3.A clinical study of total glucosides of paeony for treatment of patients with systemic lupus erythematosus accompanied by leucopenia

Objective To investigate the therapeutic efficacy and safety of using total glucosides of paeony (TGP)for treatment of patients with systemic lupus erythematosus(SLE)accompanied by low white blood cell(WBC) count. Methods According to the revised SLE Classification Standard of American College of Rheumatology in 1982,62 cases of SLE accompanied by low WBC consistent with the standard were randomly divided into experimental group(31 cases)and control group(31 cases). The conventional therapy plus prednisone of 0.5-1.0 mg?kg-1?d-1 was applied for treatment in both groups,and in the experimental group,additionally TGP(0.6 g orally each time,three times a day)was given. The therapeutic course in both groups was 12 weeks. The changes in following parameters of two groups were noted at baseline and after treatment:WBC,complement C3,erythrocyte sedimentation rate (ESR),C-reactive protein(CRP),liver and kidney function,etc;the SLE disease activity index(SLEDAI)score, the therapeutic effects and incidence of adverse reactions were evaluated in two groups. Results After 12 weeks of treatment,WBC and C3 were significantly increased,and ESR,CRP,SLEDAI were significantly decreased in the two groups,the improvement being more significant in the experimental group〔WBC(×109/L):5.38±1.32 vs. 4.16±1.39,complement C3(g/L):1.28±0.32 vs. 0.86±0.26,ESR(mm/1 h):22.36±5.26 vs. 28.85±8.56, CRP(g/L):23.25±8.26 vs. 28.32±8.52,SLEDAI(score):2.58±1.86 vs. 4.18±2.31,P<0.05 or P<0.01〕. Compared with the control group,the total effective rate was significantly higher in experimental group〔93.55%(29/31) vs. 70.97%(22/31)〕,the average daily dosage of prednisone and its total dosage in 12 weeks were significantly less in experimental group〔prednisone dosage(mg/d):16.26±9.85 vs. 25.32±9.68,the total dosage of prednisone (mg)2 016.5±125.6 vs. 2 983.6±152.8,both P<0.01〕. The incidence of infection in experimental group was obviously lower than that in the control group(9.68% vs. 35.48%,P<0.05),there was no significantly statistical difference in incidence of adverse reactions between experimental group and control group(12.90%vs. 16.13%,P>0.05). Conclusion TGP is a safe and effective treatment for patients with SLE accompanied by low WBC,and it also can significantly reduce the use of glucocorticoid dosage and the incidence of infection in such patients.
4.The correlation between interleukin-4,15 as well as 18 and telomerase activity in myasthenia gravis

Objective:To detect the serum levels of IL-4,IL-15 and IL-18 in myasthenia gravis( MG) and investigate the correlations between telomerase activity of CD4~+ T lymphocytes and these interleukins.Methods:The serum levels of IL-4,IL-15 and IL-18 were detected with ELISA and telomerase activity was detected by polymerase chain reaction enzyme-linked immonosorbent assay(PCR-ELISA) in 30 MG patients and 30 negative controls.Results:The levels of IL-4,IL-15,IL-18 and telomerase of CD4~+ T lymphocytes in MG were significantly higher than those in negative controls (P<0.01).There were positive correlations between the serum levels of IL-4,IL-15,IL-18 and telomerase activity of CD4~+ T cell in MG patients (P<0.01).Conclusion:IL-4,IL-15,IL-18 and telomerase activity of CD4~+ T lymphocytes are involved in the pathogenesis of MG;IL-4,IL-15 and IL-18 may upregulate the telomerase activity of CD4~+ T cells.

6.Antiproliferative Effect of Curcumine on Human Prostatic Hyperplasia Cell Line BPH1

OBJECTIVE:To investigate the inhibitory effect of Curcumine on the proliferation and gene expression of human prostatic hyperplasia cell line BPH1. METHODS: MTT were used to examine the cell growth of Human prostatic hyperplasia cell line BPH1 after treated with different doses of Curcumine. The expressions of PCNA mRNA and Bcl-2 at transcription were detected by RT-PCR. RESULTS: Curcumine inhibited cell growth time or dose-dependently and down-regulated the expressions of PCNA and Bcl-2 in BPH1 cells dose-dependently within 24 h after being treated by Curcumin.CONCLUSI-ON: The antiproliferative effect of Curcumine on BPH1 might be associated to the downregulation of PCNA and Bcl-2 levels.
7.Mechanism and application of gene interference in cell signal transduction

RNA interference is a newly emerging gene block technology. Exogenous and endogenous double stranded RNA in cells can restrain specific sequence gene expression. RNA interference can block the specific gene expression efficiently, and identify the upper or lower reaches of the relationship of the different genes in the complex signaling pathway. It is a powerful tool to study signal transduction pathway. RNA interference can inhibit targeted gene expression of mRNA and protein products. It could simply, specifically, efficiently and stably reduce targeted gene expression, and it could remove the target gene without affecting other materials. RNAi technology is better than any previous methods in gene function's study and application. But, it is hard to transfect the RNA into mammalian cell lines, and mammalian cells also generate antagonism to the double stranded RNA or small interfering RNA. Thus, we are looking forward that using highly effective and safety small interfering RNA in medical care.
8.Influence of Safflower(Carthamus tinctorius)on Ca~(2+) Transmembrane Influx in Rat Aorta

