Objective To study the influence of artifical liver support system (ALSS) on bone marrow stem cell (BMSC) differentiation factors in patients with chronic severe hepatitis B.Methods Fifty patients with chronic severe hepatitis B were divided into ALSS treatment group (n=25) and control group (n=25).The patients in control group received combined medical treatment and those in ALSS treatment group received ALSS treatment within 1 week of admission on the basis of combined medical treatment.The concentrations of hepatocyte growth factor(HGF),fibroblast growth factor-4 (FGF-4),epidermal growth factor (EGF) and basic fibroblast growth factor (bFGF) were detected before and 2 weeks after therapy.The data were analyzed by t test.Results The serum levels of HGF,FGF-4,EGF and bFGF of patients before ALSS treatment in treatment group were (689.10± 337.68) ng/L,(124.88±87.67) ng/L,(323.85±44.40) ng/L and (9.29± 1.38) ng/L,respectively; while the levels of those cytokines after ALSS treatment were (1081.50±356.66) ng/L,(110.76±79.71) ng/L,(347.80±71.73) ng/L and (9.57±1.15) ng/L,respectively,among which HGF level increased significantly after ALSS treatment (t =10.042,P＜0.01) and was higher than control group(t=6.670,P＜0.01).However,the levels of HGF,FGF-4,EGF and bFGF were not significantly different from those in the control group.And levels of all HGF,FGF-4,EGF and bFGF in control group were not statistically different before and after treatment.ConclusionALSS treatment can increase the serum HGF level but not FGF-4,EGF and bFGF,which may contribute to BMSC transdifferentiation that is involved in the hepatocyte repair and regeneration in chronic severe hepatitis B.

Objective To investigate the expression of NAP in patients with acute myeloid leukemia (AML). Methods Expression of NAP of peripheral blood was assessed by flow cytometry. The serum levels of G-CSF, NE and IL-18 in 53 patients with AML and 50 healthy controls were examined by ELISA. Results The expression of NAP in AML patients [1 159(563 ~ 2 033)AB/c] was decreased than that of healthy controls [1 862 (1 345 ~ 2 382)AB/c], (P < 0.05). The serum levels of G-CSF, NE and IL-8 from AML patients [34.36 (25.17 ~ 47.24) ng/L, 397.56 (324.63 ~ 563.84) pg/mL, 585.34 (403.61 ~ 866.54) pg/mL] were significantly lower than those of healthy controls[54.52(46.27 ~ 77.14) ng/L, 689.74(496.78 ~ 899.14) pg/mL, 832.36(625.17 ~ 998.24) pg/mL] (P < 0.05). The expression of NAP in AML patients was positively correlated with the serum level of G-CSF (r = 0.621, P = 0.007). Conclusions The decreased expression of NAP, G-CSF, NE and IL-8 in the peripheral blood with AML could speculated that neutrophils immune function in patients with AML was restrained.

ObjectiveTo investigate the effect of neoadjuvant chemotherapy in locally advanced cervical cancer on cell cycle,proliferation,and evaluate the feasibility of proportion of cell in different phase 　and proliferating cell nuclear antigen(PCNA) as sensitive indices to assess chemotherapeutical sensitivity and therapeutical effect.MethodsForty-nine cases of locally advanced cervical cancer were divided into response group and no-response group according to the clinical efficacy of neoadjuvant chemotherapy.Compared the proportion of cell in different phase and proliferation index of PCNA between two groups.The clinical therapeutic effect was evaluated after 4 weeks in the second neoadjuvant chemotherapy regimen.Results In 49 patients with locally advanced cervical cancer,clinical effective of 39 cases (response group),no effective of 10 cases(no-response group).The S-phase proportion of cell in the pre-neoadjuvant chemotherapy and post-neoadjuvant chemotherapy of response group[ (21.47 ± 5.21 )％ and(18.32 ±5.07)％] were higher than those of no-response group [ (9.63 ± 2.58)％ and ( 10.14 ± 2.32)％ ] (P ＜ 0.05 ).The proliferation index of PCNA of cervical cancer in the pre-neoadjuvant chemotherapy of response group [ ( 81.67 ± 7.14)％ ] was higher than that of no-response group [ (66.99 ± 2.29 )％ ] (P ＜ 0.05 ).Conclusion The S-phase proportion of cell and proliferation index of PCNA in the pre-neoadjuvant chemotherapy are important indexes to assess chemotherapeutical sensitivity and therapeutical effect.

