2.Clinical study analysis on ulinastatin for treating systemic inflammatory response in patients with serious blood infection

Objective To investigate the clinical effect of ulinastatin treating systemic inflammatory response in the patients with serious blood infection .Methods The patients with serious blood infection in our hospital were selected and divided into the control group and observation group .The control group was treated with the conventional treatment ,while on this basis the observation group was added with ulinastatin .The body temperature ,heart rate ,respiratory rate ,WBC ,Scr ,CRP ,PCT ,TNF-α,APACHE Ⅱscore and mortality rate were compared between the two groups .Results The body temperature ,heart rate ,respiratory rate and A-PACHE Ⅱ score in the observation group were lower than those in the control group ,moreover ,the levels of WBC ,CRP ,PCT , TNF-αand Scr ,and mortality rate in the observation group were lower than those in the control group ,the differences were statisti-cally significant (P<0 .05) .Conclusion Uinastatin has good effect for treating systemic inflammatory response in the patients with serious blood infection ,which can decrease the mortality and is worth clinical promotion .
3.Comparative of dynamic enhanced CT,MRI differential diagnosis of adrenal tumors

Objective To compare the clinical value of dynamic enhanced CT,MRI in differential diagnosis of adrenal tumors.Methods 62 patients with adrenal tumor by pathologically confirmed in our hospital were select-ed.All patients underwent unenhanced and dynamic enhanced CT,MRI examination.The qualitative and adrenal tumor localization diagnostic accuracy,the adrenal adenomas and non adenoma tumor clearance rate of the two meth-ods were compared.Results The accuracy of enhanced CT positioning,qualitative examination were 96.77%, 90.57%,respectively,which were slightly higher than those of the MRI enhancement(91.94%,86.79%),but the differences were not statistically significant (χ2 =0.479,0.376,all P >0.05).53 adrenal adenomas and 9 cases non-adenomas absolute and relative clearance rate clearance rate in enhanced CT 5 min delay time,the differences were statistically significant (t =2.962,2.018,all P <0.05).53 cases adrenal adenomas and 9 cases non adenoma clear-ance of correction in the enhanced MRI delay 5min,respectively (33.41 ±20.17)%,(14.74 ±12.16)%,the differ-ence was statistically significant (t =2.684,P <0.05).Conclusion Dynamic enhanced CT,MRI are good methods for differential diagnosis of adrenal tumors,two methods have location and qualitative diagnosis correct rate.So dynamic enhanced CT,MRI are reliable means of diagnosis of adrenal tumors.

5.Development of the positive personality traits scale for the university students

Objective To develop positive personality scale for the university students (PPTSUS).Methods According to the hypothesis lexicology,interviewing,literature study and questionnaire survey were used to develop the structure and items of the PPSUS.Using cluster random sampling method,total 1100 undergraduates were tested with PPTSUS.Exploratory factor analysis and confirmatory factor analysis were used to evaluate the reliability and validity of the scale.Results Exploratory principal component analysis of the items identified that the scale had 11 factors.The reliability test,Cronbach'α coefficient,split-half and test-retest correlation coefficients were 0.931,0.900 and 0.835 respectively for the total scale.The correlations between the factors and total scores were all bigger than 0.499 (P< 0.01).And in the validate factor analysis,x 2/df was 1.91,RMSEA was 0.045,NNFI was 0.742,and CFI was 0.856.Conclusion The PPSUS has good construct validity,excellent levels of internal consistency and test-retest reliability.And it achieves the basic demand of psychometrics.
6.Protective effect of ginsenoside Rb1 on ultraviolet B-induced damage and its possible mechanisms

