1.Rehabilitation nursing on hepatolenticular degeneration patients complicated by dysphagia

Objective To explore the effect of integrated nursing on hepatolenticular degeneration patients with dysphagia. Methods A total of 87 patients with hepatolenticular degeneration complicated by dysphagia were divided into the intervention group and the control group according to patients' swallowing capacity. The intervention group performed integrated nursing measures and the control group conducted routine nursing measures. Patients' swallowing capacity was compared between two groups. Results There was a significant difference in improvement of swallowing function between two groups. Conclusions Integrated nursing measures can benefit swallowing function and improve quality of life for patients with hepatolenticular degeneration complicated by dysphagia.
2.Effect of psychological intervention on the mental status and life quality of hepatolenticular degeneration patients during the perioperative period of splenectomy

Objective To explore the effects of psychological intervention on the mental status and life quality of hepatolenticular degeneration patients during the perioperative period of splenectomy.Methods 64 cases hepatolenticular degeneration patients during the perioperative period of splenectomy were randomly divided into two groups,the intervention group and the control group,each with 32 cases.Both groups received conventional comfortable nursing,and the intervention group had psychological intervention for eight weeks in addition.Symptom checklist (SCL-90) was used to assess their changes of mental health,while the World Health Organization Quality of Life Scale (WHOQOL-100) was used to assess their quality of life before and after psychological intervention.Results Before psychological intervention,there was no statistically siguificant difference between two groups' SCL-90 scores (P > 0.05).After eight weeks of psychological intervention,the sores of somatization,force,interpersonal relationship,depression,anxiety,hostility,paranoid and psychotic factors in SCL-90 of the intervention group were respectively (1.26 ± 0.43),(1.72 ± 0.44),(1.74 ± 0.45),(1.59±0.43),(1.58±0.52),(1.66±0.74),(1.44±0.57),(1.61±0.68),(1.51±0.43),all of which were lower than those of the control group [(1.60 ± 0.49),(2.06 ± 0.50),(1.74 ± 0.45),(1.85 ±0.49),(1.90±0.55),(2.07±0.66),(1.71 ±0.57),(1.95 ±0.64),(1.82 ±0.56)],and the differences were statistically significant (t =2.950,2.888,2.466,2.256,2.386,2.339,2.060,2.484,respectively; P > 0.05).Before psychological intervention,there was also no statistically significant difference between two groups' WHOQOL-100 scores (P > 0.05).And after eight weeks of psychological intervention,the score of every dimension was higher in the intervention group than in the control group,and the differences were statistically significant (t =4.401,4.694,3.242,5.410,4.576,4.847,3.834,respectively;P <0.01).Conclusions Psychological intervention can effectively improve the mental health and quality of life of hepatolenticular degeneration patients during the perioperative period of splenectomy,and thus should be strengthened.
3.Rehabilitation nursing research of hepatolenticular degeneration patients with dysarthria

Objective To explore the efficacy and the clinical practice value of rehabilitation nursing to hepatolenticular degeneration patients with dysarthria. Methods 136 hepatolenticular degeneration patients with dysarthria in hospital were enrolled in this study, and they were randomly assigned to rehabilitation nursing group and usual nursing group. Both groups were treated formally by sodium dimercaptosulphonate, the 68 patients of usual nursing group were given general nursing, the 68 patients of rehabilitation nursing group were given general nursing and rehabilitation nursing of dysarthria. Frenchay dysarthria measurement, Phonolaryngeal graph,Microspiro, Activity of Daily Living Scale and WHOQOL-BREF were used to evaluate and compare the therapeutic effect after 8 weeks. Results The total effect rate of rehabilitation nursing group was 95.6%, and usual nursing group was 72. 1%, and the difference was significant (X2 = 13.88, P < 0. 01). MPT in rehabilitation nursing group was different before and after treatment [(6. 12 ± 5.36) s vs (8.94 ± 6.43) s, t =2. 78, P <0. 01] and FVC was also different (t =2.46, P <0.05). The MPT and FVC in the two group were different (P < 0. 01). WHOQOL-BREF results have reach the statistically significance in the two group after treatment (P < 0. 01 or P <0. 05). Conclusions Dysarthria is a common language barrier of hepatolenticular degeneration; it seriously affects the physical and mental health of patients. Early speech rehabilitation training,with the treatment of hepatolenticular degeneration patients by sodium dimercaptosulphonate, can greatly promote sound organ function recovery, can obviously improve the speech function of patients and can improve daily life ability and the life quality of them, it has high value of clinical applications.
4.Attenuation of Peripheral Regulatory T-Cell Suppression of Skin-Homing CD8+T Cells in Atopic Dermatitis.

