1.Experimental effect of Puerarin on the formation of advanced glycation end products and expression of RAGE in the arota of diabetic rats

AIM To investigate the effect of puerarin on the formation of advanced glycation end products (AGEs) and RAGE(receptor for advanced glycation end products)mRNA expression in the arota of diabetic rats induced by streptozotocin (STZ). METHODS Rats were injected streptozotocin into intraperitonel with a dose of 60 mg?kg -1 . Four days later, blood glucose was measured using glucose oxygen kit. The rats with over 16 7 mmol?L -1 blood glucose were considered as diabetic ones, and then divided randomly into five groups: diabetic mellitus(DM), aminoguanidine(AG 0 1 g?kg -1 , ig), pue1(0 5 g?kg -1 , ig), pue2(0 25 g?kg -1 , ig) and pue3(0 125 g?kg -1 , ig). In order to set up a chronic model, we observed 12 weeks according to our past experiments. The fasting blood glucose(FBG) was and the contents of serum fructosamine (FMN) were measured using the test kit. The quantity of AGEs accumulation, the expression of RAGE in the arota was detected using fluorescence method and reverse transcription polymerase chain reaction(RT PCR). AIM: To investigate the effect of puerarin on the alteration of ET and ANF in plasma, NO and NOS in serum of diabetic rats induced by streptozotocin(STZ). METHODS: Rat modeling was set up by means of intraperitoneal injection of ST2 with a dose of 60mg?kg -1. Four days later, blood glucose was measured by glucose oxygen kit. On the basis of present documents, the rats that blood glucose was over 16.7mM were accepted as diabetic ones, and then were divided randomly into five groups: diabetic mellitus (DM), aminoguanidine (AG 0.1g?kg -1,ig.), puel (0.5g?kg -1,ig.),pue2(0.25g?kg -1,ig.), pue3(0.125g?kg -1,ig.) and a normal group additionally. In order to set up a chronic model, we observed 12 weeks according to our past experiments. The FBS was measured at the twelfth week using the test kit. 4.Effects of puerarin on renal protein kinase C activity in diabetic rats

Object To investigate the effect of puerarin (Pue) on renal protein kinase C (PKC) activity, kidney structure and function in diabetic rats. Methods STZ-induced diabetic rats were randomly divided into five groups: Diabetic rats model group (DM), Pue (500, 250, 125 mg/kg) treatment group, and VitE group, in addition, normal rats for control group. All rats were given by ig for 12 weeks. Kidney function and kidney index were determined; The PKC activity was measured by ELISA. The excretion of microalbuminuria (MAU) was measured by radio-immunoassay, and kidney tissue was observed by light-microscope and transmission electron microscope. Results The excretion of MAU, kidney index (kidney weight/body weight) and PKC activity in diabetic rats were significantly increased. The excretion of MAU, and PKC activity were markedly decreased in Pue treatment group, and kidney pathologic changes of diabetic rats in Pue treatment group were improved. Conclusion Pue can ameliorate early kidney hy-perdynamic abnormality in diabetic rats, possess protective effect on kidney of diabetic rats, whose mechanism may be associated partly with a down-regulation of PKC activity.
6.Influence of insulin sensitivity on insulin resistance rats treated by puerarin

Aim To investigate the effect of puerarin(Pue)on the experimental insulin resistance(IR)and the mechanism of Pue-treated metabolic syndrome(MS).Methods IR rat model was induced by i.v.small dosage streptozotocin(STZ)and high fatty diet.The effect of Pue on insulin sensitivity was studied by reformed hyperinsulinism euglycemia clamp technique(HEPC)on IR rats.Results The basic blood glucose(BBG),basic blood plasma insulin(BINS)and stable basic blood plasma insulin(SINS)of IR rats induced by high-fat feed were higher than those of control group,and those of the group treated by Pue were observably lower than those of IR model group.In HECT text,the average rate of glucose injection of IR model group was lower than that of control group from 60 min to 120 min,but the rate of high dose and middle dose of Pue group was significantly higher than that of IR model group.Conclusion The results suggest that Pue can improve the insulin sensitivity,and reduce basic blood glucose and blood plasma insulin of the experimental insulin resistance rats.

Aim To investigate the protecting role and mechanisms of crude flavones of pueraria loblata(FP) on human umbilical vein endothelial cells(HUVEC) in high glucose medium.Methods ①HUVECs were incubated in 20% FCS RPMI-1640 culture medium containing 0,5,10,30,60,90,120 mmol?L~(-1) glucose for 48 hours.The proliferation of HUVEC was analyzed with MTT method(490 nm).② HUVECs were incubated in 20% FCS RPMI-1640 culture medium containing 0 or 30 mmol?L~(-1) glucose,either alone or in the presence of FP(final concentration 10~(-3),10~(-4),10~(-5),10~(-6) mol?L~(-1) respectively) for 48 hours.At the same time,30 mmol?L~(-1) mannitol was added.The proliferation of HUVEC was analyzed using MTT method.The cell cycle of HUVEC was analyzed using flowcytometry.The contents of SOD,NO and ICAM-1 in HUVEC supernatant were measured using test kits.Results HUVEC in 5 mmol?L~(-1) glucose medium proliferated well,but the proliferation of HUVEC in 10,30,60,90 and 120 mmol?L~(-1) glucose media was disturbed,showing a negative correlation.Proliferation of HUVEC was inhibited in 30 mmol?L~(-1) glucose,compared with that in 0 mmol?L~(-1) glucose and 30 mmol?L~(-1) mannitol(P
8.Application of preoperative assessment of record sheet in neurosurgical procedures

Objective To study the application of preoperative assessment of record sheet on surgical patients in neurosurgery department .Methods Totals of 80 surgical patients were randomly divided into observation group and control group .Patients in observation group were given preoperative assessment record sheet, and patients in control group were given routine preoperative visit .The curative effect and postoperative nursing effects were compared .Results The intraoperative blood loss , operative time , length of hospital stay and patients increasing blood pressure in observation group were (268.88 ±38.38) ml, (142.1 ±10.10) min, (12.88 ±0.56) days,2, respectively, which were significantly lower than (407.50 ±54.05)ml, (178.40 ± 11.83)min, (25.63 ±1.02) days, 8 in control group.The differences were statistically significant (t=2.176, 2.288,10.960, respectively; χ2 =4.114;P <0.05).HAMA and HAMD scores of patients in observation group after operation were (10.42 ±2.64),(9.44 ±2.65),respectively, which were lower than (13.53 ±4.63), (12.32 ±2.44) before operation, and lower than (13.12 ±3.34),(11.82 ±2.54) in control group after operation too, the differences were statistically significant (t=3.043,3.856,3.544,2.643, respectively;P<0.05).Patient satisfaction scores was (7.64 ±1.65) and the number of satisfied patients was 23 in observation group, which were significantly higher than (6.55 ±1.63) and 14 in the control group.The differences were statistically significant difference (t=3.643,Z=4.073;P<0.05).Conclusions Neurosurgery patients with preoperative assessment can better improve the patient ’ s emotional impact , the curative effect of surgical treatment and reduce the risk of intraoperative complications .
