1.Preparation nucleic acid vaccine of brucella melitensis and its immune effect

Abstract Objective:To construct bmcella melitensis Br.melitensis genomic DNA expression library and study its immune effect.Meth-ods: Hind Ⅲ digested Br.melitensis genomic DNA fragments were cloned into pSV-?-Gal plasmds.The recombinant plasmids were transformedinto JM109 host bacteria. After large-scale isolation, the recombinant plasmids were inoculated into the quadriceps muscle of mice. In the 9thday after third boost immunization .agglutination antibodies against Br. melitensis and lymphocyte mitogenic ability of mice were determined. Re-sults : Expression library nucleic acid vaccine can induce the production of agglutination antibodies against Br. melitensis and stimulate the lym-phocyte porliferation primed by ConA. Conclusion:The genomic DNA expression library may provide a new vaccine against Br. melitensis infec-tion.
2.Prevalence and correlation of anemia in kidney transplant recipients

Objective To evaluate the prevalence,management and risk factors of post transplantation anemia (PTA) in a group of Chinese kidney transplant recipients after a long-term follow-up. Methods The clinical data of 154 adult kidney transplant recipients,who were followed up for at least 5 years,were retrospectively studied,including yearly hemoglobin (Hb) level,the prevalence and treatment of anemia,state of renal function,use of immunosuppressive agents and other medications. Patients were divided as anemic and non-anemic groups,and comparison of clinical data was carried out between the groups. The incidence of anemia was calculated and management of anemia was recorded. Monofactorial correlation analysis was used to determine the factors associated with Hb level,and multi-variation logistic regression was used to identify the factors related to the onset of anemia at 1 and 5 years post-transplant. Results The annual incidence of PTA at the current year of and 1 to 5 years after transplantation was 45.5%,10.7%,9.6%,14.8%,13.5% and 19.5%,respectively. About 38.3% of the patients suffered from anemia at least once during the 5 years of follow-up period,and 42% of these patients developed anemia recurrently. Correlation analyses indicated that Hb levels were associated with graft function (P0.05). 3.Effects of hydrogen-rich saline on early diabetic neuropathic pain in rats

Objective To evaluate the effects of hydrogen-rich saline on the early diabetic neuropathic pain (DPN) in rats.Methods Healthy male Sprague-Dawley rats,aged 8 weeks,weighing 180-220 g,were used in the study.DPN model was made by intraperitoneal injection of 1% streptozocin (STZ) 65 mg/kg.Twelve diabetic rats were randomly divided into 2 groups (n =6 each) using a random number table:DPN group and hydrogen-rich saline group (HRS group).Another 6 normal rats were randomly collected as control group (group C).At 14 days after STZ injection,hydrogen-rich saline 5 ml/kg was injected intraperitoneally once a day for 14 consecutive days in HRS group,while C and DNP groups received the equal volume of normal saline.Mechanical paw withdrawal threshold to yon Frey stimuli (MWT) and paw withdrawal latency to nociceptive thermal stimulation (TWL) were measured at 2 days before STZ injection (To) and 7,14,21 and 28 days after STZ injection (T1-4).The motor nerve conduction velocity (MNCV) of the right hindlimb and distal motor latency was measured after pain threshold was measured at T4.After measurement of neurological function was completed,tumor necrosis factor-α (TNF-α) and i nterleukin-6 (IL-6) contents (by ELISA) and nuclear factor kappa B (NF-κB) activity (by immuno-histochemistry) were detected.Results Compared with group C,MNCV was significantly decreased,the motor latency was prolonged,MWT was decreased at T1-T4,TWL was shortened at T2-T4,TNF-a and IL-6 contents were increased,and NF-κB activity was enhanced in DNP and HRS groups (P < 0.05).Compared with group DNP,MNCV was significandy increased,the motor latency was shortened,MWT was increased at T3.4,TWL was prolonged at T4,TNF-a and IL-6 contents were decreased,and NF-κB activity was weakened in group HRS (P < 0.05).Conclusion Hydrogen-rich saline can relieve the early DPN through inhibiting NF-κB signaling pathway in rats.

