1.Study on effect of Cassia Seed Budding transforming Organic Vanadium

Objective To Explore the effect of Cassia Seed budding transforming organic vanadium. Methods Separate organic vanadium from inorganic vanadium in Cassia Seed sprouts by dislysis bag. The content of total vanadium and inorganic vanadium were determined by using the method of Graphite Atomic Absorption Spectrophotometry. The transformation efficiency of organic vanadium in the Cassia Seed spouts was calculated. Results The seed coat of Cassia Seed could accumulate massive inorganic vanadium existing in environment during the process of its natural growing period,but the transformation efficiency of organic vanadium mainly occurred inside seed coat of Cassia Seed. Condusion Cassia Seed budding is one pathway of the biologic organification of vanadium.
3.Protective effect of brain-derived neurotrophic factor for rat hippocampal neurons cultured with high glucose concentrations

Objective To investigate the effect of brain-derived neurotrophic factor on protection of rat hippocampal neurons after cultured with high glucose concentrations.Methods Hippocampal neurons of neonatal SD rat were cultured in serum-free medium and divided into normal group,high glucose group and high glucose BDNF group.The morphological changes of the neurons were observed under inverted fluorescence microscope.Immunofluorescence stain for Map2 was performed to identify the purity of neurons.The survival of the neuron were tested by PI/Hoechest33342 staining.Results Compared with the normal group [survival rate (96.11 ± 1.63) %],the number of PI positive neurons were significantly increased in the high glucose group [survival rate (76.29 ± 6.74) %].Treatment with BDNF [survival rate (93.47 ±2.43) %] significantly reduced the number of PI positive cells.Conclusion BDNF can protect rat hippocampal neurons from apoptosis induced by high glucose.

5.Role of ultrasonography and Ki-67 expression in evaluation of neoadjuvant chemotherapy for breast can- cer

Objective To analyze the changes of ultrasonography and Ki-67 before and after neoadjuvant chemotherapy for breast cancer ,and to assess the value of ultrasonography and Ki-67 in the evaluation of neoadju-vant chemotherapy for breast cancer. Methods The focus changes of 122 cases of breast cancer before and after neoadjuvant chemotherapy were observed by Color Doppler ultrasonography. The therapeutic effect was evaluated by RECIST standard,and the changes of Ki-67 before and after chemotherapy were observed. Results There were significant differences in size and internal blood flow signal of breast cancer before and after chemotherapy (P <0.05). The sensitivity of ultrasonography in the evaluation of neoadjuvant chemotherapy for breast cancer was 89.3%and specificity 53.8%. The clinical efficiency of neoadjuvant chemotherapy for breast cancer with high Ki-67 expression was higher than that with low Ki-67 expression. Conclusion Ultrasonography shows high clinical value in the evaluation of neoadjuvant chemotherapy for breast cancer ,and the expression of Ki-67 could predict the effi-cacy of neoadjuvant chemotherapy.

7.Quality Standard of Yanyan Granules of Concentrated Sugar-free Type

OBJECTIVE:To establish the quality standard of Yanyan granules of concentrated sugar-free type. METHODS:Scrophularia ningpoensis,Menispermum dauricum,Radix et Rhizoma Glycyrrhizae in the granules were identified by TLC,and the content of linarin in Yanyan granules were determined by HPLC.RESULTS:The TLC spots were clear with high resolution.The linear range of linarin was 4.64~69.6 ?g(r=0.999 9) and the average recovery rate was 98.65%(RSD= 0.79%,n=6). CONCLUSION:The established standard is suitable for the quality control of the Yanyan granules of concentrated sugar-free type.
8.Audiologic Evaluations of Newborns After Failure of Universal Neonatal Hearing Screening

