2.Involvement of Polyamine in Growth Hormine Secretion from the GH3 Cells.

BACKGROUNDS: Polyamines are known to be essential for cell growth and differentiation. Recently, possible roles of the polyamine in signal transduction as neurotransmitter, modulator, or second messenger are suggested in many studies. Furthermore, it is widely studied that possible roles of polyamine are involved in the action of hormone. Thus, it was to investigate the effect of polyamines in the cell proliferation and secretion of GH from the GH cells. METHODS: Cells(5*10 cells/mL) were incubated for 3 days in DMEM containing test drugs and labeled with 20pCi/mL of [S]-methionine for 2 hr. Proteins secreted into the medium were separated by 13% SDS-gel electrophoresis, then autoradiography was performed to identify radiolabeled proteins. [S]-methionine labelled GH was identified by radioimmuno-precipitation. Total protein synthesis was determined from the radioactivity of the cell homogenate by liquid scintillation counter. The intracellular polyamine content was determined by HPLC. RESULTS: Externally added polyamines(putrescine, spermidine, spermine) induced cell proliferation in a dose-dependent manner at proper concentrations, specifically 50pM putrescine increased GH secretion, DFMO or MGBG, which is polyamine biosynthetic inhibitor, inhibited GH secretion in a dose-dependent fashion, In the cells treated with 20mM or 0.01mM MGBG, total protein synthesis were decreased only to 90 or 76% of the control levels and cell proliferation was also slightly inhibited. However the secretion of GH was severely blocked to 37% or 35% of the control. Hydrocortisone at 5 pM stimulated the secretion of GH to 153% of basal secretion, also doubled intracellular putrescine content. CONCLUSION: The present data show that externally added polyamines induced cell proliferation and GH secretion. Also, extemally added putrescine stimulated GH secretion significantly. GH secretion was inhibited by polyamine metabolic inhibitor in a dose-dependent manner and polyamine metabolic inhibitors, at proper concentrations, specifically blocked GH secretion without any significant influence on the total protein synthesis. The above results imply the involvement of polyamine in GH secretion.

3.Treatment of Unstable Fracture Dislocation of the Spin

The results of treatment of injuries of the thoracic and thoracolumbar spine with neurological involvement have been reviewed in a retrospective study of 14 patients who received surgical treatment. Seven patients were treated with open reduction and Harrington instrumentation and the others were treated with anterior fusion, laminectomy only, Roger wiring with H-graft and posterior fusion respectively. In the former group, distraction rods were used in all patients. Three patients with incomplete paraplegia and of the 4 patients with complete paraplegia, two patients showed marked neurological improvement. While, in the latter group, only two patients with incomplete paraplegia showed marked neurological improvement. Harrington instrumentation allowed easier postoperative nursing and early mobilization of the patients.

The angiolipoma is well circumscribed, benign neoplasm, composed of mature fat cells and well developed arterioles, venules and capillaries, in which fibrin thrombi often form. We experienced 4 adult male patients who had free movable, subcutaneous nodular masses on the upper extremities, the abdomen, the back and the buttock. All of them were confirmed as angiolipoma by the clinical and histopathological findings.

8.A Case of Metastatic Basal Cell Carcinoma.

Basal cell carcinoma is the most common skin cancer, but it rarely metastasizes. Metastatic basal cell carcinoma (MBCC) is rare in terms of incidence as well as absolute numbers. We present a case of 67-year-old woman who had a dirty oozing, ulcerated, tumor mass on the vertex and suffered from dyspnea and mild lumbago. The diagnosis of MBCC was based on clinical, radiological, and histopathological findings. To our knowledge, the occurrence of MBCC has not been reported in the Korean literatures. So we reported herein this case of MBCC of the lung and the bone.

10.Two cases of atypical fibroxanthoma of the skin.

Atypical fibroxanthoma(AFX) occurs most on sun-exposed area of the head and neck of elderly person. It has an excellent prognosis after conservative, but complete, excision. However, because of its potential, albeit small, for metastasis, it is widely regarded as a low-grade sarcoma. We present herein two cases of atypical fibroxanthoma. The case 1 was a 86-year-old female who had a small egg-sized, dome shaped nodule with eroive surface on the left cheek. The other case was a 60-year-old male who had solitry bean-sized, nodulo-ulcerative lesion on the vertex. Immunohistochemical studies revealed positive reaction for vimntin and a-antichymo trypsin. These patients have received completely total surgical ecis on and remained free of recurrence for a period of about, 2 years follow up.
