1.Evaluation on Clinical Application of Three Testing Methods for Human Cytomegalovirus Infection in Pregnancy

Objective To evaluate the testing value of ELISA, N-PCR and RT-PCR in clinical practice for pregnant women with HCMV infection. Methods 5581 pregnant women were screened by ELISA. 100 cases with positive IgM (group 1), 69 positive IgM combined with positive serous DNA (group 2) and 69 with positive mRNA (group 3) were studied on maternal-fetal transmission and pregnancy outcome. Results The accordance rate of group 3 and group 2 with group 1 are 56.25% and 43.75%, respectively. The maternal-fetal transmission rate in three groups is 19.00%,40.58% and 46.15%, respectively. A significant difference existed between group 2, 3 and group 1(P0.05). The total rate of spontaneous abortion, fetal death, fetal abnormality and neonatal death are 10.00%,15.94% and 30.77% in different groups, respectively and that of group 3, 2 are 4 and 2 times as much as that of group 1, respectively (OR=4.00, P

Objective:To investigate the role of extracellular signal-regulated protein kinase(ERK) in estrogen-induced proliferation and cell cycle transformation of breast cancer cell line MCF-7 and the related mechanisms.Methods: Estrogen receptor-positive breast cancer cell line MCF-7 was used in our study.The effects of 17?-E2 on the proliferation of MCF-7 cells was investigated by MTT assay to determine the optimal concentration of 17?-E2 for the following experiment.The effect of PD98059 on 17?-E2-induced proliferation of MCF-7 cells was measured by MTT assay to determine the intermediate concentration of PD98059.The cell cycle was analyzed by flow cytometry and telomerase activity was determined by Telomerase repeat amplification protocol PCR(TRAP-PCR) silver staining.The expression of wild-type p53 and phosphorylated ERK1/2 protein was determined by Western blotting and the expression of wild-type p53 mRNA was detected by RT-PCR.Results: ERK phosphorylation inhibitor PD98059 inhibited the proliferation of MCF-7 cells treated with 17?-E2 in a time-and dose-dependent manner(P
3.Observation and Nursing of thread-burying in acupoint in treating exercise-induced fatigue

Objective To study the observation and nursing for exercise-induced fatigue by thread-burying in acupoint. Methods 41 athletes-in-training were treated by thread-burying in acu-point for 2 courses of treatment, choosing main acupointa such as Guan yuan, Shen yu, Pi yu, Ming men,Zu sanli, San yinjiao. Then the changes of athletes' training and hemoglobin were observed. Results After two courses of treatment, athletes' quality of sleep, mental condition in the morning, diet, desire to train, physical fitness, training quality and concentration improved siguificantly.Level of hemoglobin in-creased distinctly. Conclusions The treatment of thread-burying in acupoint could improve sport fa-tigue. Combination with Chinese and western integrative medicine, dialectical treatment, emotion and diet nurse, western integral appraisal procedure to observe and asepsis operation played a positive part in the recovery from sport fatigue.

OBJECTIVE:To investigate the curriculum setting of clinical pharmacy in higher institution of traditional Chinese medicine.METHODS:Questionnaire survey was applied to investigate the work of clinical pharmacy in hospital of traditional Chinese medicine in Jiangsu province.The problems of clinical pharmacy were analyzed to explore training target and curriculum system of clinical pharmacy major with characteristics of traditional Chinese medicine.RESULTS:The curriculums of clinical pharmacy major in higher institution of traditional Chinese medicine were determined preliminary.CONCLUSION:Scientific and reasonable curriculum setting lay a strong foundation for the students to engage in the work of clinical pharmacy.
5.Prevention and control of nosocomial infection from the large number of cataract surgery at Life-line Express

Objective To investigate the prevention measure of infections from the large number of cataract surgery at the Life- line Express. Methods From September to November 2013, a total of 1 069 cases (1 070 eyes) completed cataract surgery in Songyuan City of Jilin Province. A series of peri- operative prevention measures for nosocomial infection were established and implemented according to the characteristics of cataract surgery carried out in Life- line Express and local environment. Results The measures to prevent the nosocomial infection were fulfilled quite well, 1 069 patients had received cataract surgery successfully, no case occurred entophthalmia. Conclusions Effective prevention of the nosocomial infection can be achieved through establishing a sound system and measures, which can ensure the effect of surgery.
6.The effect of transitional care on the quality of life in patients with maintenance hemodialysis

Objective To evaluate the effect of transitional care on the quality of life in patients with maintenance hemodialysis (MHD).Methods 101 MHD patients with symptoms of kidney < 80 points were assigned into the interventional group (47 of 51 cases completed the study) and the control group (45 of 50 cases completed the study) by random number table method.The patients in the interventional group received transitional care for 6 months,while control group received routine care.The effects on function index,satisfaction index and clinical index were evaluated at the 3rd month and 6th month after intervention.Results 6 months after intervention,the scores of physical composite,mental composite,burden of kidney disease,symptoms/problems list,effects of kidney disease in the interventional group were 51.78±5.91,55.32±5.36,29.79±17.64,79.39±6.73,68.02±11.38,while the scores of the control group were 38.22±9.46,42.03±8.32,19.58±17.25,69.49±10.10,52.09±16.78,and the difference between the two groups was statistically significant (t=5.308-9.063,Z=-3.07,P < 0.01).The scores of self-management behavior,health locus of control in the interventional group were 71.30-±5.42,25.74±2.42,while the scores of the control group were 65.0±8.74,22.47±3.97 (t=4.132,4.759,P < 0.01).The nursing satisfaction in the interventional group scored 91.06±7.29,which was significantly higher than the control group scored 86.22±10.72 (Z=-2.45,P< 0.05).The level of serum phosphorus (PO4),Ca × PO4 products,parathormone (PTH) in the interventional group were (1.76±0.35) mmol/L,(51.02±10.04) mg2/dl2,464.56 ng/L,which were significanlty lower than (2.01±0.54) mmol/L,(57.41±17.38) mg2/dl2,625.78 ng/L in the control group (t=-2.691,-2.167,-2.000 in tum,P < 0.01 or 0.05).Conclusions Transitional care achieves good effectiveness on the quality of life in patients with MHD.
7.Effect of Bailing Capsules on the Expressing of TGF-β1 in the Peritoneal Dialysis Solution for Peritoneal Di-alysis Patients

