1.IMMUNOHISTOCHEMICAL STUDY OF A- AND B-CELLS OF RATS' PANCREATIC ISLETS DURING EXPERIMENTAL PEPTIC ULCER

Forty rats were selected for the experiment, which were divided into three groups including experimental ulcer group, normal saline and normal control groups. The pancreatic tissue were prepared for immunohistochemical staining at 6th, 10th, 14th, and 21st day after operation. Sternberger's PAP method was used to show the glucagon cell (A cell) and insulin cell (B cell) in order to observe their changes during the experimental peptic ulcer. In normal control group, the percentage of A cells is 23.31?1.91 (X?SD) and that of B cells is 73.1?4.01. But in experimental ulcer group, the A cell percentage is raised up to 42.6?5.59 and the B cell percentage is lowered down to 57.5?4.55 on the 10th day after operation. Comparing with normal saline group and normal group their differences show statistical outstanding differences. The result may point out that both A and B cells of pancreatic islet take part in the body's natural resistance against the disease, especially A cells.
2.Prediction of neonatal hyperbilirubinemia by transcutaneous bilirubin nomograms

Objective To predict the risk of neonatal hyperbilirubinemia by transcutaneous bilirubin (TcB) nomograms and clinical risk factors.Methods Healthy term and late-preterm newborns (≥ 35 gestational weeks,and birth weight ≥ 2 000 g) born in Guizhou Maternal and Child Care Hospital between January 1,2013 and December 31,2013,were included.TcB levels were continuously recorded within 168 hours after birth.The value of hour-specific TcB nomogram combined with receiver operating characteristic (ROC)curves and Logistic regression model for predicting risk of hyperbilirubinemia was evaluated.Pearson's Chisquare test was also used for statistical analysis.Results A total of 5 250 cases were enrolled.TcB increased rapidly in the first 40 hours after birth,slowly increased between 40 to 96 hours,and reached a high level after 96 hours.Among them,the 95th percentile TcB stablized at 96 hours after birth.The 40th,75th and 95th percentile TcB peak levels were 173,217 and 248 μmol/L.Among the 5 250 neonates,there were 277 cases (5.3%) in the high-risk zone within 72 hours.The positive predictive value (PPV) was 22.02%;1 087 cases (20.7%) and 1 854 cases (35.3%) were in the medium-high risk and medium-low risk zones along with the PPV of 10.58%and 3.72%,respectively.There were 2 032 cases (38.7%) in the low-risk zone with the PPV of 1.38%.Multivariate analysis showed that the TcB high-risk zone after 72 hours was associated with gestational age,delivery mode,feeding mode and TcB level of risk zones within 72 hours.Compared to those born at ≥ 40 gestational weeks,those born at ≥ 37-<40 gestational weeks were more likely in the TcB high-risk zone after 72 hours (OR=1.80,95%CI:1.29-2.51).The likelihood was reduced by 42% among neonates born with cesarean section compared to those delivered vaginally in term of the TcB high-risk zone after 72 hours.Infants who received mixed feeding were less likely to be in the TcB high-risk zone after 72 hours when compared to breastfed infants (OR=0.51,95%CI:0.29-0.88).With the reduction of the high-risk zone level within 72 hours,the likelihood in the TcB high-risk zone after 72 hours was also decreased.ROC curve showed that the area under the curve (AUC) for predicting hyperbilirubinemia was 0.75 and its 95%CI was 0.72-0.78,with a sensitivity of 90.00% and specificity of 40.00%.The AUC of a combination of predictive results obtained by the Logistic regression model with significant variables in univariate analysis and high-risk zone after 72 hours was 0.66,and its 95%CI was 0.62-0.69.AUC estimated by Logistic regression model according to the TcB levels of risk zones within 72 hours combining with clinical risk factors was 0.79,and its 95%CI was 0.76-0.82 (P<0.01).Conclusions Hour-specific TcB nomograms of newborns in our hospital have been obtained,which facilitates the prediction and early intervention of neonatal hyperbilirubinemia.
3.Effects of pulmicort inhalation on serum sICAM-1, SP-A and IGF-1 levels in children with mycoplasma pneumonia

Objective To observe the effect of atomized inhalation on the levels of serum sICAM-1, SP-A and IGF-1 in children with mycoplasma pneumonia.Methods 108 children with mycoplasma pneumonia in our hospital from January 2015 to September 2016 were selected as the study object, and they were randomly divided into control group and observation group with 54 cases in each group, the control group were treated with conventional treatment, the observation group were treated with pulmicort respules by aerosol inhalation on the basic treatment of control group , then the clinical effective rates and signs disappearing time of different severity degree, serum sICAM-1, SP-A and IGF-1 levels before and after the treatment of two groups were compared.Results The clinical effective rates of observation group with mild, moderate and severe disease were respectively 100.00%, 100.00% and 93.33%, and they were all higher than 84.21%, 80.00% and 73.33% of control group, and the cough, fever and pulmonary rales disappearing time were respectively (4.54 ±0.66)d, (1.84 ±0.18)d and (3.76 ±0.52)d, and they were all shorter than (7.10 ±0.82)d, (3.25 ± 0.30)d and (5.88 ±0.75)d of control group, the serum sICAM-1and SP-A at first, third and fifth day after the treatment were lower than those of control group, serum IGF-1 level were higher than those of control group, the difference was statistically significant (P<0.05), the differences were all significant.Conclusion The clinical effect of pulmicort respules by aerosol inhalation is better, and it has active adjustion role for the serum sICAM-1, SP-A and IGF-1.
4.The clinical analysis of IL-6,IL-15 in peritoneal fluid and serum in patients with endometriosis

