2.Losartan improves human islet ?-cell function damaged by angiotensin Ⅱ

Objective This study aimed to investigate the effects of angiotensin Ⅱ and Losartan pretreatment on regulating insulin secretion in human islet ? cells.Methods We measured changes in intracellular calcium by confocal laser scanning microscopy using Flou3-AM-loaded human islet cells.RT-PCR was used to measure changes in intracellular CaM.Dynamic insulin secretory responses were determined by chemiluminescence following perfusion of human islets.Results Exposure of the isolated islets to angiotensin Ⅱ induced glucose-stimulated insulin release coupled with intracellular calcium ascending in first phase and descending in second phase.Intracellular CaM concentration could not be affected by angiotensin Ⅱ.Conclusion The change of free Ca2+is induced by the combination of AngⅡ with ATI receptors of islet B cells,which results in the damage to islet B cells.Losartan pretreatment protects the islet B-cell function by inhibiting calcium overload.
3.Medial and lateral plating treatment for Schatzker type V tibial plateau fractures with dual incisions

0.05).The clinical outcomes were evaluated according to improvement HSS knee joint function scoring,and the score showed an average of 87.5 points (range,71-97).[Conclusion]The dual-incision technique resulted in accurate fracture reduction with low tissue complication rate,and the medial and lateral plating treatment achieved improved stabilization which enabled mobilization of the knee joint to start immediately after surgery.Medial and lateral plating treatment with dual incisions is a safe and effective technique for Schatzker type Ⅴ tibial plateau fractures.
4.Mechanism of CCCP-induced non-canonical autophagy

Objective To identify the factors those regulate CCCP-induced non-canonical autophagy.Methods Different cells expressing GFP-LC3 were treated with or without CCCP (30μM) for 6 h.Fluorescent images were taken and cell lysates were analyzed by western blot assay.Real-time PCR was used to measure the mRNA levels of LC3B.FIP200-KO MEF cells were cultured and treated by 30 μM CCCP with or without water channel inhibitors, for 6 h.Cell lysates were analyzed by Western blot assay.Results CCCP could not induced autophagy in Atg5-KO MEF cells.CCCP could induce non-canonical autophagy in ULK1-KO MEF, FIP200-KO MEF, and Beclin1-KD U251.CCCP treatment in FIP200-KO MEF cells had no effect on the expression level of LC3B mRNA.We also found two distinct aquaporin water channel inhibitors could inhibit the generation of LC3 which was induced by CCCP.Conclusion CCCP induced non-canonical autophagy was Atg5-dependent, but Beclin1-, ULK1-and FIP200-independent.Osmotic imbalance could regulate CCCP-induce non-canonical autophagy.
5.Retrospective study of the effect of C4d deposition in peritubular capillary in chronic allograft nephropathy on the prognosis of renal allografts

Objective To investigate the effect of C4d deposition in peritubular capillary (PTC)in chronic allograft nephropathy (CAN) on prognosis and intervention of renal transplantation recipients. Methods All the cases who received the renal graft biopsy due to diagnosis of CAN from January 2000 to August 2008, and had the 2-year follow-up data were included in the study. The clinical data were analyzed according to the C4d deposition in PTC. Results Among 86 cases 39 cases were C4d positive (C4d+ group) and the remaining 47 cases were negative (C4d group). There was no significant difference in sex, age, donor source, transplant times, time after biopsy, the panel reactive antibodies (PRA) level between two groups (P>0. 05). Before intervention, there was no significant difference in serum creatinine (Scr) and 24 h urinary protein between two groups (P>0. 05). At the end of 2-year followed-up period, graft loss rate and urinary protein levels in C4d+group were significantly higher than in C4d- group (P<0. 05). Before intervention, the incidence of blood lipid disorder and hypertension was higher in C4d- group (P < 0. 05 ), but no significant difference was found in uric acid and blood sugar levels (P>0. 05). At the end of 2-year followed-up period, there was no significant difference in blood glucose, uric acid, blood pressure and lipid profile (eliminating renal lost cases) between two groups (P>0. 05). Conclusion The patients with CAN and C4d+ means the involvement of chronic humoral rejection and have poor clinical results. Effective intervention against humoral immune response can improve renal allograft survival.
6.Expression and clinical significance of cathepsin K in intervertebral disk degeneration in humans

