1.STUDY OF HIGH-RESOLUTION G-BANDING PATTERN OF RABBIT(ORYCTOLAGUS CUNICULUS)

Since rabbit (Oryctolagus cuniculus) possess many fine biological characteristics, they have been used in the studies of tumour, gene mapping and evolution. All of these need us to know the high-resolution G-banding pattern of rabbit chromosomes. Primary lung fibroblasts of new born rabbits were cultivated in RPMI 1640 medium supplemented with 20% new born bovine serum. In order to get the synchronized cells, chemical(MTX, TdR) method and physical (Cold Shock) method were used. By using trypsin-Giemsa banding-technique, we obtained fine high-resolution G-banding karyotypes of rabbit. The bands of rabbit chromosomes were analyzed and described. An idiogram which contains 583 bands of high-resolution G-banding chromosomes of rabbit has been made. The feature of rabbit choromosome bands and the method uesd to get the high resolution banding chromosomes were briefly discussed.
2.Effect of subchronic exposure to fluoride on mRNA expression of estrogen receptor in female mice

Objective To study the subchronic toxicity of sodium fluoride on mRNA expression of estrogen receptor(ER) in female mice.Methods Forty female mice were randomly divided into 4 groups according to body mass,10 in each group,and exposed to sodium fluoride solution(0,1,5,25 mg/L) through drinking water in control,low-dose,medium-dose and high-dose groups for 12 weeks.The expression levels of ERα,ER3 mRNA in ovarian tissues were determined by RT-PCR method.Results The relative expression levels of ERα,ERβ mRNA in control,low-,medium-and high-dose groups were 0.7028 + 0.0474,0.7195 ± 0.0552,0.6479 ± 0.0590,0.5684 ± 0.0513 and 0.8418 ± 0.0131,0.7729 ± 0.0974,0.7610 ± 0.0984,0.8026 ± 0.0234,respectively.The difference between high-dose and control groups of the expression level of ERα was statistically significant (P < 0.05).Conclusions The subchronic toxicity of fluoride can decrease the expression of estrogen receptor in ovarian tissue,which may have a certain effect on reproductive development of female mice.
4.Experimental study on the protective effect of lentivirus-mediated SIRT6 overexpression on radioactive lung injury in rats

Objective To investigate the protective effect of lentivirus-mediated silencing information regulation protein 6(SIRT6)overexpression on radioactive lung injury in rats.Methods 72 male 150-200 g Wistar rats were randomly divided into three groups(n=24).The 6 MV-X-ray linear accelerator was used to irradiate the lungs.The rats in each group were injected with normal saline(TNF-α)were measured at 1, 2, 4, and 8 weeks after radiotherapy(Lent-SIRT6), and the levels of tumor necrosis factor-α(TNF-IL-6 and IL-1β were measured by HE staining.The levels of TNF-α, IL-6 and IL-1β in liver tissue were detected by HE staining.The levels of TNF-α, IL-6 and IL-1β in liver tissue were detected by HE staining.Results The alveolar wall and alveolar stroma were normal in the control group.The alveolar wall of the irradiated group was thickened and fibrosis, and a large number of hypertrophic fibrous tissues were found in the alveolar stroma.The alveolar wall of Lent-SIRT6 group was thickened And alveolar stromal fibrosis symptoms were lighter than the irradiation group.Compared with the control group, the levels of serum TNF-α and IL-6, neutrophil count and the levels of TNF-α, IL-6 and IL-1β in the liver tissue were significantly higher than those in the control group (P<0.05).The lentiviruses overexpressing SIRT6 could alleviate the abnormalities caused by radiation (P<0.05).The difference was statistically significant (P<0.05).Conclusion SIRT6 can effectively inhibit the inflammatory reaction, reduce the lung injury symptoms of radiation pneumonia, and have some protective and prevention effect on lung injury.
5.The mechanism of Er-xian Decoction in regulating lipid metabolism disorders on bilateral ovariectomized rats based on metabolomics study

Studies have shown that women's menopause caused by permanent cessation of ovarian function is closely related to lipid metabolism disorders. Er-xian Decoction has been used in the clinical treatment for gynecological diseases and has a good effect on diseases related to reduced sex hormone function. In this study, metabolomics was performed on bilateral ovariectomized model rats within 12 weeks after modeling to mimic the physiological state of menopausal women in different menopausal stages and Er-xian Decoction dosed model rats. The results of liver oil red O staining sections showed lipid metabolic disorder of bilateral ovariectomized model rats and the regulating effects of Er-xian Decoction. 46 potential biomarkers (6 steroid hormones, 3 sphingolipids, 11 phospholipids and 26 glycerides) in plasma and 32 potential biomarkers (1 steroid hormones, 20 phospholipids and 11 glycerides) in liver were obtained based on lipidomics analysis. Then, we analyzed the differential metabolic pathways and construct the lipid metabolism network significantly regulated by Er-xian Decoction. The results provided valuable information for in-depth understanding of the gradual changes on lipid metabolism disorders under menopausal conditions and the characteristics and mechanisms of compound Er-xian Decoction's regulatory effects. The study complied with the procedures established by the Animal Experiment Ethics Committee of the Institute of Materia Medica, Chinese Academy of Medical Sciences and passed the animal experiment ethics examine (No. 00000918).
7.Using inverse distance weighting in studying the distribution of endemic fluorosis in Jiangsu Province

Objective To use inverse distance weighting(IDW)in studying the distribution of endemic fluor.0sis in Jiangsu Province and evaluate the value of IDW in endemic fluorosis surveillance.Methods A geographic information system(GIS)database of endemic fluorosis was established in Jiangsu Province from the data of endemic fluorosis survevs conducted during 1982-1985.With the help of Arc View 3.3 system,IDW was applied to forecast the distribution of fluoride concentration in water and the distribution of the prevalence rate of dental fluorosis in Jiangsu Province based on the electronic map of Jiangsu Province.Results IDW was applied to forecast the distribution of endemic fluorosis in Jiangsu Province.By comparing with the result of endemic investigation in the 1980's.the forecasting Was proven to be accurate,exact and detailed.Conclusion With the application of IDW and stratified sampling,it is feasible to describe the spatial distribution of endemic fluorosis in Jiangsu Province in endemic fluorosis surveillance.

