1.Effect of gene abaI expression on acinetobacter baumannii biofilm formation

Objective To investigate the influence of abaI expression on acinetobacter baumannii biofilm formation.Methods Acinetobacter baumannii strain S isolated from bums patients was collected for the study,while the standard strain ATCC19606 was served as the control.At 6,24 and 48 hours,the gene expressions of abaI,pgaA,pgaB and pgaC were detected by real-time fluorescent quantitative PT-PCR,secretion of N-acyl-homoserine lactones (AHLs) by biological sensor and biofilm formation by MTT method.Results (1) Gene expressions of abaI,pgaA,pgaB and pgaC at 6 hours were 8.63 ±5.93,1.98 ± 1.93,1.01 ± 1.32 and 2.67 ± 3.46 respectively,which showed a quick increase at 24 hours (22.81 ± 17.60,5.13 ± 4.32,5.66 ± 3.97,11.97 ± 7.75 respectively),followed by a rapid decline in 48 hours (3.43 ± 0.88,1.30 ± 0.24,3.01 ± 3.00,3.02 ± 3.29 respectively).Gene expressions of pgaB and pgaC at 6 hours and that of pgaA and pgaC at 48 hours revealed statistically significant differences from those at 24 hours (P < 0.05).(2) AHLs showed a level of 18.49 ± 11.03 at 6 hours,reached a peak of 52.23 ± 15.95 at 24 hours,then descended to 5.53 ± 0.94 at 48 hours.AHLs level at 24 hours showed statistically significant difference from that at 6 hours and 48 hours (P < 0.05).(3)Biofilm formation at 24 hours and 48 hours was 2.83 ±0.44 and 2.71 ±0.15 respectively,far higher than that at 6 hours (0.49 ± 0.11,P < 0.05).(4) In the correlation analysis among AHLs,biofilm formation and gene abaI,pgaA,pgaB and pgaC expressions,significant positive correlation was found between abaI and pgaA and between AHLs and pgaC expression (P < 0.05).Conclusion Acinetobacter baumannii may regulate gene expressions of pgaA and pgaC responsible for biofilm formation to adjust to the external environment by means of changing abaI gene expression and AHLs secretion.
2.Application of doctor-nurse integration combined with WeChat platform in thrombolytic therapy for acute cerebral infarction

ObjectiveTo explore the application effect of doctor-nurse integration combined with WeChat platform in thrombolytic therapy for acute cerebral infarction.Methods Totally 72 patients with thrombolytic treatment for acute cerebral infarction in our hospital from October 2013 to December 2015 were selected and divided into the observation group(36 cases)and the control group(36 cases)according to the patient′s admission sequence. The primary nursing team model was applied in the control group,and the doctor-nurse integration treatment model combined with WeChat platform was applied in the observation group. The thrombolytic preparation time,the national institutes of health stroke scale(NIHSS)score after treatment,Barthel index,the average hospitalization time and other indicators between two groups were compared.Results The thrombolytic preparation time(78.86±15.34) min and the average hospitalization time(10.08 ±3.11) d in the observation group were significantly shorter than the data in the control group [(93.47±15.58) min,(13.92±4.73) d;(P<0.05)];the NIHSS score after treatment(2.25±2.59)in the observation group was significantly lower than that(3.69±3.26)in the control group(P<0.05);the Barthel index(82.78±24.80)in the observation group was higher than that(70.00±28.44)in the control group(P<0.05);Compared with the patient satisfaction(88.9%) and medical staff satisfaction(98.6%)in the observation group,the data in the control group(69.4%,72.9%) were lower(P<0.05).Conclusions The doctor-nurse integration treatment model combined with WeChat platform in patients with acute cerebral infarction thrombolysis is conductive to implement the standardization of specialist treatment in patients with acute cerebral infarction.
3.Application of empathy technique in patients with first benign paroxysmal positional vertigo

Objective To explore the effects of empathy technology on emotion regulation, sleep improvement and quality of life improvement in patients with first benign paroxysmal positional vertigo (BPPV). Methods A total of 100 patients with the first BPPV in The Second Affiliated Hospital of Naval Medical University from December 2023 to November 2024 were selected, and were divided into control group and observation group by random number table method. The patients in both groups received routine reduction treatment and outpatient follow-up every 3 months after discharge, with a total of 4 follow-up visits. The patients in control group received routine health and psychological education; on this basis, the patients in observation group received empathy technique intervention before, during, after reposition and during follow-up. The emotion, sleep and quality of life scales were evaluated before intervention and at the 4th outpatient follow-up. The recurrence rates of BPPV within 1 year after discharge were compared between the two groups. Results One year after discharge, self-rating anxiety scale (SAS), self-rating depression scale (SDS), symptom checklist 90 (SCL-90) and Pittsburgh sleep quality index (PSQI) scores in the observation group were lower than those in the control group, and multi-dimensional quality of life scores in the observation group was higher than that in the control group. These differences were statistically significant (P<0.05). The recurrence rate of BPPV in the observation group was lower than that in the control group 1 year after discharge (P<0.05). Conclusions Empathy technology intervention can further improve emotion, sleep quality, and quality of life, but reduce disease recurrence rate in first BPPV patients receiving routine treatment and health education, so it can be widely used in clinical practice.
4.Morphological study of afferent connections of VGLUT2 positive neurons of mediodorsal thalamic nucleus in mice

Objective:Using retrogradely trans-monosynaptic tracing method mediated by rabies virus(RV)to ob-serve presynaptic neuronal distributions of vesicular glutamate transporter 2(VGLUT2)-positive neurons of the med-iodorsal thalamic nucleus(MD)in whole brain.Methods:The helper viruses of RV were injected into the right MD of VGLUT2-ires-Cre mice,and two weeks later RV was injected into the same area.After another one week,perfusion was taken and whole brain scanning was performed to observe the distributions of RV-labeled presynaptic neurons throughout the brain.Results:After RV virus was injected into the MD of VGLUT2-ires-Cre mice,dense presynaptic neurons were observed in the cortex and brainstem.RV labeled neurons were mostly distributed in primary motor cortex(M1),sec-ondary motor cortex(M2),medial prefrontal cortex,orbitofrontal cortex and insular cortex at the cortical level.Tha-lamic retrogradely labeled neurons were mostly found in reticular thalamic nucleus and lateral hypothalamic area,and retrogradely labeled neurons in the brainstem were mainly distributed in lateral parabrachial nucleus,periaqueductal grey matter and dorsal raphe nucleus.Conclusion:The results in the present experiment suggest that VGLUT2-positive neurons within the MD can receive ascending information transmission from the brainstem,or regulation from the reticu-lar nucleus of thalamus.As a higher-order thalamus,MD can also receive descending projections from the cortex,and is involved in a variety of functions.Our results provide morphological basis for the study of the function and the related neural circuit of the MD.
