2.EFFECTS OF FRUCTOOLIGOSACCHARIDE ON CHOLESTEROL METABOLISM IN FINISHING PIGS

Objective: To study the effects of dietary fructooligosaccharide (FOS) on cholesterol metabolism in finishing pigs. Methods: A total of 96 finishing pigs (Duroc?Landrance?Jia) were randomly alotted to four groups each of which was replicated three times with eight pigs per replicate. The groups received the same basal corn-soybean meal diet and FOS was added to the basal diet at 0,0.5%,1 0%,1.5% at the expense of corn, respectively. At the end of the feeding trial, eight pigs with similar body weights from each treatment were slaughtered. Results: (1) Supplementation with FOS significantly improved average daily gain and feed efficiency. (2) Pigs fed 1.0% and 1.5% FOS had significantly lower serum TC and LDL-C concentrations and greater HDL-C/LDL-C ratio compared with the control. Pigs fed 1.5% FOS had significantly lower serum VLDL-C and ApoB concentrations and Apo B/Apo A-1 ratio. (3) Supplementation with 1.0% and 1.5% FOS significantly decreased the activities of HMG-CoA reductase. (4) Supplementation with 1.5% FOS significantly increased the colonic acetate and butyrate and plasma acetate. Supplementation with 0.5%,1.0% and 1.5% FOS significantly increased colonic propionate. (5) Fecal cholesterol excretion was significantly greater in the pigs fed 1.0% and 1.5% FOS and fecal bile acid excretion was significantly greater in the pigs fed 1.5% FOS than the control. Conclusion: FOS may improve cholesterol metabolism by alteration in the absorption and synthesis of cholesterol resulted from the changes in colonic fermentation, and by increase in the fecal excretion of cholesterol and bile acid.
3.DYNAMIC EFFECT OF ADMINISTERING DIETHYLSTILBESTROL EXTERNALLY TO NEONATAL FEMALE BALB/c MICE ON THE EXPRESSION OF ESTROGEN RECEPTOR IN MICE SPLEENS

Objective To investigate the dynamic changes of estrogen receptors (ER) in spleen of female BALB/c mice after injected with diethylstilbestrol (DES) beginning in neonatal period. Methods Neonatal female BALB/c mice were injected subcutaneously on cervix-backside with DES within 24 hours after birth at intervals of 24 hours for 5 times. Control groups were injected with olive oil with the same method. Mice were killed on 7 days, 14 days, 21 days, 35 days and 49 days after birth separately, the spleen was taken out, imbedded with paraffin and sectioned serially in 5??m. The expression of ER in slice of spleen was detected by immunohistochemistry technique. Results ER positive cells were present in both DES-injected groups and control groups. ER positive cells were mostly lymphocytes. A correlation between age and the intensity of positive reactions was showed in both groups. The intensity of ER immunohistochemistry positive reaction in DES group was much stronger than that in control group, and there was a significant statistic difference between the two groups. Conclusion The ER expression exists in spleen of female BALB/c mice and increases with the age of mice. The contact of DES in neonatal female BALB/c mice will lead to an increased ER expression in their spleens and this effect can continue to their manhood at least.
4.The effect of cardiac massage by subdiaphragmatic compression on hemodynamics and apotosis of myocardial cells of rabbit with cardiac arrest during abdomen operations