Objective To investigate the relationship between the levels of intedeukin(IL)-6, IL-15 and endometriosis (EM). Method The levels of IL-6, IL-15 in the peritoneal fluid (PF) and serum of 74 patients with EM (EM group) and 46 patients without EM (control group) were measured by double-an-tibody ELISA. Results Higher levels of IL-6 in PF and serum were observed in EM group [(1017.81±361.98) ng/L,(455.47±161.52) ng/L]than those in control group [(284.63±70.50) ng/L,(149.37± 43.09) ng/L], and there was significant difference (P<0.01). The levels of IL-6 in PF and serum in EM group with stage Ⅲ-Ⅳ [(1253.44±189.63) ng/L, (556.50±93.34) ng/L]were significantly higher than those in patients with stage Ⅰ-Ⅱ [(582.81±107.75) ng/L, (268.96±63.48) ng/L](P < 0.01). There was positive relationship between the levels of IL-6 in PF and serum (r=0.950, P=0.01). The levels of IL-15 in PF in EM group [(333.45±63.94) ng/L]were significantly higher than those in control group[(203.85± 70.52) ng/L](P<0.01). No significant difference was found in the levels of serum IL-15 between EM group and control group (P>0.05). No significant difference was observed either in the levels of IL-15 in PF and serum between patients with stsge Ⅲ-Ⅳ and stage of EM Ⅰ-Ⅱ (P>0.05). Conclusions The increase of IL-6, IL-15 in PF may contribute to the development of EM. Serum IL-6 levels are of clinical diagnostic value in patients with EM.
5.Studies on the Internal Relationship between Traditional Identification Term in Chinese Medicine and Phar-maceutical Botany

OBJECTIVE:To enrich the identification diversity of traditional Chinese medicine(TCM),and provide theoretical guidance for the quality evaluation of TCM. METHODS:According to literature references and traditional identification experienc-es,characteristics including medicinal shape,size,color and lustre,surface,texture,section,odor and other aspects were identi-fied by sense organs such as eyes,hands,nose and mouth. The vivid traditional identification term were obtained through systemat-ic summarization in order to explore the internal relationship with pharmaceutical botany. RESULTS&CONCLUSIONS:As the sim-plest identification method,traditional identification method can rapidly identify the species and quality of TCM,evaluate the quali-ty,and has great significance to solve the security issues of clinical medication and health care in daily life. There was a correlation between the traditional identification and botanical research,which could be able to provide theoretical guidance to characters identi-fication and quality evaluation of TCM.
6.Relationship of 25-hydroxy vitamin D level and severity of diabetic retinopathy in type 2 diabetes mellitus

Objective To discuss the relationship between serum 25-hydroxy vitamin D[25(OH)D] level and the severity of diabetic retinopathy in type 2 diabetes mellitus.Methods 1474 cases of type 2 diabetic patients were classified into three groups according to their severity of diabetic retinopathy, the non-diabetic retinopathy group(NDR group, n =640), the background diabetic retinopathy group(BDR group, n =494) and the proliferative diabetic retinopathy group(PDR group, n=340).640 healthy people with normal level of glycosylated hemoglobin(HbA1c) were selected as control group(NDM group, n=640).Disease duration, body mass index(BMI), systolic pressure, low density lipoprotein-cholesterol(LDL-C), HbA1c and 25(OH)D were detected and compared among groups.Results Disease duration and HbA1c were increasing significantly, and the levels were NDM group
7.The expression of IgG4 in salivary glands and the clinical analysis of 58 cases of Sj(o)gren's syndrome

