1.Investigation in climacteric symptoms and depression of gynaecology outpatients and inpatients

Objective To investigate the climacteric symptoms and depressive related factors of climacteric females in gynecology outpatients and inpatients,to provide evidence for effective health promotion.Methods 416 climacteric women were investigated by using improved menopause Kuppermen Scales and Center for Epidemiological Studies Depression (CES-D).The results underwent analysis.Results There were 93.99% of patients suffering from climacteric syndrome,49.52% with depression symptom.The main factors of climacteric syndrome were eating breakfast according to the timetable every day,nutritionally balanced diet,controlling of salt intake,regular body exercises and to maintain a proper body weight.Factors related to depression symptoms were sore breasts,constipation,dry skin,lower abdominal pain.The correlation between climacteric syndrome and depressive symptom was significant.Conclusions The main influencing factor of climacteric syndrome was healthy lifestyle.The climacteric syndrome increases the risk of depressive symptoms.
2.Study of postpartum depression state and its related factors among Chinese and Japanese parturients

ObjectiveTo explore the related factors of postpartum depression among Chinese and Japanese women and analyze the cause of it.Methods587 delivery women who were from Tokyo,Japan and Henan China were investigated by using the Edinburgh Postnatal Depression Scale(EPDS) and 12-item General Health Questionnaire (GHQ-12), 352 Chinese women included and 235 Japanese women included.ResultsThe incidence of postpartum depression of Chinese women was 31 cases accounting for 8.81% and Japanese women was 7 cases accounting for 2.98%. The number of GHQ- 12 cases were higher in Chinese women than in Japanese women. Through Logistic regression analysis, dehvery experience, having job or not, having family support or not, having someone looking after baby or not, the degree of physical pain are related factors were influencing factors of postpartum depression.Conclusions The incidence of postpartum depression of Chinese women was higher than Japanese women. Having no job is the risk factor of postpartum depression. The factors of eutocia, having someone looking after baby, the lower degree of physical pain were protective factors.
3.Effects of Notch1 gene on the proliferation of and apoptosis in xenografted human cutaneous squamous cell carcinoma cells in nude mice

ObjectiveTo investigate the role of Notch1 gene in xenografted human cutaneous squamous cell (SCL-1) carcinoma. MethodsFifteen nude mice were divided into three groups, including untreated group(inoculated with SCL-1 cells treated with phosphate buffered saline), empty vector group (inoculated with SCL-1 cells transfected with empty vector) and Notch1 group(inoculated with SCL-1 cells transfected with Notch1 expression vector). All the mice were inoculated with SCL-1 cells(1 x 108/ml) of0.2 ml. Then, the growth of xenografted tumor was observed every other day. Fifteen days later, the mice were sacrificed, tumor tissue was dissected and subjected to terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling (TUNEL) assay for the detection of cell apoptosis, reverse-transcription(RT)-PCR and Western blot for the examination of mRNA and protein expressions of Notch1, bcl-2 and bax, respectively. ResultsThe proliferation of xenografted tumor in Notch1 group was obviously inhibited compared with the untreated group. The weight of xenografted tumor in Notch1 group was significantly lower than that in the untreated group and empty vector group (0.574 ± 0.219 g vs. 2.642 ± 0.404 g and 2.606 ± 0.512 g, F= 26.642, P< 0.01). TUNEL assay demonstrated that the number of apoptotic cells per 500 cells in tumor tissue specimens was(87 ± 9) in Notch1 group, evidently higher than that in the untreated group(8 ± 2) and empty vector group(10 ± 3) (F = 194.266, P < 0.05 ). Further, RT-PCR and Western blot revealed that the mRNA and protein expressions of Notch1 and bax were significantly upregulated, but those of bcl-2 were markedly downregulated in the Notch 1 group, with significant difference among the three groups(all P < 0.05). ConclusionsNotch 1 gene can inhibit the growth of xenogra ffted human cutaneous squamous cell(SCL-1) carcinoma and induce SCL-1 cell apoptosis likely by upregulating bax expression and downregulating bcl-2 expression.
4.Detection and analysis of protein and mRNA expressions of pituitary tumor transforming gene (PTTG) and basic fibroblast growth factor (bFGF) in cutaneous squamous cell carcinoma tissue

