1.Suppression Effect of Di-n-butyl Phthalate in vitro on Superoxide Dismutase of Various Organs of Mice

Objective The present study was conducted to study the oxidation damage effects of DBP on organs of mice and the molecular mechanisms of the effects. Methods The cells of the organs (liver,kidney,testis,lung) were exposed to different concentration of DBP (0?mol/L,5?mol/L,20?mol/L,80?mol/L) for one hour in vitro,and then the superoxide dismutase (SOD) activities of those cells of different organs were measured. Results With statistical significance,the SOD activities in liver,kidney,testis,lung cells were increased gradually with the increase of DBP concentrations. Conclusion The results suggested that DBP could induce the oxidative damage in liver,lung,kidney and testis cells of mice.
2.Study on relieving effects of exogenous SNP, Spd on Belamcanda chinensis under salt-alkalline stress.

The study is aimed to provide the theoretical basis for exploiting and utilization of salt-alkaline soil and cultivating Belamcanda chinensis. In this study, we exerted exogenous substances SNP, Spd to relieve the damage of the mixing salt-alkaline stress on B. chinensis seedling which is NaCl, Na2SO4, NaHCO3 and Na2CO3 four kinds of salt molar ratio of 9: 1: 9: 1, salt concentration of 100 mmol x L(-1). The result illustrated that high pH stress is a major factor caused the salt-alkaline stress, the interaction between time and the concentration of each, treatment was observed, what is more, there are synergies between the salt and alkali stress. The content of B. chinensis seedling leaves' membrane peroxidation index (MDA, O2-*) and metabolites (soluble protein, soluble sugars, organic acids) are showing an upward trend in varying degrees under 100 mmol x L(-1) salt-alkaline stress. It is effective to reduce the content of MDA and O2-*. and improve the levels of metabolites, in which the SNP (0.05 mmol x L(-1)) and Spd (0.5 mmol x L(-1)) to alleviate damage effects is the best. Therefore we can hold the conclusion that SNP and Spd can effectively mitigate the damage of B. chinensis seedling on salt-alkaline stress, improve the resistance ability of B. chinensis seedling which can provide the scientific basis for the utilization of salt-alkaline soil, and the cultivation of B. chinensis.

3.Effects of angiotensin Ⅱ on NF-κB binding activity in alveolar macrophage

Objective To determine the effects of angiotensin Ⅱ (Ang Ⅱ) on NF-κB DNA binding activity in alveolar macrophage. Method Human alveolar macrophages were isolated and made homogeneous from alveo-lar lavage fluid, and cuhtured in DMEM. Alvcolar macrophages were treated with AugⅡ (10-6M) for 15 min, 30 min, 60 min and 120 min, respectively. Moreover, alveolar macmphages were pretreated with irbesartan (AngⅡ type 1 receptor blocker) for Ⅰ hour before stimulated with Angiotensin Ⅱ for Ⅰ hour. Electrophoretic gel mobility shift assay (EMSA) was used to detect NF-κB DNA binding activity. The protein expression of IκBα was examined by Western blot. Expressions of TNF-α and ICAM-1 mRNA were detected by using RT-PCR. Results EMSA re-vealed that there was a increase in up-regulation of NF-κB DNA binding activity after alveolar macrophages were treated with Ang Ⅱ for 15 rain and peaked at 60 min. Irbesartan treatment reduced DNA binding activity. Com-pared with control group, the protein expression of IκBα decreased in Ang Ⅱ treatment group(0.29±0.11, P= 0.013), and Irbesartan treatment significantly increased protein expression of IκBα(0.83±0.12, P=0.001). The expressions of TNF-α and ICAM-1 mRNA were up-regulated by AngⅡ in comparison with the control group (TNF-α:1.13±0.17 vs. 0.42±0.099; ICAM-1 0.55±0.08 vs. 0.16±0.050, P=0.003). Irbesartan inhibited the expressions of TNF-α (0.77±0.15 vs 1.13±0.17, P=0.02; ICAM-1(0.32±0.07 vs 0.55±0.08, P =0.001). Conclusions Ang Ⅱ is capable to stimulate NF-κB signal pathway in alveolar macrophages.