Objective To investigate the hearing and audiologic aspect of children who failed the universal newborn hearing screening.Methods A prospective analysis of 136 consecutive records of neonates referred to Chengdu women and children health hospital due to universal neonatal hearing screening failure. The diagnostic evaluation was performed at 3 and 6 months old and the protocol consisted of a standard otologic examination, diagnostic auditory brainstem response(ABR), transient evoked otoacoustic emissions (TEOAE), and middle ear assessment.Results In the initial 3 months old diagnosis, bilateral hearing loss was confirmed in 72 infants and unilateral hearing loss in 64 infants.Among 136 cases,mild hearing loss was found in 109,moderate in 9,severe in 7,and profound in 11 infants. In 6 months old confirmed diagnosis,permanent hearing loss was confirmed in 114 children with normal hearing in 22 infants. Bilateral hearing loss was diagnosed in 52 infants and unilateral hearing loss in 62 infants.Among these 114 cases,mild hearing loss was observed in 87,moderate in 11,severe in 4,and profound in 12.Conclusion The hearing level of the infants continues to change and those referred from hearing screening are in need of an in-depth evaluation and a comprehensive follow-up program.
9.Differences on the parameters of the optic disc between the amblyopic eye and non-amblyopic eye in adolescent with anisometropic amblyopia

AIM: To compare the peripapillary retinal nerve fiber layer ( RNFL ) thickness and peripapillary topographic map's parameters between amblyopic eyes and non -amblyopic eyes in adolescent with anisometropic amblyopia by optical coherence tomography(OCT).
METHODS: Thirty - four juveniles with anisometropic amblyopia were selected. Peripapillary RNFL thickness and peripapillary topographic map were measured by frequency domain OCT with both eyes in all participants, and the differences between amblyopic eyes and non -amblyopic eyes were compared.
RESULTS: There was no significant difference in average thickness of peripapillary RNFL and in any other region of peripapillary RNFL. The disc area in amblyopic eyes was bigger than that in non - amblyopic eyes ( t =2. 8054,P= 0. 0263). The disc area in amblyopic eyes were significantly related to the thickness of nasal RNFL and the rim area(r= 0. 7592,0. 7501;P= 0. 0289,0. 0321).
CONCLUSION: There existed some difference in peripapillary structure between amblyopic eyes and non-amblyopic eyes in adolescent with anisometropic amblyopia.
10.Construction and immunoscreening of cDNA library of Armillifer agkistrodontis nymphs

Objective To construct cDNA entry library and cDNA expression library of Armillifer agkistrodontis (A.) nymphs and make a preliminary immunoscreening for the cDNA expression library.Methods The nymphs were collected from the Kunming mice infected experimentally with A.agkistrodontis eggs and the total RNA were extracted from the nymphs using TRIzol Reagent.After purifying the mRNA,the synthesized cDNAs were cloned into the donor vector pDONR222 by BP reaction of Gateway technology and the recombinants were transformed into the DH10B cells by electroporation,the cDNA entry library was obtained.Next,the expression vector pDEST17 was ligated with entry clones by LR reaction,and the recombinants were transformed into the BL21 (DE3) cells.Hence,the cDNA expression library was constructed.Then,the expression library was immunoscreened with the mixed sera of mice infected with A.agkistrodontis,and the insertions of positive clones were sequenced.After that,the open reading frame(ORF) of positive slone sequence,the homology of the screened genes and their encoded proteins were analyzed by Finder and BLAST (basic local alignment search tool) program of National Center of Biotechnology Information(NCBI),and the discovered new genes were submitted into the GenBank.Besides,the physico-chemical properties,secondary structure and B cell epitopes of encoded proteins were also analyzed by bioinformatics software.Results The average titer and total clones of the cDNA entry library were 1.45 × 105 CFU/ml(colony-forming unit,CFU) and 1.74 × 106 CFU,respectively,and the range of fragment length of the inserted cDNA was between 0.2-4.0 kb,with an average of 1.4 kb.The total clones of cDNA expression library were 1.00 × 105 CFU,and the fragment length of the inserted cDNA was between 0.3-2.2 kb,with an average of 1.0 kb.Five positive clones,coded S1,S5,A1,D1 and F1,respectively,were obtained through preliminary immunoscreening.The sequence and homology of the five positive clones were sequenced and analyzed by BLAST program.No significant similarities were found in pentastomida species,which meant that they were all novel genes of A.agkistrodontis.The gene sequences were submitted to GenBank,with the accession number from JQ180451 to JQ180455.Also,results obtained by bioinformatics software showed that the predictive encoding proteins were all potential to be valuable recombinant diagnostic antigens.Conclusions The cDNA library of A.agkistrodontis nymphs is successfully constructed,and five new genes of A.agkistrodontis are discovered.The establishment of cDNA library and the discovery of the new genes will lay a foundation for further studying the gene functions and screening the immunodiagnostic antigens.
