1.Correlation of ER with Bcl-2 in cervical cancer and pre-cancerous lesion

Objective:This study aimed to investigate the relationship between estrogen receptor (ER) and Bcl-2 in cervical can-cer and precancerous lesions. Methods:ER and Bcl-2 were evaluated by immunohistochemistry in 20 non-tumor tissues, 30 low-grade cervical intraepithelial neoplasias, 70 high-grade cervical intraepithelial neoplasias, and 80 cervical cancers. Results:The positive rate of ER was 75%in non-tumor tissues;this rate decreased with disease severity (χ2=24.266, P<0.01). The overexpression of ER protein was remarkably lower in cervical intraepithelial neoplasias (CIN) Ⅱ-Ⅲ and carcinomas than in non-tumorous cervices. Differences were not significant between CINⅠand non-tumorous cervices (χ2=0.751, P=0.386). Furthermore, ER expression was not correlated with histological type and tumor grade. The positive rate of Bcl-2 was 75.38%in squamous cell carcinoma. This rate increased with dis-ease severity (χ2=27.715, P<0.01). The Bcl-2 protein overexpression was remarkably higher in CINⅠ, CINⅡ-Ⅲ, and carcinomas than in non-tumorous cervices. Bcl-2 expression was also not correlated with histological type (P=0.500);by comparison, Bcl-2 expression was correlated with tumor pathological grade (χ2=4.862, P=0.027). The co-expression of ER and Bcl-2 was 38.57%and 15.38%in CINⅡ-Ⅲand squamous cell carcinoma, respectively;the differences between these two values were significant (χ2=9.108, P=0.003). A cor-relation between ER and Bcl-2 was observed in CINⅡ-Ⅲ(r=0.506, P<0.01);no correlation was observed between other groups. Con-clusion: ER expression was inversely correlated with Bcl-2 in cervical cancer progression. The co-expression of these markers may have an important function in the course of high-grade cervical intraepithelial neoplasia.
2.Development of RT-PCR-ELISA assay for detecting Cryptosporidium hominis

To establish a highly sensitive and specific method to detect the presence of Cryptosporidium homini, the RT-PCR-ELISA assay was tried, in which the primer with a biotin-labeled probe was designed to amplify fragment containing the highly variable region by multiple alignment between p23 gene of C.hominis and other Cryptosporidium spp. The RT-PCR was used to amplify the target fragment, and the amplified product was used to hybridize with the probe primer. The hybridized product was then captured on micro-plate wells coated with streptavidin and reacted with anti-digoxin antibody labeled with horse-radish peroxidase. This method of testing was then used for the detection of C.hominis in 22 clinical specimens and compared with the conventional methods of testing. It was demonstrated that the RT-PCR--ELISA for the detection of C.hominis was proved to be quite sensitive and specific. Its sensitivity was 100 times higher than that of the general PCR. From the result of clinic detection, the detection rate of RT-PCR-ELISA assay attained to 86%(19/22), while those of RT-PCR, sucrose floating method and anti-acid staining were 27%, 27% and 50% respectively. This result indicates that the RT-PCR-ELISA assay is more sensitive to detect C.hominis than the other three methods of testing.
3.Comparative study on the application of self-made hemostatic device in minimally invasive surgery

Objective To explore the operation of grasping forceps and nursing staff made telescopic injection device were used for the clinical effect of Hemostasises thoracoscopic lobectomy. Methods 60 cases of thoracoscopic lobectomy for patients according to the date of surgery were divided into operation group and telescopic grasping forceps injection device group 30 patients were given hemostasia operation grasping forceps extraction hemostatic powder and hemostatic powder into the injection device for telescopic wound hemostasis treatment and preventive treatment, were compared between the two groups in the instrument body, completely hemostatic powder cover the wound time, evacuation time of endoscopic instruments, single hemostatic powder wound coverage, wound bleeding rate, the incidence of bleeding again installed. Results The telescopic injection device group and operation time grasping forceps group into cavity instruments, endoscopic evacuation equipment time showed no significant difference (P>0.05). Hemostatic powder completely cover the wounds of time (24.90 ± 5.90) s and (55.60 ± 25.60) s, there was significant difference between two groups (t=6.401, P<0.01). The single ended wound coverage of blood meal was 86.67%(26/30) and 76.67%(23/30), there was significant difference between two groups (χ2=2.016, P<0.05). Wound bleeding rate was 10.00%(3/30) and 33.33%(10/30), there was statistically significant difference between the two groups (χ2=4.812, P<0.05). The incidence of bleeding again to install powder was 10.0%(3/30) and 93.3%(28/30), there was significant difference between two groups (χ2=41.713, P <0.01). Conclusions The nursing staff made telescopic injection device assisted surgery were thoracoscopic lobectomy of the wound to stop bleeding and prevent rebleeding, wound coverage than grasping forceps, completely cover the wound operation less time than grasper, re install a low incidence of hemostatic powder, the whole process to shorten the operation time.