Objective To study the effect of controlled ovarian Hyperstimulation(COH)on expression of AQP3 mRNA in mouse oocytes at metaphase Ⅱ. Methods Twenty female mice(6-7 weeks) were randomly allocated into 2 groups, mice in COH group were superovulated by intraperitoneal injection of 7.5 IU pregnant mare's serum gonadotropin(PMSG) followed by 5 IU human chorionic gonadotropin(HCG) after 46-48?h. Nothing was given to mice in control group and the estrus cycle was observed at 9?am everyday. 12-16?h following hCG injection (COH group) or at 8?am next day after the estrus (control group), mice were killed by cervical dislocation. The oviducts were excised.Cumulus masses were recovered from the dilated ampullae under a dissecting microscope,digested granulosa cells using hyaluronidase. Semi-quantitative real-time PCR of AQP3 mRNA in mouse MⅡoocytes was investigated with ?-actin as the internal control. 5.Accuracy evaluation of CyberKnife system

Objective To evalutate the accuracy of CyberKnife system and measure its relative output factor.Methods The accuracy of target localization of the system was measured with E2E test,film dosimetry,6D-skull tracking mode and fiducial tracking mode.Six kinds of detectors were used to detect the collimators of the system.Results The positioning errors were (0.531 ±0.293) and (0.482±0.297) mm respectively by 6D-skull tracking mode and fiducial tracking mode.The detectors proved the relative output factors of 5-mm collimator were 0.133 ±0.006,0.320±0.010,0.635 ±0.010,0.656±0.009,0.777± 0.013 and 0.680±0.014,and those of 35-mm collimator were 0.983±0.002,0.990±0.002,0.987±0.001,0.992±0.002,0.990± 0.010 and 0.981 ±0.010.Conclusion The total error of CyberKnife system was less than 1 mm.Large collimater has high consistencies between the output factors of the detectors,while there are significant differences between the output factors in case of the collimater with the diameter less than 15 mm.
6.Pathologically confirmed histoplasmosis: analysis of 14 cases.

OBJECTIVETo summarize the features of disease history, clinical manifestations, adjuvant examination results, diagnosis, treatments and outcome of patients with histoplasmosis.
METHODSThe clinical data of 14 patients with biopsy-confirmed histoplasmosis between 2000 and 2012 in Nanfang Hospital were analyzed retrospectively.
RESULTSThe clinical manifestations of histoplasmosis included fever, productive cough, chest pain, and abdominal pain, accompanied occasionally by neurological symptoms, lymph node enlargement or surface mass. Seven out of the 14 o patients had underlying immunosuppressive conditions, 9 had chest imaging changes, and 2 showed reduced white blood cells, red blood cells and platelets. The cases were initially diagnosis as tuberculosis, malignant tumor, or malignant lymphoma before the definite diagnosis was established pathologically. Ten patients received treatments with itraconazole, amphotericin B, fluconazole or voriconazole, and 9 of them responded favorably to the treatments.
CONCLUSIONHistoplasmosis, with a low incidence and diverse clinical manifestations, presents with no specific imaging features to easily cause misdiagnosis and missed diagnosis, and its definite diagnosis relies on pathological examination.

Objective To investigate the processing characteristics of locative prepositions in patients with Chinese aphasia,and to provide the theoretical evidence for the rehabilitation of aphasia.Methods Twenty aphasic patients caused by left-hemisphere stroke and twenty matched normal controls were studied.Using the locative prepo- sition repeating task(single words,locative preposition phrases and words in sentences),the comprehension task, filling-gap task,the visual-spatial function task and the short-term memory task,we compared the performance be- tween these two groups.Results The aphasic patients had more difficulty in repeating locative prepositions in sen- tences,in comprehension task and filling-gap task,their short term memory was impaired.Both groups did well in re- peating single words and phrases.Conclusion The processing of locative prepositions was impaired in Chinese aphasics.The repetition of locative prepositions was more difficult than that of phrases and single words.The preposi- tions were often omitted.It might be due to the impairment of their short-term memory,or it might have something to do with role they played in the syntactic structure.The latter might also impact the comprehension and filling-gap score.We should make plans before rehabilitation therapy.
8.Efficacy and safety of fixed combination of nitrendipine and atenolol in treatment for patients with mild to moderate essential hypertension and their optimal dosage matching

