorionic gonadotropin (β‐HCG) returning to normal level ,the time of endo‐uterine mass disappearing ,the patient's hospital stay and adverse effects ,were compared between the patients in the two groups .Results:The cure rate of patients in the observation group (100% ) was significantly higher than that (64% ) in the control group(P<0 .05);the blood loss (47 .1 ± 14 .2)mL ,the time of endo‐uterine mass disappearing (23 .6 ± 23 .7) d and hospital stay (13 .2 ± 4 .3) d of patients in the observation group were significantly lower than those in the control group [(322 .1 ± 105 .2) mL ,(80 .4 ± 78 .3) d ,(20 .1 ± 7 . 8)d ,P<0 .05)];there was no occurrence of severe complications in the two groups .Conclusions:Vascular intervention in the treatment of uterine scar pregnancy after cesarean section can effectively kill the embryo tissues ,reduce the blood loss ,and retaln the womb as well as the fertility .
8.Application of Mini-CEX in standardized training of the first year for new nurses

Objective:To improve the standardized training effects of the first year graduate new nurses by the mini clinical evaluation exercise (Mini-CEX) .Methods:By cluster sampling, a total of 91 new nurses hired in 2017 of Xinqiao Hospital Affiliated to Army Medical University were in control group; and 120 new nurses hired in 2018 were in experimental group. Control group carried out routine training method. On the basis of that, experimental group implemented the Mini-CEX based clinical nursing core competence training and operational skill training once a month after changing routine teaching rounds and operational training, lasting 12 times. The scores of Mini-CEX for the first and twelfth time of experimental group were compared by paired sample t-test, the theoretical achievements and skill performances of new nurses between two groups were compared by independent sample t-test. Results:The scores of Mini-CEX of clinical nursing core competence training and operational skill training in experimental group for the twelfth time were (6.27±0.46) and (6.73±1.10) respectively, higher than those for the first time with statistical differences ( t=-8.48, -7.64; P<0.01) . Among new nurses in standardized training of the first year, the theoretical achievements and skill performances of department in experimental group were (87.70±11.32) and (92.90±8.22) , those of hospital were (85.50±11.83) and (89.67±8.99) respectively, higher than those in control group with statistical differences ( t=-2.71, -2.09, -2.24, -3.22; P<0.05) . Conclusions:Mini-CEX has significant effects in training for the first year among new nurses .

We report a complete genomic sequence of rare isolates (minor genotype) of the SARS-CoV from SARS patients in Guangdong, China, where the first few cases emerged. The most striking discovery from the isolate is an extra 29-nucleotide sequence located at the nucleotide positions between 27,863 and 27,864 (referred to the complete sequence of BJ01) within an overlapped region composed of BGI-PUP5 (BGI-postulated uncharacterized protein 5) and BGI-PUP6 upstream of the N (nucleocapsid) protein. The discovery of this minor genotype, GD-Ins29, suggests a significant genetic event and differentiates it from the previously reported genotype, the dominant form among all sequenced SARS-CoV isolates. A 17-nt segment of this extra sequence is identical to a segment of the same size in two human mRNA sequences that may interfere with viral replication and transcription in the cytosol of the infected cells. It provides a new avenue for the exploration of the virus-host interaction in viral evolution, host pathogenesis, and vaccine development.
