Objective To study the clinicalpathological characteristics,surgical treatment andprognosis of the gastrointestinal stromal tumor(GIST).Methods The clinical data of 160 cases of pathologically confirmed GIST were retrospectively reviewed.Clinical features,pathological characteristics andimmune markers were studied by multiple factor analysis.Results There were no intraoperative or deathsin 140 patients who underwent radical resection.Univariate analysis of survival revealed significant differences in the main tumor pathological characteristics,including mitotic figure and metastasis and Fletcher'sclassification(P <0.05).Immunohistochemical results showed that the positive rates of KIA and CD133were 97.6% and 81.3% respectively,and their influences to survival prognosis were of statistical significance(P <0.05).A total of 145(90.6%)patients finished the follow up and 53 cases of them had multiple liver metastasis.These patients received imatinib mesylate(400 mg/d)and the efficacy was significant(P <0.05).Multivariate regression analysis showed that Fletcher's classification and imatinib mesylatewere the independent risk factors affecting prognosis.Conclusion Fletcher's classification is an effectivemethod for evaluating the prognosis of GIST.Radical resection and imatinib mesylate can reach a remarkable curative effect in treating original and metastatic lesions of the GIST patients.

Objective To compare the consistency and accuracy of determination of peripheral blood lymphocyte(PBL)interleukin-2(IL-2)with ELISA and evaluation of the expression of PBL IL-2 mRNA with real time PCR(RT-PCR).Methods In 20 kidney recipients,the PBL IL-2 level was determined with ELISA and the mRNA expression in PBL IL-2 was evaluated by real time fluorescence quantitative PCR(RT-FQ-PCR)before operation and C0/C2 time on the 7th day after operation.The same detection and evaluation were also done in 15 chronic allograft nephropathy(CAN)patients and 5 non CAN patients.Results IL-2 and IL-2 mRNA expression showed positive correlation before operation and 1 year later after operation,R2=0.58 and 0.4 respectively,both P0.05.On the 7th day after operation,IL-2 and the inhibition rate of IL-2 mRNA expression were(16.31?9.59)% and(69.92?7.10)%,respectively.A positive correlation existed between C2 IL-2 concentration and the inhibition rate of IL-2 mRNA expression,R2=0.56,P0.05.Also,IL-2 inhibition and PBL IL-2 mRNA inhibition showed no linear correlation.Conclusions The technique of determining PBL IL-2 by ELISA can be used for evaluating immunity status and immunosuppressive tolerance in patients with renal transplantation before the operation and at stable stage after operation.However,it should not be used for assessing perioperative immunity status and evaluating effect of cyclosporine.

Objective To study the effect of poly(ADP-ribose)polymerase(PARP)inhibitor 5-AIQ on the adhesion,migration and invasion of mouse colon adenocarcinoma cell line(CT26).Methods The expression of PARP after 5-AIQ treatment was detected Western blot.The cell adhesion,migration and invasion of CT26 after 5-AIQ treatment were observed by cell-matrix adhesion assay,cell migration assay and matrigel invasion assay.Results The expression of PARP in 5-AIQ-treated CT26 was weaker than that in 5-AIQ-untreated cells(P

BACKGROUND: Several studies have confirmed that activation of intervertebral disc enzymes is closely related to matrix degradation. Matrix metalloproteinase and tissue inhibitor of metalloproteinase have been shown to exert important roles in the process of extracellular matrix degeneration in intervertebrel disk. Besides these two enzyme systems, whether other proteeses that exhibit degrading effects on extracellular matrix are involved in the intervertebral disk degeneration remains poorly understood.OBJECTIVE: To detect the cathepsin K expression in normal and degenerated human intervertebrel disc cells and investigate the correlation between cathepsin K and intervertebral disc degeneration.METHODS: Cathepsin K expression was detected in intervertebral disc tissue from 30 patients with lumbar intervertebral disc protrusion using immunohistochemistry SP method and ELISA. At the same time, the intervertebral disc tissue from 15 healthy adult cadavers and/or spine fracture patients was taken as control, Cathepsin K protein expression in normal and degenerated human intervertebral disc tissues were compared.RESULTS AND CONCLUSION: Cathepsin K expression was observed in normal and degenerated intervertebral disc tissues.The expression level was significantly higher in degenerated tissue than normal tissue (P ＜ 0.05). These findings demonstrate that Cathepsin K possibly participates in the intervertebral disc degeneration.

Objective To develop an uncertainty study model of the enzymological reference method to systematically research the uncertainty influencing factors of 6 enzymological reference methods ALT,AST,LDH,AMY,GGT and CK for determining the key factors influencing the reference method and better guiding the establishment and operation of the enzymological reference methods.Methods The uncertainty of the six enzymological reference methods was evaluated by the theoretical evaluation com-bined with the experiment design according to series of standards including JJF1059-1999 Evaluation and Expression of Uncertainty in Measurement,JJF1135-2005 Evaluation of Uncertainty in Chemical Analytical Measurement,CANA-GL06 Guidance on Evalua-ting the Uncertainty in Chemical Analysis and CNAS-CL33 Guidance on the Application of Testing and Calibration Laboratories Competence Accreditation Criteria in the Field of Clinical Enzymology Reference Measurement.Results The established enzymo-logical uncertainty study model was C=ΔAΔt · 1ε · 1L ·V 1 +V 2 +V SV ·fT ·f pH ·fλ·fkit ·fresult .Conclusion The uncertainty e-S valuation model is set up and successfully applied in the uncertainty evaluation of the six enzymological reference methods.

Objective To investigate the diagnostic and therapeutic characteristics of prostate cancer with prostate specific antigen(PSA)4-10?g/L. Methods The data of prostate biopsies for the patients with PSA 4-10?g/L from May 1998 to February 2004 and the treatment and prognosis of these patients were retrospectively analyzed. Results In the 141 cases, 34 were diagnosed as prostate cancer, including 3 cT1, 21 cT2, 6 cT3 and 4 cT4. Mean Gleason score was 5.8, and the mean Gleason scores of cT1 and cT2 were significantly lower than those of cT3 and cT4. Twenty-four cases of cT1 and cT2 underwent radical prostatectomy. Four cases of cT3 recieved radical prostateetomy after 4 months of neoadjuvant endocrine therapy. Two cases of cT3 and 4 cT4 received castration. Pathological section from radical surgeries showed that 21 cases were organ confined and 7 were locally invasive. The difference of mean Gleason score between these two groups was significant in statistics. After 18 to 69 months (mean 42 months)follow-up, 21 organ confined cases were free survived, 4 locally invasive cases had biochemical recurrence and 1 case had multifocal bone metastasis. No cancer-specific death happened. Two of 4 cT4 cases were still alive and 2 died of the tumor. Conclusions Routine needle biopsy is necessary for the cases with“grave area”PSA of 4～10 ?g/L. Prostate cancers with PSA 4～10/?g/L are not always early diseases. Gleason score is a very important index for determine the stage of pathology after surgery. Radical prostatectomy is an effective treatment for organ confined disease.
