1.Analysis of thyroid function screening results in 776 cases of early pregnant women

Objective To explore the incidence and characteristics of thyroid disease in early pregnancy and to provide a scientific basis for eugenics .Methods The chemiluminescence immunoassay was used to detect the concentrations of serum thyrotropin (TSH) ,free thyroxine (FT4) ,triiodothyronine (FT3) and thyroid peroxidase antibody (TPOAb) in 776 early pregnant women (<12+6 weeks ,experiment group) and 100 non‐pregnant child‐bearing women (control group) .The test results were analyzed accord‐ing to the different diagnostic criteria of early pregnancy and non‐pregnant childbearing women .Results The serum concentrations of TSH ,FT4 and TPO‐Ab had statistical differences between the experimental group and the control group (P<0 .05) .The inci‐dence rate of thyroid diseases in early pregnant women was 35 .05% ,which was significantly higher than 15 .00% in non‐pregnant childbearing women ,and the difference was statistically significant (P<0 .05) .The thyroid diseases in pregnant women of the ex‐perimental group were mainly hypothyroidism (9 .28% ) and sub‐clinical hypothyroidism (22 .94% ) ,compared with the control group ,the difference was statistically significant(P<0 .05) .Conclusion Early pregnancy has high incidence rate of thyroid disease , which can lead to birth defects .Therefore ,screening early gestation thyroid diseases should be paid attention to provide theoretical basis for eugenics .
2.Effects of miR-506 on malignance phenotypes of hepatocellular carcinoma cells

Objective To investigate effects of microRNA-506 (miR-506) on malignant phenotypes of hepatocellular carcinoma (HCC) cells, including cellular viability, proliferation and invasion. Methods HCC cell lines HepG2 and QGY-7703 were served as model. Five experimental groups were established in this study, including cell control, pcDNA 3 blank vector control, miR-506 over-expression, pSIH1 blank vector control and miR-506 suppression groups. Real-time reverse transcription PCR assay was performed to measure miR-506 level. CCK-8, colony formation and Transwell assays were performed to detect viability, colony formation activity and invasion activity of HCC cell lines, respectively. Effects of miR-506 on these indexes were evaluated. Results In HepG2 and QGY-7703 cell lines, miR-506 level increased in the miR-506 over-expression group (P<0.01), and its level decreased in the miR-506 suppression group (P<0.05) compared with the related blank vector control groups. In the miR-506 over-expression group, cellular viability was significantly reduced (P<0.01), cell colony number decreased, and number of cell penetrating Transwell microporous membrane was also decreased (P<0.01). In the miR-506 suppression group, cellular viability significantly increased (P<0.01), and both colony number and penetrating cell number increased (P<0.05). Also, there were no effects on the above indexes in pcDNA3 and pSIH1 blank vector control groups compared with those of cell control group (P>0.05). Conclusion miR-506 plays a tumor suppressor role in HCC cells by inhibiting cell viability, colony formation and invasion.
3.MicroRNA-506 inhibits malignancy of colorectal carcinoma cells by targeting LAMC1

Objective To investigate the effects of microRNA-506 (miR-506) on malignant phenotype of colorectal carcinoma cells. To identify the target gene of miR-506 in colon carcinoma. Methods SW480 cells were divided into five groups, known as normal cell group, miR-506 overexpression and, miR-506 inhibition groups with their vehicle groups.The migration and invasion abilities of SW480 cells were measured with Transwell migration assay. Cell viability and colony forming activities were measured by CCK8 and colony formation assays, respectively. Furthermore, bioinformatic method, green fluorescent protein (GFP) reporter assays, quantitative reverse transcription polymerase chain reaction (qRT-PCR) and Western blot were applied to predict potential target genes of miR-506. Results The number of migrated and invasive cells, viability and clonality in the miR-506 overexpression groups reduced. LAMC1 mRNA and protein levels in the miR-506 overexpression groups were lower than those in the control groups. Conclusion LAMC1 is a direct target gene for miR-506 and miR-506 could inhibit the cell migratioin and invasion.
4.Observation on curative effect of ozone combined with nifuratel nystatin vaginal soft capsules on the treatment of vaginitis

Objective To explore the clinical effect of ozone combined with nifuratel nystatin vaginal soft capsules on the treatment of vaginitis, and to provide the basis for clinical application. Methods 204 patients with vaginitis in our hospital from January 2013 to December 2016 were divided into the observation group (114 cases) and the control group (90 cases) according to the different treatment methods. The control group was treated with nifuratel nystatin vaginal soft capsules, and the observation group was treated with ozone combined with nifuratel nystatin vaginal soft capsules. The clinical efficacy, symptom relief time and adverse reaction were compared between the two groups. Results After treatment, the total effective rate in the observation was 97.37%, which was significantly higher than 85.56% in the control group(P<0.05). The rapid remission rate of the clinical symptoms in the observation group was significantly higher than that in the control group (P<0.05). There was no significant difference in the incidence of adverse reactions between the two groups.Conclusion The effect is significant that ozone combined with nifuratel nystatin vaginal soft capsules is used on the treatment of vaginitis, which can quickly relieve symptoms, and will not significantly increase the adverse reactions, is worthy of application.
5.Comparison of the effects of different doses of methylprednisolone therapy for children with severe hand,foot and mouth disease