BACKGROUND: Several studies have confirmed that activation of intervertebral disc enzymes is closely related to matrix degradation. Matrix metalloproteinase and tissue inhibitor of metalloproteinase have been shown to exert important roles in the process of extracellular matrix degeneration in intervertebrel disk. Besides these two enzyme systems, whether other proteeses that exhibit degrading effects on extracellular matrix are involved in the intervertebral disk degeneration remains poorly understood.OBJECTIVE: To detect the cathepsin K expression in normal and degenerated human intervertebrel disc cells and investigate the correlation between cathepsin K and intervertebral disc degeneration.METHODS: Cathepsin K expression was detected in intervertebral disc tissue from 30 patients with lumbar intervertebral disc protrusion using immunohistochemistry SP method and ELISA. At the same time, the intervertebral disc tissue from 15 healthy adult cadavers and/or spine fracture patients was taken as control, Cathepsin K protein expression in normal and degenerated human intervertebral disc tissues were compared.RESULTS AND CONCLUSION: Cathepsin K expression was observed in normal and degenerated intervertebral disc tissues.The expression level was significantly higher in degenerated tissue than normal tissue (P < 0.05). These findings demonstrate that Cathepsin K possibly participates in the intervertebral disc degeneration.
7.Modified rat model of subarachnoid hemorrhage

Aim To develop a modified model of subarachnoid hemorrhage in rats. Motheds The SAH model was induced by injecting autologous blood into the cisterna magna without exposing the occipitoaxial ligament.The main modified point was a hole bored by a dental drill at the middle point of the boundary between the parietal bone and the occipital bone,then a catheter was inserted 8 mm to cisterna magna and the autologous blood was injected . Hemorrheological parameters were measured after the model was created.The intrinsic oversxidase method was used to show the microvessel of CA_1 region. The diameter, density ,and the aera of microvessel were measured and ananlyzed by Image-Pro Plus image analysis system. Results The total blood viscosity(?b) and erythroagglutation index(AL) in SAH model group increased significantly,however,the microvesse area and microvesse density in SAH model group decreased obviously. Conclusion The modified model of subarachnoid hemorrhage in rats is reliable and easy to operate on.
8.Effects of ginsenoside Rg1 on free amino acids in brain of sports fatigue-mice

Objective: To study the effect and possible mechanism of Ginsenoside Rg1 in sports CNS fatigue and dose-response relationship.Methods: Kun Ming male mice were divided randomly into 6 groups: control group and five different doses groups(5,10,20,40,80 mg/kg).They were treated orally with Ginsenoside Rg1 for 2 weeks.The levels of amino acids of brain tissue were tested after swimming for 90 min and the exhaustive swimming time were recorded.Results: The levels of BCAA and the ratio of BCAA/AAA in groups 5,10 and 20 mg/kg were significantly higher than control group and the levels of AAA in groups 5,10,20 and 40 mg/kg were significantly decreased.The exhaustive swimming time in groups 5,10 and 20 mg/kg were much longer than in control group.Conclusion: Through adjusting the ratio of BCAA and AAA and decreasing the level of excitatory amino acids neurotransmitter,Ginsenoside Rg1 can elevate movement ability and resist against CNS fatigue.The roles of mid-low and low doses may be better.
9.Determination of astragaloside Ⅳ in different medicinal parts of Astragalus membranaceus (Fisch.) Bge.var.mongholicus (Bge.) Hsiao

Objective To determine and compare the contents of astragaloside Ⅳ in different medicinal parts of Astragalus membranaceus var. mongholicus.Methods The contents of astragaloside Ⅳ in fibrous roots, root heads, taproots and whole roots of A.membranaceus var.mongholicus were determined by HPLC.Results The order of contents of astragaloside Ⅳ in A.membranaceus var.mongholicus was fibrous roots >whole roots >taproots >root heads;The content of astragaloside Ⅳ in A.membranaceus var.mongholicus from Inner Mongolia was 1.85 to 2.7 times of the standard which prescribed in 2010 edition of Chinese Pharmacopoeia.Conclusion Fibrous roots, which were discarded in traditional processing methods, can be used as raw material to extract astragaloside Ⅳ.This study may provide a reference for the harvest and produce of A.membranaceus var.mongholicus.
10.Indications for early surgical treatment of thoracolumbar burst fracture

Objective To observe the indications for early surgical trea tment of thoracolumbar burst fracture (TLBF). Methods 139 cases who had been def initely diagnosed as TLBF were analyzed retrospectively. They were divided into 2 groups: operation and non-operation. 47 patients had non-surgical treatment, such as bed-rest plaster brace. 92 patients had surgical treatment with plate and root screw. Spinal cord functions were evaluated according to the ASIA metho d. Results All patients were followed up for 6 months to 10 years (averaged 6 y ears and 2 months). Their average kyphosis angle measured with Cobb method was 2 3.5? before the operation and 8.5? after the operation. The average degree o f middle column posterior convexity was 25%~85% before the operation (averag ed 33%) and 0~53% after the operation (averaged 12%). The average height of anterior column was 45% before the operation and 85% after the operation. T he average degree of spinal stenosis was 30%~85% before the operation (avera ged 55%) and 0-50% after the operation (averaged 20%).