ObjectiveTo investigate the role and clinical pathological significance of PTTG and bFGF in cutaneous squamous cell carcinoma(CSCC).MethodsTissue specimens were collected from the lesions of 42 patients with CSCC and normal skin of 42 normal human controls.The protein and mRNA expressions of PTTG and bFGF were detected by immunohistochemistry and in situ hybridization in these specimens respectively.ResultsA significant increase was observed in the positive expression rates of PTTG and bFGF proteins[64.3%(27/42) vs.11.9%(5/42),73.8%(31/42) vs.21.4%(9/42),both P< 0.05] and mRNA [59.5%(25/42) vs.7.1%(3/42),75.0%(29/42) vs.16.7%(7/42),both P< 0.05] in the CSCC tissue specimens than in the control specimens.The protein and mRNA expressions of PTTG were positively correlated with those of bFGF(both P < 0.05),and closely correlated with histological grade of CSCC (both P <0.05).ConclusionThe high expression of PTTG and bFGF may be associated with the initiation of CSCC.
5.Effect of PTTG siRNA on MMP-2 and MMP-9 expression in nude mice bearing xenografted human cutaneous squamous cell carcinoma

Objective To investigate the effect of pituitary tumor-transforming gene (PTTG)siRNA on the growth,invasion of,and expression of metastasis-related cytokines including matrix metalloproteinase-2 (MMP-2)and MMP-9 in xenografted human cutaneous squamous cell carcinoma in nude mice.Methods SCL-1 cells were subcutaneouslv inoculated into Balb/c nude mice to establish a xenograft model of human cutaneous squamous cell carcinoma.Then,15 mice bearing xenografted carcinoma were equally divided into 3 groups to be inoculated with phosphate buffer saline (PBS),control siRNA,and PTTG siRNA of 50 nmoI/L,respectively,ever),other day for 2 weeks.The size of xenograted carcinoma in these mice was measured every other day.At the end of 2-week treatment.the mice were killed followed by the evaluation of tumor weight,as well as the quantification of mRNA and protein expression of PTTG,MMP-2 and MMP-9 by reverse transcription (RT)-PCR and Western-blot,respectively.Results The xenograft model of human cutaneous squamous cell carcinoma was successfully established.The treatment with PTTG siRNA obviously inhibited the growth of the xenografted tumom and the expression of PTTG mRNA and protein compared with PBS and control siRNA (all P<0.05).In addition,the expression of MMP-2 and MMP-9 in xenografted tumors in PTTG siRNAtreated mice were significantly lower than those in PBS and control siRNA-treated mice.suggesting that PTTG siRNA evoked the decrease in invasive and metastatic ability of xenografted tumors.Conclusions PTTG siRNA can inhibit the growth of human cutaneous squamous cell carcinoma xenografts in nude mice,and downregulate the expression of invasion-and metastasis-related cytokines,including MMP-2 and MMP-9.
6.Bactericidal effect of ozonated camellia oil on Staphylococcus aureus in vitro

Objective:To explore a new method for detecting the bactericidal effect of oiling agent in vitro,and to determine the disinfectant effecacy ofozonated camellia oil on Staphylococcus aureus.Methods:Suspension of Staphylococcus aureus was prepared and innoculated on the LB plate by plate scribing method.After culture overnight,21 bacterial monoclones with the same diameter were selected and divided into 3 groups:A negative control group,a baseoil (camellia oil) group and an ozonated camellia oil group.We used a ring to isolate the single clone and added oil inside the ring,cultured the whole plate over night,picked out each single clone (with gel) to 5 mL LB medium and cultured it for 12 h.The final concentration of the LB medium was detected by plate count method and turbidimetry.Results:According to the plate count method and turbidimetry,the bacterial concentration in the ozonated camellia oil group was lower than that in the negative control group and base oil group Conclusion:Bacterial monoclone culture method shows that ozonated camellia oil can significantly kill Staphylococcus aureus,and this method is an effective method for evaluating the bactericidal function of the oiling agent in vitro.
7.Investigation of self-acceptance status and its influencing factors of elderly people in nursing home

