2.The effects of cryopreservation on the immunogenicity of HUVEC

Objective To study the effects of cryopreservation on the immunogenicity of human umbilical vein endothelial cells(HUVEC).Methods HUVEC were isolated ex vivo and cryopreserved.Lymphocyte stimulation index(SI)was analyzed by MTT in lymphoeyte-endothelial cell co-culture.Both HLA-ABC and HLA-DR antigen expression on fresh or cryopreserved HUVEC,and the effects of IFN-γ treatment on HLA antigen expression in both groups were determined by using flow cytometry.Results No difference in SI was observed between fresh prepared and cryopreserved HUVEC(1.716±0.181 vs 1.686±0.145,P>0.05).The percentage of HLA-ABC expression was(96.6±1.9)%and(96.0±1.4)%in fresh and cryopreserved HUVEC(P>0.05),and the mean intensity for HLA-ABC expression was 84.1±5.7 and 82.4±4.8 in fresh and cryopreserved HUVEC(P>0.05),respectively.However,no HLA-DR expression was observed in both groups.When treated with IFN-γ,HLA-ABC expression was significantly up-regulated,and HLA-DR expression was induced in a dose-dependent manner.No significant difference was found in the HLA-ABC expression between fresh and cryopreserved HUVEC(P>0.05),while the HLA-DR expression in cryopreserved HUVEC was remarkably lower than in fresh HUVEC with the increase of IFN-γ(P<0.01).Conclusion The immunogenicity of HUVEC remains stable by cryopreservation without IFN-γtreatment or treated with low concentration of IFN-γ(≤50 U/ml).However,the HLA-DR expression in HUVEC was remarkably reduced in eryopreserved cells treated with a high concentration of IFN-γ(≥100 U/ml).These data indieated that the effects of cryopreservation on immunogenicity of HUVEC may result from the decreased responses of HLA-DR expression by the stimulation of IFN-γ treatment.
3.Effectiveness of metronidazole combined with cefuroxime sodium in the laparoscopic treatment of patients with acute non-simple appendicitis

Objective To investigate the application value of metronidazole combined with cefuroxime sodium in the laparoscopic treatment of patients with acute non-simple appendicitis.Methods The subjects were 80 patients with acute non-simple appendicitis who underwent laparoscopic appendectomy from July 2020 to June 2023.Among them,41 patients with acute non-simple appendicitis who were treated with metronidazole combined with cefuroxime sodium for laparoscopic surgery were used as the observation group,and 39 patients with acute non-simple appendicitis who were treated with metronidazole combined with ceftriaxone sodium for laparoscopic surgery were used as the control group.The intraoperative situation,inflammation control before and after surgery,postoperative recovery status,and adverse reactions after medication were compared between the two groups.Results There were no statistically significant difference between the two groups in terms of periappendiceal abscess/peritonitis,surgical time,and intraoperative bleeding,the differences were not statistically significant(P>0.05);The inflammatory indexes of acute non-simple appendicitis in both groups gradually returned to normal after surgery,and the white blood cell count,procalcitonin(PCT)and C-reaction protein(CRP)levels in the observation group showed greater decrease than those in the control group,the differences were statistically significant(P<0.05)on the first day and the third day after surgery.The duration of analgesics use and hospital stay in the observation group were(1.8±1.2)and(3.5±0.7)d,respectively,which were significantly shorter than those of(2.7±1.4)and(4.6±0.4)d,the differences were statistically significant(P<0.05).The incidence of intra-abdominal infection in the observation group was 7.7%,which was significantly lower than that of 24.4%in the control group,the difference was statistically significant(P<0.05).The adverse reactions in the two groups were 4.9%and 7.7%,respectively,and the difference was not significant(P>0.05).Conclusion Metronidazole combined with cefuroxime sodium combined with laparoscopic appendectomy can significantly reduce the incidence of abdominal infection in patients with acute non-simple appendicitis,it has high safety.
4.Identification of host proteins interacting with African swine fever virus inner envelope protein p17.

African swine fever virus (ASFV) infection leads to a mortality rate of up to 100%, causing devastating disasters to the pig industry. Understanding the ASFV infection and replication is therefore of great importance. ASFV has more than 150 open reading frames, among which the inner coat protein p17 encoded by the D117L gene is involved in the formation of the icosahedral structure of the virus. However, little is known about the mechanism how p17 regulates host cell function. In this study, the potential host proteins interacting with ASFV p17 were screened by immunoprecipitation technique combined with protein profiling analysis. The interactions of p17 with mitochondrial membrane protein TOMM70 and heat shock protein HSPA8 were confirmed by co-immunoprecipitation technique and laser confocal experiments. This study provides important information for further exploring the function of p17 during ASFV infection.
