1.Experimental study on expressing GM-CSF in human bone marrow stromal cells by total saponins of Panax ginseng (TSPG)

Objective: To study the relationship between effect of total saponins of Panax ginseng(TSPG) on expression of GM-CSF in human bone marrow stromal cells and modulation of human granulocytopoiesis and monocytopoiesis.Methods:The techniques of culture of hematopoietic progenitor cells and human bone marrow stromal cells in vitro,immunocytochemistry, nucleic acid in situ hybridization were used.Results:TSPG could markedly promote the colony formation of CFU-GM; human bone marrow stromal cells conditioned media prepared with TSPG could significantly enhance the proliferation and differentiation of CFU-GM,and expression of GM-CSF in protein and mRNA level in human bone marrow stromal cells induced by TSPG were much higher than those of the control group.Conclusion:TSPG may directly and/or indirectly promote stromal cells in hematopoietic inductive microenvironment to produce GM-CSF, which further prowote the proliferation and differentiation of human CFU-GM.
2.The Activation of ERK3 Signaling Pathway Blocks Cell Proliferation

Extracellular signal-regulated kinase3 (ERK3) is distinguished from other ERK family members especially in its molecular biological characteristics including the big intron between exons in its gene structure, the serine189 mono-phosphorylated site and C-terminal extention of its kinase structure. The specially activating phosphorylation site of serine189 indicates that all MEKs, which phosphorylate serine/threonine double phosphorylation sites of MAPKs, are unable to activate ERK3. The C-terminal extension involves both subcellular localization of ERK3 and binding to intact cyclin D3, which can profoundly affect cell cycle regulation. According to update reports, ERK3 signal pathway in the regulation of cell cycle might be as follows: Ras→B-Raf→ERK3kinase→ERK3→decrease of CDK compounds of G1-phase→increase of the inhibiting factor (retinoblastoma protein) of S-phase→blockage of S-phase of cell cycle→cell differentiation entry while cell proliferation arrest. Moreover, the activation of ERK3 signaling pathway is also associated with cell differentiation, embryonic development, insulin secretion and cancer diseases.
3.Expression and significance of MMP-2 and VEGF in retinal neovascularization

Objective Researches showed that matrix metalloproteinase-2 (MMP-2) has a critical role in the neovascularization of tumor,and vascular endothelial growth factor(VEGF)is a promoting factor of new blood vessel formation.There still is little literature about the effect of MMP-2 in retinal neovascularization up to now.This study tried to explore the expression and significance of MMP-2 and VEGF in retinal neovascularization.MethodsA retinal neovascularization model was established in 30 7-day-old cleaning C57BL/6J mice exposed to an environment of high concentration of oxygen for 5 days,and 30 matched mice were raised in normal air environment.Fifteen mice from hyperoxic group and control group were sacrificed in 10 days after treatment and the eyeballs were enucleated to make retinal stretched preparation.Adenosine diphosphate-ase (ADPase) stained retina flat-mounts was performed to assess the retinal vascular profiles,and H&E staining was applied to count the number of new vascular cell nuclei.The expression of MMP-2 and VEGF was detected using immunohistochemistry by calculating the intergrated value of positive cells.ResultsThe retinal stretched preparation presented more neovascularization in mice from the hyperoxic group compared with control group.The number of nuclei from the vascular endothelial cells in the new vessels breaking through the internal limiting membrane in the hyperoxic group was 33.51±2.55,indicating a significant increase in comparison with control group (7.27±0.20)(t=9.345,P<0.05).There were stronger expression of MMP-2 protein and the VEGF protein in the ganglion cell layer,inner plexiform layer and inner nuclear layer,and neovascularization breaking through the internal limiting membrane in the hyperoxic group compared with control group (t=4.25,P<0.05;t=6.38,P<0.05).Expression of MMP-2 showed the positive correlation with the expression of VEGF(r=0.825,P<0.05).ConclusionBoth MMP-2 and VEGF promote retinal neovascularization.The overexpression of MMP-2 and VEGF play a synergistic role during the formation of neovascularization.
4.ICF Based Disability and Rehabilitation Information Standard and Its Applications

The applications of ICF based infor-mation standard had been discussed in the areas of disability and rehabilitation assessment, clinical and rehabilitation information, Function-al Diagnosis Related Groups (fDRGs) and disability survey statistics and policy development.
5.Analysis of allergen skin test results in 1124 patients with allergic rhinitis in the junction of Guangdong and Guangxi provinces.

