1.Preliminary study on efficacy and mechanism of Atractylodes Macrocephelae Rhizoma extracts in metabolic hyperlipidemia rats.

Hyperlipidemia is a major factor causing coronary heart disease and atherosclerosis. The high-density lipoprotein cholesterol (HDL-C) is a major indicator for measuring lipid levels. However, there is no an effective medicine that can obviously increase HDL-C at present. According to previous laboratory studies, atractylodes macrocephalae extracts could significantly increase HDL-C level. In this study, the metabolic hyperlipidemia rat model was established by feeding high-sugar and fat diets and alcohol-drinking to explore the effect and mechanism of atractylodes macrocephalae extracts on hyperlipidemia rats. According to the findingins, different doses of atractylodes macrocephalae extracts could reduce the levels of TC, TG, LDL-C, ACAT and increase the contents of LCAT, HDL-C. Particularly, the atractylodes macrocephalae extracts (100 mg · kg(-1) group showed increase in HDL-C by about 50% and significant declines in HMG-CoA reductase, TC, TG. In conclusion, Atractylodes Macrocephelae Rhizoma extracts could effectively regulate the dyslipidemia of hyperlipidemia rats, especially on HDL-C. Its mechanism may be related to reduction in cholesterol synthesis by inhibiting HMG-CoA reductase in livers and increase in lipid metabolism and transport by regulating LCAT and ACAT levels.

2.Effect of improper diets on blood viscosity in SD rats in high-salt and fat diet and alcohol abuse simulation model.

OBJECTIVETo observe the effect of composite factors, like long-term high-salt & fat diet and alcohol abuse on blood viscosity and blood pressure in rats, and compare with a model induced by high molecular dextran, in order to build a chronic hyperviscosity aminal model which is similar to human hyperviscosity in clinic and lay a foundation for efficacy evaluation on traditional Chinese medicines.
METHODMale SD rats were randomly divided into the normal group, the high molecular dextran (HMD) group and the high salt & fat and alcohol (HSFA) group. The HMD group was given normal diet and water for 23 day and then 10% HMD through tail vein for 5 days. The HSFA group was fed with high salt and high fat diets every day and alcohol for 20 h x d(-1) for 13 weeks. After the modeling, whole blood viscosity and plasma viscosity were measured in the 5th, 8th and 11th week. Blood pressure was measured in the 5d, 7h, and 10th week. Red cell count (RBC) and hematocrit (HCT) were measured in the 11th week. PAgT, Fb, ET-1, NO, PGI, TXA2 contents of the normal group and the HSFA group were measured in the 13th week, and IECa21 content was measured with flow cytometry. Result: After the modeling, the HMD group was in good conditions with glossy hairs and active behaviors. The HSFA group was depressed with withered hairs and less activities. During the 5th-11th weeks, the HMD group and the HSFA group showed higher values in high and low shear whole blood viscosity (WBV) than the normal control group. The plasma viscosity (PV) of HMD rats was significantly increased only in the 5th week, and that of HSFA rats significantly increased in the 8"' and 11th week, particularly in the 11'h week. In the 111h week, the HSFA group showed significant increases in RBC and HCT. After the modeling, the blood pressure of HMD rats showed no significant changes, but the blood pressure of HSFA rats significantly increased during 7' and 101h weeks, particularly in the 10"' week. In the 13th week, PAgT, IECa2+, Fb, ET-1 of HSFA rats significantly increased, but with decreases in NO and PGI2.
CONCLUSIONLong-term high salt & fat and alcohol diets can cause abnormal blood viscosity in rats. WBV significantly increased since the 5th week in rats, and PV increased since the 8th week. The mechanism for increasing BV may be: (1) increases in RBC, HCT, and IECa2+, (2) PAgT increase, (3) Fb content increase, or (4) TXA2/PGI2, ET-1/NO imbalance. Although the modeling time with the method is longer than that with the HMD method, the model is more stable and moderate, and could lead to abnormal increases in WBV and PV; Whereas the HMD method only induced transient increase in plasma viscosity and abnormal increase in SBP. The model is more similar to traditional Chinese medicine syndromes and pathogenesis, with higher value for studies on efficacy of traditional Chinese medicines.

