1.EXPRESSION AND DISTRIBUTION OF AROMATASE mRNA IN THE BRAIN OF THE MOUSE

Objective\ To exam the expression and the distribution of the aromatase mRNA in the brain of the mouse. Methods\ RNA dot\|blotting as well as in situ hybridization technique were used. Results\ (1)There were aromatase specific mRNA expression in the brain tissue during the period from E16 to P300,the highest levels of mRNA were detected at postnatal 6 days,and the lowest levels were found at adulthood.(2)The location of the aromatase mRNA was confined to neuronal(but not glial)cell bodies and their processes.(3)The mainly distribution of aromatase mRNA was detected in the regions of the cerebral cortex,thalamus,hypothalamus and limbic system.Many heavily labeled cells were found in the layer of pyramidal cells of cerebral cortex,medial preoptic area.medial septal nucleus,pyramidal layer of hippocampus,amygdaloid nuclei.cingulate cortex,piriform cortex and periamygdaloid cortex.The moderately dense signal was present in several thalamic and hypothalamic nuclei such as ventromedial nucleus,ventrolateral nucleus,laterodorsal thalamic nucleus,paraventricular nucleus,etc. Conclusion\ There was relationship between the gene expression of aromatase with the development of brain,there was good agreement between the distribution of aromatase mRNA and aromatase activity as previously reported.The high levels of aromatase mRNA in the region of hippocampus and cerebral cortex suggested that aromatase may implicate for sex dimorphism in cognition as well as learning and memory.\;
2.Present and future for associating liver partition and portal vein ligation for staged hepatectomy

Compared to portal vein embolization and traditional two-stage hepatectomy,associating liver partition and portal vein ligation for staged hepatectomy (ALPPS) can induce the proliferation of liver rapidly,concomitantly with high incidence of complications and mortality in the perioperative period.The feasibility and safety have been improved gradually as the improvement of technology and the accumulation of experience.But it is still controversial on its efficacy for malignant tumors,especially with insufficiency of medium-and long-term out-comes.The mechanism of rapid proliferation induced by ALPPS needs more studies with further steps.
3.STUDY ON GENE EXPRESSION AND REGULATION OF AROMATASE IN MALIGNANT GLIOMA CELL LINE SHG-44

Objective To explore the gene expression of aromatase and estrogen receptor (ER-?) in malignant glioma cell line SHG-44. Methods Cell culture, immunocytochemistry, in situ hybridization and RT-PCR techniques were used. Results Aromatase and estrogen receptor gene expressions were detected in SHG-44 cells.The aromatase gene in these cells was expressed by means of the multi promoters (1^3, 1^4 and P Ⅱ).Conclusion It may provide some new data for the hormone regulation in carcinoma of nerve system.
4.Effect of different dose ofsustained-release theophyline on prealbumin, C-reactive protein and white blood cell count in patients with pediatric asthma

Objective To analyse effect of different dose of sustained-release theophyline on prealbumin, C-reactive protein and white blood cell count in patients with pediatric asthma.Methods 58 children with acute asthma attack in our hospital were collected.All children were randomly divided into high dose group and low dose group,29 cases in each group.On the basis of conventional treatment, high dose group were treated with high dose theophylline sustained release tablets 5 mg/kg, one times per eight hours,orally,and low dose group was treated with low dose theophylline sustained release tablets,2 mg/kg, one times per eight hours,orally.Two groups were treatment for two weeks.After the treatment, the serum levels of pre albumin, C-reactive protein and veinal blood level of white blood cell count were detected in all children.Results Compared with high dose group post-treatment, the serum level PA was higher in low dose group (P<0.05);the serum level of CRP was lower in low dose group (P<0.05);the veinal blood level of WBC, percentage of neutrophils and percentage of eosinophils was lower in low dose group (P<0.05) .Conclusion Compared with high dose of sustained-release theophyline,the low dose of sustained-release theophyline can significantly reduce the serum CRP level and veinal blood level of WBC, percentage of neutrophils and percentage of eosinophils, improve serum PA level in patients with pediatric asthma, reduce inflammation in patients.
5.Long-Term Effect of Topiramate Treatment on 24 Cases of Lennox-Gastaut Syndrome

