In this paper,the recent advances in both biomedical sciences and higher medical education reform were reviewed and analyzed.Furthermore,we proposed and reconstructed the teaching system of biochemistry and molecular biology course in our university,including its teaching content,teaching methods,teacher team,teaching management,etc.The preliminary practice of this system has obtained significant positive effects on teaching quality and student performance.
2.Effect of chronic intermittent hypobaric hypoxia on reactivity of adrenergic receptors in rabbit sino-atrial node

AIM: To investigate the effect of chronic intermittent hypobaric hypoxia (CIHH) on the activity of adrenergic receptor(AR) in rabbit sino-atrial node (SAN) by intracellular recording.METHODS: New Zealand rabbits were randomly divided into three groups: control group (Con), 14 d CIHH treatment group (CIHH14) and 28 d CIHH treatment group (CIHH28). The CIHH rabbits were exposed to a simulated 5 000 m (oxygen 11.1%) hypobaric hypoxia in hypoxic chamber for 14 d or 28 d (6 h/d), respectively. SAN preparation was used and the transmembrane action potential was recorded by micropipettes. Isoproterenol hydrochloride (ISO, agonist of β-AR) and phenylephrine (PE, agonist of α_1-AR) at different concentrations (0.01, 0.1 and 1 μmol/L) were applied cumulatively to investigate the electrophysiological effect of the drugs on the rabbit SAN among Con, CIHH14 and CIHH28 groups, respectively. RESULTS: (1) CIHH didnt change the parameters of action potential of SAN recorded under basic condition. (2) ISO changed some parameters of AP significantly in a dose-dependent manner, including increases in the amplitude of AP (APA), maximal rate of depolarization (V_(max)), the velocity of diastolic (phase 4) depolarization (VDD), and rate of pacemaker firing (RPF). (3) The response of AP to ISO in CIHH rabbits was decreased significantly compared to that in Con animals. Under 1 μmol/L of ISO, the increases in VDD, RPF, APA and V_(max) in CIHH animals were smaller than those in Con animals (P<0.05). (4) No effect of PE (0.01, 0.1 and 1 μmol/L) on the parameters of action potential was observed. CONCLUSION: CIHH decreases the reactivity of β-AR, but has no effect on α_1-AR in SAN of rabbit.
3.Exploration on graduate management mode based on scientific research innovation platform

In order to strengthen the graduate management in scientific research platform and to ensure the quality of graduate training,the ideological and moral education was invigorated through establishing virtual party branch,the behavior was regulated through establishing and amplifying the daily management system,the student interests were protected through establishing financial management system and the cultivation quality was guaranteed through perfecting the academic management system.Satisfactory results were achieved in molecular medicine and cancer research center in Chongqing Medical University.
4.Construction of Saccharopolyspora erythraea M synthesizing a novel ketolide 3-deoxy-3-oxo-erythronolide B.

Genetic engineering on macrolide antibiotics was a new field in recent years and more than 100 novel polyketides had been produced until then. Using genomic DNA of S. erythraea A226 as a template, about 3.2 kb DNA fragment without KR6 domain was amplified by overlapping PCR technique and cloned into pWHM3 carrier, which resulted in the construction of homologous recombinant plasmid pWHM2201. Plasmid pWHM2201 was introduced into protoplasts of S. erythraea A226 by PEG-mediated transformation and integrated into the gene locus for erythromycin biosynthesis. After integrants grew for two generations on R3M media without Tsr, they were protoplasted and grown on R3M plates. By PCR identification, 8 mutants without KR6 domain were selected out and named S. erythraea M(1-8). With the identification of mass spectrometry, it was proved that S. erythraea M1 synthesized a novel ketolide compound 3-deoxy-3-oxo-erythronolide B.

7.Shielding property of different prosthetic materials to shield radiation of (125)I seed.

OBJECTIVETo investigation the shielding property to (125)I of several different prosthetic materials used in clinical when prostheses are used as carriers of (125)I seed in tumor treatment.
METHODS(125)I seeds were taken as the radiation sources to establish a model in vitro and the radiation doses were measured by thermoluminescent dosemeters (TLD). The shielding property of titanium plate, cochrome plate, and poly methyl methacrylate (PMMA) plate were detected, calculated and compared with that of plumbum plate in the control group.
RESULTSThe radiation doses of the radiation source at 10 mm distance through the following materials were measured, and the results are 2.805 mGy (0.09 mm thick titanium plate), 1.082 mGy (0.48 thick titanium plate), 0.390 mGy (0.41 mm thick cochrome plate), and 0.261 mGy (0.67 mm's cochrome plate), and 2.885 mGy (1.685 mm thick PMMA plate).
CONCLUSIONSThe shield property of cochrome is optimal. The poly methyl methacrylate behaves to some extend as a shield to the (125)I seed which can shield the radiation as the effects of plumbum when adopted together with the 0.557 mm's (or more) titanium plate.

