1.Quantification of neomangiferin in rat plasma by liquid chromatography-tandem mass spectrometry and its application to bioavailability study$

Neomangiferin, a natural C-glucosyl xanthone, has recently received a great deal of attention due to its multiple biological activities. In this study, a rapid and sensitive ultra-high performance liquid chroma-tography tandem mass spectrometry (UHPLC–MS/MS) method for the quantification of neomangiferin in rat plasma was developed. Using chloramphenicol as an internal standard (IS), plasma samples were subjected to a direct protein precipitation process using methanol (containing 0.05% formic acid). Quan-tification was performed by multiple reactions monitoring (MRM) method, with the transitions of the parent ions to the product ions of m/z 583.1-330.9 for NG and m/z 321.1-151.9 for IS. The assay was shown to be linear over the range of 0.2–400 ng/mL, with a lower limit of quantification of 0.2 ng/mL. Mean recovery of neomangiferin in plasma was in the range of 97.76%–101.94%. Relative standard deviations (RSDs) of intra-day and inter-day precision were both o 10%. The accuracy of the method ranged from 94.20%to 108.72%. This method was successfully applied to pharmacokinetic study of neomangiferin after intravenous (2 mg/kg) and intragastric (10 mg/kg) administration for the first time. The oral absolute bioavailability of neomangiferin was estimated to be 0.53%7 0.08%with an elimination half-life (t1/2) value of 2.74 7 0.92 h, indicating its poor absorption and/or strong metabolism in vivo.
2.Clinical observation of arthroscopic debridement with injecting chuanxiongqin in treating knee osteoarthritis

Objective To study the clinical therapeutic effect of arthroscopic debridement cooperated with injecting chuanxiongqin in treating knee osteoarthritis. Methods Sixty cases of knee osteoarthritis were randomly divided into 2 groups: treating group and control group, 30 cases in each group. The treating group were treated by arthroscopic debridement cooperated with injecting chuanxiongqin;the control group were given hyaluronate-Na after received arthroscopic debridement. The operation evaluation criteria of JOA was taken to value the therapeutic effect at 1 week, 3 months, 6 months and 12 months postoperation. At the same time, nitric oxide (NO) content in joint fluid were measured at preoperation, 1 week, 4 weeks, 3 months postoperation. Results At 1 week postoperation, there were no statistically significant differenced > 0.05) between the two groups,at 3, 6 and 12 months postoperation, the therapeutic effect of the treating group were much better than those of the control group (P < 0.01);NO content in joint fluid of preoperative treatment group [(90.32 ± 22.24) μmol/L]and control group [(90.31 ±22.20) μ mol/L]was similar, there was no significant difference (P >0.05). At 3 months postoperation, NO content in joint fluid of the treating group [(85.63 ± 30.63) μ mol/L]decreased significantly compared with preoperative (P < 0.05 ) , and the content was much lower than that of the control group [(89.66 ± 20.63 ) μ mol/L], there was significant difference between the two groups (P < 0.05). Although NO content in joint fluid of the control group was decreased, but the difference was not statistically significant (P >0.05). Conclusions Arthroscopic debridement with injecting chuanxiongqin in treating knee osteoarthritis can control inflammatory reaction of cartilage and synovia]in effect, improve the therapeutic effect. Therefore, it should be avalid method in treating knee osteoarthritis.
3.The effect of Qibaipingfei capsule on right ventricular hypertrophy index(RVHI)of rats with phlegm and blood stasis of COPD

Objectlve To explore the effect of Qibai Pingfei capsule on the right ventricular hypertrophy index (RVHI)of rats with phlegm and blood stasis of COPD.Methods 60 rats were randomly divided into 6 groups,a control group,a model group,a Ligustrazine group,a Nifedipine group,a high dose Qibai Pingfei capsule group and a low dose Qibai Pingfei capsule group.Composite factors method was adopted to establish phlegm and blood stasis COPD rat model.At the same time of modeling.Qibai Pingfei capsule,Ligustrazine,Nifedipine were also given to these rats.Observed the changes of RVHI in each group.Results RVHI did not show statistic difierence between high dose of the Qibai Pingfei capsule group and the control group(P>0.05).and between the Nifedipine group and the model group(P>0.05).RVHI manifested significant diffbrencc between the Ligustrazine group and the low dose Qibai Pingfei capsule group(P<0.01),and between the Ligustrazine group and the control group(P<0.01).Conclusion High dose Qibai Pingfei capsule can effectively prevent the right ventricular hypertrophy of model rats with the phlegm and blood stasis of COPD.Ligustrazine has certain effects but not as good as high dose Qibai Pingfei capsule.Nifedipine can not prevent the right ventricular hypertrophy of model rats.
4.Effects of rehabilitation on patients dependent on mechanical ventilation

Objective To explore weaning dependent patients off mechanical ventilation through rehabilitative treatment. Methods Eighty-four patients dependent on mechanical ventilation were randomly divided into a rehabilitation group (n =44) and a control group (n =40). All were treated with routine medication, but in addition patients in the rehabilitation program followed a rehabilitation program. Three weeks later, the success rates and the time of weaning off mechanical ventilation were observed in the two groups. Results In the rehabilitation group,88.6% of the patients were successfully weaned off mechanical ventilation, versus 60% of the control group. The respective average weaning times were 6.8 ± 3.8 days and 14.3 ± 5.2 days. Both differences were statistically significant. Conclusions Rehabilitative treatment improves the weaning success rate and reduces the time needed for weaning mechanical ventilation-dependent patients.
5.Lidamycin inhibits the proliferation of HERG K+ channel highly expressing cancer cells and shows synergy with anticancer drugs.