PURPOSE: Cutaneous lymphocyte-associated antigen (CLA)-expressing CD8+T cells have been known to play an important role in the pathogenesis of atopic dermatitis (AD). However, the mechanisms underlying the loss of self-tolerance remain unclear. Regulatory T cells (Tregs) play a key role in the development of homeostasis in the immune system. We, therefore, hypothesized that a reduced ability of Tregs to inhibit autologous CD8+CLA+T cells might be underlying mechanism in AD. MATERIALS AND METHODS: CD8+CLA+T cells and Tregs were obtained from the peripheral blood of AD patients and control volunteers. The frequencies of CD8+CLA+T cells were evaluated. The proliferative responses of CD8+CLA+T cells were assessed by flow cytometry, and the levels of transforming growth factor-beta1 (TGF-beta1) and interleukin-10 (IL-10) in culture supernatants were detected by enzyme-linked immunosorbent assay. RESULTS: Our results revealed higher frequency and increased expression of perforin and granzyme-B in peripheral CD8+CLA+T cells in AD, and lower inhibitory ability of Tregs on proliferation of CD8+CLA+T cells in AD. Meanwhile, the levels of TGF-beta1 produced by Tregs were significantly lower in AD, and anti-TGF-beta1 abolished such suppression. CONCLUSION: The attenuated inhibitory ability of Tregs on hyper-activated autologous CD8+CLA+T cells, mediated by TGF-beta1, plays an important role in the pathogenesis of AD.

5.Correlation analysis between cytokines levels in serum and bronchoalveolar lavage fluid, blood T cell subsets and pneumoconiosis severity.

OBJECTIVETo analyze the correlation between the pneumoconiosis severity and the cytokines levels in serum and bronchoalveolar lavage fluid (BALF) or blood T cell subsets.
METHODSThe subjects were divided into five groups: control group (6 cases), group exposed to dusts (6 cases) and 3 pneumoconiosis groups (36 in stage I, 12 in stage II and 10 in stage III). ELISA was used to detect IL-6, sIL-2R and TNF-α levels in serum and BALF. The subsets of blood T cells were classified by flow cytometer.
RESULTSAs compared with control group and group exposed to dusts, the levels of serum IL-6 and sIL-2R in patients with II or III stages significantly increased, which were positively correlated with pneumoconiosis stages (r(1) = 0.74, r(2) = 0.81, P < 0.05). The level of serum TNF-α significantly decreased in patients with III stages, as compared with control group and group exposed to dusts. There was a negative correlation between serum TNF-α level and pneumoconiosis severity (r = -0.58, P < 0.05). There was a positive correlation between the levels of IL-6, sIL-2R and TNF-α in BALF and the levels of IL-6, sIL-2R and TNF-α in serum (r(1) = 0.77, r(2) = 0.96 and r(3) = 0.88, P < 0.05). The proportion of CD(4)(+)T cells and the ratio of CD(4)(+)/CD(8)(+) decreased dramatically in patients with II and III stages. But there was no correlation between these values and disease severity.
CONCLUSIONThe immune function in Th cell was inhibited. The levels of IL-6, sIL-2R and TNF-α in serum and BALF were associated with the severity of pneumoconiosis.

6.Analysis of human papilloma virus infection of women in early pregnancy and postpartum.

OBJECTIVETo explore the clinical value of examination of cervical HPV infection in women in early pregnancy and postpartum.
METHODSUsing flow-through hybridization and gene chip techniques, we examined 3 806 cervical specimens of pregnant and postpartum women of different ages with different cervical diseases. The women were grouped into different age groups by every 5 years for HPV DNA genotyping of the specimens, with another 4080 women without pregnancy serving as the control.
RESULTSOf the total of 7886 specimens, high-risk HPV infection was detected a the rate of 12.5%. In pregnancy, postpartum and nonpregnancy, the infection rate was 14.3%,, 10.5%, and 11.7%, respectively. In the 4 age groups, the infection rate was 16.9%, 12.1%, 13.8%, and 22.1%, respectively.
CONCLUSIONThe high-risk HPV infection rate in pregnancy differs significantly from that in nonpregnancy and postpartum. The infection rate also differs with age during pregnancy and postpartum. Examination of HPV infection during pregnancy is safe and feasible.

7.Effects of agonist and antagonist of cysteinyl leukotriene receptors on differentiation of rat glioma C6 cells.

OBJECTIVETo investigate the role of cysteinyl leukotriene (CysLT) receptors in the differentiation of rat glioma C6 cells.
METHODSRat glioma C6 cells were treated with the agonist LTD(4), the CysLT(1) receptor antagonist montelukast and the differentiation inducer forskolin. Cell morphology and GFAP protein expression were determined after treatments.
RESULTForskolin (10 μmol/L) induced morphological changes and GFAP protein expression (cell differentiation) in C6 cells, but LTD(4) (0.1-100 nmol/L) did not induce these changes. Montelukast (1 μmol/L) alone did not affect C6 cell differentiation, while it induced the differentiation when combined with the LTD(4) (100 nmol/L).
CONCLUSIONThe CysLT(2) receptor may modulate the differentiation of rat glioma C6 cells.