Objective:To investigate the effects of Bailing capsules on the expression of TGF-β1 in the peritoneal dialysis solution for peritoneal dialysis patients. Methods: Totally 40 patients treated with peritoneal dialysis ( PD) were randomly divided into two groups (20 cases in the control group and 20 cases in the experimental group). All the patients were treated with PD by 1. 5% perito-neal dialysis effluent (6 000 ml everyday), and the patients in the experimental group were additionally treated with Bailing capsules (5 capsules each time, three times a day after meals) for 6 months. The adequacy of PD (including Kt/V and Ccr) of the two groups was examined after the one-month treatment. The renal function and the level of TGF-β1 in the effluent in 1, 3 and 6 month were com-pared between the two groups. Results:The adequacy of PD ( Kt/V and Ccr ) had no significant difference between the two groups (P<0. 05). The level of serum creatine was decreased significantly in both groups after the one-month treatment (P<0. 05), and no significant change was shown in 3 and 6 month (P>0. 05). In the control group, the expression of TGF-β1 in the effluent in 1, 3 and 6 month was increased gradually with significant difference (P<0. 05). In the experimental group, the expression of TGF-β1 in the effluent had no significant difference in 1, 3 and 6 month (P>0. 05), which was lower than that in the control group (P<0. 05). Conclusion:Bailing capsules can decrease the expression of TGF -β1 in the effluent for the patients treated with PD and inhibit the peritoneal fibrosis.
8.The effects of murine cytomegalovirus on the maturation, fertilization, cleavage and blastula formation of mouse oocytes in vitro.

To study the effects of mouse cytomegalovirus (MCMV) on the in vitro maturation, fertilization, cleavage and blastula formation of mouse oocytes, the immature oocytes were infected in vitro by MCMVs of different dosages (100 TCID(50), 10 TCID(50) and 1 TCID(50)). The oocytes were then observed for in vitro maturation, fertilization, cleavage and blastula formation and the ultrastructural changes after the culture with the viruses. Our results showed that no significant differences were found in IVM, IVF, cleavage and blastula formation among the groups treated with of virus of various dosages. And ultrastructural abnormality was observed in the oocytes treated by 100 TCID(50) of viruses. It is concluded that MCMV did not have any conspicuous effects on IVM, IVF, cleavage and blastula formation of murine immature oocytes.

9.The Construction of Human Cytomegalovirus pp65 Prokaryotic Expressing Vector and the Inducement of Specific Immune Response

Objective To construct human cytomegalovirus pp65 prokaryotic expressing vector and induce specific T lymphocyte immune response with recombinant pp 65 protein of human cytomegalovirus(HCMV) to observe the effect of CTL on infected HEL cells. Methods The whole cDNA of HCMV pp65 was amplified by PCR and inserted into prokaryotic expressing vector pRSET by gene engineer technique. The product was purified by affinity chromphotography and identified with Western blot after that recombinant plasmid was expressed by the induction of IPTG. With MTT technique, we observed the stimulating and proliferating effect of recombinant HCMV pp65 protein on PBMC in vitro. Cytotoxicity of PBMC on HCMV-infected HEL cells was detected. Results The pp65 prokaryotic expressing vector was successfully constructed and could express in engineering bacteria DE3. High dose of pure recombinant protein was acquired and had been identified. The rhHCMV pp65 protein can activate PBMC and cause the proliferation of it in vitro. The proliferated PBMC have the specific cytotoxicity to HEL cells infected by HCMV. Conclusions The acquired recombinant HCMV pp65 protein could induce specific T cells immune response in vitro to kill the HCMV infected HEL cells. And it is very important for the immune therapy of the HCMV infections.
10.Clinical and laboratory features of severe cases of hand, foot and mouth diseases in Wenzhou

Objective To investigate the clinical and laboratory features of severe cases of hand,foot and mouth disease (HFMD) in Wenzhou, Zhejiang Province. Methods Clinical data of 107 children with HFMD, including 97 severe and 10 critical cases treated in Children' s Hospital Affiliated to Wenzhou Medical College during January and May 2010 were retrospectively analyzed. One hundred and fifty children with mild HFMD were also selected as the controls. Clinical features and laboratory results were compared between the two groups. Results Fever, rash and infection in central nervous system were observed in all patients with severe HFMD, and symptoms on respiratory, digestive and cardiovascular systems were more serious than those of mild HFMD cases. White blood cell counts (WBC) were higher in severe group than those in controls (t = 12.72, P <0.01). Hyperglycemia (9. 2 mmol/L) and abnormal troponin (0. 3 -9. 0 ng/mL) were presented in all the critical patients. Cerebrospinal fluid WBC counts were raised in 97 severe HFMD patients (98.5 × 106/L for average) with predominance of lymphocytes. Among 107 severe patients, EV71 was positive in 70, including all 10 critical cases. Conclusion Involvement of nervous,respiratory and digestive symptoms is common in severe cases of HFMD, and EV71 is the predominant pathogen.
