1.Correlation between 1H-MRS and the expression of VEGF,MMP-9 and uPA in brain glioma

Objective To evaluate the relationship between 1H-MRS features of brain glioma and its pathological grade and invasiveness, and the correlation between those features and the expression of VEGF, MMP-9 and uPA.Methods The brain gliomas in 55 patients confirmed by pathology were divided into two groups: high grade and low one.An analysis of intragroup and intergroup differences in some metabolite ratios of 1H-MRS in the tumor parenchyma was conducted.The expression of MMP-9,VEGF and uPA in brain glioma was detected,and the correlation between the above three parameters and changes in metabolite ratios of 1H-MRS were explored.Results There was significant difference and correlation in Cho/Cr and Cho/NAA ratio in the tumor parenchyma between high and low grade(P<0.01).The expression of VEGF and MMP-9 in tumor was significantly different(P<0.01) but with significant correlation(P<0.01),and there was no significant difference and correlation in NAA/Cr ratio and uPA expression(P>0.05).Conclusion VEGF and MMP-9 play an important role in the development of brain glioma.Cho/Cr and Cho/NAA ratios provide an important reference for preoperative grading of brain gliomas and indirectly reflect the expression of VEGF and MMP-9.
2.Yupingfeng granule combined with citric acid and Desloratadine Citrate Disodium tablets in the treatment of allergic rhinitis and its effects of immune function

Objective To investigate the effect ofYupingfeng granule combined with Desloratadine Citrate Disodium tablets on the clinical efficacy and immune function in patients with allergic rhinitis.Methods A total of 112 patients with allergic rhinitis were randomly divided into two groups according to their order of treatment. The patients in the control group (n=56) were treated with Desloratadine Citrate Disodium tablets, and the patients in the experimental group (n=56) were treated withYupingfeng granule and Desloratadine Citrate Disodium tablets. The efficacy of clinical, the rate of adverse reaction, the improvement of immune and the rate of recurrence were compared between the two groups. Results The total effective rate of the experimental group was 94.64%, it is higher than the control group 85.71%, and the difference was statistically significant (Z=-3.254,P<0.01). After the treatment, runny nose (1.2 ± 0.1vs. 2.0 ± 0.2,t=6.420), nasal itching (1.2 ± 0.3vs. 1.7 ± 0.3,t=7.003), sneezing (0.8 ± 0.1vs. 1.3 ± 0.1,t=6.716), stuffy nose (1.1 ± 0.1vs. 1.6 ± 0.3,t=7.256)scores in the experimental group were significantly lower than those in the control group (P<0.05). There was no significant difference in the total IgE (114.48 ± 9.85 IU/mlvs. 130.64 ± 10.51 IU/ml,t=6.994) and VCAM-1(512.56 ± 13.84 μg/Lvs. 624.98 ± 7.06 μg/L,t=7.803) between the two groups (P<0.05). The levels of IL-4(16.25 ± 1.53 ng/Lvs. 22.38 ± 3.16 ng/L,t=5.458), IL-10 (15.98 ± 2.43 ng/Lvs. 21.82 ± 1.75 ng/L, t=6.149), LTB4 (59.14 ± 0.96 ng/Lvs. 67.46 ± 3.04 ng/L,t=5.342) in the experimental group were significantly lower than those in the control group (P<0.05). Six-month after the treatment, the relapse rate of the experimental group was 5.36%, while the control group was 16.07%, and the difference between two groups were significant (P<0.05).Conclusions The combination ofYupingfeng granule and Desloratadine Citrate Disodium tablets in the treatment of allergic rhinitis can significantly improve the clinical symptoms, adjust the immune function and reduce the recurrence of the disease.
3. Clinical significance of cytogenetic monitoring in chronic myeloid leukemia

Objective:
To analyze the association of cytogenetic abnormalities with the prognosis of chronic myeloid leukemia (CML) patients in tyrosine kinase inhibitors (TKI) era.
Methods:
Karyotype analysis of chromosome G-banding was carried out in 387 newly diagnosed CML patients by short-term culture of bone marrow cells. The correlation of cytogenetic abnormalities and CML progression was explored in combination with ABL tyrosine point mutations.
Result:
Of 387 patients with positive BCR-ABL fusion gene assayed by fluorescence in situ hybridization (FISH) technique, 94.1% (364/387) patients were Ph positive and 5.9% (23/387) Ph negative; 320 patients (87.9%) had a translocation t (9;22) (q34;q11) and 5 (1.4%) a variant translocation t (v;22) . Additional cytogenetic aberrations (ACA) at diagnosis were found in 10.7% (39/387) Ph+ patients, major route ACA in 22 (56.4%) cases and minor route ACA in 15 (38.5%) cases and 2 patients (5.1%) lacked the Y chromosome (−Y) ; 23.4% (71/303) patients occurred ACA during TKI treatment and the most frequent abnormalities were abnormal chromosome numbersd, which were likely associated with high proportion of disease progression (
4.Effects of Medial Protrusio Technique of Total Hip Arthroplasty on Acetabular Cup in Adult Patient with Developmental Dysplasia of the Hip:A Finite Element Analysis