This study is to investigate inhibitory effects of lidamycin (LDM) on the proliferation of HERG K+ channel highly expressing cancer cells and its synergy with anticancer drugs. MTT assay was used to examine the inhibitory effects of lidamycin combined with various anticancer drugs on the proliferation of human lung cancer A549 cells, human colon cancer HT-29 cells and herg-stably-transfected A549 cells. Using the xenograft model of subcutaneously transplanted HT-29 in nude mice, inhibitory effect was appraised in vivo. The coefficient of drug interaction (CDI) was used to evaluate the synergistic effect of drug combination. LDM significantly inhibited the proliferation ofA549 cells and HT-29 cells with IC50 values of 2.14 and 4.64 ng mL(-1), respectively. The efficacy in HT-29 cells with high HERG potassium expression level is less potent than that in A549 cells with low expression level. In terms of IC50 values, LDM suppressed the growth of herg-stably-transfected A549 cells less potently than pCDNA3.1-stably-transfected A549 cells. There existed synergistic effects in the combinations of fluorouracil (5-FU) and LDM, doxorubicin (DOX) and LDM, or hydroxycamptothecine (HCPT) and LDM. CDI values of the combinations of 5-FU and LDM were more than 0.75. CDI values of LDM and DOX were more than 0.70, but some CDI values of LDM and HCPT were less than 0.70. As for the CDI values, synergistic effects of the combination of LDM and HCPT were the most potent of the three groups. There is no relationship between the inhibitory effect of the growth of cancer cells by 5-FU and HERG potassium expression level. HERG expression level negatively correlated with inhibitory effect on the proliferation of cancer cells by DOX. HERG expression levels and chemosensitivity were positively correlated for HCPT. In the model of subcutaneously xenograft transplanted HT-29 in vivo, LDM and/or HCPT effectively inhibited the growth of HT-29 in nude mice, and the optimum CDI of the combination of LDM and HCPT was less than 1. HERG expression level negatively correlates the chemosensitivity of cancer cells to LDM. There exist synergistic effects in vitro and in vivo in the combination of LDM and HCPT, which inhibitory effects of the proliferation of cancer cells positively modulated by HERG potassium expression level. HERG K+ channel may become a target of combined therapy for choosing anticancer drugs.

6.Microscopic study on skin and soft tissue after repeated expansion.

OBJECTIVEThe study was to evaluate the microscopic changes on skin and soft tissue after repeated expansion for clinical work.
METHODSSix little pigs were divideded as: conventional expansion group, repeated expansion group, and blank control group. Histologic, ultrastructure and bFGF of the skin were observed and measured in each group after samples had been made.
RESULTSThe skin and soft tissue after repeated expansion were healthy on the whole. Compared with the conventional expansion group, there was more microscopic change in the repeated expansion group. Collagen fibers were injured evidently. Cells were injured slightly and proliferated much more, and moreover, they were more activated. The content of bFGF was more higher.
CONCLUSIONSThe skin and soft tissue after repeated expansion are healthy on the whole by more growth and more repair though repeated expansion may result in more injuries. So repeated expansion is safe and feasible.

7.Analysis of surgical diagnosis and treatment of 150 cases with mammographic calcifications

Objective To investigate the diagnosis and treatment of mammographic calcifications found by mammary molybdenum target inspection.Methods 150 women who were found out with breast calcifications by mammary molybdenum target inspection and had been treated with surgical excision biopsy were selected.Their clinical data were retrospectively analyzed.Results In 150 cases of mammographic calcifications,102 cases were benign lesions by pathologic diagnosis,and 48 cases were malignant.Compared the morphological characteristics of the benign and malignant breast calcifications,the nature of the lesion was associated with lesion size (x2 =5.55),the density of calcification (x2 =7.591) and the distribution of calcifications (x2 =11.859).Conclusion Mammographic calcifications found by mammary molybdenum target inspection has a certain clinical value for diagnosis and treatment of breast cancer,so such patients should be encouraged to do biopsy to diagnose or exclude breast cancer.
8.Correlation between genotype and HCV RNA in chronic hepatitis C patients

