1.Impact of paricalcitol on proteinuria in diabetic nephropathy rats

ObjectiveTo study the effect of 1,25-(OH)2D3 analogs paricalcitol on proteinuriaindiabeticnephropathy (DN)rats, andtoinvestigateitspossiblemechanism.Methods DN model rats were established by intraperitoneal injection with streptozotocin.All the DN rats were randomly divided into the paricalcitol group(group P ) and DN group(group D).Healthy rats were chosen as healthy control group(group N).24-h urinary protein and serum biochemical indicators were examined after 12 weeks.ELISA was applied to detect the level of renin and Ang Ⅱ in the kidney.Immunohistochemistry and real-time PCR were used to detect the protein and mRNA expression of heparanases(HPA)and podoein.Results Compared with group N,24-h urinary protein,serum creatinine,renin and Ang Ⅱ in group D and group P were markedly increased,and they were significantly higher in group D as compared to group P (all P<0.05).Compared with group N,the expression of HPA protein and mRNA in group D and group P increased markedly,and higher expression was found in group D(all P<0.05).The expression of podocin protein and mRNA in group D and group P decreased markedly,and lower expression was found in group D(all P<0.05).Renin level was positively correlated with HPA protein expression (r=0.78,P<O.OS),negatively correlated with podocin protein expression(r=-0.63,P<O.05),and not correlated with their mRNA expression.Conclusion Paricalcitol can significantly reduce the proteinuria,which may be associated with the inhibition of renin by down-regulating protein expression of HPAin glomerular basement membrane and up-regulating protein expression of podocin in podocyte.
2.Clinical Effect Observation of Alendronate Combined with Jintiange Capsules in the Treatment of Postm-enopausal Osteoporosis

To explore the clinical effect of alendronate combined with Jintiange capsules in the treatment of postmeno-pausal osteoporosis ( PMOP) . Methods:Ninety-eight cases of PMOP patients were randomly divided into three groups. The combina-tion treatment group (34 cases) was treated with alendronate tablets combined with Jintiange capsules, while the control group A (32 cases) and the control group B (32 cases) was respectively treated with alendronate tablets and Jintiange capsules, and the treatment course was six months. The changes of the clinical symptom score, bone mineral density ( BMD) , serum calcium, serum phosphorus, AKP, hepatorenal function and so on before and after the treatment were recorded. Results:After the six-month treatment, the clinical symptom score of the treatment group was much better than that in the two control groups(P<0. 05). BMD in the three groups was in-creased after the treatment(P<0. 05), and the increase in the treatment group was more notable than that in the two control groups(P<0. 05). Conclusion:Alendronate combined with Jintiange capsules can effectively improve BMD and relieve clinical symptoms in the patients with PMOP, which is worthy of promoted application.
3.Influence of paricalcitol on renal tubulointerstitial fibrosis in diabetic ne-phropathy

[ ABSTRACT] AIM:To investigate the effect of paricalcitol ( P) on renal tubulointerstitial fibrosis and the under-lying mechanisms in diabetic nephropathy ( DN) .METHODS:DN rat model was induced by a single intraperitoneal in-jection of streptozotocin after fasting.The animals were randomly divided into 2 groups: the DN rats in paricalcitol-inter-vened group ( group P) were injected intraperitoneally with paricalcitol dissolved in propylene glycol after the day when the model was induced successfully at a dose of 0.4μg/kg (3 times a week);the DN rats in DN group ( group D) were given isopyknic propylene glycol.Normal control group ( group C) was also set up.The samples of blood, urine and renal tissue were collected after intervention of paricalcitol for 12 weeks.The biochemical indexes were measured.The renal tissues were used for pathologic observation and determining the expression of transforming growth factor-β1 (TGF-β1), Wnt-4,β-catenin and Klotho by immunohistochemistry and Western blotting.In addition, the correlation among the above indexes was analyzed.RESULTS:(1) Scr, BUN and 24 h urine protein increased significantly in group D compared with group C, while decreased in group P compared with group D ( P<0.05 ) .( 2 ) The area of renal tubulointerstitial fibrosis in-creased in group D compared with group C, while decreased in group P compared with group D (P<0.05).(3) The ex-pression of Klotho decreased, while the expression of TGF-β1, Wnt-4 and β-catenin increased in group D compared with group C (P<0.05).Compared with group D, the expression of Klotho increased, while the expression of TGF-β1, Wnt-4 andβ-catenin decreased in group P (P<0.05).(4) The expression of Klotho was negatively correlated with the fibrosis area, TGF-β1, Wnt-4 andβ-catenin (P<0.05).CONCLUSION:Paricalcitol inhibits renal tubulointerstitial fibrosis in DN by promoting the expression of renal Klotho, and inhibiting Wnt/β-catenin signaling pathway activation and TGF-β1 synthesis.
4.Antibacterial Effect of Foot-ulcer-cure Ointment on Ulceration of Diabetic Rat:An Experimental Study