8.Construction of HEK293 cell lines expressing hCysLT₂ receptor and its application in screening of antagonists.

OBJECTIVETo construct HEK293 cell lines stably expressing hCysLT(2) receptor, and to evaluate its application in screening of synthetic compounds with antagonist activity.
METHODSThe recombinant plasmid pcDNA3.1(+)-hCysLT(2) was transfected into HEK293 cells using Lipofectamin 2000. The transfected HEK293 cells were selected in 96 well plates by limiting dilution with 600 μg/ml C418 for 8 weeks. The expression of human CysLT(2) receptor was detected by RT-PCR and immunofluorescence staining. In HEK293 cells stably transfected with hCysLT(2), the agonist LTD(4)-induced elevation of intracellular calcium concentration ([Ca2(+)]i) was measured as the index for screening compounds with antagonist activity.
RESULTAfter selection in 96 well plates by limiting dilution, 12 monoclones were obtained and 11 of them highly expressed hCysLT(2) receptor. The positive control ATP at 50 μmol/L and LTD(4) at 100 nmol/L elevated [Ca2(+)]i in hCysLT(2)-HEK293 cells. AP-2100984 inhibited LTD(4)-induced [Ca2(+)]i elevation, but selective CysLT(1) receptor antagonists did not exert such an effect. The newly synthesized compounds DXW2, DXW3, DXW4, DXW5, DXW9, DXW25, DXW26, DXW29 and DXW35 at 1 μmol/L significantly inhibited LTD(4)-induced [Ca2(+)]i elevation. The IC(50) values of DXW4 and DXW5 were 0.25 μmol/L and 7.5 μmol/L.
CONCLUSIONHEK293 cell lines stably expressing hCysLT(2) receptor have been successfully constructed, and can be used to screen compounds with CysLT(2) receptor antagonist activity.

Gorham-Stout disease(GSD)is a sporadic chronic disease characterized by progressive bone dissolution,absorption,and disappearance along with lymphatic vessel infiltration in bone-marrow cavities.Although the osteolytic mechanism of GSD has been widely studied,the cause of lymphatic hyperplasia in GSD is rarely investigated.In this study,by comparing the RNA expression profile of osteoclasts(OCs)with that of OC precursors(OCPs)by RNA sequencing,we identified a new factor,semaphorin 3A(Sema3A),which is an osteoprotective factor involved in the lymphatic expansion of GSD.Compared to OCPs,OCs enhanced the growth,migration,and tube formation of lymphatic endothelial cells(LECs),in which the expression of Sema3A is low compared to that in OCPs.In the presence of recombinant Sema3A,the growth,migration,and tube formation of LECs were inhibited,further confirming the inhibitory effect of Sema3A on LECs in vitro.Using an LEC-induced GSD mouse model,the effect of Sema3A was examined by injecting lentivirus-expressing Sema3A into the tibiae in vivo.We found that the overexpression of Sema3A in tibiae suppressed the expansion of LECs and alleviated bone loss,whereas the injection of lentivirus expressing Sema3A short hairpin RNA(shRNA)into the tibiae caused GSD-like phenotypes.Histological staining further demonstrated that OCs decreased and osteocalcin increased after Sema3A lentiviral treatment,compared with the control.Based on the above results,we propose that reduced Sema3A in OCs is one of the mechanisms contributing to the pathogeneses of GSD and that expressing Sema3A represents a new approach for the treatment of GSD.
10.Method for screening cysteinyl leukotriene receptor 2 antagonists and preliminary screening of compounds.

OBJECTIVETo establish a method for screening cysteinyl leukotriene receptor 2 (CysLT(2)) antagonists and to preliminarily screen a series of synthetic compounds.
METHODSRat glioma cell line (C6 cells) highly expressing CysLT(2) receptor was used. Intracellular calcium concentration was measured after stimulation with the agonist LTD(4),which was used to screen compounds with antagonist activity for CysLT(2) receptor. Bay u9773, a CysLT1/CysLT(2) receptor non-selective antagonist, and AP-100984, a CysLT(2) receptor antagonist, were used as control.
RESULTPT-PCR showed a higher expression of CysLT(2) receptor in C6 cells. LTD(4) at 1 mumol/L significantly increased intracellular calcium in C6 cells; the maximal effect was about 37.5% of ATP, a positive stimulus.LTD(4)-induced increase of intracellular calcium was blocked by CysLT(2) receptor antagonists, but not by CysLT(1) receptor antagonists. Among the synthetic compounds, D(XW-)1,2,13,23,29 and 30 inhibited LTD(4)-induced increase of intracellular calcium.
CONCLUSIONLTD(4)-induced change in intracellular calcium in C6 cells can be used as a screening method for CysLT(2) receptor antagonists. The compounds, D(XW-)1,2,13,23,29 and 30, possess antagonist activity for CysLT(2) receptor.