Objective To determine the distribution of HCV genotypes in patients with chronic hepatitis C,study the distribution of genotype in different gender and the relationship between genotypes and serum HCV-RNA levels.Methods Two hundred and six cases of HCV RNA positive patients(all with relevant clinical data) receiving pegylated interferon therapy were collected from May to December 2010.HCV RNA was detected in 206 hepatitis C patients from 40 hospitals in China by Roche Cobas AmpliPrep/Cobas TaqMan HBV test,and genotype was determined by Abbott RealTime HCV G enotype Ⅱ .The distribution of genotypes in the gender was analyzed by chi-square test analysis.The relationship between genotypes and serum HCV RNA levels was detected by single factor analysis and two independent sample t test analysis.Results There were seven different subtypes of HCV in 206 samples,including genotype 1,7 cases(3.4% ,7/206); genotype 1a,2 cases(1.0%,2/206); genotype 1b,123 cases (59.7 %,123/206); genotype 2,32 cases(15.5 %,32/206); genotype 3,27 cases(13.1%,27/206); genotype 6,6 cases(2.9% ,6/206) ;genotype 1/6,5 cases(2.4% ,5/206) ;genotype 2/4,1 cases(0.5%,1/206).There was no significant difference between HCV genotype and gender in 132 cases with genotype 1 and 65 cases with non-genotype 1(genotype 2,3,6) (x2 = 0.000,P > 0.05).There was significant association between quantity of HCV RNA and genotype in 188 patients with HCV(F = 3.371,P< 0.01).The 197 patients with HCV single genotype were divided into five groups in terms of region(East,South,West,North and Center).There was no significant difference between HCV genotype 1 and non-genotype 1 in the five groups(x2 = 5.840,P > 0.05).Conclusions It is suggested that HCV 1 b is the most prevalent type in China,followed by HCV 2.There is no significant difference between HCV genotype and gender.The levels of HCV RNA with genotype 1b are significantly higher than those with genotype 3.The levels of HCV RNA with genotype 2 are significantly higher than those with genotype 3.The levels of HCV RNA with genotype 6 are significantly higher than those with genotype 3.
9.Retros Flt-1 decelerates the growth of a murine experimental osteosarcoma.

OBJECTIVETo examine the influence of vascular endothelial growth factors (VEGF) in controlling the growth of an experimental osteosarcoma in mice by performing retrovirus-mediated sFlt-1 gene modification.
METHODSFrom March to October 2010 human osteosarcoma G-292 cells were in vitro infected with retroviral vectors encoding soluble Flt-1 or LacZ gene before transplanted into proximal tibiae of immune deficient SCID mice to establish experimental orthotopic osteosarcoma. Daily observation and biweekly microCT were performed to monitor tumor development and progression till sacrifice at 8 weeks after tumor cell inoculation for histological and molecular analyses.
RESULTSSuccessful transgene expression was confirmed in the culture media of sFlt-1 transduced G-292 cells using ELISA, and with positive X-gal staining of the LacZ transduced cells. Noteworthy tumors were grown in all mice on the tibiae receiving G-292 cell inoculation, with clear detection on microCT images starting 2 weeks after inoculation. Over the time period, tumors derived from sFlt-1 transduced G-292 cells were distinctively smaller in size compared to the ones from wide-type G-292 and G-292-LacZ cells. Histology showed typical osteosarcoma characteristics including severe cellular pleomorphism, bone erosions, and neo-vascularization. Real-time polymerase chain reaction indicated significantly higher sFlt-1 expression in sFlt-1 transduced groups than the wild-type G-292 or LacZ treated groups. Strong expression of oncogenes c-myc and c-fos were also obvious, along with the expression of VEGF in the primary tumor tissue.
CONCLUSIONRetrovirus-mediated sFLT-1 gene modification decelerates the osteosarcoma tumor growth in this murine model.

10.Preliminary molecular analysis of bacterial composition in periapical lesions with primary endodontic infections of deciduous teeth.

BACKGROUNDThe bacterial composition of periapical lesions in deciduous teeth has not been well documented. This study was designed to explore the bacterial compositions, especially the dominant bacteria in periapical lesions using 16S rRNA sequencing.
METHODSTissue samples were collected from 11 periapical lesions in deciduous teeth with primary endodontic infections. DNA was extracted from each sample and analyzed using 16S rRNA cloning and sequencing for the identification of bacteria.
RESULTSAll DNA samples were positive for 16S rRNA gene PCR. One hundred and fifty-one phylotypes from 810 clones were identified to eight phyla, and each sample contained an average of 25.9 phylotypes. In addition, 59 phylotypes were detected in more than two samples, and Fusobacterium (F.) nucleatum (8/11), Dialister (D.) invisus (8/11), Campylobacter (C.) gracilis (7/11), Escherichia (E.) coli DH1 (6/11), Aggregatibacter (A.) segnis (6/11), and Streptococcus (S.) mitis (6/11) were the most prevalent species. Furthermore, 45 as-yet-uncultivated phylotypes were also identified.
CONCLUSIONSChronic periapical lesions in deciduous teeth contained polymicrobial infections. F. nucleatum, D. invisus, C. gracilis, E. coli DH1, A. segnis, and S. mitis were the most prevalent species detected by 16S rRNA sequencing.