Objective:To understand the present situation of self-acceptance of elderly in nursing home and explore its influencing factors, so as to provide basis for improving self-acceptance level of elderly in nursing home.Methods:Totally 415 elderly people in 3 Medical and nursing homes in Fuyang city of Anhui province were selected by Self-Acceptance Scale, and influencing factors were analyzed.Results:The total score of Self-Acceptance Scale of the elderly in nursing home was (44.11±7.25) points, the score rate was 67.86% (44.11/65), in the middle level; the results of multiple linear stepwise regression showed that the relationship with children, hobbies, number of children, length of stay in nursing home, frequency of visits and age were the main influencing factors ( F value was 48.228, P<0.001). Conclusion:The self-acceptance of the elderly in nursing home is in the middle level. The nursing home or social should pay attention to the higher age group and the newly admitted elderly in nursing home, give such people timely psychological guidance, help them to plan their later life, cultivate their interests and hobbies through recreational activities, and take corresponding measures to harmonize their relationship with their children in order to improve their self-acceptance level and promote mental health.
9.Effects of different intensity of high frequency repetitive transcranial magnetic stimulation on upper limb function in patients with early post-stroke

Objective:To evaluate the effects of high-frequency repetitive transcranial magnetic stimulation(HF-rTMS)of different intensities on the recovery of upper limb motor dysfunction in patients during the early recovery stage of cerebral infarction. Method:Sixty patients with cerebral infarction at early recovery stage were randomly divided into the HF-rT-MS 80%RMT group,the HF-rTMS 100%RMT group and the sham stimulation group,with 20 patients in each group.All three groups were also given basic treatment(medication and routine rehabilitation training).Be-fore and after 4 weeks treatment,the Fugl-Meyer Assessment-Upper extremity(FMA-UE)and Modified Barth-el Index(MBI),the latency of motor evoked potential(MEP).central motor conduction time(CMCT)and blood flow velocity of the ipsilateral middle cerebral artery(MCA)were recorded. Result:Before treatment,there were no significant differences in FMA-UE score,MBI score,MEP latency,CMCT and the blood flow velocity of MCA among the three groups(P>0.05).After four weeks of treatment,the above indicators of the three groups were significantly higher than those before treatment(P<0.05).More-over,the improvement of the indicators in the 80%RMT group and 100%RMT group were significantly higher than those in the sham stimulation group(P<0.05).Except for MBI score,the improvement in all indicators in the 100%RMT group were the most significant(P<0.05). Conclusion:HF-rTMS can improve upper limb function and activity of daily living during the early recovery stage of cerebral infarction.Compared with 80%RMT,100%RMT HF-rTMS has more advantages in improv-ing function,motor nerve excitability and cerebral blood flow.
10.Simultaneous Determination of 6 Flavonoids in the Roots of Tetrastigma hemsleyanum by HPLC

OBJECTIVE: To establish a method for simultaneous determination of rutin, isoquercitrin, kaempferol-3-O- rutinoside, daunorubicin, quercetin and kaempferol in the roots of Tetrastigma hemsleyanum. METHODS: HPLC method was adopted. The determination was performed on Alliance SilGreen C18 column with mobile phase consisted of 0.2% phosphoric acid solution-acetonitrile solution (gradient elution) at the flow rate of 1.0 mL/min. The detection wavelength was 360 nm and the column temperature was at 35 ℃. The sample size was 15 μL. RESULTS: The linear range of rutin, isoquercitrin, kaempferol-3- O-rutinoside, daunorubicin, quercetin and kaempferol were 21.77-217.77, 12.37-123.75, 13.23-132.31, 4.63-46.30, 5.75-57.50, 3.36-33.66 μg/mL (all r=0.999 9), respectively. limit of detection of them were 0.217 8, 0.123 8, 0.066 2, 0.046 3, 0.191 7, 0.112 3 μg/mL, respectively. limit of quantitation of them were 0.435 6, 0.247 5, 0.165 4, 0.154 3, 0.575 0, 0.421 2 μg/mL, respectively. RSDs of precision (n=6), stability (24 h, n=7) and reproducibility tests (n=6) were lower than 3.20%. The average recoveries of them were 96.23%, 86.88%, 97.51%, 97.67%, 97.50%, 87.46%, RSDs were 1.85%, 1.90%, 1.84%, 1.87%, 1.25%, 2.01% (n=9), respectively. CONCLUSIONS: The method is fast and simple, and could be applied for simultaneous determination of rutin, isoquercitrin, kaempferol-3-O-rutinoside, daunorubicin, quercetin and kaempferol in the roots of T. hemsleyanum.