OBJECTIVE To observe the antibacterial effect of the foot-ulcer-cure ointment a compound traditional chinese materia medica(TCMM) preparation manufatured on ulceration of(diabetic) rat,in order to give the basis for clinical treatment.METHODS The total(amount) of bacteria and amount of G~-bacteria and the quota of wound healing after the ointment given to the ulceration surface of(diabetic) rats were observed.RESULTS At the third and seventh day,the total amount of(bacteria) and amount of G~-bacteria of the experiment group were remarkably less than the first day and the control group;the speed rate of the wound healing was faster than the control one.CONCLUSIONS The foot-ulcer-care ointment of TCMM has excellent antibacterial functions to diabetic ulceration.It is worth for popularization.
5.Leptin receptor of the hind brain nuclei is involved in the conditioned taste preference of rats.

OBJECTIVEConditioned taste preference (CTP) is a taste learning reflex by which an animal learns to prefer a substance which tastes not well and has been studied with much interest in recent years. However, the neural substrates of CTP are less known. This study aimed to determine the possible neural path- ways of CTP and whether serum leptin level and the leptin receptor (OB-Rb) in the hind brain are involved following CTP formation.
METHODSWe established CTP of quinine in rats with a 2-bottle preference test. The serum leptin concentrations were detected, the expression of c-fos in the rat brain was tested to determine the nuclei in relation with establishment of CTR Finally, the OB-Rb mRNA expression was examined by RT-qPCR assay in parabrachial nucleus (PBN) and the nucleus of the solitary tract (NST) of the hind brain.
RESULTSCompared with control group, the level of serum leptin was higher in the CTP group (4.58 ± 0.52 vs 1.67 ± 0.25 µg/L, P < 0.01); increased c-fos positive cells were found in the anterior hypothalamus (AH, 221.75 ± 4.96 vs. 178.50 ± 6.63 cells/mm², P < 0.05), the basal lateral amygdala (BLA, 70.75 ± 6.17 vs 56.50 ± 3.62 cells/ mm², P < 0.05) and the nucleus of the solitary tract (NST, 41.25 ± 1.32 vs 32.50 ± 1.02 cells/mm², P < 0.05). But in ventromedial nucleus of the hypothalamus (VMH, 20.75 ± 2.73 vs 38.5 ± 1.54 per 1 mm², P < 005), PBN (21.50 ± 2.24 vs 36.25 ± 1.49 cells/mm², P < 0.05) and the central nucleus of the amygdala (CeA, 22.25 ± 1.53 vs 35.50 ± 2.11 cells/mm², P < 0.05), the number of c-fos positive cells was decreased in the CTP group. In addition, we found OB-Rb mRNA expression in PBN of CTP group rats was higher than that of control group (0.95 ± 0.055 vs 0.57 ± 0.034, P < 0.05), while there was no significant difference of OB-Rb mRNA expression in NST between the two groups.
CONCLUSIONNuclei AH, BLA, NST, VMH, PBN and CeA participate in the formation of CTP. Leptin and its receptor in PBN may be involved in the formation and maintenance of CTP.

