1.Influence of miRNA-155 on lymphoma.

Lymphomas are the most common ones of hematologic tumors. In China, Hodgkin's lymphoma accounts for 9% - 10% of lymphomas, which has a good response to chemotherapy, while non-Hodgkin's lymphoma accounts for nearly 90% of lymphomas, and the incidence of which tends to rise in recent years. At present, it was realized that miRNA and lymphomas are closely related to each other. More attention has been paid to the effects of miRNA on the pathophysiological process of lymphoma. This review is focused on miRNA-155, one of the miRNA family members, and its action mechanism in diffuse large B-cell lymphoma, Burkitt lymphoma and Hodgkin's lymphoma, expecting to shed light on the future therapy.

2.Methylation of Id4 gene and inhibitive effect of arsenic trioxide on it in Raji cells.

OBJECTIVETo study methylation of Id4 gene and demethylation effect of arsenic trioxide (ATO) in Raji cells.
METHODSHuman Burkitt's Raji lymphoma cells were cultared and treated with ATO at different concentrations and different time points. Methylated degree of Id4 gene was detected by methylation specificity polymerase chain reaction (MS-PCR), Id4 mRNA expression in Raji cell by reverse transcription polymerase chain reaction (RT-PCR), the growth of cell by MTT assay, and cell apoptosis and cycle distribution by Flow Cytometry (FCM).
RESULTS(1) The Id4 gene exhaustive methylation in control group, and hypermethylation in experimental group were reversed by ATO in a dose-dependent manner. (2) Id4 mRNA expression in Raji cells treated with ATO for 48 h increased gradually with ATO concentration increasing in experimental group. (3) Raji cell growth inhibited rates after different concentrations of ATO treatment for 24, 48, 72 h were 12.15% ∼ 92.17% in the experimental group (P < 0.05). (4) Apoptosis peak emerged after ATO treatment for 48 hours in experimental group, while a much lower apoptosis in control group. (5) After ATO treatment for 48 h in experiment group, the cells were arrested at G(0)/G(1) phase in a dose-dependent manner.
CONCLUSIONId4 gene presents exhaustive methylation in Raji cells. ATO can reverse the hypermethylation of Id4 gene and recover the expression of Id4 mRNA. Hypermethylation of Id4 gene is one of the reasons of Raji cells malignant proliferations.

3.Prognostic significance of IKZF1 gene deletions in patients with B-cell acute lymphoblastic leukemia.

Objective: This study aimed to investigate the prognostic significance of IKZF1 gene deletion in patients with acute B lymphoblastic leukemia (B-ALL) . Methods: The clinical data of 142 patients with B-ALL diagnosed in Nanfang Hospital between March 2016 and September 2019 were analyzed. Results: IKZF1 deletion was found in 36.0% of the 142 patients with B-ALL, whereas exon 4-7 deletion was found in 44.0% . White blood cell counts were higher in patients with the IKZF1 deletion (52.0% and 28.3% , P=0.005) ; these patients also experienced worse effects of mid-term induction therapy (40.0% and 70.7% , P<0.001) and had a higher proportion of Philadelphia chromosome-positive (52.0% and 21.7% , respectively, P<0.001) . Univariate analysis revealed that the 3-year overall survival rate (OS) and event-free survival rate (EFS) in the IKZF1 deletion group were significantly lower than the IKZF1 wild-type group [ (37.1±7.3) % vs (54.7±5.4) % , (51.8±7.9) % vs (73.9±4.7) % ; P=0.025, 0.013, respectively]. Multivariable analysis showed that harboring IKZF1 deletion was an adverse factor of EFS and OS (HR=1.744, 2.036; P=0.022, 0.020, respectively) . Furthermore, the IKZF1 deletion/chemotherapy group had significantly lower 3-year OS, EFS, and disease-free survival rates than other subgroups. In the IKZF1 deletion cohort, allo-hematopoietic stem cell transplantation (HSCT) significantly improved OS and EFS compared to non-allo-HSCT[ (67.9±10.4) % vs (31.9±11.0) % , (46.6±10.5) % vs (26.7±9.7) % ; P=0.005, 0.026, respectively]. Conclusion: Pediatric-inspired chemotherapy was unable to completely reverse the negative effect of IKZF1 deletion on prognosis. Pediatric-inspired regimen therapy combined with allo-HSCT, in contrast, significantly improved the overall prognosis of IKZF1 deletion B-ALL.

4.Characteristics of two cases of Burkitt lymphoma/leukemia with concurrent t(8;14) and t(14;18).

