1.Experimental Study on Effect of Huangqi Danshen Drop Pill on Pituitary - Induced Acute Myocardium Ischemia

Objective To observe the effect of Huangqi Danshen Drop Pill(HDDP) on pituitary (Pit) induced acute myocardium ischemia. Methods Acute myocardium ischemia rat model was established by sublingual venous injection of Pit(0. 35U/kg). The changes of the T wave ( II channel ECG) and serum contents of lactate dehydrogenase (LDH), CK, superoxide dismutase (SOD) and malondialdehyde (MDA) were observed in various times after Pit injection. Results HDDP could significantly lower the height of T waves, increase SOD activity and decrease MDA content. Conclusion HDDP can improve acute myocardium ischemia induced by Pit.
2.Effect of gax gene on the expression of PCNA and CDK2 in vascular smooth muscle cells

Objective To investigate the role of proliferating cell nuclear antigen (PCNA) and cyclin dependent kinase 2 (CDK2) in inhibition of vascular smooth muscle cells (VSMCs) proliferation by gax gene. Methods After being transfected by AdCMV-gax, the expression of gax, PCNA and CDK2 in VSMCs were assessed. The effect of gax overexpressions on VSMCs proliferation was observed by 3 H-thymidine incorporation. Results After AdCMV-gax was transfected to VSMCs, the level of Gax protein expression was significantly higher than that before transfection. PCNA and CDK2 expressions were decreased after VSMCs were transfacted with AdCMV-gax. The level of 3 H-thymidine incorporation was decreased significantly in with AdCMV-gax transfected VSMCs compared with that in non-transfected VSMCs. Conclusion The mechanism of inhibition of VSMCs proliferation by gax gene is related with depression of expression of PCNA and CDK2.
3.The calculating method of space magnetic induction of circular permanent magnet in the field of magnetic biologY efiect.

Formulas, which are applied to the space magnetic induction of circular permanent magnet, are deduced from BiotSavart-Laplaces law in electromagnetics. The calculating methods of the formulas are set up with numerical integral and computer program. The space quantity distribution of magnetic induction of a circular permanent magnet is obtained through an objective example. These provide theory basis for the magnet application in the field of magnetic biology effect.
5.Control study on skull to boneless head CTA by surface shaded display technique and volume computed tomographic digital subtraction angiography in diagnosing AVM

Objective To investigate the diagnostic value of technologies for arteriovenous malformation(AVM ) ,which are skull to boneless head CTA by surface shaded display technique (hereinafter referred to as the SSD‐CTA technology) .Methods Totally 27 patients of the First Affiliated Hospital of Chongqing Medical University were selected ,which were diagnosed with AVM by VCTDSA .The original data to our hospital CT workstation were imported ,SSD‐CTA technology to bone reconstruction were performed ,and the data of the two groups were compared .Another 5 patients in our hospital which were diagnosed with AVM by SSD‐CTA examination were selected ,and compared the results with DSA .Results The quality score of SSD‐CTA reconstructed image was less than that of VCTDSA ,but the diagnosis of the two technologies for AVM had no significant statistical difference (P>0 .05) .The number of AVM patients from our hospital wsa too less to do the statistical analysis .The detail resolution of SSD‐CTA was less than DSA ,but the diagnosis of SSD‐CTA was consistent with DSA .Conclusion SSD‐CTA has clinical practical val‐ue ,can be used as a check ,intracranial venous malformation diagnosis .

An ultra-performance liquid chromatography coupled with time-of-flight mass spectrometry (UPLC/Q-TOF-MS)-based chemical analytic technology was used to evaluate the chemical constitution of Radix Aconiti Lateralis Preparata in the process of decocting, so as to provide a scientific basis for processing Radix Aconiti Lateralis Preparata.
7.Study on the application of bio-heat transfer science to the quantitative analysis of tongue inspection

OBJECTIVE: To research the mechanism of the tongue inspection in traditional Chinese medicine and to study the special relationship between temperature distribution of the tongue and the tongue color. METHODS: The temperature field of human and pig's tongue was measured by infrared thermal vision system and thermal-couple probe. The blood perfusion rates in the human tongue and the pig's tongue were obtained with laser Doppler rheometer and CBI-8000 physiology research system. The temperature field of the profile of tongue was computed by numerical method. RESULTS: The quantitative curve of the temperature distribution on the surface of tongue and the relationship between temperature and blood perfusion rate were obtained. And the temperature in the profile of tongue was obtained by finite element method. CONCLUSION: The research involves that the different tongue color can reflect different tongue temperature and blood perfusion rate. This proved that it is acceptable to apply the bio-heat transfer theory to the tongue inspection research. The results provided a new method to research the tongue inspection in traditional Chinese medicine.
8.Effects of berberine on the tumor-associated macrophages of intestinal polyps in Apc (Min/+) mice