OBJECTIVE:
To investigate the characteristics and influence factors of sensitization hy allergens in patients with allergic rhinitis in the junction of Guangdong and Guangxi provinces.
METHOD:
One thousand one hundred and twenty-four patients who were diagnosed with allergic rhinitis hy clinical history and symptoms underwent a skin prick test. The percent of allergens with positive skin response and Multiple Allergens Positive Scores were recorded, and their influence factors, included age, gender and test period were evaluated.
RESULT:
The percent of top three inhalant allergens with positive skin tests were as follow: dermatophagoides pteronyssinus (77.0%) ,dermatophagoides farina (77.1%), blattella germanica (42.3%); the percent of top three ingested allergens with loositive skintests were as follow: crab ( 39.5%), shrimp (31.1%), ribbonfish (10.8%). The average Multiple Allergens Positive Score was (3.6 +/- 2.7). The percent of allergens with positive skin response and Multiple Allergens Positive Score of different allergens changed as age, gender and text period changed, and the changing trend of them was accordant. The percent inhalant allergens with positive skin tests in dermatophagoides pteronyssinus, dermatophagoides farinae and cat fur in different gender showed a significant difference.
CONCLUSION
Acarine allergens were the most common allergens in patients with allergic rhinitis in the junction of Guangdong and Guangxi provinces. The percent of allergens with positive skin response and the severity of multiple allergy were influenced by age, gender and text period. The severity of multiple allergy can be measured by Multiple Allergens Positive Score.

7.Development of Classification Markup Language of International Classification of Functioning, Disability and Health

Management information platform of International Classification of Functioning, Disability and Health (ICF) was developed based on the Classification Markup Language (ClaML). The standard representative and management of the ICF structure, encoding system,classes and semantics had developed, and the functions such as the classification editing, modification, search, viewing and checking had been realized. This system can be used in classification import, export and application in other systems.
8.Inhibitory effect of captopril on retinal neovascularization in mice

AIM: To study the inhibitory effect of captopril on retinal neovascularization (RNV).METHODS: Sixty seven-day-old mice were randomly divided into treated group and control group with thirty mice in each group. These mice were exposed to 750 50mL/L oxygen for 5 days and then to room air.The treated group had been injected captopril (2.7mL/kg), while control group had been injected 9g/L sodium chloride (2.7mL/kg) by intravitreal for 5 days.The mice were sacrificed at the 17th day after birth and the eyes were enucleated. Adenosine diphosphate-ase(ADPase) stained retina flat-mounts was performed to assess the retinal vascular profiles, Hematoxylin Eosin (HE)staining method was applied to count the number of new vascular cell nuclei and the expression of matrix metalloproteinase-2(MMP-2)and pigment epithelium derived factor (PEDF)was detected by immunohistochemical method.RESULTS: Comparing with control group,regular distributions and good branch and reduced density of RNV were observed in the treated group. The number of nucleus of new vessels vascular endothelial cells breaking through the internal limiting membrane was less in the treated group than in the control group (P<0.05). Stain of retinal MMP-2 was weaker in the treated group than in the control group and stain of retinal PEDF was stronger in the treated group than in the control group.CONCLUSION: Intravitreal injection of captopril (2.7mL/kg) may block the RNV in the oxygen-induced mouse model and may provide an effective method for prevent-ing RNV.

10.Triple-Endobutton technique for the treatment of Tossy type III acromioclavicular joint dislocation.

OBJECTIVETo evaluate the clinical outcomes and complications of Triple-Endobutton plates in treating Tossy type III acromioclavicular joint dislocation.
METHODSFrom January 2011 to January 2013,45 patients with Tossy type III acromioclavicular joint dislocation were treated with Triple-Endobutton plates. There were 35 males and 10 females with an average age of 30.5 (ranged from 19 to 60) years old. At the final follow-up, VAS, DASH, Constant-Murley criterion were used to evaluate shoulder function.
RESULTSAll patients were followed up from 15 to 36 months. No neurovascular injury, wound infection and stress fractures were found,but 3 patients had a re-dislocation. At the final follow-up,the mean VAS score was decreased from (5.7±1.6) preoperatively to postoperative (0.2±0.1); DASH score was significantly decreased from (19.6±4.3) preoperatively to (0.3±0.1) postoperatively; Constant-Murley score was improved from (34.4±4.3) before operation to (94.8± 3.5) after operation.
CONCLUSIONClinical outcomes of treating Tossy type III acromioclavicular joint dislocation with Triple-Endobutton plates is satisfactory. However, re-dislocation is still the most common complication. Careful perioperative management is an important factor in preventing re-dislocation.
