1.The Combined Effect of Fast Neutron and hyperthermia according to the Sequence and Interval in MKN-45 Cells.

PURPOSE: It has been well established that response of cells and tissues to low LET radiations(X- or grmma-ray) can enhanced by comdining with hyperthermia. However, There has been relatively little of hyperthermia on the possible modification of either cellular or tissue responses to other types of radiation. So, We investigated the combined effect of fast neutron irradiation and hyperthermia according to the sequence and time interval of the two MATERIALS AND METHODS: In MKN-45 cells, a human stomach cancer cell line, Surviving fractions were measured according to the sequence treatment of 6,4,2,0 hour interval for fast neutron irradiation(1.5Gy) combined with hyperthermia(41 degrees C for 30 min or 43 degrees C for 30 min). RESULTS: D(0) and n of MKN-45 for neutron were 0.8Gy and 2.5, respectively. The surviving fraction by 1.5 Gy of neutron was 0.36+/-0.34. Interacting powers were mostly. The surviving fraction by 1.5 Gy of neutron was 0.36+/-0.34. Interacting powers were mostly ranged between 1 and 2, bur they were 3.0Gy 2.7, respectively for hyperthermia (41 degrees C for 30 min) followed by neutron irradiation 6 and 4 hours later. CONCLUSION: The combined effect of fast neutron (1.5Gy) and hyperthermia (41 degrees C or 43 degrees C for 30min) is largely independently additive. Preceding mild hyperthermia (41 degrees C for 30 min) 4 or 6 hours before neutron may cause decreased sensitivity to subsequent neutron irradiation.

OBJECTIVE: Considering the different prevalence rates of diseases such as asthma and chronic obstructive pulmonary disease in Asians relative to other races, Koreans may have unique airway structure and lung function. This study aimed to investigate unique features of airway structure and lung function based on quantitative computed tomography (QCT)-imaging metrics in the Korean Asian population (Koreans) as compared with the White American population (Whites). MATERIALS AND METHODS: QCT data of healthy non-smokers (223 Koreans vs. 70 Whites) were collected, including QCT structural variables of wall thickness (WT) and hydraulic diameter (Dh) and functional variables of air volume, total air volume change in the lung (ΔVair), percent emphysema-like lung (Emph%), and percent functional small airway disease-like lung (fSAD%). Mann-Whitney U tests were performed to compare the two groups. RESULTS: As compared with Whites, Koreans had smaller volume at inspiration, ΔVair between inspiration and expiration (p < 0.001), and Emph% at inspiration (p < 0.001). Especially, Korean females had a decrease of ΔVair in the lower lobes (p < 0.001), associated with fSAD% at the lower lobes (p < 0.05). In addition, Koreans had smaller Dh and WT of the trachea (both, p < 0.05), correlated with the forced expiratory volume in 1 second (R = 0.49, 0.39; all p < 0.001) and forced vital capacity (R = 0.55, 0.45; all p < 0.001). CONCLUSION: Koreans had unique features of airway structure and lung function as compared with Whites, and the difference was clearer in female individuals. Discriminating structural and functional features between Koreans and Whites enables exploration of inter-racial differences of pulmonary disease in terms of severity, distribution, and phenotype.

6.3-Carene, a Phytoncide from Pine Tree Has a Sleep-enhancing Effect by Targeting the GABA(A)-benzodiazepine Receptors

3-Carene, a bicyclic monoterpene, is one of the major components of the pine tree essential oils. It has been reported that, in addition to its known properties as a phytoncide, 3-carene has anti-inflammatory, antimicrobial, and anxiolytic effects. We have previously demonstrated that α-pinene, the major component of pine tree, has a hypnotic effect through GABA(A)-benzodiazepine (BZD) receptors. However, a hypnotic effect of 3-carene has not been studied yet. Here, we report that oral administration of 3-carene increases the sleep duration and reduces sleep latency in pentobarbital-induced sleep test. 3-Carene potentiates the GABA(A) receptor-mediated synaptic responses by prolonging the decay time constant of inhibitory synaptic responses. These enhancing effects of 3-carene are reproduced by zolpidem, a modulator for GABA(A)-BZD receptor, and fully inhibited by flumazenil, an antagonist for GABA(A)-BZD receptor. The molecular docking of 3-carene to the BZD site of GABA(A) protein structure, suggests that 3-carene binds to the BZD site of α1 and ϒ2 subunits of GABA(A)-BZD receptor. These results indicate that, similar to α-pinene, 3-carene shows a sleep-enhancing effect by acting as a positive modulator for GABA(A)-BZD receptor.