4.Establishment of Abcb1 knock out rat and Abcb1 humanized rat models

Objective To knock out the Abcb1 gene of rat,and establish the Abcb1 humanized rat model based on the Abcb1 knock out rat.Methods The animal model was established using BAC and CRISPR/Cas9 technology,and was analyzed by PCR, RT-PCR and real-time PCR.Results Establishing a rat model expressing human Abcb1 stably by transfer the 153 kb BAC containing human Abcb1 promoter and cDNA into rat genome, and establishing the Abcb1 knock out rat at the same time.Establishing the Abcb1 humanized model by crossing these two strains together.The expression pattern of Abcb1 in Abcb1 humanized rat is different from the wild type rat.The Abcb1 humanized model express not only the human Abcb1 gene but also has similar expression pattern as human.Conclusions The Abcb1 knock out rat and the Abcb1 humanized rat were successfully established, and this model is close to human concerning about the drug metabolism related to Abcb1.
5.Analysis of the relationship between interleukin SNPs and breast cancer susceptibility

Objective To investigate the relationship between IL‐10 SNPs and breast cancer susceptibility .Methods Collect‐ed 156 blood samples of breast cancer patients as case group and 156 blood samples of health as control .Genotype SNPs of IL‐10‐592 ,‐1082 were analyzed by genomic DNA extraction ,PCR amplification and TOF mass spectrometry .The data was used by statis‐tical analysis .Results IL‐10‐592 C/C ,C/A and A/A site the genotype frequencies were not diversity between two groups(χ2 =3 .26 ,P>0 .05) .IL‐10‐1082 G/G、G/A、A/A site the genotype frequencies were statistically significant between two groups(χ2 =14 .07 ,P<0 .01) .In Logistic multivariate regression analysis found that :be compared with IL‐10‐592 A/A ,carrying IL‐10‐592 C/A ,C/C group respectively the risk of breast cancer were OR=1 .039(95% CI:0 .484 -2 .233) ,P=0 .922 ,OR= 1 .635(95% CI:0 .683-3 .915) ,P =0 .270 ,which had no statistical significance .In Logistic multivariate regression analysis found that :be com‐pared with IL‐10‐1082 A/A ,carrying IL‐10‐1082 G/A、G/G group respectively the risk of breast cancer were OR=0 .424(95% CI:0 .210-0 .855) ,P=0 .016 ,OR=0 .455(95% CI:0 .178 -1 .163) ,P= 0 .100 .Conclusion IL‐10‐592 may be not associated with breast cancer susceptibility .IL‐10‐1082 G/A may be a protection factor with breast cancer susceptibility .

Pharmaceutical preparations, particularly as a "secret recipe" of traditional Chinese medicine in medical institutions, are the product of China's medical and health industry, and they are also an important means of competing of different medical institutions. Although pharmaceutical preparations have advantages and characteristics than institutes for drug and pharmaceutical companies, the quality standards of pharmaceutical preparations in medical institutions has not reached the desired level over the years. As we all know, the quality of pharmaceutical preparations is important to ensure the efficacy, especially under the environment of people pay more sttention on drug safety and effectiveness and contry increase emphasis on the stste of pharmaceutical preparations. In view of this, we will improve the grade, stability, and clinical efficacy of pharmaceutical preparations by the advanced equipment, testing instruments and the process dynamic quality control technology. Finally, we hope we can provide new ideas for the quality control of pharmaceutical preparations.

7.Gene promoter methylation in glucose-6-phosphate dehydrogenase deficiency.