This article aimed to report two cases of Burkitt lymphoma/leukemia with concurrent t(8;14) and t(14;18). Morphology, immunophenotype, cytogenetics and molecular biology (MICM) methods were applied to diagnosis. The results showed that the two cases were both acute lymphocytic leukemia L3 type according to FAB criteria. Conventional cytogenetic technique or interphase fluorescence in situ hybridization (FISH) demonstrated that t(8;14) and t(14;18) were detected concurrently in both patients. CD20, CD10, FMC7, CD38 and CD19 were expressed in both patients by immunophenotyping. According to MICM, they were both diagnosed as Burkitt lymphoma/leukemia. The first patient died in one month after chemotherapy, and the second patient survived 19 months after rituximab- combined high-dose chemotherapy and subsequently allogeneic hematopoietic stem cell transplantation (HSCT). In conclusion, t(8;14) and t(14;18) may present simultaneously in Burkitt lymphoma/leukemia and indicate poor prognosis. Rituximab-combined chemotherapy and subsequently HSCT could improve the outcomes of such cases.

5.Association of lymphoid malignancy incidence with EB Virus and IL-10 gene polymorphism.

EBV (Epstein-Barr Virus) is a common herpes virus in patient with lymphatic system tumor, which firstly discovered in the cell line of Burkitt's lymphoma. 90% people worldwide had been infected by EB virus before grown-up, but not all people have the EBV-related disease or tumor. Most EBV infection can not elicit clinical symptoms. EBV infection in tumor of lymphatic system is common. It was early known that the EBV existence may cause the immortalization of normal B cells, which like the tumor cells. It means that EBV plays an important role in the tumorigenesis. EBV Bcrf1 code frame is homology to human IL-10, which is also called viral IL-10, showing immunosuppressive effect similar to the IL-10. IL-10 is also an important immunoregulatory factor, the secretory level of which influences the occurrence and development of lymphatic system diseases; the genotype of SNP site in IL-10 promoter region also associates with secretory level of IL-10. This review discusses the close relation between tumor of lymphatic system, EBV infection and gene polymorphism of IL-10.

6.PHI regulates histone methylation and acetylation in Burkitt lymphoma Daudi cell line.

This study was purposed to investigate the effects of phenylhexyl isothiocyanate (PHI) on Burkitt lymphoma Daudi cell line and regulation of histone acetylation and methylation in Daudi cells, and to explore the potential mechanism. The apoptotic rate of Daudi cells treated with PHI was measured by flow cytometry, the changes of histone H3 and H4 acetylation, histone H3K9 and H3K4 methylation in Daudi cells treated with PHI were detected by Western blot. The results showed that PHI could induce apoptosis of Daudi cells, increased the acetylation level of H3 and H4, enhanced the methylation of H3K4, but reduced the methylation of H3K9. It is concluded that the PHI can up-regulate the acetylation level of histone H3 associated with transcription stimulation and the methylation of histone H3K4, down-regulate the methylation on histone H3K9 associated with transcription inhibition, promotes the apoptosis of Daudi cells. PHI may be a potential agent for target therapy of lymphoma.

7.Expression of retinoblastoma protein in child acute leukemia cells and its clinical significance.

The research was aimed to detect the expression levels of retinoblastoma protein (pRb) in child acute leukemia cells, and to explore its possible association with leukemia cells cycle, the risk of disease, minimal residual disease (MRD) monitoring and prognosis of B-ALL. Flow cytometry (FCM) was used to detect the expression of pRb in 89 cases of acute leukemia (including 25 AML, 10 T-ALL and 54 B-ALL) and bone marrows from 7 normal children (control group). Meanwhile the cell cycle in some cases was analyzed. The results showed that (1) the FCM could accurately detect the expression of pRb in acute leukemia cells; (2) the high level of pRb expression was frequent in all types of child acute leukemias. In the same case, the expression of pRb was significantly increased in leukemia cells when compared with non-leukemia cells. And no detectable pRb protein was found in partial cases of acute leukemia; (3) there was a close relation between expression of pRb and the cell cycle of leukemia cells, the number of G(1) phase cells in pRb positive case of B-ALL was more than that in pRb negative case (92% vs 77%); (4) in B-ALL, the level of pRb expression in MRD positive group was significantly lower than that in MRD negative group (P < 0.05), but pRb expression was stable in non-leukemia cells during therapy; (5) pRb expression was related to the early response to therapy in B-ALL, the expression of pRb was significantly increased in sensitive group when compared with insensitive group (P < 0.05). It is concluded that high level or absence of pRb expression can be found in child acute leukemia cells. The expression of pRb is positively related to cell cycle of leukemia cells, MRD monitoring and the early response to therapy. In short, the detection of pRb expression level can guide the therapy and the evaluation of prognosis in B-ALL.