Objective To investigate the effects of berberine on tumor-associated macrophages (TAM)and the expression of cyclooxygenase-2 (COX-2)of intestinal polyps in Apc(Min/+) mice.Methods A total of 20 Apc(Min/+) mice,four weeks old,were equally divided into the control group and the berberine group,10 in each group.The mice of the control group drank plain water,while the mice of berberine group drank water with 0.1 % berberine.After 12 weeks,all the mice were sacrificed.The intestine and colon were isolated,and the numbers of polyps were counted.The expression of F4/80,inducible nitric oxide synthase-2 (iNOS),macrophage mannose receptor (MR)and COX-2 was detected by immunohisto-chemistry method.The relative expression of COX-2 at protein level was measured by Western blotting. The t test was performed for comparison between two independent groups.Results The total number of intestinal polyps,the number of small intestinal polyps and the number of colon polyps of the berberine group (11 .50±2.05 ,10.50±1 .77 and 1 .00±0.46,respectively)were all less than those of the control group (30.63±1 .69,28.00±2.00 and 2.63±0.74,respectively),and the differences were statistically significant (t=16.727,16.952 and 3.162,P =0.001 ,0.001 and 0.010,respectively).The percentage of F4/80 positive cells in the stroma of polyps of the berberine group ((17.40 ±4.23 )%)was less than that of the control group ((31 .24±6.34)%),and the difference was statistically significant (t =5 .327, P =0.043).The percentage of iNOS positive cells in the stroma of polyps of the berberine group ((7.43± 1 .78 )%) was higher than that of the control group ((2.72±0.68)%), and the difference was statistically significant (t=7.335 ,P =0.004).The percentage of MR positive cells in stroma of polyps of the berberine group ((19.52±1 .54)%)was less than that of the control group ((12.63±0.68)%),and the difference was statistically significant (t=5 .634,P =0.016).The percentage of COX-2 positive cells in stroma of polyps of berberine group ((3.38 ± 0.51 )%)was less than that of the control group ((7.60±0.57 )%),and the difference was statistically significant (t = 7.234,P = 0.001 ).The relative expression of COX-2 at protein level of polyps of the berberine group was lower than that of the control group. Conclusion Berberine may take the role in inhibiting the growth of intestinal polyps in Apc(Min/+) mice through interfering the differentiation of TAM in polyps and suppression the expression of COX-2.
9.Elucidation of compatibility principle and scientific value of Chinese medical formulae based on pharmacometabolomics.

By using UPLC coupled with MS, the analysis of the constituents absorbed into blood after oral administration of Yinchenhao decotion with different combination had been carried out, the body fate of constituents detected in the blood was elucidated also; At the same time, the overall body respones after multi-input of the medical formule were evaluated by metabolomic analysis. These data explained the scientific values in both drug metabolisim and metabonomics, the results had given some new evidences for the potentialities of pharmacometabonomics to elucidate the compatibility principle of Chinese medical formule. As a conclusion, concept of "Pharmacometabolomics of Chinmediformulae" has been defined.

10.Study on the induction of drug metabolizing enzyme CYP1 A1 activity by Ginsenoside Rc,Re,Rf and Rg1

Aim To investigate the induction effect of ginsenoside Rc, Re, Rf and Rg1 on CYP1A1, and further validate the role of aryl hydrocarbon receptor in CYP1A1 expression. Methods Dual luciferase re-porter gene system was performed. Four kinds of gin-senoside were screened for aryl hydrocarbon receptor activation by reporter assays, and TCDD as the positive control. Further with different concentrations of ginsen-oside Rc, Re, Rf and Rg1 treated on LS174T cells, RNA and total protein were extracted to detect the reg-ulating effect of ginsenosides on CYP1 A1 mRNA and protein expression with Real-time PCR and Western blot technology respectively. Results Reporter gene screening showed that the ginsenoside Rc, Re, Rf and Rg1 could activate AhR and had potential effects on the induction of CYP1A1 enzyme. Meanwhile, dose-de-pendent induction of the gene expression were observed in response to ginsenoside Rc, Re, Rf and Rg1 and the levels of CYP1 A1 protein expression were increased by ginsenoside Rc, Re, Rf and Rg1 in varying de-grees. Conclusion Ginsenoside Rc, Re, Rf and Rg1 can up-regulate the gene and protein expression of CYP1 A1 possibly via the AhR-mediated CYP1 A1 path-way.