Objective To investigate the biomechanical effects of the medial protrusio technique on the acetabular cup in adult patients with developmental hip(DDH)after total hip arthroplasty.Methods The CT scanning data of bilateral hips from an adult patient with unilateral DDH were obtained further to develop a finite element model of the affected hemipelvis.The medial protrusio technique with various levels of medial protrusio was simulated,and the biomechanical differences between the medial protrusio and non-protrusio groups were evaluated.Results In the simulated pull-out test,the maximum anti-pull-out load strength of the non-protrusio group was 1 166 N.Compared with the non-protrusio group,the anti-pull-out load strength of the 4 mm and 8 mm medial protrusio groups increased by 45.8%and 57.1%,respectively.The peak micromotion at the cup-bone interface for the non-protrusio group was 166.4 μm in the standing phase of the gait cycle,and that of the 4 mm and 8 mm medial protrusio groups was decreased by 46.2%and 62.1%,respectively.Regarding the immediate stress distributions of periacetabular bone tissues following cup implantation,the differences between the groups were not significant.Under the loading condition of the standing phase,the non-protrusio group yielded the lowest average and peak stresses.The average stress increased with the level of medial protrusio,and the highest peak stress was observed in the 4 mm medial protrusio group.Conclusions The medial protrusio technique can improve the initial stability of the acetabular cup,and the initial stability is positively proportional to the protrusio level.However,owing to the concentration of marginal stress at the cup-bone interface,a minor medial protrusio cup with insufficient bone coverage might increase the risk of various prosthesis-related complications.
5.Cloning and Functional Characterization of Farnesyl Diphosphate Synthase Gene in Biosynthesis of Terpenoid Components in Chinese Materia Medica

ObjectiveThis study aims to enhance of the farnesyl pyrophosphate(FPP) pool in Saccharomyces cerevisiae by heterologously expressing different farnesyl diphosphate synthases(FPSs) from various plants, thereby increasing the production of terpenoid compounds by the engineered yeast. MethodsRNA from mixed samples of roots, stems, and leaves of seven plants including Arabidopsis thaliana, Rosa rugosa, Artemisia annua, Centella asiatica, Humulus lupulus, Medicago sativa, and Panax ginseng was extracted by column chromatography and reverse transcribed into the first strand of complementary DNA(cDNA), and based on the transcriptome data of the seven species of plants, sequence-specific primers were designed for CaFPS, RrFPS, MsFPS, HiFPS, PgFPS, AtFPS, and AaFPS, the full-length of the genes was cloned, and the genes were analyzed for bioinformatics in order to construct a pESC yeast shuttle vector. These seven plant-derived FPSs were further heterologously expressed in the previous constructed β-elemene-producing yeast, and the yield of β-elemene was indicated for their catalytic acivities. ResultsThe coding sequences of CaFPS, RrFPS, MsFPS, HiFPS, PgFPS, AtFPS, and AaFPS were all of 1 021 bp in length and encoding 301 amino acids, all of which were similarly related to the endogenous FPS-encoding gene(ERG20) in S. cerevisiae. After heterologous expression, RrFPS was identified as the most effective in catalyzing the synthesis of FPP from isopentenyl pyrophosphate(IPP) and dimethylallyl pyrophosphate(DMAPP). Compared to the control strains, the RrFPS overexpressed yeast strains YB-1-Rr and YB-3-Rr increased the production of β-elemene by 231.25% and 189.3%, respectively. ConclusionBy comparing the functions of FPS-encoding genes from seven different plant sources, it is determined that the protein encoded by the RrFPS from R. rugosa has the best catalytic ability, which can provide key genetic elements for the construction of engineered yeast strain constructs with high terpenoid production.
6.Effect of poria cocos on gastrointestinal motility in mice

Objective To investigate the regulatory effect of poria cocos on gastrointestinal motility in mice. Methods A total of 130 Kunming mice were randomly divided into negative control group, low-dose and high-dose groups of raw poria cocos powder, low-dose and high-dose groups of cooked poria cocos powder, low-dose and high-dose groups of poria cocos surrogate culture powder, low-dose, medium-dose and high-dose groups of poria cocos water extract, and low-dose, medium-dose and high-dose groups of poria cocos alcohol extract, with 10 mice in each group. The animals were administered by gavage for 7 days, once a day. After the last administration, the intestinal propulsion function test and gastric solid emptying test were conducted to observe the regulating effect of poria cocos on gastrointestinal motility of mice. Results Compared with the negative control group, the small intestine propulsion rate in the low-dose group of poria cocos surrogate culture powder was significantly increased (P<0.01). Except the high-dose group of raw poria cocos powder, the other poria cocos groups had higher gastric residual rate (P<0.05). Conclusion Poria cocos does not promote intestinal propulsion of mice under normal physiological condition, but it can inhibit gastric empting and exert a moderating effect on gastrointestinal function in normal mice.