8.The cytogenetic features of Burkitt leukemia.

OBJECTIVETo evaluate the incidence of chromosomal abnormalities in Burkitt leukemia (BL).
METHODSConventional cytogenetics (CC) was carried out to detect the karyotypes. Meanwhile, three color interphase fluorescence in situ hybridization (FISH) was used to detect the t(8;14)(q24;q32) in 17 newly diagnosed BL.
RESULTSThe results showed that the incidence of chromosomal abnormalities was 41.2% (7/17) by CC technique, including one of each for simple t(8;14)(q24;q32), complex chromosomal abnormality containing t(8;14)(q24;q32), t(8;22)(q24;q11), the complex chromosomal abnormality of t(12;22)(?;?), t(2;13)(?;?) with + 12, and one with two marker chromosomes. FISH method detected eight cases of t(8;14)(q24;q32), including the two detected by CC technique. Five samples (5/8) showed 2A1G1O2F (two blue, one green, one orange and two yellow signals in interphase nuclei), while three samples (3/8) showed 2A1G1O1F(two blue, one green, one orange and one yellow signals in interphase nuclei).
CONCLUSIONTwo different breakpoints have been identified on the c-Myc locus on 8q24. Interphase FISH was more sensitive in detecting t(8;14)(q24;q32), and it is an important complement to CC. It should be used as a routine method for diagnosis of BL.

9.Effect of bcl-2 siRNA on apoptosis of Burkitt's lymphoma cell line CA46.

The aim of this study was to investigate the effect of bcl-2 siRNA on bcl-2 gene expression and apoptosis of lymphoma cell line CA46. A siRNA was designed and synthesized. Then siRNA was transfected into CA46 cells by cationic liposome. At 48 hours after transfection, apoptosis and mitochondria transmembrane potential of CA46 cells were detected by flow cytometry, the expression of bcl-2 mRNA and BCL-2 protein in CA46 cells were detected by RT-PCR and flow cytometry respectively. The results showed that at 48 hours after transfection, apoptosis of CA46 cells occurred, mitochondria transmembrane potential changed. The expression of bcl-2 mRNA and BCL-2 protein in CA46 cells decreased significantly. In conclusion, bcl-2 siRNA depresses the expression of bcl-2 gene in CA46 cells specifically, then changes the mitochondria transmembrane potential, resulting in apoptosis of CA46 cells.

10.Clinical study of mature B-cell lymphoma in 11 children with chromosome 11 long-arm abnormalities.

Objective: To explore the clinical, pathological, diagnostic, treatment, and prognostic features of children with mature B-cell lymphoma (MBCL) . Methods: This retrospective study included pediatric patients with MBCL with chromosome 11 long-arm abnormalities who were diagnosed and treated at our hospital from December 2018 to February 2023. Results: Among the 11 pediatric patients with MBCL, nine were male and two were female, with a median age of 9 (2-13) years and a median disease course of 1.8 (0.5-24) months. The clinical manifestations were cervical lymph node enlargement in four patients, nasal congestion and snoring in four patients, abdominal pain in two patients, and difficulty breathing in one patient. There were seven cases of Burkitt's lymphoma, two of follicular lymphoma, and two of advanced B-cell lymphoma according to the pathological morphology examination. No patients had central nervous system or bone marrow involvement, and no extensive metastasis was observed on B-ultrasound or positron emission tomography-computed tomography (PET/CT). One patient had a huge tumor lesion. The Revised International Pediatric Non-Hodgkin Lymphoma Staging System classified four patients as stage Ⅱ, five as stage Ⅲ, and two as stage Ⅳ. 11q probe detection showed five cases of 11q gain, three of 11q loss, and three of both gain and loss. FISH showed positive MYC expression in three patients, including eight with advanced B-cell lymphoma with 11q abnormalities and three with Burkitt's lymphoma with 11q abnormalities. According to the 2019 edition of the National Health Commission's diagnostic and treatment guidelines for invasive MBCL in children, one patient was classified as Group A, two as Group B, and eight as Group C. Early evaluation of the efficacy showed complete remission. After mid-term evaluation, the intensity of chemotherapy was reduced in Group B and Group C. Among two cases of chemotherapy, the remaining nine cases had a median follow-up of 32 (6-45) months, and none had event-related survival. Conclusion: The incidence of MBCL with 11q abnormalities in children is low, clinical symptoms are mild, and progression is slow. The absence of MYC, BCL2, BCL6 rearrangements, C-MYC negative and 11q abnormalities on FISH is an important diagnostic indicator, and reducing the intensity of chemotherapy can improve prognosis.