Objective To compare the clinical effects of different doses of methylprednisolone therapy for children with severe hand,foot and mouth disease (HFMD).Methods According to different dosage methods, 240 children with severe HFMD were divided into large dose group,medium dose group and small dose group,80 cases in each group.The three groups were given different doses of methylprednisolone infusion on the basis of conventional treatment:large dose group(5 ~10mg·kg· -1 d -1 ),medium dose group(3 ~5 mg·kg· -1 d -1 ),small dose group (1 ~2mg · kg · -1 d -1 ).Results The time of fever sustaining,panic ease,mechanical ventilation,duration of hypertension and heart rate recovery of the medium -dose group were (47.93 ±4.72)h,(45.54 ±2.42)h,(51.43 ± 6.85)h,(53.66 ±7.62)h,(52.45 ±7.84)h,which were significantly shorter than those of the small -dose and large -dose group(all P <0.05).The incidence of turning to critically ill and the rate of ventilator use of the medium-dose group were significantly lower than those of the small -dose and large -dose group (all P <0.05 ).The incidence of pulmonary edema and pulmonary hemorrhage of the medium -dose group were significantly lower than those of the small -dose and large -dose group(all P <0.05).The differences among three groups were not statistically significant in the complications such as hypokalemia,hypocalcemia and gastrointestinal bleeding (all P >0.05 ). Conclusion Medium dose of methylprednisolone in the treatment of children with severe HFMD has significant effect and less adverse reactions,which is worthy of promotion.
6.Absorptive Characteristics of Ginsenoside Rb_3 in Caco-2 Cell Monolayer Model

OBJECTIVE:To study the absorptive characteristics of ginsenoside Rb3 in Caco-2 cell monolayer model. METHO-DS:The ginsenoside Rb3 cell samples underwent high speed centrifugation, then the supernatant was collected and determined by LC-ESI-MS/MS method in which the mobile phase consisted of acetonitrile-1 mmol?L-1 ammonium formate water solution (34 ∶ 66) with ginsenoside Rg2 as internal standard, and the tandem mass spectrometry was operated in negative electrospray ionization in a multiple reaction monitoring (MRM) mode, with detection ions m/z1077.7→m/z 783.4 for ginsenoside Rb3 and m/z 783.6→m/z 475.1 for ginsenoside Rg2.The concentration of ginsenoside Rb3 across the Caco-2 cell monolayer model was determined and the apparent permeability coefficient(Papp) of ginsenoside Rb3 was calculated. RESULTS: The calibration curve for ginsenoside Rb3 was linear in the range of 50~2 000 ng?mL-1,with intra-day precision and inter-day precision at less than 15%. P(AP-BL) from apical side (AP) to basolateral side (BL) was 3.22?10-6 cm?s-1, whereas P(BL-AP) from BL to AP was 6.0?10-6 cm?s-1,and the ratio of P(BL-AP)/P(AP-BL) was 1.86.CONCLUSION:The LC-ESI/MS/MS method is simple and sensitive, and it is applicable for the study of the absorptive characteristics of ginsenoside Rb3 in Caco-2 cell monolayer model.

Objective To explore the effect of ω-3 fish oil on nutrition improvement and inflammatory reaction of patients with gastrointestinal tumor after operation.Methods 60 cases with gastrointestinal tumor were divided into control group(30 cases) and study group (30 cases),both groups were provided with parenteral nutrition treatment[ 104.6 kJ · kg-1 · d-1 ].Fish-oil fatty emulsion was given to the study group.The postoperative for the first day received half of the total energy and the total energy The remaining four days.Blood samples were gained on the morning of day 1,on the morning of day 3 and day 6 after operation respectively to measure albumin ( ALB),prealbumin (PA),total protein(TP),transferrin(TRF),the neutrophilic granulocyte count,lymphocyte count (TLC),serum C-reactive protein(CRP).Results Both groups of patients was comparable(all P < 0.05 ).Both groups of patients was treated after five days of postoperative and ALB,TP,TRF were not significantly different.PA in both groups on day 6 was significantly increased,and the study group was higher than control group.there were statistical differences between them ( all P < 0.05 ).The neutrophilic granulocyte count and CRP of both groups were significant reduced,and the study group was lower than control group.There were statistical differences between them( all P <0.05 ).Conclusion ω-3 fish oil on nutrition could improve nutritional quality and modulate inflammatory reaction of patients with gastrointestinal tumor after operation.
8.The emergency epidemiologic characteristics of casualties cases with head injury in Shanghai