Objective To investigate the effect of triptolide on the proliferation, apoptosis and and H3K4me3 protein expression in multiple myeloma KM3 cell.Methods The MM KM3 cell line was cultured with triptolide.Cell proliferation was detected by MTT method .Apoptosis was evaluated by Annexin -V -FITC/PI -labeled flow cytometry and confocal microscopy.The expression of H3K4me3 in KM3cells was assayed by Western blotting and confocal microscopy.Results Triptolide had obvious inhibition on proliferation of KM 3 cells and showed a dose and time dependence.Triptolide had obvious inhibition on apoptosis of KM 3 cells,and with the increase of triptolide concentration , the apoptosis ratio increased gradually. The the total apoptosis rates were (48.97 ±1.78 )%, ( 53.72 ±2.21 )% and ( 60.75 ±2.43 )%.Confocal laser scanning microscopy showed H 3K4me3 accumulated in the nucleus, after 48 h intervention of 80 nmol· L-1 of triptolide, the mean value of fluorescence intensity of H3K4me3 in KM3 cells (21.96+0.34 ) was significantly lower than that in the control group ( 39.86 ±0.47 ) ( P <0.05 ) .Conclusion Triptolide inhibited cell proliferation, induced cell apoptosis, reduced the expression of H3K4me3 protein.
8.CD62p expression in platelet during the preparation course of Cryopreservated platelet-rich plasma.

In order to explore the factors that affect CD62p expression in platelet during the whole course of cryopreservated platelets preparation, CD62p expression of platelet was evaluated by flow cytometry assay. The whole course of cryopreservated platelets preparation in order included whole blood collection, centrifugation for fresh platelet-rich plasma preparation, addition of dimethyl sulfoxide, and freeze in -80 degrees C refrigerator and thaw in 38 degrees C water bath. Result showed that the CD62p expressed slowly from whole blood collection to addition of dimethyl sulfoxide, but expressed abruptly after freeze and thaw and it occupied 82 per cent of the whole expression. It was concluded that the whole blood collection, centrifugation for fresh platelet enriched plasma preparation and addition of dimethyl sulfoxide were optimized in the whole course, but the damage to platelets in the whole course of cryopreservation could not be avoided. It suggests that improved techniques are needed to reduce the damage to cryopreservated platelets.

9.Molecular and prenatal diagnosis for a Chinese pregnant woman with a novel mutation of β thalassemia.

OBJECTIVETo conduct molecular and prenatal diagnosis for a couple with β thalassemia.
METHODSBlood routine examination and hemoglobin analysis were used for screening of thalassemia. Seventeen common Chinese mutations of β thalassemia were detected for the carriers with β thalassemia using PCR/RDB. The unknown mutation of β thalassemia was identified by DNA sequencing and DHPLC analysis.
RESULTSThe husband was heterozygote of CD41/42 (-TCTT). The wife carried a mutation IVS-I-110 (G→A) of β thalassemia having not been reported in Chinese so far. The fetus was a double mutated heterozygote of IVS-I-110 (G→A) and CD41/42 (-TCTT). The pregnancy was terminated.
CONCLUSIONMutation IVS-I-110 (G→A) of β thalassemia in Chinese is of importance to the genetic counseling and prenatal diagnosis of thalassemia.

10.Clinical significance of large-scale screening of A1555G mutation of mitochondria DNA for neonates.

OBJECTIVETo explore the necessity of large-scale screening of mitochondria DNA (mtDNA) A1555G mutation for prevention of aminoglycoside antibiotic induced deafness in newborns.
METHODSOne thousand blood filter samples were collected from neonates born in July 2008 in Shenzhen. DNA was extracted with Chelex-100 Resin and amplified by PCR. The mtDNA A1555G mutation was determined by denaturing high-performance liquid chromatography(DHPLC) for PCR products. The positive frequency was calculated.
RESULTSThe mitochondrial DNA A1555G mutation was detected in 2 cases of 1000 neonates. The frequency of mutation was 0.2%.
CONCLUSIONThere is a high frequency of mtDNA A1555G mutation in neonates, the large-scale screening of mtDNAA1555G mutation in newborns might detect the individuals sensitive to aminoglycoside antibiotic, which is helpful to guide a rational medication for newborns and the maternal relatives at high-risk. Furthermore, it might be useful to prevent aminoglycoside antibiotic induced deafness.