Objective To observe the effect of cardiac massage by subdiaphragmatic compression (D-CPR) on the length of time required from cardiac arrest (CA) to restoration of spontaneous circulation (ROSC),hemodynamics,rate of ROSC,survival rate of 6 h,level of Caspase3 in myocardial cells and apoptosis index (AI) of myocardial cells and compare the effect of standard cardiac massage by chest compression (S-CPR) on those variables in order to choose the more effective resuscitation method for the patient with CA during abdomen operations.Methods A total of 32 healthy New Zealand rabbit were randomly (random number) divided into two groups,namely S-CPR group and D-CPR group (n =16 in each group).All of rabbits were anesthetized with ketamine and Shumianxin (a kind of hypnotics) by intraperitoneal injection,subsequently tracheotomy was made for endotracheal intubation,and right internal jugular vein was catheterized for monitoring central venous pressure (CVP) and left common carotid artery was for indwelling cannula to monitor arterial blood pressure.Lead-2 of ECG was placed.After laparotomy and vital signs of rabbits stabilized for 5 minutes,the endotracheal tube was clamped at the end expiration for 8 minutes to make asphyxial cardiac arrest model.The effects of two different methods were observed and compared in respects of changes in hemodynamics、length of time elapsed from CA to ROSC、ROSC rate and the survival rate in 6 h.The level of Caspase3 in myocardial cells and AI of myocardial cells were detected by using immunohistochemistry staining method and TUNEL,respectively 6 hours after successful resuscitation.Results ①The length of time consumed fiom CA to ROSC in D-CPR group was shorter than that in S-CPR group (P <0.05) ②Coronary perfusion pressure (CPP) and MAP 15 minutes after CPR were higher in D-CPR group than those in S-CPR group (P < 0.05).③SBP and DBP after ROSC were higher in D-CPR group than those in S-CPR group.④ROSC rate in D-CPR group was significantly higher than that in S-CPR group (81% vs.43%,P <0.05).⑤Survival rate in 6h in D-CPR group was substantially higher than that in S-CPR group (75% vs.25%,P < 0.05).⑥HE staining showed that severe myocardial damage manifesting in edema of myocardial cell,indistinguishable cell boundary,and patchy necrosis with infiltration of scanty inflammatory cells were found in S-CPR group.While in D-CPR group,mild myocardial damage in form of slight cellular edema and distinctive cell boundary was observed.⑦Level of Caspase3 in myocardial cells in terms of integrated optical density (IOD) of postive Caspase3 cells was substantially lower in D-CPR group than that in S-CPR group (P < 0.05).⑧Apoptosis index (AI) of cells was lower in D-CPR group than that in S-CPR group (P < 0.05).Conclusions ①The hemodynamics in D-CPR group was more stable than that in S-CPR.group,and D-CPR increased CPP,MAP,ROSC rate and survival rate in 6h,improving achievement of successful resuscitation.②D-CPR was more effective in terms of shortening the length of time for restoration of spontaneous circulation、decreasing level of Caspase3 in myocardial cells、decreasing apoptosis index of myocardial cells and ameliotating myocardial damage from ischemic repeffusion injury.
5.Study on flipped classroom in teaching of “auricular needling method”

Objective To explore the effect of flipped classroom in teaching auricular needling method. Methods Sophomores of acupuncture and Tuina speciality were divided into two groups based on classes, with 33 students in experimental group and 30 in control group. In teaching auricular needling method, experimental group adopted flipped classroom by online resources and classroom activities, while control group adopted traditional classroom such as teaching, watching video, demonstration. Post-test score of two groups were compared by independent samples t-test. The teaching effect of flipped classroom was evaluated by questionnaire survey of students in experimental group. Results The total score of auricular needling (88.4±13.91), score of location (55.40±7.34), and score of application (32.00±10.31) in experi-mental group were significantly higher than those in con-trol group (71.48±15.93, 47.55±12.23 and 23.94± 11.97) (P<0.05). Students' satisfaction concerning the flipped classroom in experimental group was up to 90%. Conclusion Flipped classroom can be used in teaching auricular needling method, which can improve teaching quality and promote improvement of students' self-study and communication skills.
6.Practice of Clinical Pharmacists Participating in One Case of Multiple Organ Cryptococcus Infection during Perinatal Period

Objective: To evaluate the clinical pharmacists’ role in the drug treatment of perinatal women with multiple organ cryptococcus infection. Methods:Clinical pharmacists participated in the treatment of one case of perinatal women with multiple organ cryptococcus infection and provided comprehensive pharmaceutical service, including antifungal drug selection, adverse drug reactions prevention and monitoring, glucocorticoid selection in fetal lung maturity and recommendation in lactation. Results: Clinical pharma-cists provided suggestions on the drug treatment in many ways to enhance the safety, efficiency and economic of the drug treatment, and improve the patient's compliance as well. Conclusion:Clinical pharmacists can optimize the treatment regimen and play an active role in the drug treatment of perinatal patients with cryptococcus infection.
7.Effects of caffeine citrate on neuronal proliferation and apoptosis and long-term learning ability in neonatal rats with hypoxia-ischemic brain damage