Objective To estimate the effect of ginsenoside Rb1 on the production and clearance of cyclobutane pyrimidine dimer (CPD) as well as on the expression of two nucleotide excision repair-associated proteins,xeroderma pigmentosum group C (XPC) and excision repair cross-complementing group 1 (ERCC1),by ultraviolet B (UVB)-irradiated murine epidermal cells and human HaCaT keratinocytes.Methods Totally,42 BALB/c mice were shaved on the back and divided into four groups: untreated group (n =6),UVB group irradiated with UVB only (n =12),low-dose and high-dose Rb1 group (both n =12) treated with Rb1 of 0.5 g/L and 2g/L (100 μl/cm2) respectively two hours before UVB irradiation.The dose of UVB in the animal experiment was 180 mJ/cm2.Half of the mice in each group were killed at 0.5 and 16 hours respectively after the irradiation,then,the back skin was resected and subjected to the determination of CPD levels in the epidermis by immunohistochemical SP method.Some cultured HaCaT cells were divided into several groups to be treated with different concentrations (5,20,50 mg/L) of Rb1 before or after different doses (15 and 30 mnJ/cm2) of UVB irradiation,and cells were collected at 0.5 and 12 hours after the irradiation.Subsequently,genomic DNA was extracted and CPD was detected by dot blot hybridization.Some HaCaT cells were cultured with or without the presence of Rb1 (50 mg/L) and irradiated with UVB (30 mJ/cm2),then,the cells were collected immediately or at 0.5,2,4 and 12 hours after the irradiation,and total protein was extracted and subjected to immunoblot analysis for the quantification of XPC and ERCC1 proteins.Results There was a high level of CPD in the epidermis of mice at 0.5 hour after the irradiation,with no significant differences between these groups (P > 0.05).The number of CPD-positive cells per high power field (× 400) in the murine epidermis at 16 hours was statistically lower in the low-and high-dose Rb1 group than in the UVB group (32.1 ± 8.5 and 14.6 ± 4.1 vs.67.3 ± 11.2,both P <0.01).The CPD level in HaCaT cells was similar between these groups at 0.5 hour after UVB irradiation,but was markedly decreased at 12 hours in Rb1-treated groups.After UVB irradiation,the protein expressions of XPC and ERCC1 decreased with time in untreated HaCaT cells but increased with time in Rb1 (50 mg/L)-treated HaCaT cells.In detail,the XPC/glyceraldehyde-3-phosphate dehydrogenase (GAPDH) protein ratio in untreated HaCaT cells was 0.68 ± 0.11 immediately after the irradiation,significantly higher than that at 0.5 hour (0.47 ± 0.09,P<0.05),2 hours (0.45 ± 0.08,P<0.05),4 hours (0.37 ± 0.06,P<0.01),and 12 hours (0.18 ± 0.03,P <0.01),and that in Rb1-treated HaCaT cells was 0.56 ± 0.07 immediately after the irradiation,compared to 0.48 ± 0.14 at 0.5 hour (P> 0.05),0.68 ± 0.15 at 2 hours (P> 0.05),0.97 ± 0.20 at 4 hours (P<0.01),and 0.79 ± 0.12 at 12 hours (P <0.05).The ERCC1/GAPDH protein ratio in untreated HaCaT cells was 0.28 ± 0.03 immediately after the irradiation,higher than that at 0.5 hour (0.25 ± 0.03,P > 0.05),2 hours (0.21 ± 0.02,P<0.05),4 hours (0.14 ± 0.02,P<0.01) and 12 hours (0.11 ± 0.01,P<0.01),and that in Rb1-treated HaCaT cells was 0.27 ± 0.04 immediately after the irradiation,compared to 0.24 ± 0.04 at 0.5 hour (P> 0.05),0.29 ± 0.05 at 2 hours (P> 0.05),0.35 ± 0.05 at 4 hours (P<0.05),0.39 ± 0.05 at 12 hours (P <0.01).Conclusions Ginsenoside Rb1 shows no obvious effect on the UVB-induced production of CPD,but markedly accelerates the clearance of CPD,which may be partly associated with the upregulation of XPC and ERCC1 protein expression.
7.Changes of metabolic syndrome related indexes before and after blood donation in blood donors

Objective To investigate the significance of blood donation in preventing metabolic syndrome (MS)by analyzing the changes of the blood pressure and the related biochemical indexes before and after blood donation in the blood donors with high risk of MS.Methods The blood specimens of 102 blood donors,aged 45 -55 years old with the systolic blood pressure of 110 -140 mmHg and the diastolic blood pressure of 75 -90 mmHg,the body mass index> 24 kg/m2 ,were selected as the subjects.The blood pressure,heart rate,iron reserves,blood glucose,blood lipids,and the other indicators were determined before blood donation. All the above indicators were tested again on 42 d after blood donation.Results The blood pressure,iron reserves,blood glucose and LDL/HDL ratio after donation showed significant changes compared with before blood donation(P <0.05 ).Conclusion The changes of the MS related blood indexes exist before and after blood donation,blood donation has a certain significance for preven-ting MS in the blood donors with high risk of MS.
8.The role of IL-23/IL-17 inflammatory axis in viral myocarditis in mice

Objective To observe the dynamic changes of interleukin-23 (IL-23)/interleukin-17 (IL-17) inflammatory axis expression in the myocardium of mice with viral myocarditis (VM) and further to explore its significance. Methods A total of 60 BALB/c mice were randomly divided into VMC group (n=50) and control group (n=10). Mice in VMC group were intraperito-neally inoculated with 0.1ml Eagle's solution containing coxsackievirus B3 (CVB3) to establish VM model and 10 mice were sacri-ficed each time at days 7, 14 and 28 after inoculation. Mice in control group were treated with 0.1ml Eagle's solution without CVB3 and all the 30 mice were killed on day 28 after inoculation. Myocardial histopathologic changes were detected with hema-toxylin and eosin (HE) staining and myocardial histopathological scores were calculated. The expression levels of myocardial IL-23/IL-17 mRNA and protein were determined by real-time quantitative PCR and immunohistochemistry, respectively. Serum IL-23 and IL-17 concentrations were examined using ELISA. Also, the relationship between serum IL-23 and IL-17 concentra-tions and myocardial histopathological scores was analyzed in VMC group. Results At days 7, 14 and 28 after inoculation, the myocardial histopathological scores, mRNA and protein expressions of myocardial IL-23/IL-17 and serum IL-23 and IL-17 con-centrations were significantly higher in VMC group than those in control group (P<0.01). Each of the above indicators reached its highest level on 7th day after inoculation (P<0.01) and then gradually decreased. There were significant differences among dif-ferent time points (P<0.05). Serum IL-23 and IL-17 concentrations showed significantly positive correlation with the myocardi-al histopathologic scores in VMC group, respectively (r=0.73 and 0.76, P<0.05). Conclusions IL-23/IL-17 inflammatory axis may play an important role in the process of VMC and is closely associated with the severity of VMC.