Objective To explore the effect and adverse reaction of topiramate(TPM) on treating Lennox-Gastaut syndrome(LGS)(including therapeutic alliance or single ). Methods Twenty-four cases with LGS whose attacks could not be controlled by re-(gular) therapy were selected.TPM was gradually increased from low dosage till its showing effect or untolerant adverse reaction.Results Two cases were excluded because of adverse reaction and increase of attacks. The remained cases were followed up from 6 months to 15 months (average: 9 months). The total effective rate was 82.6%, 11 cases accounting for 45.8% free of attack. The tonic-clonic seizure reduced more than 50% accounting for 82.2%, the full control accounting for 66.7%. The myoclonic seizure reduced more than 50% accounting for 81.8%,the full control accounting for 58.8%.The atypical absence seizure reduced more than 50% accounting for 81.8%, the full control accounting for 63.6%. The maximum effect occurred about 2-40 weeks following TPM used, the dosage about 2-10 mg/(kg?d).The adverse reaction included anorexia (8 cases), language disorder (5 cases), drowsiness (4 cases), decrease of anamnesis (3 cases), weight loss or unchanged(3 cases), inattention (3 cases), depression (3 cases), mental bradypraxia (2 cases ), skin damage (1 case), stupor (1 case), gross hematuria(1 case).The hepatic and renal function were normal during therapy. Conclusion TPM is a new, broad-spectrum, effective and safe antiepileptics drug on treating LGS.
7.Simultaneous determination of five main index components and specific chromatograms analysis in Xiaochaihu granules.

Reversed phase high performance liquid chromatography with diode array detector was employed for simultaneous determination of five main index components and specific chromatograms analysis in Xiaochaihu granules with a linear gradient elution of acetonitrile-water (containing 0.1% phosphoric acid) as mobile phase. The results showed that five main index components (baicalin, baicalein, wogonoside, wogonin, enoxolone) were separated well under the analytical condition. The linear ranges of five components were 0.518 - 16.576, 0.069 - 2.197, 0.167 - 5.333, 0.009 - 0.297 and 0.006 - 0.270 mg x g(-1), respectively. The correlation coefficients were 0.999 9, and the average recoveries ranged from 95% to 105%. Twelve common peaks were selected as the specific chromatograms of Xiaochaihu granules with baicalin as the reference peak. There were good similarities between the reference and the ten batches of samples. The similarity coefficients were no less than 0.9. The analytical method established is highly sensitive with strong specificity and it can be used efficiently in the quality control of Xiaochaihu granules.
8.Construct eukaryotic expression vector with aFGF gene and transfect muscle satellite cells

Objective To construct human acidic fibroblast growth factor (aFGF) recombinant eu karyotic expression vector and transfect it into muscle satellite cells(MSCs) of rat,in purpose of further study the method to set up cell bank.Methods The aFGF gene was cloned from human total RNA which was obtained from human skeletal muscle tissue by RT-PCR method.Human interleukin 2 (IL-2) signal peptide sequence (SPS) was obtained by direct chemosynthesis method.Then aFGF and SPS were fused to obtain SPS-aFGF.Finally,directional cloning SPS-aFGF into pEGFP-N1,the recombinant (pEGFP-N1-SPS-aFGF) was obtained.The recombinant was confirmed by endonuclease digestion and DNA sequencing.MSCs were purified by difference-speed adherence method and were ideontified by immunofluorescence assay.The correct cells were divided into 3 groups:Experimental group (aFGF +N 1),control group (N 1),blank group (blank).All the groups were transfected by Lipofectamine 2000TM Reagent,and pEGFP-N1-SPS-aFGF,pEGFP-N1 were respectively added in experimental group and control group while blank group was added none plasmid.Fluorescence microscope was employed to detect transfection efficiency tendency along with time changes.The expression of target gene was detected by fluorescent quantitation PCR and Western blot.Results (1) The sequencing of pEGFP-N1-SPS-aFGF was completely correct and the outcome of endonuclease was equal to actual ban s-ize.(2)The expression of GFP in transfected cells were observed by fluorescencemicroscope and transfection efficiency reached the peak at 72 h.(3)Real-time fluorescent quantitation PCR proved strong aFGF mRNA expression in transfected cells (the average relative expression of experimental group was 1464.95)with aFGF gene,while it was detected a little in the other groups (the average relative expression of control group was 1.016 and blank group was 1.000) (P < 0.05).Western blot also proved strong expression in Experimental group then the other two groups.Conclusion aFGF eukaryotic expression vector was successfully constructed and transfected into MSCs.This study may be expected to obtain some specific functions cells.