ObjectiveTo investigate the effects of caffeine citrate (CC) on the neuronal proliferation and apoptosis and long-term learning ability in neonatal rats with hypoxia-ischemic brain damage (HIBD).MethodsForty-eight 7-day-old Sprague-Dawley neonatal rats were randomly divided into 3 groups: sham operation group (n=16), HIBD group (n=16), HIBD + caffeine citrate group (CC group,n=16). Rats in HIBD and CC groups received ligation of left common carotid artery followed by 2 hours of hypoxia to establish HIBD model. Rats in CC group were injected intraperitoneally with CC (20 mg/kg) before and at 0 min, 24 h, 48 h, and 72 h after hypoxia-ischemic (HI), and rats in the other two groups were injected intraperitoneally with an equal volume of normal saline at the corresponding time. Meanwhile, from postnatal day 10, each rat was injected intraperitoneally with 5-bromo-2’-de-oxyuridine (BrdU) (50 mg/kg) for 5 consecutive times, once every 12 h. On postnatal day 12, BrdU in the hippocampal dentate gyrus and cleaved caspase-3 in the hippocampal CA1 area were detected by immunohistochemistry, and neuronal apoptosis in hippocampal CA1 area were detected by TUNEL staining. On postnatal day 28, long-term learning and memory ability of rats was tested by Y maze.ResultsThere was signiifcant difference in the number of BrdU-positive cells in brain tissues of rats among three groups (F=101.38,P<0.01). The BrdU-positive cells in HIBD group and CC group were signiifcantly more than those in sham operation group (P<0.05). There was signiifcant difference in the number of cleaved caspase-3-positive cells in hippocampal CA1 area among three groups (F=379.77,P<0.01). The cleaved caspase-3-positive cells in CC group were signiif-cantly fewer than those in HIBD group but signiifcantly more than those in sham operation group (P<0.05). The TUNEL-pos-itive cells in hippocampal CA1 area were signiifcantly different among three groups (F=505.92,P<0.01) which was most in HIBD group and fewest in sham operation group and signiifcant difference was found through multiple comparison (P<0.05). The total learning number of avoiding electric shock tested by Y maze was signiifcantly different among three groups (F=32.05, P<0.01) which was most in HIBD group. Correct response rate was significantly different among three groups (F=24.99, P<0.01) which was lowest in HIBD group.ConclusionsCaffeine citrate can improve the ability of long-term learning and memory in neonatal rats with hypoxia-ischemic brain damage, the mechanism of which may be related to reducing the neuronal apoptosis after hypoxia ischemia.
8.Application of direct observation of procedural skills (DOPS) in teaching acupuncture manip-ulation skills

Objective To explore the effect of direct observation of procedural skills (DOPS) in teaching acupuncture manipulation skills. Methods Sophomores of acupuncture and Tuina spe-ciality were divided into two groups based on their classes, 36 students in experimental group and 31 in control group. In experimental group, DOPS on filiform needling was done twice in the middle and at the end of the semester respectively. In control group, DOPS was done once at the end of semester. The final exam's DOPS score and satisfaction degree of teachers and students concerning the results of the assessment in two groups were compared by independent samples t-test. P<0.05 indicates signifi-cant differences. The teaching effect of DOPS was also evaluated by questionnaire survey of students in experimental group. Results The total score of DOPS (4.26 ±0.24), scores of needle insertion (4.18±0.42), and scores of reinforcing and reducing method (4.03±0.28) in experimental group were higher than those in control group ((4.14±0.25), (4.00±0.29) and (3.87±0.37), P<0.05). The teachers' satisfaction (8.38±0.51) and students' satisfaction (8.47±1.03) concerning the results of the assessment in experimental group were higher than those in control group ((8.05±0.78) and (7.94±1.15), P<0.05). More than 80% of students in experimental group supported the saying of‘DOPS promoting students' skills training and enhancing the ability of handwork ’ . Conclusions DOPS can be used as an effective teaching and examination method and can improve students' skills of acupuncture manipulation.
9.Establishment of an Examination and Assessment System of Filiform Needle Manipulation

Filiform needle acupuncture technique is the core content of needling and moxibustion method. The examination and assessment of filiform needle manipulation are the most important measure for testing the effect of filiform needle acupuncture technique teaching. The establishment of a scientific examination and assessment system of filiform needle manipulation should involve comprehensive examination and assessment contents, definite and detailed evaluation rules and quantified scoring criteria to make the examination and assessment system be able to comprehensively and objectively assess the students' skills of filiform needle manipulation to achieve the purpose of improving the quality of filiform needle acupuncture technique teaching.