9.Application of 3% hypertonic saline in patients with severe craniocerebral injury after operation

Objective To study the application of 3% hypertonic saline (HS) in patients with severe craniocerebral injury after operation.Methods Sixty-eight cases with severe craniocerebral injury after operation were divided by random digits table method into HS group and control group with 34 cases each.The patients in control group were treated with 125 ml 20% mannitol every 6 hours or 8 hours.The patients in HS group were given conventional mannitol and added with 130 ml 3% HS every 12 hours or 8 hours through fast intravenous drip altemated with mannitol.The levels of intracranial pressure (ICP),mean arterial pressure (MAP),central venous pressure (CVP) were recorded within 6 hours,12 hours,24 hours of the 1st day,24 hours of the 2nd day,24 hours of the 3rd day,24 hours of the 5th day.The Na+ level of blood serum and the plasma osmotic pressure were recorded and calculated within 12 hours,24 hours of the 1st day,24 hours of the 2nd day,24 hours of the 3rd day,24 hours of the 5th day.The Glasgow coma scale( CCS) score was accounted within 24 hours of the 1 st day,24 hours of the 3rd day,24 hours of the 5th day.Results Compared with control group,the level of ICP in HS group decreased at every time point (P < 0.05).The tendency of ICP in two groups was not significant on the 1st and 2rid day (P > 0.05 ),but the level of ICP within 24 hours of the 3rd and 5th day obviously increased compared with those of the 1st and 2nd day (P<0.05).The levels of MAP and CVP in HS group were significantly higher than those in control group within 6 hours,12 hours and 24 hours of the 1 st day and 24 hours of the 2nd day [ ( 87.98 ± 5.03 ),(88.56 ± 5.36),(87.04 ±6.90),(90.03 ±5.19) mm Hg (1 mm Hg =0.133 kPa) vs.(77.98 ±5.09),(79.98 ±6.09),(80.98 ± 5.27),(81.98 ± 4.32) mm Hg and (9.23 ± 1.24),(9.67 ± 1.35),( 10.21 ± 1.38 ),( 10.56 ± 1.96)mm Hg vs.(7.15 ± 2.01 ),(8.32 ± 1.53),(8.67 ± 1.89),(9.22 ± 2.03) mm Hg] (P < 0.05),but the values within 24 hours of the 3rd and 5th day between two groups had no significant differences (P> 0.05 ).The Na+ level of blood serum and the plasma osmotic pressure in HS group at every time point was obviously higher than that in control group (P< 0.05).The GCS scores within 24 hours of the 1st,3rd and 5th day in HS group were (4.21 ± 2.31 ),(5.44 ± 2.46 ),(7.23 ± 1.64 ) scores,respectively,while the scores in control group were (4.14 ± 2.10),(5.15 ± 2.31 ),(7.31 ± 2.12) scores,respectively ;the score within 24 hours of the 5th day in two groups was obviously higher than that of the 1st day respectively (P< 0.05 ),but the scores between two groups had no statistical significance (P> 0.05 ).Conclusions Adding 3% HS is more effective to decrease ICP,improve the brain perfusion and reduce the adverse reactions.3% HS can be used as the first-line therapy for patients with severe craniocerebral injury after operation.
10.Effect of mild hypothermia induced by endovascular cooling with heating exchange catheters on severe traumatic brain injury in patients

Objective To investigate the effects of mild hypothermia induced by endovascular cooling with heating exchange catheters on severe traumatic brain injury in patients. Methods Twenty patients with severe traumatic brain injury aged 18-60 yr were randomly divided into 2 groups (n=10 each):ice bay cooling group (group IBC) and endovascular cooling group (group EVC).The state of consciousness was scored on a Glasgow coma scale (GCS).The patients had GCS scores of 3-8. The patients underwent emergency surgery.A probe of intracranial pressure monitor was placed during operation.In group EVC intravascular heat exchange catheters were inserted via femoral vein and connected to intravascular heat exchange system (CoolGard 3000, Alsius, USA). In group EVC body temperature was reduced to 34℃ and maintained at this level for 48 h. MAP, HR, body temperature and intracranial pressure (ICP) were continuously monitored and cerebral perfusion pressure (CPP) was calculated. Blood samples were taken from peripheral vein for determination of serum concentrations of neuron specific enolase (NSE), myelin basic protein (MBP) and S-100B (by enzyme linked immunosorbent assay) and GCS scores were assessed at 10 min before (baseline) and 12, 24, 48 and 72 h after operation. The state of consciousness was again assessed 3 months after operation and scored on Glasgrow outcome scale(GOS). Results ICP was significantly lower and CPP was higher after operation in group EVC than in group IBC. Serum concentrations of NSE, MBP and S-100B were significantly lower after operation in group EVC than in group IBC. Conclusion Mild hypothermia induced by endovascular cooling with heating exchange catheters can effectively reduce severe traumatic brain injury in patients.