Objective To evaluate efficacy and safety of fixed combination of nitrendipine and atenolol in treatment for patients with mild to moderate essential hypertension and their optimal dosage matching.Methods Totally,275 patients with essential hypertension were selcted from seven hospitals in Shanghai,Nanjing and Suzhou,China and randomized into five groups with same proportional probability in a double-blind,double-dummy,parallel active-controlled,multi-center clinical trial,receiving fixed combination of nitrendipine and atenolol at three different dosage matching (nitrendipine/atenolol 5/12.5 mg,5/10 mg,5/7.5 mg for groups 1,2 and 3),and nitrendipine (10 mg for group 4) or atenolol (25 mg for group 5),respectively for eight weeks.Results Mean reduction of diastolic blood pressure (DBP)was (17±7) mm Hg,(18±9) mm Hg and (17±7) mm Hg for groupl,2 and 3,respectively from the baseline,significantly greater than that in groups 4 and 5[(13±7) mm Hg and (12±6) mm Hg,respectively].Mean reduction of systolic blood pressure (SBP) was (21 ±11)mm Hg,(24±12) mm Hg,(23±11) mm Hg,(19±13) mm Hg and (18±9) mm Hg,respectively for the five groups from the baseline,and the reduction in group 2 was significantly greater than that in group 5,with an overall efficacy of 94.4%,98.1% and 88.2% for groups 1,2 and 3,respectively,all statistically higher than that in group 5 (71.4%) with P<0.01,eight weeks after treatment.The ratio of patients with increased dose of antihypertensive agents in week 5 was lower in group 2 than that in the other four groups,with mild adverse reaction only,no obvious change in laboratory biochemical examinations,and no needs in special management.Conclusions Fixed combination of atenolol and nitrendipine with an optimal doses of 5 mg and 10 mg respepctively was effective and safe for mild and moderate hypertension with good tolerance.
9.Effects of Lycium barbarum polysaccharides on blood indexes andliver tissue morphology in rats with intrahepatic cholestasis

Objective:
To study the effects of Lycium barbarum polysaccharides ( LBP ) on blood indexes and liver tissue morphology in rats with intrahepatic cholestasis.
Methods:
Sprague-Dawley rats were randomly divided into the control group, the model group, and LBP low, medium and high dose group. The rats in the model group and LBP dose groups were given 60 mg/kg alpha-naphthylisothiocyanate ( ANIT ) by gavage every three days of the experiment, and the rats in the control group were given salad oil instead of ANIT. From the third day, the rats in each dose group were given 40, 150 and 600 mg/kg LBP, and the rats in the model group were given distilled water. After four weeks, the blood and urine indexes were measured, and the morphological changes of liver tissue were observed.
Results:
From the third day of the experiment, the activity of rats in the model group and LBP dose groups decreased, and the color of urine changed to dark yellow. There was no abnormality in the group. In the model group, the levels of serum total bilirubin, direct bilirubin, total bile acid ( TBA ), alkaline phosphatase ( ALP ), γ-glutamyltransferase(γ-GGT), cholesterol, alanine aminotransferase ( ALT ), aspartate aminotransferase ( AST ), white blood cell ( WBC ), percentage of granulocyte, urinary bilirubin, urinary bile acid, liver mass and liver to body ratio were higher than those in the control group, while red blood cell and percentage of lymphocyte were lower than those in the control group ( all P<0.05 ). Pathological changes of liver tissue were observed. The levels of serum TBA, ALP, γ-GGT, ALT, AST, WBC and liver to body ratio in LBP high dose group were lower than those in the model group ( all P<0.05 ). The infiltration of inflammatory cells, proliferation and expansion of bile duct, degeneration and necrosis of liver cells were alleviated.
Conclusions
LBP can improve the blood indexes and pathological changes of liver tissue in rats with intrahepatic cholestasis at the dosage of 600 mg/kg. Inhibition of inflammatory response and reduction of oxidative stress injury may be the mechanism for alleviating cholestatic liver injury.
10.Inhibitory effect of pigment produced by Fusarium sp JN158 on MCF-7 cell proliferation

Aim To explore new ways for developing anticancer drugs by the separation of pigment from Fu-sarium species JN158 ( Fusarium sp JN158 ) , the iden-tification of its structure, the screening of anticancer components and the study of its partial mechanism. Methods Pigment separation was done by HPLC, structural analysis by UV, IR, NMR, the screening of anticancer activity by MTT. Western blot was used to analyze the protein expression of CyclinD1, NF-κB, VEGF in tumor cells. Results The results showed that the pigment from Fusarium produced a total of six different peaks, of which peak Ⅵ was the anthocya-nins. Its molecular weight is about 382, molecular for-mula is C17 H18 O10 . According to investigation, this pig-ment was probably a new compound, which could in-hibit the proliferation of MCF-7 cells markedly ( IC50:0.011mmol·L-1 ,P<0.05;the control medicine ube-nimex IC50:10 mmol · L-1 ) in a concentration-de-pendent manner, and had no effect on human umbilical cord intravenous endotheliocyte ( HUVEC ) . The influ-ence on the gene expression of CyclinD1, NF-κB, VEGF in MCF-7 cells varied with the concentration of this compound. The Western blot results showed that VI pigment compound inhibited CyclinD1, NF-κB, VEGF gene expression (P<0.05 or 0. 01),compared with the control group. Conclusion The Ⅵ pigment compound from Fusarium sp JN158 could inhibit MCF-7 proliferation by inhibiting CyclinD1, NF-κB, VEGF gene expression. The compound may be a promising compound against breast cancer.
