1.Fluence of Glucosidorum Tripterygii Totorum on joint and serum soluble interleukin-2 receptor in patients with ankylosing spondylitis

Objective The current study was designed to find out the effect of Glucosidorum Tripterygii Totorum(GTT) on the serum level of soluble interleukin-2 receptor(sIL-2R) in patient with ankylosing spondylitis(AS).Method 29 patients with active AS were selected to take GTT(1mg per kg) three times a day for one year.After that,its curative effect was evaluated.The serum level of sIL-2R of these patients was measured by sandwich ELISA method and was compared with that of normal subjects.Result The serum level of sIL-2R in active AS patients was obviously higher than that of the non-active AS patients.(P<0.01).The total effective rate of GTT on AS was 89.6%,while clinical relief rate 27.6%,obvious effective rate 44.8%,effective rate 17.2% and non-effective rate 10.4%.The patients' serum level of sIL-2R after therapy was significantly lower than that before therapy except patients with no effect.(P<0.05).Conclusion GTT has positive curative effect on active AS patients and could cause obvious decrease of the serum level of sIL-2R.The serum level of sIL-2R can be used as an important index of activity of AS and as a guide of therapy.
3.Control study of high-frequency ultrasound and three-dimensional computed tomography in diagnosis of developmental dysplasia of the hip in infant

Objective To observe the feasibility and value of ultrasound in diagnosis of developmental dysplasia of the hip (DDH) in infant. Methods A total of 84 hips of 42 infants aged 6 to 36 months who were suspected of DDH were included in this study. All patients were examined by ultrasonography with the Graf's method, and were scaned by three-dimensional computed tomography (3D CT). The results of ultrasounogrphy were compared with that of 3D CT, and the availability of ultrasound in diagnosing DDH in infant was evaluated. Results Twenty-eight normal hips, 12 dysplasia hips, 15 subdislocation hips and 29 dislocation hips were diagnosed in the 84 hips. The accuracy of ultrasound in diagnosis of DDH was 97.62%. Conclusion High-frequency ultrasound scan is a valuable method in diagnosing the DDH in infants, and can be used as a routine diagnostic method.
4.Therapeutic Observation of Acupuncture-moxibustion for Chronic Non-bacterial Prostatitis

Objective To observe the clinical efficacy of acupuncture-moxibustion in treating chronic non-bacterial prostatitis (CNBP), and its effect on the inflammatory cytokines in prostatic fluid. Methods A hundred CNBP patients were randomized into a treatment group and a control group, 50 in each group. The control group was intervened by oral administration of Tamsulosin Hydrochloride sustained-release capsules, while the treatment group was additionally treated with acupuncture-moxibustion. After 28-day treatment, changes of IL-6, IL-8, IL-10, IFN-γ, COX-2, PGE2, TNF-α, MIP-2, and iNOS contents were observed, and the clinical efficacies were compared. Results The IL-6, IL-8, IL-10, IFN-γ, COX-2, PGE2, TNF-α, MIP-2, and iNOS contents were significantly changed after intervention in both groups (P<0.05). The contents of IL-6, IL-8, IFN-γ, COX-2, PGE2, TNF-α, MIP-2, and iNOS contents in the treatment group were significantly different from those in the control group after intervention (P<0.05). The total effective rate was 89.4% in the treatment group versus 78.3% in the control group, and the difference was statistically significant (P<0.05). Conclusions Acupuncture-moxibustion is an effective method in treating CNBP, and can improve the associated indexes of prostatic fluid.
5.Gene expression of MMP-9 in MCAO rats and the influence of of Buyanghuanwu Decoction on it

Objective: The experiment was undertaken to explore the blood-brain barrier permeability and the gene expression of matrix metalloproteinase-9 (MMP-9) in ischemic cerebra and the impact of Buyanghuanwu Decoction on them. Methods: The (middle cerebral artery occlusion, MCAO) rat model was used. At 1h, 3h, 24h, 48h, 5d after reperfusion. They were measured. Results: In treatment group, the BBB permeability and the MMP-9 gene expression decreased. Conclusion: Buyanghuanwu Decoction can protect the BBB by inhibiting the MMP-9 gene expression.
6.The Molecular Mechanism of LPS-induced CHI3L1 Expression