Objective To analyze the emergercy epidemiological characteristics of coeualties with head in-jury in Shanghai. Method By a prospective study,the data of 18 076 casualties with head injury during the whole year 2004 collected from 12 joint hospitals in Shanghai were documented well in the unified survey tables with wide-range items failed in by the attending physician, who got the firsthand information from patients and witness.The data were analyzed by using SPPS version 11.5 software. Results Of 18 076 casualties with head injury,the ratio of male to female was 2.11: 1.The mean age of female was older than that of male (t=10.575, P<0.01).The highest incidence of casualties occurred in people of twenties (24.7 % ). The local residents of Shanghai ac-counted for 34.2% of casualties. More casualties often occurred in December,January,Mareh and August than in he rest. Of 5.1% casualties with head injury were assochted with labour work.The leading cause of injury was dif-ferent in patient cohorts of different ages. Fall was the most main cause of trauma in children cohort (aged<14)and the senile patients cohort (aged > 60). The violert assault and traffic accident caused the most head injuries in the adolescent people cohort ( aged 15~34 years old) and the young people cohort ( aged 35~59 year old). The majority of casualties (85.5%) received CT scan.The scalp laceration (40.2% of patients) was seen more often than other types of injury . The mortality of easualties with traumatic brain injury was 0. 5 % . Conclusions The kmowledge of epidemiologieal aend of emergency deparhnent visitors with head injury is amportant guidance to physicians arranging emergency medical resources rationally and formulating a comprehesive prevention stategy of castahies with head injury.
9.Synthesis of N-(2-18F-fluoropropionyl)-L-glutamine and its biodistribution study in mice

Objective To synthesize 18F labeled N-(2-18 F-fluoropropionyl)-L-glutamine (18 F-FPGLN) for tumor PET imaging,and to perform its biodistribution study on normal mice and PC-3 tumorbearing nude mice.Methods 4-nitrophenyl-2-18F-fluoropropionate (18F-NFP) was synthesized on the MF2V-IT-I synthesizer and was purified by semi-preparative HPLC.Anhydrous 18F-NFP was added to a solution of L-glutanine t-butyl ester to synthesize 18F-FPGLN t-butyl ester,which was hydrolyzed by HCl (3 mol/L) and neutralized with NaOH (2 mol/L) solution.18F-FPGLN product was collected for further study.Biodistribution study was performed on normal Kunming mice and PC-3 prostate cancer tumor-bearing nude mice,respectively.Results 18F-FPGLN was synthesized with 10%-15% (decay uncorrected) overall radiochemical yield after 130 min of radiosynthesis.The radiochemical purity was higher than 96%.Rapid and high uptake of radiotracer was observed within the kidneys,and was quickly excreted through the urinary bladder.The uptake in kidney reached (35.0±1.2) %ID/g at 5 min post-injection,and descended to (1.5±0.3) %ID/g at 120 min.The liver,lung,heart and small intestine showed relatively moderate uptake of radioactivity.The uptake in the pancreas,muscle,spleen,stomach and brain was low,and the lowest uptake of (1.5±0.3) %ID/g was found in the brain at 30 min post injection.High accumulation of 18F-FPGLN in PC-3 xenograft was observed,and the tumor/muscle ratio reached 2.07 at 60 min post injection.Conclusion A novel N-position 18F-labeled glutamine analogs 18F-FPGLN,with favorable pharmacokinetic characteristics,is synthesized successfully,which makes it possible to perform tumor PET imaging using 18F-FPGLN subsequently.
10.Diagnostic value of CEA, CA125, CYFRA21-1, and NSE in lung cancer

Objective To explore the diagnostic value of four tumor markers analyzed with Logistic regression and receiver operator characteristic (ROC) curve in patients with lung cancer.Methods The serum levels of carcinoembryonic antigen (CEA) 、carbohydrate antigen 125 (CA125),cytokeratin 19 fragment (CYFRA21-1) and neuron specific enolase (NSE) were determined by radioimmunoassay in 112 patients with lung cancer and 74 patients with benign pulmonary disease.Four tumor markers were analyzed by Logistic regression and ROC curve.Results The serum levels of CEA,CA125,CYFRA21-1 and NSE in lung cancer patients were [4.53(2.22-11.53)ng/ml,28.97 (11.39-62.10) U/ml,4.05(2.29-8.18)ng/ml,14.11 (11.35-24.12) ng/ml],respectively,which were significantly higher than those in health adults[2.08 (1.45-2.52) ng/ml,12.90 (9.80-19.44) U/ml,1.53 (1.21-2.17) ng/ml,11.38 (9.07-12.80) ng/ml] (all P < 0.01).According to regression equation Y =1/[1 + EXP (4.902-0.394X1-0.627X2-0.165X3)],the area under the ROC curve (AUC),sensitivity,specificity,and accuracy of new variable Y were 0.915 ± 0.020,79.46%,93.24%,and 84.95%,respectively.Conclusions Application of logistic regression and ROC curve increases diagnostic accuracy in lung cancer.
