Objective To investigate the relationship between p38MAPK and astrocytes activation by detecting the changes of p38MAPK,TNF-?,NO and NOS in the substance P-induced activation of spinal cord astrocytes in vitro.Methods Astrocytes,cultured from spinal cord of SPF rat and identified by immunofluorescence method with GFAP antibody,were grouped into the control group,the SP stimulus group(SP group) and the SB203580 interrupt p38MAPK group(interrupt group) in which SP(10-7 mol/L) and SB203580(10 ?mol/L) were added to the supernatant for 12 h.The WB method,RT-PCR and ELISA were used to determine the changes of p38MAPK,phosphorylated p38MAPK,GFAP mRNA,TNF-?,NO and NOS in the astrocytes or supernatant at 1,3,12,24 and 36 h respectively.Results The GFAP positive rate of the cultured cells was higher than 95%.p38MAPK level in the SP group did not have any change,whereas p-p38 level increased significantly after 1 h and GFAP mRNA,NO,NOS and TNF-? level increased after 3 h.When p38MAPK pathway was inhibited by SB203580 in the SP+SB group,GFAP mRNA,NO,NOS and TNF-? were significantly reduced compared with those in the SP group.The level of p38MAPK,p-p38,GFAP mRNA,NO,NOS and TNF-? in the SB group did not have changes compared with those in the normal group.Conclusion These results indicate that p38MAPK signal pathway contributes to substance P induced activation of spinal cord astrocytes according to inflammatory factors attenuation after p38MAPK signal pathway interruption by SB203580.

Objective To investigate the expression of p38 mitogen-activated protein kinase(p38MAPK) in posterior horn of L5-S2 spinal cord in analgesia for chronic prostate pain with intrathecal injection of SB203580(p38MAPK inhibitor activity) in a murine model.Methods Forty-five male SPF SD rats were randomly divided into control group(n=5),prostatitis group(n=10) and SB203580 analgesia group(n=30,SB203580 was intrathecally injected).In the SB203580 analgesia group,complete Freund's adjuvant was injected into the prostate and SB203580 of 0.5?g/10?l,2.5?g/10?l and 12.5?g/10?l was intrathecally injected respectively.Five days and ten days after drug administration or simple modeling,phosphorylated p38MAPK(p-p38MAPK),glial fibrillary acidic protein(GFAP),tumor necrosis factor-?(TNF-?),nitric oxide(NO) level and induced nitric oxide synthase(iNOS) activity of L5-S2 spinal dorsal horn of rats were determined by Western blotting and ELISA assay,respectively.Results In the prostatitis group,the p-p38MAPK,GFAP,TNF-?,NO level and iNOS activity of L5-S2 spinal dorsal horn of rats at the 5th day and the 10th day were significantly higher than those of the control group and increased with time.The p-p38MAPK,GFAP,TNF-?,NO levels and iNOS activity at the 5th day and 10th day in SB203580 analgesia group were lower than those in the prostatitis group.Conclusion P38MAPK signaling pathway was involved in the mechanism of pain conduction and analgesia by SB203580 after prostatitis.The level of inflammatory factors in spinal cord can be effectively reduced by blocking the p38MAPK signaling pathway.

Objective To investigate the effect of up-regulation of tetraspanin-5 (Tspan-5) expression on the metastatic ability of colorectal cancer (CRC) cells. Methods The coding sequence of Tspan-5 mRNA was amplified from total RNA of LoVo cells by RT-PCR and cloned into pEGFP-C1 to construct Tspan-5/pEGFP-C1,a eukaryotic expression vector. The expression of Tspan-5 in LST-R1 cells was up-regulated by gene tranfection. Western blotting was used to detect the expression level of Tspan-5/EGFP fusion protein,and laser scanning confocal microscopy (LSCM) was used to observe the distribution of Tspan-5/EGFP in LST-R1 cells. The changes in adhesion,migration and invasion ability of LST-R1 cells were determined by adhesion,migration and invasion experiment in vitro,respectively. Liver metastasis model of nude mice was used to test the changes in liver metastasis ability of LST-R1 cells in vivo. Objective To study of the imaging diagnosis and differential diagnosis of cystitis glandularis. Methods The clinical data and imaging findings of 8 cases with cystitis glandularis proved by pathology were retrospectively analyzed with literaures re-view. Results The lesions on images appeared as nodular mass in 4 cases, diffuse in 2 cases and mixed type in 2 cases,1 case with cystic degeneration. The lesions in 2 cases obviously reduced in size after treatment. The CT value of the lesions post contrast-en-hanced scan averagely increased up to 15.8 HU compared with that of plain scan. The lesions were localized in 6 cases and diffuse in 2 cases. Conclusion cystitis galandularis has some imaging characteristics, but the final diagnosis is depended on pathology.