OBJECTIVETo investigate the features of methylation in the promoter region of glucose-6-phosphate dehydrogenase (G6PD) gene and the association between gene promoter methylation and G6PD deficiency.
METHODSFluorescent quantitative PCR was used to measure the mRNA expression of G6PD in 130 children with G6PD deficiency. Sixty-five children without G6PD deficiency served as the control group. The methylation-sensitive high-resolution melting curve analysis and bisulfite PCR sequencing were used to analyze gene promoter methylation in 22 children with G6PD deficiency and low G6PD mRNA expression. The G6PD gene promoter methylation was analyzed in 44 girls with normal G6PD mRNA expression (7 from G6PD deficiency group and 37 from control group).
RESULTSTwenty-two (16.9%) children with G6PD deficiency had relatively low mRNA expression of G6PD; among whom, 16 boys showed no methylation, and 6 girls showed partial methylation. Among the 44 girls with normal G6PD mRNA expression, 40 showed partial methylation, and 4 showed no methylation (1 case in the G6PD group and 3 cases in the control group).
CONCLUSIONSGene promoter methylation is not associated with G6PD deficiency in boys. Girls have partial methylation or no methylation in the G6PD gene, suggesting that the methylation may be related to G6PD deficiency in girls.

8.Chemical constituents from roots of Illicium majus.

Ten compounds, including seven sesquiterpenes, two phenols and one phenylpropanoid, were isolated from the roots of Illicium majus by means of silica gel, ODS, Sephadex LH-20, and preparative HPLC. On analysis of MS and NMR spectroscopic data , their structures were established as cycloparviflorolide (1), cycloparvifloralone (2), tashironin (3), tashironin A (4), anislactone A(5), anislactone B (6), pseudomajucin (7), syringaldehyde (8), methyl-4-hydroxy-3, 5-dimethoxybenzoate (9), and (E)-3-methoxy-4,5-methylenedioxycinnamic alchol (10), respectively. Compounds 1-4 and 8-10 were first isolated from this plant. In the in vitro assays, at a concentration of 1.0 x 10(-5) mol x L(-1), compounds 5 and 6 were active against LPS induced NO production in microglia with a inhibition rate of 75.31% and 53.7%, respectively.

9.Evaluation of thin prep cytology test in transbronchial needle aspiration for diagnosis of lung cancer

Objective To explore the diagnostic value of thin prep cytology test(TCT) in central lung cancer. Methods All 127 patients suspected central lung cancer were detected by transbronchial needle aspiration(TBNA). The patients were randomly assigned into two groups, the group of conventional smear,CS (62 cases) and the group of CS+TCT(65 cases). The sensitivity and specificity of two methods were compared. Results The sensitivity and specificity of CS were 75.9%and 100.0% respectively. The sensitivity and specificity of CS+TCT were 96.2% and 100.0% respectively. The sensitivity of CS+TCT was significantly higher than sensitivity of CS in diagnosis of lung cancer (P=0.000046<0.01). Conclusion Combination of CS and TCT techniques can improve the TBNA diagnostic sensitivity of lung cancer. TBNA-TCT detection methods should be applied to clinical activites.
10.Study on distribution status of Oncomelania hupensis-intermediate host of Schistosoma japonicum in Jiangxi Province I Analysis of distribution of O. hupen-sis in Poyang Lake area

Objective To grasp the distribution status of Oncomelania hupensis snails in Poyang Lake area,so as to provide the evidence for formulating and adjusting the schistosomiasis prevention and control strategy in lake areas. Methods The vec-tor grid was created and sampled randomly by 200 m × 200 m in the spatial database of grassland,and the distribution of snails was investigated in the selected grid by using the method of mechanical sampling by 50 m × 50 m. At the same time,the eleva-tion of investigation points was extracted based on the topographic map of Poyang Lake. Results Totally 949 and 210 investiga-tion points were collected from the south and north of Poyang Lake areas,accounting for 3.04%and 3.21%of all the investiga-tion points in the respective region. The number of investigation points,the appearance rate of snail frame,and the average den-sity of alive snails were 15231,8.15%,and 0.463/0.1 m2,respectively. The elevation of snail distribution area of the south and north Poyang Lake areas were 11-16 m and 9-16 m respectively. The elevation of concentrated snail belts of the south Poyang Lake area were 12-13 m and 15-16 m,and the elevation of concentrated snail belts of the north Poyang Lake area was 12-14 m. Conclusions The distribution of snails is in the range of 9-16 m. The suitable habitats of snail breeding are moving from the south Poyang Lake area to the north Poyang Lake area,and from high elevation to low elevation. In the future,the schistosomia-sis prevention and control measures could be formulated based on the geographical characteristics of current snail distribution in order to consolidate the achievements of schistosomiasis control.