Influence of Carthamus tinctorius L. has been studied on Ca2+ transmembrane influx in rat aorta when its extract was used at 5, 50, 500?g/ml. It was shown that the resting 45Ca influx in rat aorta ring was not altered by C. tinctorius but the 45Ca influx evoked by norepinephrine (1.2 x 10-6 mol/L ) and KCl solution (100mmol/L) was inhibited in a concentration-dependent manner. The results demonstrated that the receptor-operated Ca2+ channel (ROC) and potential-dependent Ca2+ channel (PDC) in cell membrane of smooth muscle could be blocked by C.tinetorus as well as by verapamil.
9.Two cases of Castleman disease in the neck.

To investigate the clinical manifestation, clinicolpathological diagnosis and treatment of Castleman disease (CD) in the neck. The data of 2 patients with CD in the neck were reviewed retrospectively, together with literature summary, in order to analyze its clinical manifestation, pathological characteristics, clinical and pathological types, imaging characteristics and treatment. Two cases presented as solitary, painless neck masses and met the criteria of localized CD, and also had histological evidence of the hyaline vascular variant. Complete surgical resection was the first choice of treatment. One patient was alive without evidence of recurrence for ten years. The another patient had masses recurred in the neck one month after operation and followed to undergo curative chemotherapy, no recurrence in one-year follow-up. CD in the neck is a rare disease. The diagnosis for the disease is mainly based on pathological examination. Surgical resection is the main treatment, if necessary, need combination with radiotherapy and chemotherapy.

10.Clinical study on auricular plaster therapy in the auxiliary treatment of insomnia caused by liver fire disturbance in a sub-healthy population

Objective:To investigate the clinical efficacy of psychotherapy combined with auricular plaster therapy in the auxiliary treatment of insomnia caused by liver fire disturbance in a sub-healthy population.Methods:Eighty sub-healthy patients with insomnia caused by liver fire disturbance who received treatment in Wenzhou Hospital of Traditional Chinese Medicine from June 1, 2017 to December 31, 2018 were included in this study. These patients were randomly assigned to receive cognitive behavioral therapy (control group, n = 40) or cognitive behavioral therapy combined with auricular plaster therapy (observation group, n = 40) for 4 weeks. Polysomnography was used to evaluate the sleep condition before and after treatment. The Pittsburgh Sleep Quality Index was used to evaluate the severity of insomnia. The World Health Organization Quality of Life Scale Brief Version (WHOQOL-BREF) was used to evaluate the quality of life of patients. The therapeutic effect was compared between the two groups. Results:After treatment, total sleep time [(421.25 ± 11.58) min vs. (456.25 ± 11.23) min], sleep efficiency [(70.32 ± 4.44)% vs. (76.58 ± 5.89)%], non-rapid eye movement stage 2 (N2) sleep [(43.25 ± 4.44)% vs.(45.62 ± 4.36)%], N3 sleep [(14.25 ± 4.52)% vs. (18.52 ± 5.55)%], rapid eye movement (R) sleep [(21.98 ± 5.21)% vs. (24.66 ± 4.87)%] in the control group were significantly shorter or lower than those in the observation group ( t = 13.723, 5.367, 9.035, 2.408, 3.772, 2.376, all P < 0.05). After treatment, sleep quality [(1.85 ± 0.54) points vs. (1.10 ± 0.42) points], sleep-onset latency [(1.52 ± 0.51) min vs. (1.08 ± 0.35) min], sleep duration [(1.30 ± 0.41) min vs. (0.75 ± 0.41) min], sleep efficiency [(1.53 ± 0.44) points vs. (1.02 ± 0.33) points], sleep disturbance [(1.20 ± 0.41) points vs. (0.78 ± 0.31) points], hypnotics [ (0.11 ± 0.02) points vs. (0.04 ± 0.01) points], daytime dysfunction [(1.65 ± 0.47) points vs. (1.03 ± 0.28) points] and total scores [(9.11 ± 1.33) points vs. (5.80 ± 1.01) points] in the control group were significantly higher than those in the observation group ( t = 6.933, 4.499, 5.999, 5.864, 5.167, 19.789, 7.167, 12 535, P < 0.05). After treatment, the scores in physical health [(13.85 ± 2.22) points vs. (17.86 ± 2.01) points], psychological domain [(15.52 ± 1.89) points vs. (16.35 ± 1.33) points], social relationships [(16.01 ± 1.58) points vs. (19.52 ± 1.85) points] and environment domain [(17.52 ± 1.90) points vs. (20.89 ± 2.22) points] in the control group were significantly lower than those in the observation group ( t = 8.468, 4.499, 9.124 and 7.294, all P < 0.05). After treatment, total effective rate in the observation group was significantly higher than that in the control group [95.0% (38/40) vs. 72.5% (29/40), χ 2 = 7.440, P < 0.05]. Conclusion:Psychotherapy combined with auricular plaster therapy can significantly increase the sleep duration and sleep efficiency and improve the quality of life of sub-healthy patients with insomnia caused by liver fire disturbance, which is worthy of clinical promotion.