8.Expression and significance of CD147 and E-cadherin in human gastric carcinoma.

OBJECTIVETo investigate the expression of CD147 and E-cadherin in gastric carcinoma and their correlation with clinicopathological features.
METHODSThe expression of CD147 and E-cadherin in gastric cancer tissue chip (TC) was detected by in situ hybridization (ISH) and immunohistochemistry in 220 cases of gastric carcinoma and 31 cases with normal gastric mucosa.
RESULTSThe expression rates of CD147 mRNA, E-cadherin mRNA and E-cadherin protein were 50.5%, 17.7% and 15.5%, respectively. The CD147 expression was negatively correlated with E-cadherin expression. There was a significant relationship between CD147 mRNA expression and tumor diameter, TNM stage, invasion depth, vessel invasion, lymph node and distant metastasis. The E-cadherin mRNA expression was closely related with TNM stage, invasion depth and vascular invasion. There was a significant relationship between E-cadherin protein expression and tumor diameter, TNM stage and vascular invasion. The mean survival time in cases with CD147-positive expression was shorter than that in negative cases, while E-cadherin was in an opposite relationship.
CONCLUSIONIn gastric carcinoma, up-regulation of CD147 and down-regulation of E-cadherin are in a negative correlation. The examination of both these two factors together is useful for predicting the invasion and metastasis of gastric cancer, therefore, can be used as a significant marker to direct clinic therapy and estimation of prognosis.

9.Study on the expression and significance of Galectin-3 and CDC25B mRNA in human gastric carcinoma.

OBJECTIVETo study the expression of Galectin-3 and CDC25B mRNA in gastric carcinoma and their correlation with clinical-pathological features and the survival time.
METHODSTissue microarray (TMA) technique and in situ hybridization were used to detect the expression of Galectin-3 and CDC25B mRNA in 220 gastric carcinoma specimens and 31 normal gastric mucosa samples.
RESULTSIn situ hybridization results revealed that from the 220 cases, the positive expression rate of Galectin-3 and CDC25B mRNA were 58.6% and 54.1%, respectively. There was significant relationship between the Galectin-3 mRNA expression and tumor diameter, advanced TNM stage, invasion depth, vessel invasion, lymph node and distant metastasis. There was significant relationship between CDC25B mRNA expression and tumor diameter, advanced TNM stage, vessel invasion, lymph node and distant metastasis. In addition, there was apositive relationship of Galectin-3 and CDC25B mRNA expression. Finally, the mean survival time in cases with Galectin-3 and CDC25B mRNA positive expression was significantly shorter than those without Galectin-3 and CDC25B expression.
CONCLUSIONThe expression of Galectin-3 and CDC25B mRNA appears to act as a promoting factor in the onset and development of gastric cancer. It can be used as a marker of prognosis of gastric carcinoma in clinical practice.

10.Polydatin attenuates ischemia/reperfusion-induced apoptosis in myocardium of the rat.

The aim of the present study was to investigate the effect of polydatin on apoptosis induced by ischemia/reperfusion (I/R) in rat myocardium and to explore the underlying mechanism. Adult male Sprague-Dawley (SD) rats were randomly divided into control, I/R and polydatin (50 mumol/L) groups. On the Langendorff apparatus, isolated rat heart was subjected to 30-min global ischemia followed by 60-min reperfusion. TUNEL labeling and flow cytometric techniques were used for the measurement of apoptosis and the expression of Bcl-2 and Bax protein in cardiomyocytes of rat. The results showed: (1) Compared with those in the control group, the number of TUNEL-positive cells and apoptosis rate were increased in I/R group; (2) Compared with that in the I/R group, the number of TUNEL-positive cells was significantly decreased in the polydatin group [(18.1+/-4.0)% vs (35.1+/-5.4)%, P<0.01]; (3) Apoptosis rate assayed by flow cytometry in I/R group was significantly higher than that in polydatin group [(15.43+/-4.55)% vs (8.66+/-3.18)%, P<0.01]; (4) Expression level of Bax protein was higher in I/R group than that in polydatin group (P<0.05), while the level of Bcl-2 protein and Bcl-2/Bax ratio were higher in polydatin group than those in I/R group (P<0.05, P<0.01), respectively. The results obtained suggest that polydatin exerts an inhibitory effect on I/R-induced apoptosis through increasing Bcl-2 protein expression and decreasing Bax protein expression in myocardium of the rat.