7.Management of zygomatic complex fractures

砄bjective: To study the cause,clinical characteristics and treatment of zygomatic complex fractres(ZCF). Methods: Medical records of 79 cases with ZCF and followed up for 3 months to 2 years were reviwed and analysed. Reduction and cosmatic outcome were assessed by clinical evaluation and postoperative images. Results: Of the 79 patients, 87.3% were male, with the peak incidence in the age group from 20 to 40 years old(73.5%). 58.2% were due to traffic accidents, 15.2% to assalts. Incidence of malar asymmetry, enophthalmos, diplopia,limited mandiblar movement and malocclusion were 83.5%,24.1%,19.0%, 60.8% and 35.4% respectively. Of the nerve disturbances, 41.8% of the infraorbital and 13.9% of the facial nerve were involved. For the treatment the most freqently used approach was the coronal incision in combination with the maxillary vestibular incision(30.4%), followed by coronal incision alone(22.8%). 40.5% of the patients were treated by rigid internal fixation with titanium miniplates alone or combined with wire fixition(20.3%). All patients but five regained satisfactary facial appearance. Three patients still showed slightly post surgical enophthalmos and diplopia at a 6 month follow up. All of the patients with malocclusion regained their functional occlusion after treatment. Conclsions:A variety of techniques can be used in the treatment of ZCF. Among them the coronal incision in combination with other incisions or alone and rigid internal fixation can produce a satisfactary outcome.
8.Changes of Activated T Cell Disfunction in Children with Refractory Mycoplasma Pneumoniae Pneumonia

Objective To study disfunction of activated T cell from peripheral blood and explore its role on pathogenesis in patients with refractory mycoplasma pneumoniae pneumonia (MPP).Methods T cell subsets and expression of membrane molecule on activated T cell in peripheral blood were measured in 26 refractory MPP patients by using immunofluorescence and flowcytometry,and compared with those of 18 control healthy children.Results The percentage of CD4~+ cell from peripheral blood in refractory MPP children was significantly lower than that in control group(P0.05).Conclusions There are disfunctions of T cell in refractory MPP patients in acute period,and the abnormal T cell activated disfunction may play an important role on pathogenesis.
9.Clinical efficiency of transurethral non-muscle invasive resection or electrocautery of bladder tumor under local anesthesia

Objiective To assess the feasibility of transurethral non-muscle invasive resection or electrocautery of bladder tumor (TURBT) under local anesthesia. Methods Ten patients with non-muscle invasive bladder cancer (NMIBC) history, who were confirmed NMIBC relapse by imaging and cystoscopy were selectively implemented TURBT or electrocautery under local anesthesia. Patients′anesthesia and tolerance under different local anesthesia were observed. Results All surgeries were completed successfully with clear base and without residual tumor. Tumor tissues were pathologically confirmed NMIBC. Patients were well tolerated to local anesthesia. The situation of pain scores were 0 point in 2 cases, 1 point in 4 cases, 2 points in 3 cases and 3 points in 1 case. Conclusions For NMIBC patients with poor surgery tolerance and high anesthetic risk and on the basis of low pressure on the bladder, selectively implementing TURBT or electrocautery under sufficient urethral anesthesia is feasible from the perspective of cancer cure and patients′tolerance.
10.Study on the characteristics and hepatotropism of the negative regulatory element of the HBVnt453-250

Objective To analyze the characteristics and hepatotropism of this negative element HBVnt453-250 sequence.Methods pHBv453-250,pHBV250-453.plucHBV453-250 and plucHBV250-453 were constructed,with luciferase and enhanced green fluorecence protein(EGFP)gene as the reporter gene,respectively.After transfection of HepG2 cells with these plasmids,luciferase assays,real-time PCR and western blot assays were used to detect the gene transcription and expression level.The SV40 promoter of pGL3 control and pHBV453-250 were replaced by the cytomegalovirus early promoter,resulting in plasmids pCMVcontrol(luc)and pCMV453-250(luc).Results Compared with pHBV453-250,the mutant plasmids.with the inhibitory element inserted in difierent site or inverted orientation.exerted similar downregulation of Juciferase gene transcription and expression.Western blot analysis demonstrated the similar repression when EGFP was used as the reporter gene.By transfeeted to HepG2 cell line,the plasmid pCMV453-250(1UC)could reduce lneiferase activity(36.56%)compared with pCMLcontrol(luc).When the plasmids plueHBV453-250 and plucHBV250-453 were transfected to non-liver cell lines(A549,HeLa),luciferase gene was expressed weakly,compared with that of pGL3control(P<0.05).Conclusion The inhibitory effect of HBVnt453-250 sequence acted in both orientation-and position-independent manners,and had no promoter selectivity and funotioned in hepatocyte-independent manner.