Treatment of chronic osteomyelitis has become a difficult problem in clinical medicine due to its extended pathological process,high occurrence of complication and recurrence of disease.The major pathogenesis mechanism is Gram-negative bacteria infection,such as staphylococci.LPS is an important substance found in the cell wall of Gram-negative bacteria and administration of LPS to skeleton relevant cells in vitro could simulate the pathological characteristics of osteomyelitis patients.The results of quantitative real-time PCR and Western blot showed that CHI3L1 was up-regulated obviously in the infected bone tissues of osteomyelitis patients and LPS-stimulated osteoblasts.Analysis of the luciferase activity of NF-?B reporter gene vector revealed that LPS could activate NF-?B.Bay11-7082,an inhibitor of NF-?B activation,suppressed the elevation of CHI3L1 expression induced by LPS.Pre-incubation of osteoblasts with anti-TNF-? antibody or silencing TNF-? receptor expression by siRNA inhibited the induction effect of LPS on CHI3L1.Inhibition of NF-?B activation also prevented up-regulation of TNF-? induced by LPS.In conclusion,LPS stimulated TNF-? expression through activating NF-?B,then TNF-? induced CHI3L1 expression.It was demonstrated for the first time that CHI3L1 expression is promoted in osteomyelitis and LPS-treated osteoblasts and investigates the molecular mechanism of LPS-induced CHI3L1 expression in osteoblasts.
7.Clinical Observation of Lung Protective Ventilation Strategy in Gynecologic Laparoscopic Surgery

Objective:By monitoring the lung compliance,oxygenation index(OI),CC16、IL-8 in plasma,observe whether the protective lung ventilation in patients undergoing gynecological laparoscopic surgery can improve the respiratoryw function and reduce lung injury.Methods:40 cases of ASA grade Ⅰ-Ⅱ were selected,and 40 patients were treated by elective laparoscopic radical resection of cervical cancer.The patients were divided into two groups according to the random number table method in 40 cases.Mechanical ventilation in general anesthesia,A group used traditional intermittent positive pressure ventilation mode,tidal volume is set to 10ml/KG (PBW);B group used lung protective ventilation strategy,tidal volume is set to 6ml/KG (PBW) + 5cmH2O PEEP + recruitment manoeuvres.For patients before pneumoperitoneum(T0),2 hours after pneumoperitoneum (T1),4 hours after pneumoperitoneum (T2)MAP,HR,CVP,RR and C were observed.In pneumoperitoneum (T0),2 hours after pneumoperitoneum(T1),4 hours after pneumoperitoneum (T2),2 hours after operation(T3),24 hours after surgery(T4) venous,IL-8,CC16 content detection.Results:Before pneumoperitoneum,two groups of patients with pulmonary compliancethe difference was statistically significant(P <0.05).Compared T1 、T2 with T0 in A group and B group,the pulmonary compliance all decreased and there existed significant deference(P<0.05).Notably,A group decreased evidently(P<0.05).T0,T1,T3,T4 time points in two groups of patients with oxygenation index,the difference was statistically significant (P < 0.05).Two groups of patients with CC16,IL-8content in T1,T2,T3,T4 time point,the difference was statistically significant (P < 0.05).Conclusions:Lung protective ventilation strategy can improve respiratory function in patients with gynecological laparoscopic surgery,and reduce the incidence of lung injury.
8.Endoplasmic reticulum stress mediates the neurotoxic effect of misfolded proteins in neurodegenerative diseases

Endoplasmic reticulum(ER)stress can be caused by disturbances in the function of the ER with the accumulation of misfolded proteins.The ER response is characterized by unfolded protein response(UPR)causing translational attenuation,induction of ER chaperones and degradation of misfolded proteins.In case of prolonged or aggravated ER stress,cellular signals leading to apoptosis are activated.ER stress has been suggested to be involved in human neurodegenerative diseases,such as Alzheimer's disease,Parkinson's disease,and amyotrophic lateral sclerosis,as well as other disorders.Here we will discuss the neurotoxic effect of ER stress in these three major neurodegenerative diseases,and highlight current knowledge in this field that may reveal novel insight into disease mechanisms and help to design better therapies for these disorders.
9.Matrix metalloproteinase-9 and inflammatory reaction

MMP-9 is one of the family of matrix metalloproteinases, which degrades extracellular matrix. MMP-9 is induced by many inflammatory factors, including IL-1?,IL-8 and TNF-?. Meanwhile, MMP-9 potentiates inflammatory factors by aminoterminal processing. The signal transduction by which inflammatory factors induce MMP-9 is also an important part in recent research. This review will give a summary in all these fields.
