1.The clinical analysis of ganciclovirum for children infectious mononucleosis treatment

Objective To observe the clinical effects and safety of ganciclovirum for children infectious mononucleosis.Methods 56 cases diagnosed children were divided into ganciclovirum group(n=30) and virazole group(n=26) randomly.The two groups accepted ganciclovirum and virazole treatment respectively.The clinical effects and side effects were compared.Results The fever period,the time of lymph node,liver,spleen enlargement,pharyngitis of ganciclovirum group were shorter than those of virazole group.The difference was significant.No obvious side effect was observed during ganciclovirum treatment period.Conclusion Ganciclovirum is more effective and safe for children infectious mononucleosis.

Objective To analyze the blood pressure,heart rate,the US national institutes of health stroke scale(NIHSS) score,Glasgow coma scale(GCS) score for cerebral infarction patients with cerebral hemorrhage after recombinant human tissue-type plasminogen activator (rt-PA) therapy.Methods From April 2011 to April 2013,48 cases with cerebral infarction after intravenous rt-PA thrombolysis were collected.These cases were divided into two groups:the first group (32 cases):without cerebral hemorrhage after thrombolysis; the second group (16 cases):with cerebral hemorrhage after thrombolysis.The systolic blood pressure,diastolic blood pressure,heart rote,NIHSS score and GCS score were compared between the two groups before and after thrombolysis.Results After thrombolysis,the systolic and diastolic blood pressure were significantly increased in cerebral hemorrhage cases compared with before thrombolysis and patients without cerebral hemorrhage.After thrombolysis,the NIHSS score was significantly reduced in the cases without cerebral hemorrhage compared with that before thrombolysis,while the NIHSS score was significantly higher in the cases with cerebral hemorrhage compared with that before thrombolysis.After thrombolysis,the heart rate and GCS score showed no significant difference between the patients with and without cerebral hemorrhage.Conclusions The rising of systolic blood pressure,diastolic blood pressure and NIHSS score helps us determine the early cerebral hemorrhage after thrombolysis in cerebral infarction patients.
4.Construction of eukaryotic expression vector of pre-miR-15a and its inhibitory effect on Raji cells proliferation

Objective To construct an eukaryotic expression vector of pre-miR-15a,and to investigate the inhibitory effect of pre-miR-15a to Raji cells. Methods The pGCSIL-GFP vector encoding pre-miR-15a nucleotides was transfected into the bacterial competent cells,and then confirmed by PCR and sequencing analysis. The identified vector was transfected into Raji cells with oligofectamine 2000. The cells were divided into 3 groups,blank,negative control and pre-miR-15a group. Semi-quantitative RT-PCR was used to detect the expression of Bcl-2 mRNA,and immunofluorescence indirect for Bcl-2 protein expression. The growth of Raji cells was assayed by trypan blue dye exclusion method. Results PCR and sequences analysis indicated that the recombinant clones was identical with target sequences. Many green fluorescent cells were observed under fluorescent microscopy. The levels of Bcl-2 mRNA at every group had no obviously difference. Bcl-2 protein expression was obviously decreased at pre-miR-15a group compared with the other groups. Trypan blue dye exclusion method showed the cell growth was inhibited at 48 h and 72 h post-transfection. Conclusion We successfully construct the eukaryotic expression vector of pre-miR-15a,and it can inhibit the growth of Raji cells.
5.The gene expression profiles through activating endothelial target for acetylcholine

Aim To investigate the effects of gene expression through activating endothelial target for acetylcholine (ETA).Methods Comparative studies were carried out to explore the distinct gene expression of carbachol and pilocarpine.Results Human coronary aortic endothelial cells were incubated with carbachol which activates ETA and muscarinic receptors and with pilocarpine which activates muscarinic receptors distinctly for 10 hours at the concentration of 100 ?mol?L -1.The endothelial gene expression was detected by BiostarH-14112S cDNA expression profiling microarrays containing 14 000 human unigenes. There were 801 differential genes totally (491 differentially expressed both by carbachol and by pilocarpine, 310 differentially expressed by carbachol exclusively). We found a significant increase in expression of 119 and a significant decrease in expression of 191 genes after treatment by carbachol preferentially. And these genes were not affected by pilocarpine. Further globally functional catagorization indicates that there are seven types of distinct expressed genes induced by carbachol selectively including membrane receptors and G proteins, ion channels, G protein coupled receptors, transcriptional factors, apoptosis-related genes, cell adhesive factors, thrombosis and atherosclerosis-related genes.Conclusion The differentially expressed genes evoked by carbachol exclusively are closely associated with ETA and its effectors.

Objective Through the statistical analysis on literature on asthenia cold syndrome in recent ten years,to comprehend the present situation of studies on the feature and objectivization of symptom distribution of asthenia cold syndrome.Methods Collect literature on asthenia cold syndrome from the periodicals of traditional Chinese medicine and integrated traditional and western medicine from the year 1996 to 2006.The described features and objective indices of symptom distribution of this syndrome were systematically analyzed and summarized.Results In the literature,the diagnostic criteria of asthenia cold syndrome were made mainly by the authors themselves,few referring to others',being different in the descriptions of symptom distribution.The researches on objectivization indices were scattered,most of which took the single or multiple index(es) as subject(s).Conclusion Cold limbs,tiredness,aversion to cold,and loose stools are possibly significant in the diagnosis of asthenia cold syndrome.The way of the studies on diagnostic criteria and its objectivization of asthenia cold syndrome need to be improved.

8.Study on quality standard for Jianweishu Granule

Objective: To establish the assay methods for paeoniflorin and baicalin in Jianweishu Granules (Radix Scutellariae, Radix Paeoniae Alba, Radix Astragali seu Hedysari, Radix Notoginseng, etc.). Methods: Radix Astragali, Radix Notoginseng; Radix Paeoniae Alba and Herba Dendrobii in Jianweishu Granule were identified by TLC. The paeoniflorin and baicalin in Jianweishu Granule were determined by HPLC on shim pack CLC ODS C 18 Column (6.0mm?150mm,10?m); using methanol water phosphoric acid (47∶53∶0.2) and methanol water (30∶70) as mobile phases, respectively; the flow rate were at 1.0 mL?min -1 ; the detection wavelength were 280nm and 230nm, respectively; the column temperature were at room temperature and 40?C respectively. Results: The average recoveries are 98.94% and 98.67%. RSD are 2.31% ( n =5) and 1.36% ( n =5). Conclusion: The methods are simple, sensitive, accurate and reproducible.

There is another factor involving in endothelium dependent relaxation besides NO, which is independent of cGMP, EDHF. Its hyperpolarization mechanism may be related to the open of calcium dependent K + channels(K + Ca). NO, PGI 2,H 2O 2 and cytochrome P450 derived metabolite of arachidonic acid are all the candidates of EDHF. In the normal blood vessels, the influence of EDHF on endothelium dependent relaxation is little. But in some diseases, when the production and effect of NO is disturbed ,the mechanism of EDHF may play a specially important role.