Objective To investigate the changing characteristics of muscle mass by dual energy X-ray absorptiometry (DEXA) in middle aged and elderly people with different ages.Methods This study included 343 healthy people aged 50-89 years (178 males and 165 females).Subjects were divided into 4 groups:aged 50-59,60-69,70-79,and 80-89 years group.Fat-free body mass (FFM),the appendicular lean body mass (aLM),trunk lean mass (tLM) were measured by DEXA.The aLM/body weight (Wt％) and aLM index (ASMI) were calculated by DEXA.Results The tLM in both gender was decreased in aged 80-89 years group than in aged 50-59 years group [male (10.6± 1.8) kg vs.(18.4±2.9) kg,female (7.1±1.2) kg vs.(14.8±2.1) kg,F=5.7 and 7.8,P＜ 0.01].The average ASMI in both gender was decreased in aged 70-79 and 80-89 years groups than in aged 50-59 years groups [male (5.5±0.4) kg/m2,(4.9±0.7) kg/m2 vs.(6.1±0.5) kg/m2,female (4.3±0.4) kg/m2,(3.9±0.5) kg/m2 vs.(4.8±0.6) kg/m2,F=24.9 and 9.6,both P＜0.01].The quality of muscle mass was decreased with aging in the elderly.Conclusions Ageing is an important risk factor for sarcopenia in the elderly.DEXA can be used to evaluate the muscle mass in middle aged and the elderly,and ASMI can be used as a reliable measurement index.

AIM: To investigate the effects of fluvas-tatin on expression of intercellular adhesion molecule-1 after myocardial ischemia-reperfusion in rabbits with hyper-lipidemia. METHODS: 30 rabbits which were gastric perfusion administered intralipid were randomly divided into 3 groups ( n - 10 in each) : IR ( ischemia-reperfusion) group, S (sham-operation) group and F (fluvastatin 10 mg?kg-1 ) group. Electrocardiography and cardiac function were recorded during the experiment. At the end of reperfusion, ischemic area and infarct size were defined by Evans blue and triphenyltetrazolium chloride staining. The expression of ICAM-1 in myocardium was measuredby RT-PCR. RESULTS: After ischemia-reperfusion, the expression of ICAM-1 mRNA in myocardial and infarct size decreased and cardiac function significantly improved in F group compared with IR group. CONCLUSION: The increase of expression of ICAM-1 mRNA in myocardial may be one of the important factors in inducing myocardial ischemia-reperfusion injury. The myocardial protective mechanism of fluvastatin maybe attribute to its effect on decreasing the expression of ICAM-1 mRNA in myocardial .

Objective To investigate the feasibility of differentiating human umbilical cord blood stem cells into hepatocytes. Methods Thirty-six BALB/c nude mice were randomly divided into experimental group and control group(18 in each of the group), and experimental group was again randomly divided into group A, B and C (six in each of the group). The mice in experimental group and control group were exposed to 350 cGy radiation produced by 60Co. After 3 h, karyocytes at different concentrations in the fresh human umbilical cord blood were injected into the mice in experimental group A, B, C via their tail veins, and the equal volume of normal sodium (NS) was also injected into control group via tail veins. After one month, carbon tetrachloride (CCl4) was injected into experimental group A, B and control group via abdominal cavity, and the equal volume of normal sodium was injected into experimental group C. After two months, immunohistochemistry and reverse transcriptase polymerase chain reaction (RT-PCR) were used to detect the expressions of human cytokeratin-18 (CK18), cytokeratin-19 (CK19) and albumin (ALB) in liver tissues of all mice. Objective To investigate the effects and mechanism of different dosage of fluvastatin on the prevention of heart muscle ischemia reperfusion injury in rabbits.Method Thirty-five rabbits were randomly divided into 5 groups with 7 rabbits in each: sham group,myocardial ischemia reperfusion control group,low dosage of fluvastatin pretreatment group (2 mg/kg,Group F1),middle dosage of fluvastatin pretreatment group(5 mg/kg,Group F2) and large dosage of fluvastatin pretreatment group(20 mg/kg,Group F3).The left ventricular systolic pressure(LVSP),the max rate of rise of left ventricular pressure(?dp/dt_(-max)) and left ventricular end-diastolic pressure(LVEDP) were detected during the experiment.At the end of reperfusion,the infarct size and area at risk were defined by Evans blue and TTC staining,and the levels of myocardial nitrogen monoxidum(NO) and nitricoxide synthase(NOS) were measured.Result Compared with the ischemia reperfusion group,the indexes of heart function improved significantly,the level of myocardial NO was increased significantly and the myocardial infarct size was decreased significantly in the groups F2 and F3.There was no significant difference between the group F1 and ischemia reperfusion group.Conclusion Fluvastatin exerts a cardioprotective effect against myocardial ischemia reperfusion injury in rabbits.NO is likely involved in this protective mechanism.