BACKGROUND: Studies have shown that stability of artificial joint prosthesis is highly correlated with medullary cavity matching.However, whether medullary cavity appearance and diameter changes with age, ultimately affecting prosthesis and medullary cavity match remains unclear.OBJECTIVE: To evaluate the effect of age on femoral medullary cavity morphology and geometric parameters, and observe medullary cavity changes with age so as to guide prosthesis selection and operation and improve long-term survival of prosthesis.DESIGN, TIME AND SETTING: Comparative observation of morphology. The experiment was performed at the Department of Orthopedic Surgery, Beijing Friendship Hospital Affiliated to Capital Medical University from August 2007 to April 2008. PARTICIPANTS: A total of 100 healthy female and male people were selected from Orthopedics Outpatient Department.Informed consent was obtained from all subjects.METHODS: All patients underwent a left full-length femoral CT scanning, 150 kV, 100 mA. The images were reconstructed and measured at the PICKER 2000 Voxel Q station.MAIN OUTCOME MEASURES: The contents of 36 data collected from the patients included general information of the patients,the external parameters of femur, radius-length parameters of femoral medullary cavity and morphological parameters of femoral medullary cavity.RESULTS: Among all femoral parameters, the site of isthmus altered significantly with aging (P< 0.05). The anterior and posterior cortical thicknesses of isthmus and medullar anteroposterior diameters of isthmus changed significantly with age in female group (P < 0.05), but were not changed significantly in male group (P > 0.05).CONCLUSION: As for the prosthetic design, the shape design of prosthesis needs not to change with aging for male or female. Properly lengthening the femoral stem will help to get best fit of prosthesis. Proximally fixed femoral prosthesis seems to have superiority theoretically for elderly female patients.

Objective To explore the feasibility and value of preservation of left colonic artery (LCA) in dealing with inferior mesenteric artery (IMA) in laparoscopic anterior resection of rectal carcinoma.Methods The clinical data of 72 cases of laparoscopic anterior resection of rectal carcinoma from April 2010 to October 2013 were retrospectively analyzed including 32 cases with preservation of LCA (observation group) and 40 cases without preservation of LCA (control group).The blood loss,operative time,postoperative exhaust time,terminal ileum stoma,the number of lymph nodes removed around the root of IMA,the rate of lymph node metastasis around the root of IMA and prognosis were compared between two groups.Results There was no significant difference in the blood loss,operative time,postoperative exhaust time,terminal ileum stoma,the number of lymph nodes removed around the root of IMA,the rate of lymph node metastasis around the root of IMA between two groups (P ＞ 0.05).No case in observation group needed to free the splenic flexure of colon and to make the terminal ileum stoma,while 3 cases in control group needed to free splenic flexure of colon because of blood supply disorder in the proximal intestine (P =0.046),and 4 cases underwent terminal ileum stoma following anastomosis (P =0.042).No anastomotic leakage occurred in observation group,while 2 cases of anastomotic leakage occurred in control group(P =0.090).After followed up for 6-48 months,no significant difference was found in local recurrence and liver metastasis in two groups (P ＞ 0.05).Conclusion Laparoscopic anterior resection of rectal carcinoma with preservation of LCA in dealing with IMA can effectively retain the blood supply of proximal intestine.

Objective To study the influence of systematic respiratory training of patients with chronic ob- structive pulmonary disease(COPD)on their off-respirator pulmonary function and emotional depression.Methods Sixty COPD patients were divided into a treatment group and a control group.The patients in both groups were given routine treatment,while those in the treatment group were also given respiratory training in addition for 9-12 months after ceasing the mechanical ventilation.The index of pulmonary function such as FEV1 and depression(SDS score) were evaluated before and after the respiratory training.All data was analyzed by using SPSS 10.0.Results The FEVI of all patients was improved after 9-12 months of respiratory training(P