Objective Mikulicz's disease (MD) was considered to be a subtype of Sj(o)gren's syndrome (SS) due to the clinical and histological similarities between them.Evidence had shown that there were differences between MD and typical SS.The purpose of this study was to investigate the correlation between MD and SS,by means of analyzing the expression of IgG4 in salivary glands and the clinical characteristics of patients who were previously considered as SS.Methods The paraffin sections of salivary glands from SS patients were stained with monoclonal antibodies to IgG4 and CD38.Patients were divided into two groups based on the pathological results.Analysis of the symptoms,the signs and the laboratory results were carried out in these patients.The difference in laboratory parameters and histopathological gradings in the two groups was analyzed.Normal and abnormal distributed data comparison was conducted using random independent samples t-test and rank sum test respectively.Two-sample rates were compared with Chi-square test.Results Based on immunohistochemistry of IgG4 distribution,the 58 patients with SS were divided into two groups:IgG4 related (9 cases) and non-IgG4 related (49 cases).Histopat-hologically,IgG4 related cases showed IgG4+ plasma cells/IgG+ plasma cells infiltration and there were more IgG4 related monoclonal antibody expressed when compared to IgG4 unrelated cases.In addition,there were also significant differences in clinical features between the two groups.IgG4 related disease was associated with male gender,higher level of plateletconnt,lymphocytes [(2.4±0.8)×109/L vs (1.4±0.7) ×109/L] count and CRP [(52±60) mg/L vs (15±17) mg/L] levels and lower titer of IgM [(1.2±0.7) g/L vs (1.8±0.8) g/L],antinuclear antibody (56% vs 87%) and anti-SSB antibodies (13% vs 54%) (P<0.05),when compared with IgG4-unrelated cases.There was no significant difference in other indicators (P>0.05).Conclusion The present study has demonstrated that some of the MD patients misdiagnosed as SS.Some of the laboratory tests such as the level of platelet and lymphocyte count,serum level of CRP,IgM,antinuclear antibody,anti-SSB antibodies,the serum levels of IgG4 and the histopathological presentations in the salivary gland are different between these two disorders.Because of good response to steroid in MD,so laboratory tests and pathological examinations for IgG4 can help to avoid misdiagnosis.
8.Therapeutic effect analysis of small bone flap craniotomy decompression of posterior cranial fossa and duraplasty for 45 patients with Chiari malformation type Ⅰ

Objective To evaluate the therapeutic effect of small bone flap craniotomy decompression of posterior cranial fossa and duraplasty in the treatment of Chiari malformation type Ⅰ.Methods The clinical data of 45 Chiari malformation type Ⅰ patients who were treated with small bone flap craniotomy decompression of posterior cranial fossa and duraplasty were retrospectively analyzed,31 cases among them with syringomyelia.Results According to Tator etc.standard,1 month after surgery,the excellent in 30 cases,good in 15 cases.Follow up from 6 months to 6 years,the excellent in 37 cases,good in 8 cases.Among 31 patients with syringomyelia,26 cases were syringomyelia subsided,5 cases were not obvious change.Conclusion The small bone flap craniotomy decompression of posterior cranial fossa and duraplasty can make the craniocervical decompression,and has obvious effect of treating syringomyelia,is safe and effective in treatment of Chiari malformation type Ⅰ.
9.Effect of ?-elemene on proliferation of human multiple myeloma cells

AIM: To investigate the effects of ?-elemene,extracted from curcuma wenyujin,on proliferation, cell cycle and apoptosis of human multiple myeloma RPMI-8226 cells. METHODS: The effect of ?-elemene on the growth of human multiple myeloma RPMI-8226 cells was studied through MTT assay,cell cycle and apoptosis was studied by combined Annexin-V protein iodide staining,The morphological changes was studied by combining Hoechst33342-PI staining. RESULTS: ?-elemene inhibited the proliferation of RPMI-8226 cells in a time-and dose- dependent manner. Compared with the control cells,the proportions of the RPMI-8226 cells in the G0 /G1 phase rose,and the proportions of the RPMI-8226 cells in the S and G2 /M phases fell decreased. RPMI-8226 cells treated with ?-elemene for 48 h induced apoptosis in a dose-dependent manner. CONCLUSION: ?-elemene is able to inhibit the proliferation of RPMI-8226 cells by arresting the cell cycle and inducing the cell apoptosis.
10.Research in skin healing and repair function and mechanism of Hibiscus rosa-sinensis Linn bud extract

Aim To test the skin healing and repairing efficacy and the mechanism of Hibiscus rosa-sinensis L bud extract by using the animal models. Methods KM mice were randomly divided into three groups:the model group, the positive control group, and the n-bu-tyl alcohol extract ( HrBN) group. Using the boils and carbuncles model, the healing condition of all the animals were observed. KM mice were kept in the SPF condition room and divided into five groups: the model group, the positive control group, and the low, middle, high dose groups. Using the full-thickness loss model, the repairing results of all the mice were ob-served. Through the antimicrobial test, the results of MIC and inhibition zone were obtained. The carbon clearance test was used to collect the blood at the time 5min and 15min, and get the liver and spleen, and the results of K andαwere obtained. Results In vivo ex-periments showed there was significant difference be-tween groups;the HrBN extract had the outstanding ef-ficacy in healing and repairing skin boils and full-thickness loss models. It had higher recovery rate than other ethanol extract, such as ethyl acetate extract and chloroform extract. In vitro experiments showed that the HrBN extract, ethyl acetate extract ( HrBE) ,AB-8 macroporous resin 30% alcohol part and 60% alcohol part had obvious antimicrobial efficacy. The carbon clearance test showed HrBN had a good effect in im-proving immune function, and it can increase the K and α. Conclusion HrBN in animal models exerts good skin healing and repairing efficacy, which might be related to its antibacterial activity and immunologic enhancement function.
